We reported that multiresponsive amphiphilic graft poly(ether amine)s (agPEAs) comprised of hydrophilic poly(ethylene oxide) (PEO) and hydrophobic octadecyl alkyl chain as side‐chain were prepared through one‐pot synthesis. In aqueous solution, these obtained agPEAs can self‐assemble into stable nanomicelles, whose aggregation can be controlled by temperature, pH, and ionic strength with tunable cloud point (CP). In the presence of these obtained agPEAs, hydrophobic dye Nile red can be dispersed into aqueous solution and hydrophilic dye methyl orange can be dispersed into nonpolar toluene. The agPEAs are expected to be potential in application such as encapsulation and controlled release of drugs, due to their simple synthesis, amphiphilicity, and multistimuli response. © 2009 Wiley Periodicals, Inc. Self‐association in aqueous solution of amphiphilic poly(acrylic acid)‐b‐poly(propylene oxide)‐b‐poly(acrylic acid) (PAA‐b‐PPO‐b‐PAA) copolymers having various outer PAA block lengths are presented. These copolymers show two thermosensitive behaviors. The first one, due to hydrogen bonds between PAA and PPO resulting in large aggregates, was observed by visible spectroscopy. The second one, due to the association of PPO middle block into aggregates, was evidenced by dynamic light scattering and pyrene fluorescence. These critical temperatures both depend on the ionization and the length of PAA blocks. The characterization of the aggregates above the critical aggregation concentration by fluorescence quenching experiments showed a very low aggregation number corresponding to dimers or trimers association depending on the conditions. © 2016 Wiley Periodicals, Inc. Aliphatic polyesters and polyphosphoesters (PPEs) have received much interest in medical applications due to their favorable biocompatibility and biodegradability. In this work, novel amphiphilic triblock copolymers of PPE and poly(L ‐lactic acid) (PLLA) with various compositions were synthesized and characterized. The blocky structure was confirmed by GPC analyses. These triblock copolymers formed micelles composed of hydrophobic PLLA core and hydrophilic PPE shell in aqueous solution. Critical micellization concentrations of these triblock copolymers were related to the polymer compositions. Incubation of micelles at neutral pH followed by GPC analyses revealed that these polymer micelles were hydrolysized and resulted in decreased molecular weights and small oligomers, whereas its degradation in basic and acid mediums was accelerated. MTT assay also demonstrated the biocompatibility against HEK293 cells. These biodegradable polymers are potential as drug carriers for biomedical application. © 2008 Wiley Periodicals, Inc. A surface‐active p‐vinyl benzyloxy‐ω‐hydroxy‐poly(ethylene oxide) macromonomer containing 22 pendant structural units of ethylene oxide (St–PEO22) was synthesized with an initiation method. Because of its solubility in a large variety of solvents, the free‐radical copolymerization with electron‐acceptor N‐phenylmaleimide (NPMI) was performed at 60 °C in benzene and tetrahydrofuran (THF) as isotropic media and in a water–THF mixture or water as a heterogeneous medium. Oil‐soluble 2,2′‐azobisisobutyronitrile and water‐soluble 4,4′‐azobis(4‐cyanovaleric acid) were used as the initiators at fixed concentrations. Two different St–PEO22/NPMI comonomer ratios (1/1 and 3/7) at a fixed total comonomer concentration in the polymerization system were used. The structures, compositions, and microstructure peculiarities of the obtained alternating, amphiphilic, comblike copolymers were determined by NMR analysis. For the copolymers synthesized in hydrophilic media, differential scanning calorimetry showed, near the endothermic peak attributed to the melting of the poly(ethylene oxide) side chains, the presence of a second peak due to the partially ordered phase that could exist between the crystalline state and the isotropic melt. A series of amphiphilic biodegradable and biocompatible poly(p‐dioxanone)‐grafted poly(vinyl alcohol) (PVA) copolymers with well‐defined structure were obtained by a three‐step synthesis based on the “grafting from” concept. The first step (protection step), called the partial silylation of PVA hydroxyl groups, was accomplished by 1,1,1,3,3,3‐hexamethyldisilazane and catalyst chlorotrimethylsilane in dimethyl sulfoxide using THF as cosolvent. The second step was the ring‐opening polymerization of p‐dioxanone (PDO) initiated from the remaining OH groups of the partially silylated PVA. Finally, a deprotection step was followed: the silylether group was deprotected easily under very mild conditions. The synthetic conditions of the first two steps were investigated, and the structures of polymers formed in each step were characterized by various analytical methods. The results showed that the molecular structure of the PVA‐g‐PPDO could be controlled easily by the degree of silylation and the feed ratio. In addition, the micellization of amphiphilic PVA‐g‐PPDO copolymers in water was proved by fluorescence spectra and dynamic light scattering, and the relationship between structural parameters of copolymers and micellar properties was studied preliminarily. © 2010 Wiley Periodicals, Inc. Complex amphiphilic polymers were synthesized via core‐first polymerization followed by alkylation‐based grafting of poly(ethylene oxide) (PEO). Inimer 1‐(4′‐(bromomethyl)benzyloxy)‐2,3,5,6‐tetrafluoro‐4‐vinylbenzene was synthesized and subjected to atom transfer radical self‐condensing vinyl polymerization to afford hyperbranched fluoropolymer (HBFP) as the hydrophobic core component with a number‐averaged molecular weight of 29 kDa and polydispersity index of 2.1. The alkyl halide chain ends on the HBFP were allowed to undergo reaction with monomethoxy‐terminated poly(ethylene oxide) amine (PEOx‐NH2) at different grafting numbers and PEO chain lengths to afford PEO‐functionalized HBFPs [(PEOx)y‐HBFPs], with x = 15 while y = 16, 22, or 29, x = 44 while y = 16, and x = 112 while y = 16. The amphiphilic, grafted block copolymers were found to aggregate in aqueous solution to give micelles with number‐averaged diameters (Dav) of 12–28 nm, as measured by transmission electron microscopy (TEM). An increase of the PEO:HBFP ratio, by increase in either the grafting densities (y values) or the chain lengths (x values), led to decreased TEM‐measured diameters. These complex, amphiphilic (PEOx)y‐HBFPs, with tunable sizes, might find potential applications as nanoscopic biomedical devices, such as drug delivery vehicles and 19F magnetic resonance imaging agents. © 2010 Wiley Periodicals, Inc. The solubility nature of many medicines presents a challenge for successful delivery of these drugs to the body. Polymeric carriers are potentially viable as vessels for both the protection and transport of these medicinal substances. In an effort to generate polymeric materials for this desired application, A‐B‐A triblock copolymers have been synthesized with a central block composed of hydrophilic poly (ethylene glycol) (PEG) and flanking hydrophobic sequences composed of five valine units terminated with end groups of varying hydrophobicity. These copolymers were constructed by adding amino acids stepwise to the hydrophilic block using solution phase chemistry. The self‐assembly behavior of all polymers was investigated using fluorimetry with a pyrene probe. In general, copolymers with more hydrophobic end groups exhibited lower critical aggregation concentrations (CACs). Fmoc‐terminated copolymers displayed the lowest CAC of 0.032 mg/mL and demonstrated little cytotoxicity when exposed to SW620 colorectal cancer cells. Transmission electron micrographs show the presence of multiple compartments within these spherical assemblies, which may prove useful in encapsulating medicinal substances. © 2008 Wiley Periodicals, Inc. Novel amphiphilic A14B7 multimiktoarm star copolymers composed of 14 poly(ε‐caprolactone) (PCL) arms and 7 poly(acrylic acid) (PAA) arms with β‐cyclodextrin (β‐CD) as core moiety were synthesized by the combination of controlled ring‐opening polymerization (CROP) and atom transfer radical polymerization (ATRP). 14‐Arm star PCL homopolymers (CDSi‐SPCL) were first synthesized by the CROP of CL using per‐6‐(tert‐butyldimethylsilyl)‐β‐CD as the multifunctional initiator in the presence of Sn(Oct)2 at 125 °C. Subsequently, the hydroxyl end groups of CDSi‐SPCL were blocked by acetyl chloride. After desilylation of the tert‐butyldimethylsilyl ether groups from the β‐CD core, 7 ATRP initiating sites were introduced by treating with 2‐bromoisobutyryl bromide, which further initiated ATRP of tert‐butyl acrylate (tBA) to prepare well‐defined A14B7 multimiktoarm star copolymers [CDS(PCL‐PtBA)]. Their molecular structures and physical properties were in detail characterized by 1H NMR, SEC‐MALLS, and DSC. The selective hydrolysis of tert‐butyl ester groups of the PtBA block gave the amphiphilic A14B7 multimiktoarm star copolymers [CDS(PCL‐PAA)]. These amphiphilic copolymers could self‐assemble into multimorphological aggregates in aqueous solution, which were characterized by dynamic light scattering (DLS), transmission electron microscopy (TEM) and atomic force microscopy (AFM). © 2010 Wiley Periodicals, Inc. Amphiphilic poly(amidoamine) (PAMAM) dendrimers consisting of a hydrophilic dendrimer core and hydrophobic aromatic dansyl or 1‐(naphthalenyl)‐2‐phenyldiazene (NPD) shells have been synthesized. These amphiphilic dendrimers from the zero generation to the third generation self‐assemble into vesicular aggregates in water. The self‐assembly behavior of these dendrimers strongly depends on their generations. The generation dependence has been further investigated by an exploration of their electrochemical properties. A novel amphiphilic biodegradable triblock copolymer (PGL‐PLA‐PGL) with polylactide (PLA) as hydrophobic middle block and poly(glutamic acid) (PGL) as hydrophilic lateral blocks was successfully synthesized by ring‐opening polymerization (ROP) of L ‐lactide (LA) and N‐carboxy anhydride (NCA) consecutively and by subsequent catalytic hydrogenation. The results of cell experiment of PGL‐PLA‐PGL suggested that PGL could improve biocompatibility of polyester obviously. The copolymer could form micelles of spindly shape easily in aqueous solution. The pendant carboxyl groups of the triblock copolymer were further activated with N‐hydroxysuccinimide and combined with a cell‐adhesive peptide GRGDSY. Incorporation of the oligopeptide further enhanced the hydrophilicity and led to formation of spherical micelles. PGL‐PLA‐PGL showed better cell adhesion and spreading ability than pure PLA and the GRGDSY‐containing copolymer exhibited even further improvement in cell adhesion and spreading ability, indicating that the copolymer could find a promising application in drug delivery or tissue engineering. © 2007 Wiley Periodicals, Inc. Reverse iodine transfer polymerization (RITP), offering the appealing potential of the in situ generation of transfer agents out of molecular iodine I2, is employed in the synthesis of anionic amphiphilic diblock copolymers of poly(styrene) and poly(acrylic acid). Starting with well‐characterized poly(styrene) as macro‐transfer agents synthesized by RITP, diblock copolymers poly(styrene)‐b‐poly(tert‐butyl acrylate) of various lengths are successfully yielded in solution with a good architectural control. These blocks are then subjected to acid deprotection and subsequent pH control to give rise to anionic amphiphilic poly(styrene)‐b‐poly(acrylic acid). Besides, homopolymers of tert‐butyl acrylate are produced by RITP both in solution and in emulsion. Furthermore, a fruitful trial of the synthesis of diblock copolymers poly(tert‐butyl acrylate)‐b‐poly(styrene) is carried out through chain extension of the poly(tert‐butyl acrylate) latex as a macro‐transfer agent in seeded emulsion polymerization of styrene. 1,4‐Dibromobenzenes carrying nonpolar hexoxy and polar oligo(ethylene glycol) side chains were subjected to Suzuki polycondensation with a benzene‐1,4‐bisboronic acid ester to produce high‐molar‐mass poly(para‐phenylene)s. The molar masses were determined with size exclusion chromatography with conventional polystyrene and universal calibration. These novel amphiphilically equipped rigid‐rod polymers have the potential to segregate lengthwise into polar and nonpolar domains, a property that has only rarely been described, and promise to exhibit novel interesting supramolecular properties. The oligo(ethylene gylcol) side chains terminate with a silyl‐protected alcohol group, and its deprotection on the polymer was proven to proceed quantitatively. This not only led to a further polarity increase but allows us to attach even more polar (e.g., charged) units in future projects. © 2003 Wiley Periodicals, Inc. Novel calixarene‐centered amphiphilic A2B2 miktoarm star copolymers composed of two PCL arms and two PEG arms with calix[4]arene as core moiety were synthesized by the combination of CROP and “click” chemistry. First, a heterotetrafunctional calix[4]arene derivative with two hydroxyl groups and two alkyne groups was designed as a macroinitiator to prepare calixarene‐centered PCL homopolymers (C4‐PCL) by CROP in the presence of Sn(Oct)2 as catalyst at 110 °C. Next, azide‐terminated PEG (A‐PEG) was synthesized by tandem treating methoxy poly(ethylene glycol)s (mPEG) with 4‐chlorobutyryl chloride and NaN3. Finally, copper(I)‐catalyzed cycloaddition reaction between C4‐PCL and A‐PEG led to A2B2 miktoarm star copolymer [C4S(PCL)2‐(PEG)2]. 1H NMR, FT‐IR, and SEC analyses confirmed the well‐defined miktoarm star architecture. These amphiphilic miktoarm star copolymers could self‐assemble into multimorphological aggregates in water. The calix[4]arene moieties with a cavity <1 nm on the hydrophilic/hydrophobic interface of these aggregates may provide potential opportunities to entrap guest molecules for special applications in supermolecular science. © 2010 Wiley Periodicals, Inc. Amphiphilic diblock copolymers of polystyrene (PS) and poly(N‐vinylpyrrolidone) (PNVP) were prepared by a combination of ATRP and MADIX. Well‐defined PS with bromine end group was synthesized by ATRP in bulk at 110 °C using (1‐bromoethyl) benzene as an initiator. The Br‐ end group was then converted to xanthate as verified by 1H NMR spectroscopy, elemental analysis, and UV‐spectroscopy. PS‐b‐PNVP copolymers were produced by MADIX of NVP in bulk at 60 °C using PS‐xanthate as a macro‐chain transfer agent and the kinetics of polymerization were investigated. The structures of PS‐b‐PNVP were characterized using GPC and 1H NMR. Amphiphilic PS‐b‐PNVP could form spherical micelles with PS cores and PNVP shells in aqueous solution as confirmed by 1H NMR and laser light scattering (LLS). The values of critical micelle concentration of PS‐b‐PNVP and the average aggregation number of PS‐b‐PNVP in the micelles were measured using pyrene as a probe and static LLS, respectively. The aggregation number increases concomitantly with temperature (10–50 °C), but the hydrodynamic radius of the micelles remains almost constant over the same temperature range, which may indicate shell dehydration at a higher temperature. © 2008 Wiley Periodicals, Inc. Thermo‐sensitive amphiphilic copolymers, PVCL‐PTrpAMT and PVCL‐PVP‐PTrpAMT of hydrophilic N‐vinylcaprolactam (VCL), N‐vinylpyrrolidone (NVP), and hydrophobic Nt‐Boc‐tryptophanamido‐N′‐methacryl thioureas (TrpAMT) monomers, were synthesized and characterized by 1H NMR, UV‐spectroscopy, and GPC‐MALLS. The cloud point (CP) measurement showed that hydrophobic PTrpAMT and hydrophilic PVP segments significantly altered the phase transition temperature of PVCL with comparable molecular weight in aqueous solution. The CP of PVP‐PTrpAMT solution was 38.0°C, lower by 5.0°C than that of unmodified PVCL. In the presence of phosphate buffer saline (PBS), the CP value of the PVCL polymer decreased by ~2.0°C in comparison to that of the aqueous solution. Fluorescent spectroscopy and TEM studies revealed that PVCL‐PTrpAMT and PVCL‐PVP‐PTrpAMT self‐assembled into the spherical micelles, 30–70 nm in diameter, at concentrations over their CMCs in an aqueous solution. Cytotoxicity tests demonstrated that the PVCL copolymers were not harmful to cell viability, which may favor the use of the copolymers as potential thermo‐sensitive polymers in pharmaceutical applications. Per‐2,3‐acetyl‐β‐cyclodextrin with seven primary hydroxyl groups was synthesized by selective modification and used as multifunctional initiator for the ring‐opening polymerization of ε‐caprolactone (CL). Well‐defined β‐cyclodextrin‐centered seven‐arm star poly(ε‐caprolactone)s (CDSPCLs) with narrow molecular weight distributions (≤1.15) have been successfully prepared in the presence of Sn(Oct)2 at 120 °C. The molecular weight of CDSPCLs was characterized by end group 1H NMR analyses and size‐exclusion chromatography (SEC), which could be well controlled by the molar ratio of the monomer to the initiator. Furthermore, amphiphilic seven‐arm star poly(ε‐caprolactone‐b‐ethylene glycol)s (CDSPCL‐b‐PEGs) were synthesized by the coupling reaction of CDSPCLs with carboxyl‐terminated mPEGs. 1H NMR and SEC analyses confirmed the expected star block structures. Differential scanning calorimetry analyses suggested that the melting temperature (Tm), the crystallization temperature (Tc), and the crystallinity degree (Xc) of CDSPCLs all increased with the increasing of the molecular weight, and were lower than that of the linear poly(ε‐caprolactone). As for CDSPCL‐b‐PEGs, the Tc and Tm of the PCL blocks were significantly influenced by the PEG segments in the copolymers. Moreover, these amphiphilic star block copolymers could self‐assemble into spherical micelles with the particle size ranging from 10 to 40 nm. A series of block copolymers comprising poly(N‐isopropylacrylamide) (PNIPAM) and poly(ethylene oxide) (PEO) end‐functionalized with a quaternary ammonium group (RQ) was synthesized by free‐radical polymerization of N‐isopropylacrylamide with well‐defined RQPEO macroazoinitiators. The radical termination occurred mainly by disproportionation, as confirmed by combining the data from size exclusion chromatography (SEC) and rheology measurements. The copolymers denoted RQExNy differ in type of the terminal group [FQ = C8F17(CH3)2N+ or MQ = (CH3)3N+] and in the length of the PEO (Ex; x = 4, 6, or 10 K) and PNIPAM (Ny; y = 7 or 17–19 K) blocks. The type of the terminal group determined the behavior of the block copolymers in the dilute and semidilute regime. Self‐assembled species formed by both FQ and MQ modified block copolymers were detected by static light scattering measurements at 25 °C and above the lower critical solution temperature (LCST). The LCST of the block copolymers depended on the type of the RQ group and the length of the blocks. FQ‐modified copolymers form elastic gels below and above the LCST. It was inferred that the FQ groups and the PNIPAM blocks form segregated microdomains that serve as junctions to maintain a viscoelastic network. © 2004 Wiley Periodicals, Inc. This article discusses an effective route to prepare amphiphilic diblock copolymers containing a poly(ethylene oxide) block and a polyolefin block that includes semicrystalline thermoplastics, such as polyethylene and syndiotactic polystyrene (s‐PS), and elastomers, such as poly(ethylene‐co‐1‐octene) and poly(ethylene‐co‐styrene) random copolymers. The broad choice of polyolefin blocks provides the amphiphilic copolymers with a wide range of thermal properties from high melting temperature ~270 °C to low glass‐transition temperature ~?60 °C. The chemistry involves two reaction steps, including the preparation of a borane group‐terminated polyolefin by the combination of a metallocene catalyst and a borane chain‐transfer agent as well as the interconversion of a borane terminal group to an anionic (? O?K+) terminal group for the subsequent ring‐opening polymerization of ethylene oxide. The overall reaction process resembles a transformation from the metallocene polymerization of α‐olefins to the ring‐opening polymerization of ethylene oxide. The well‐defined reaction mechanisms in both steps provide the diblock copolymer with controlled molecular structure in terms of composition, molecular weight, moderate molecular weight distribution (Mw/Mn < 2.5), and absence of homopolymer. © 2002 Wiley Periodicals, Inc. The present article describes the synthesis and emulsion copolymerization of a block-type amphiphilic poly(2-oxazoline) macromonomer possessing a polymerizable vinyl ester group. The macromonomer was synthesized by one-pot two-stage block copolymerization of 2-oxazolines using vinyl iodoacetate as initiator. 2-Methyl- and 2-n-butyl-2-oxazolines were employed for the construction of hydrophilic and hydrophobic segments, respectively. The surface activities evaluated by the surface tension of the macromonomer in water were fairly good. Emulsion copolymerization of vinyl acetate with the macromonomer was carried out. The macromonomer acted as a polymeric surfactant, as well as a comonomer. The resulting copolymer latex particles were spherical and their diameter was in the sub-micron range. The effects of the composition of the macromonomer on the emulsion copolymerization and the resulting latex particles were examined. © 1993 John Wiley & Sons, Inc.  相似文献

In this work, a series of PLGA‐PEG diblock copolymers were synthesized by ring‐opening polymerization of L‐lactide and glycolide using mPEG as macroinitiator and stannous octoate as catalyst. Spherical micelles were obtained from the various copolymers by using co‐solvent evaporation method. The biocompatibility of micelles was evaluated with the aim of assessing their potential in the development of drug delivery systems. Various aspects of biocompatibility were considered, including MTT assay, agar diffusion test, release of cytokines, hemolytic test, dynamic clotting time, protein adsorption in vitro, and zebrafish embryonic compatibility in vivo. The combined results revealed that the micelles present good cytocompatibility and hemocompatibility in vitro. Moreover, the cumulative effects of micelles throughout embryos developing stages have no toxicity in vivo. It is thus concluded that micelles prepared from PLGA‐PEG copolymers present good biocompatibility as potential drug carrier.  相似文献
