Three new monoterpenoid indole alkaloids, vinmajorines C–E ( 1 – 3 ), along with 18 known analogues ( 4 – 21 ), were isolated from the whole plants of Vinca major. The new structures were elucidated as (5α,15β,16R,17α,19β,20α,21β)‐10,17‐dimethoxy‐21‐methyl‐18‐oxa‐5,16‐cycloyohimban‐19‐ol ( 1 ), (5α,15β,16R,17α,20α,21β)‐10‐methoxy‐21‐methyl‐18‐oxa‐5,16‐cycloyohimban‐17‐ol ( 2 ), and (5α,15β,16R,17α,20α,21β)‐10‐methoxy‐21‐methyl‐18‐oxa‐5,16‐cycloyohimban‐17‐yl acetate ( 3 ), respectively, by extensive NMR and MS analysis and comparison with known compounds. Compounds 1 – 3 were evaluated for their cytotoxic activities against five human cancer cell lines, compounds 1 and 3 showing moderate cytotoxic activities.  相似文献

Two new compounds,8(14),15-isopimaradiene-2α,3α,19-triol(1),and 6α,7β-dihydroxy-17(20)-cis-5α-pregna-16-one(2),together with four known copounds,a oxygenated rare phyllocladane,phyllocladan-16α,19-diol(3),kaempferol-3-0-β-D-galactopyranosied,kaempferol-3-0-α-L-rhamnopyranoside and scopoletin, were isolated from the leaves of Ailanthus tripysa.Structures of 1-3 were elucidated on the basis of spectroscopic data as compared with related compounds.  相似文献

2-Alkoxy-4-heteroarylaminomethylene-5(4H)-thiazolones 4 were converted with various nucleophiles into β-heteroarylamino-α,β-dehydro-α-amino acid derivatives 11, 14, 15, 16, 17, 18 , and 19 . Reduction of 4 with sodium borohydride in ethanol saturated with gaseous ammonia afforded the corresponding β-heteroaryl-amino substituted alanyl amides 20 . Thiazoledione derivative 7a was transformed with sodium methoxide in methanol into 1-(4,6-dimethylpyrimidinyl-2)-4-mercaptocarbonylimidazol-2(3H)-one ( 8a ).  相似文献

The roots of Trachycalymma fimbriatum (WEIMARCK ) BULLOCK contain both cardenolide and pregnaneglycosides. Elimination of 2-deoxysugars by mild acid hydrolysis gave a mixture from which some anhydroderivatives and the following compounds could be isolated: uzarigenin ( l ), ascleposide = 3-O-(6-deoxy-β-D -allopyranosyl)-uzarigenin ( 4 ), coroglaucigenin ( 6 ) and two pregnane derivatives (H and J). Compound H could be identified as 3β,14β-dihydroxy5α, 17α-pregnan-20-one ( 10 ). Compound J is probably a new substance, for which we tentatively assign structure 18 , i.e. 3β8β,14β-trihydroxy-5α,17α-pregnan-20-one. We suspect H and J to be artefacts produced from the corresponding 17b-derivatives during acid hydrolysis. 17-iso-H is probably a precursor in the biosynthesis of uzarigenin. The cardenolides of Trachycalymma fimbriatum are the same as found in Asclepias glaucophylla, a closely related species, while the pregnane derivatives of the latter are distinctly higher hydroxylated.  相似文献

Three new ent‐kaurane diterpenoids, (4α)‐19‐nor‐ent‐kaurane‐4,16,17‐triol ( 1 ), (4α,16α)‐17‐(acetyloxy)‐19‐nor‐ent‐kaurane‐4,16diol ( 2 ), and 17‐hydroxy‐ent‐kaur‐15‐en‐19‐al ( 3 ), together with 11 known compounds, were isolated from the stem bark of Annona squamosa L. The structures of 1 – 3 were identified by analysis of their spectroscopic data. All compounds were evaluated for cytotoxic activity against human lung cancer (95‐D) and ovarian cancer (A2780) cell lines, and compounds 3, 5, 7, 11 – 14 exhibited promising antiproliferative activities with IC50 values ranging from 0.38 to 34.66 μM .  相似文献

New pyrido[2,3‐d]pyrimidines 11 , 12 , 13 , and 21 have been synthesized. The vasorelaxant effect on smooth muscle isolated from rat aorta, via PDEs inhibition, of these compounds along with other pyrido[2,3‐d]pyrimidines 14 , 15 , 16 , 17 , 18 , 19 , 20 reported earlier by our group, has also been determined. These pyrido[2,3‐d]pyrimidines 11 , 12 , 13 , 14 , 15 , 16 , 17 , 18 , 19 , 20 , 21 were synthesized by the reaction of ferrocenyl‐ethynyl ketones ( 1 , 2 , 3 , 4 ) or α‐alkynyl ketones ( 5 , 6 , 7 , 8 , 9 , 10 ) with 6‐amino‐1,3‐dimethyluracil using [Ni(CN)4]?4 as an active catalytic species, formed in situ in a Ni(CN)2/NaOH/H2O/CO/KCN aqueous system. Evaluation of the vasorelaxant effect of compounds 11 , 12 , 13 , 14 , 15 , 16 , 17 , 18 , 19 , 20 , 21 demonstrated that all compounds relax the tissue in a concentration‐dependent manner. The structural changes do not alter the effectiveness; however, there are differences related to potency expressed as EC50. Compounds 12 (7‐ferrocenyl‐1,3‐dimethyl‐5‐(m‐tolyl)‐pyrido[2,3‐d]pyrimidine) and 13 (7‐ferrocenyl‐1,3‐dipropyl‐5‐(4‐metoxyphenyl)‐pyrido[2,3‐d]pyrimidine) were the most potent compounds, even more than rolipram, reference drug; the EC50 was 0.41 ± 0.02 μM and 0.81 ± 0.11 μM for 12 and 13 , correspondingly. The EC50 of compounds 15 (7‐ferrocenyl‐1,3‐dimethyl‐5‐phenyl‐pyrido[2,3‐d]pyrimidine), 14 (7‐ferrocenyl‐5‐(3,5‐dimethoxyphenyl)‐1,3‐dimethylpyrido[2,3‐d]pyrimidine), and 19 (5‐n‐butyl‐7‐ethyl‐1,3‐dimethylpyrido[2,3‐d]pyrimidine) was similar to EC50 of rolipram. Compounds 11 , 12 , 13 , 14 , 15 , 16 , 17 , 18 , 19 , 20 , 21 significantly induce concentration‐dependent vasorelaxation in endothelium‐intact aortic rings. In addition, the relaxation responses to each compound in either endothelium‐intact or endothelium denuded aortic rings were comparable, suggesting that removal of the functional endothelium has no significant influence on its intrinsic vasorelaxant activity. In vitro capability of conserving cyclic‐AMP or cyclic‐GMP (adenosine and guanosine 3′, 5′‐cyclic monophosphate) via PDE inhibition for compounds 12 , 13 , 14 , 15 and 19 was evaluated. Compounds 15 and 19 show the highest percent inhibition effect (94.83% and 83.98%, respectively) for the decomposition of c‐AMP. Docking studies showed that the compound 15 was selective for the inhibition of PDE‐4.  相似文献

Enantiomerically pure cis- and trans-5-alkyl-1-benzoyl-2-(tert-butyl)-3-methylimidazolidin-4-ones ( 1, 2, 11, 15, 16 ) and trans-2-(tert-butyl)-3-methyl-5-phenylimidazolidin-4-one ( 20 ), readily available from (S)-alanine, (S)-valine, (S)-methionine, and (R)-phenylglycine are deprotonated to chiral enolates (cf. 3, 4, 12, 21 ). Diastereoselective alkylation of these enolates to 5,5-dialkyl- or 5-alkyl-5-arylimidazolidinones ( 5, 6, 9, 10, 13a-d, 17, 18, 22 ) and hydrolysis give α-alkyl-α-amino acids such as (R)- and (S)-α-methyldopa ( 7 and 8a , resp.), (S)-α-methylvaline ( 14 ), and (R)-α-methyl-methionine ( 19 ). The configuration of the products is proved by chemical correlation and by NOE 1H-NMR measurements (see 23, 24 ). In the overall process, a simple, enantiomerically pure α-amino acid can be α-alkylated with retention or with inversion of configuration through pivaladehyde acetal derivatives. Since no chiral auxiliary is required, the process is coined ‘self-reproduction of a center of chirality’. The method is compared with other α-alkylations of amino acids occurring without racemization. The importance of enantiomerically pure, α-branched α-amino acids as synthetic intermediates and for the preparation of biologically active compounds is discussed.  相似文献

Isoquinolin‐5‐ylhydrazinium chloride 13 and 5‐bromoisoquinolin‐8‐ylhydrazinium chloride 14 were converted via Fischer syntheses with 3‐methylbutan‐2‐one into indolenines, 2,3,3‐trimethyl‐3H‐pyrrolo[2,3‐f]isoquinoline 15 and 5‐bromo‐2,3,3‐trimethyl‐3H‐pyrrolo[3,2‐h]isoquinoline 16 , respectively. Exposure of the indolenines to the Vilsmeier reagent produced diformyl compounds 17 and 18 , which reacted with arylhydrazines to give the corresponding pyrazoles 19a , 19b , 19c , 19d , 19e , 19f , 19g , 19h , 19i and 20a , 20b , 20c , 20d , 20e , 20f , 20g . Reaction of 17 with thiourea gave a pyrimidine‐2(1H)‐thione 23 or with hydroxylamine hydrochloride, an isoxazole 24 .  相似文献

Abstract

In the present study, five new ent-kaurane diterpenes including 4α-hydroxy-17,19-dinor-ent-kaurane-16-one (1), 4β-hydroxy-16β-H-18-nor-ent-kaurane-17-oic acid (2), 4β,17-dihydroxy-16α-acetoxy-18-nor-ent-kaurane (3), Annosquamosin Z (4) and 16α-H-ent-kaurane-17,18-dioic acid, 17-methy ester (5) were isolated from Annona squamosa L. pericarp. The compounds were also evaluated for their cytotoxic activities against SMMC-7721 and HepG2 cell lines, among which compound 3 exhibited potent cytotoxicity with IC50 value of less than 20?μM.  相似文献

The N, N-bis(2-chloroethyl)aminobenzoate isomers and the 4-methyl-3-N, N-bis(2-chloro-ethyl)aminobenzoate of 3β-hydroxy-13α-amino-13,17-seco-5α-androstan-17-oic-13,17-lactam, 3α-hydroxy-13α-amino-13,17-seco-5α-androstan-17-oic-13,17-lactam, 3α-hydroxy-13α-amino-13,17-seco-5-androsten-17-oic-13,17-lactam and 17β-hydroxy-3-aza-A-homo-4α-androsten-4-one, have been prepared and their biological activity evaluated against P388 leukemia in vivo and Ehrlich Ascites tumor (EAT), P388 and L1210 leukemias and Baby Hamster cells (BHK) in vitro. The esters in which the alkylating congener is linked to the lactam alcohol in the axial position are inactive in vivo in P388 leukemia, while compounds 1, 4, 6, 13, 14 and the alkylating congeners 17, 18 and 20 are active. The effect of the homo-azasteroidal of N, N-bis(2-chloroethyl)aminobenzoic acid isomers and of 4-methyl-3-N, N-bis(2-chloroethyl)aminobenzoic acid on the incorporation of the radioactive precursor into the DNA of L1210, P388 leukemias, Ehrlich ascites tumor and, baby Hamster kidney cells was investigated. Higher inhibitory effects on the incorporation of the radioactive precursor was obtained with the ortho derivatives, yielding <70% inhibition of thymidine incorporation in all tumor lines tested.  相似文献

The synthesis of tetradeuterated 15-methyl-PGF and 17-phenyl-18, 19, 20-trinor-PGF, is described. Gas chromatographic -mass spectrometric methods, using the deuterated compounds as internal standards and carriers, for quantitation of corresponding unlabeled prostaglandin analogues is described. Chemical examination of a marine sponge Xestospongia sp. resulted in the isolation of 20 sterol derivatives ( 1 – 20 ), including eight new sterols namely aragusterols J – L ( 1 – 3 ), (5α,7α,12β,22E)‐7,12,18‐trihydroxystigmast‐22‐en‐3‐one ( 4 ), (5α,7α,12β,24R)‐ and (5α,7α,12β,24S)‐7,12,20‐trihydroxystigmastan‐3‐one ( 5 / 6 ), and (5α,7α,12β,22E,24R)‐ and (5α,7α,12β,22E,24S)‐7,12,20‐trihydroxyergost‐22‐en‐3‐one ( 7 / 8 ). The structures of new compounds were determined through extensive spectroscopic analyses and chemical conversion. The sterol diversity was mainly characterized by the presence of a cyclopropane unit at side chain, while compound 4 with 18‐hydroxymethyl group was found in stigmasterol family for the first time. Cytotoxic test revealed the inhibitory effects of compounds 1 , 4 , and 17 against human leukemia cell line K562 with IC50 values of 18.3, 24.1, and 34.3 μm , respectively.  相似文献

Diazodization of pyrazolo[3,4‐b]pyridine 1 afforded diazonium salt 2 that coupled with active methylene compounds such as 3a , 3b , 6 , 7 , 8 , 15a , 15b , 16a , 16b , 17 , and 24 in pyridine to give aryl hydrazone derivatives 4a , 4b , 9 , 10 , 11 , 18a , 18b , 19a , 19b , 20 , and 25 , respectively. The previous synthesized compounds underwent cyclization in acetic acid to give the corresponding pyridopyrazolotriazines 5a , 5b , 12 , 13 , 14 , 21a , 21b , 22a , 22b , 22c , 23 , and 26 , respectively. The newly synthesized compounds were characterized by elemental analysis and spectral data and screened for their antioxidant activities. The results of ABTS method showed clearly that compounds 1 , 4b , 5b , 11 , 20 , 25 , and 26 displayed promising in vitro antioxidant activities. Compounds 1 and 4a exhibited high protection against DNA damage induced by the bleomycine iron complex.  相似文献

Structures for the genins of the ester glycosides of Marsdenia erecta are suggested. They are based on the behaviour in alkaline hydrolysis of these ester glycosides, their NMR. and mass spectra and ORD. data. All genins are derived from three acyl-free pregnane derivatives, i.e. drevogenin-P ( 1 ), 17 β-marsdenin ( 3 ) and marsectohexol ( 7 ). The structure of 1 is known, 3 and 7 are new compounds, i.e. 3 = 3β,8β,11α,12β,14β-pentahydroxy-Δ5-pregnen-20-one and 7 = 3β,8β,11α,12β,14β,20ξ-hexahydroxy-Δ5-pregnene. Formulae 13–17 were attributed to the acyl-genins A-1, A-2, A-3, A-4 and A-5, but only two of them were pure compounds, i.e. acyl-genin A-3 = 11,12-di-O-tiglyl-17β-marsdenin ( 15 ) and acyl-genin A-5 = 11,12-di-O-acetyl-marsectohexoi ( 17 ). Acyl-genin A-1 is a mixture of the two esters 13a + 13b derived from drevogenin-P, and similarly acyl-genin A-2 is a mixture of the esters 14a + 14b derived from 17β-marsdenin. The poorly characterised acyl-genin A-4 is most probably a mixture of the esters 16a + 16b , also derived from 17β-marsdenin.  相似文献

Two new trichothecenes, named 8α‐hydroxyroridin H and myrothecin A, along with six known compounds, 8α‐acetoxy roridin H, isororidin K, verrucarin A, verrucarin J, verrucarin L and 8α‐acetoxy verrucarin L, were isolated from the fermentation broth of a halotolerant fungus Myrothecium sp. GS‐17, which was separated from the soil sample of a salina. Structure elucidation and NMR signal assignments were achieved on the basis of spectroscopy. Nitration of 2,3-dihydrofuro[3,2-b]- N-oxide 3b and -[2,3-c]pyridine N-oxide 3c afforded the nitropyridine compounds 4b, 5b and 6 from 3b and 4c, 5c, 5′c and 7 from 3c , while -[2,3-b]- N-oxide 3a and -[3,2-c]pyridine N-oxide 3d did not give the nitro compound. Chlorination of 3b and 3c with phosphorus oxychloride yielded mainly the chloropyridine derivatives 15b, 15′b from 3b and 15c and 15′c from 3c , whereas 3a and 3d gave pyridine derivatives formed through fission of the 1–2 ether bond of the furo-pyridines 13a , 14 and 13d . Acetoxylation of 3b and 3c gave 3-acetoxy derivatives 18b and 18c and the parent compound 1b and 1c . Acetoxylation of 3a yielded compounds formed through fission of the 1–2 bond 16 and 17 and 3d gave furopyridones 19 and 19 ′. Cyanation of 3b and 3c yielded mainly the cyanopyridine compounds 20b, 20c and 20′c . Cyanation of 3a and 3d gave the cyanopyridine compounds 20a , 20d and 20′d accompanying formation of the pyridine derivatives 21a, 21d and 21′d .  相似文献

A novel series of coumarins, thiadiazoles, thiazoles, and pyridines were synthesized via reaction of 5‐bromosalicylaldehyde with different reagents. Thus, 5‐bromosalicylaldehyde 1 was reacted with compounds 2a–d affording iminocoumarins 3a–d , which on hydrolysis with 10% hydrochloric acid, afforded coumarins 4a–d , respectively. On the other hand, reaction of 1 with benzylhydrazinecarbodithioate 5 afforded derivative 6 , which reacted with hydrazonoyl halides 7a–f , afforded 1,3,4‐thiadiazoles 11a–f , respectively. Moreover, thiazoles 15 and 16 were obtained via reaction of 1 with thiocarbohydrazide 13 and hydrazonoyl halides. However, condensation of 2‐acetyl‐5‐bromobenzofuran 17 with benzaldehyde afforded chalcone 18 , which reacted with pyridiniumbromides 19a–c , afforded pyridines 20a–c , respectively. Furthermore, pyridines 21–24 were synthesized from the reaction of chalcone 18 with different active methylene compounds. Reaction of 24 with ethylchloroacetate, chloroacetone, and chloroacetonitrile afforded thienopyridines 26a–c , respectively. The structures of the newly synthesized compounds were established based on their spectral data and elemental analyses. Also, selected newly synthesized compounds were screened for their antimicrobial activity against various microorganisms by disk diffusion method.  相似文献

A reaction of N,O-bis-(diethylhydrogensilyl) trifluoroacetamide with 5β-pregnane-17,20,21-triols afforded diethylhydrogensilyl (DEHS) cyclic diethylsilylene (DES) derivatives. 5β-Pregnane-3α,17α,20β,21-tetraol yielded the 3α,21-bis-DEHS-17α,20β-DES compound, whereas the structure of the major product from its 20α isomer was assigned as the 3α,17α-bis-DEHS-20α,21-DES derivative, which would undergo facile isomerization to the unstable 3α,21-bis-DEHS-17α,20α-DES structure under electron ionization. Fragmentations of these DEHS-DES compounds are discussed on the basis of isotope labelling experiments, linked scanning data, and accurate mass measurements.  相似文献
