Starting from a previously described aromatic oligoamide helically folded capsule that binds tartaric acid with high affinity and diastereoselectivity, we demonstrate the feasibility of the direct in situ modification of the helix backbone, which results in a conformational change that reduces its affinity for guests by two orders of magnitude. Specifically, ring contraction of the central pyridazine unit into a pyrrole in the full helical sequence was investigated by using electrochemical and chemical processes. The sequence containing the pyrrole was synthesized independently in a convergent manner to ascertain its structure. The conformation of the pyrrolic folded capsule was elucidated in the solid state by X‐ray crystallography and in solution by using 1H and 13C NMR spectroscopy. Solution studies revealed an unanticipated solvent‐dependent equilibrium between the antianti and synsyn conformations of the pyrrole ring with respect to its two adjacent pyridine units. Titrations of the pyrrole‐containing sequence monitored by 1H NMR spectroscopy confirmed the expected drop in affinity for tartaric acid and malic acid that arises from the conformation change in the backbone that follows the replacement of the pyridazine by a pyrrole. The reduction of the pyridazine to a pyrrole was characterized by cyclic voltammetry both on the entire sequence and on a shorter precursor. The lower cathodic potential of the precursor made its preparative‐scale electroreduction possible. Direct in situ modification of the pyridazine within the entire capsule sequence was achieved chemically by using zinc in acetic acid.  相似文献

A variety of unsaturated thioethers have been subjected to cross‐coupling reactions with functionalized zinc reagents in the presence of a transition‐metal catalyst. Three different catalytic systems based on Pd(OAc)2 or [Ni(acac)2] and the ligands S‐Phos or DPE‐Phos gave the best results. N‐Heterocyclic thioethers based on a pyridine, pyrimidine, pyrazine, pyridazine, triazine, benzothiazole, benzoxazole, pyrrole, or quinazoline ring, as well as thiomethylacetylenes, serve as electrophiles in this cross‐coupling reaction. Aryl‐, heteroaryl‐, benzylic, and alkylzinc halides with sensitive functionalities, such as ester, nitrile, or ketone groups react at ambient temperature with unsaturated thioethers using a Ni catalyst. The corresponding Pd‐catalyzed reactions require slightly higher temperatures. Large‐scale cross‐coupling experiments (10–20 mmol) with N‐heterocycles are also reported.  相似文献

Enantioselective synthesis of pyrrole‐annulated medium‐sized‐ring compounds by an iridium‐catalyzed allylic dearomatization/retro‐Mannich/hydrolysis sequence is presented. Various substituted pyrrole‐annulated seven‐ and eight‐membered‐ring products were obtained under mild reaction conditions with moderate to good yields and excellent enantioselectivity. Additionally, these products contain a scaffold widely distributed in natural products and biologically active compounds. The current method provides a convenient way for accessing such pyrrole‐anuulated medium‐sized‐ring compounds.  相似文献

In the isomeric compounds 2‐benzyl‐3‐methyl‐1‐phenyl­benzo­[b]­furo­[2,3‐c]­pyrrole and 2‐benzyl‐1‐methyl‐3‐phenyl­benzo­[b]­furo­[2,3‐c]­pyrrole, both C24H19NO, the pyrrole ring, although presumably somewhat strained, does not differ appreciably from N‐methyl­pyrrole except for a relatively short C—C single bond in the pyrrole ring.  相似文献

Some azido‐ and iminophosphorane derivatives of 3,6‐dichloro‐ and 3,4,5,6‐tetrachloropyridazine were synthesized and studied by means of NMR measurements. Based on multinuclear data (chemical shifts, coupling constants) for compounds containing the azide group, no potentially possible tetrazole–azide equilibrium can be observed, even under acidic conditions. An unusual substitution of a chlorine atom (in position 4) of tetrachloropyridazine in the reaction with hydrazine was demonstrated by NMR measurements of two newly synthesized compounds containing azido‐ and iminophosphorane groups. Using multinuclear magnetic resonance data, the sites of ethylation and protonation of azido‐ and iminophosphorane derivatives of chloropyridazines were established. In the case of the tetrazolopyridazines, ethylation occurs at the N1′ and N2′ atoms, whereas for monocyclic compounds it takes place at the N1 and/or N2 atoms of the pyridazine ring. Preferred sites of protonation are the N1′ atom of the tetrazole ring and the N1 atom of the pyridazine ring. Moreover, the structures of potassium salts of 6‐(3‐cyano‐1‐triazeno)tetrazolo[1,5‐b] pyridazine and its amido derivative were established using NMR data, especially 15N NMR chemical shifts. 2‐Phenyl‐1,1,3‐tricyanopropene[α‐(cyanomethyl)benzylidene‐malononitrile] undergoes bromination with N‐bromosuccinimide (NBS) to afford 2‐phenyl‐1,1,3‐tricyano‐3‐bromopropene: [α(bromocyanomethyl)benzylidene malononitrile]. This bromo derivative undergoes reactions with sodium hydrogen sulfide, hydrazine hydrate, phenyl hydrazine, hydroxylamine hydrochloride, ethyl thioglycollate, urea derivatives, and cyanacetohydrazide to afford thiophene, 4H‐pyridazines, 4H‐oxazine and 4H‐thiopyran, N‐substituted pyrrole, and pyrrolo[1,2‐b]pyridazine derivatives respectively.  相似文献

Abstract

Owing to increasing demand for phenothiazine compounds both in medicine and industry, novel rings in these series have been prepared and evaluated. The benzene rings have been replaced with pyrrole, pyrazole, pyridine, pyridazine, pyrimidine, pyrazine, thiazine and triazine rings or their combinations leading to twelve new polyaza- and thia-phenothiazines. Expansion of the central thiazine ring in these systems has also been carried out. This article therefore provides a survey of the chemistry and applications of new polyaza- and thia-phenothiazine systems and their related thiazepines and thiazocines.  相似文献

(+)‐Ryanodine ( 1 ) is the ester derivative of 1H‐pyrrole‐2‐carboxylic acid and the complex terpenoid (+)‐ryanodol ( 2 ), which possesses eleven contiguous stereogenic centers on the ABCDE‐ring system. Compound 1 is known to be a potent modulator of intracellular calcium release channels, whereas the activity of 2 is significantly weaker. To chemically construct 1 , the multiple oxygen functional groups must be installed on the fused pentacycle in stereoselective fashions and the extremely hindered C3‐hydroxy group must be acylated in a site‐selective manner. First, the total synthesis of 2 was accomplished by introducing the five stereocenters from the previously prepared enantiopure ABDE‐ring 7 . Stereoselective construction of the C3‐secondary, C2‐ and C6‐tertiary alcohols was achieved by three nucleophilic reactions. The C9‐ and C10‐trisubstituted carbon centers were regio‐ and stereoselectively introduced by hydroboration/oxidation of the six‐membered C‐ring, which was formed by the ring‐closing metathesis reaction. Direct esterification of the C3‐alcohol with pyrrole‐2‐carboxylic acid proved unsuccessful; therefore, we developed a new, two‐step protocol for attachment of the pyrrole moiety. The C3‐hydroxy group was first converted into the less sterically cumbersome glycine ester, which was then transformed into the pyrrole ring through condensation with 1,3‐bis(dimethylamino)allylium tetrafluoroborate. Reaction of unsaturated fulvene endoperoxides with dimethyl 1,2,4,5‐tetrazine‐3,6‐dicarboxylate gave saturated fulvene endoperoxides containing the 1,2‐dihydropyridazine ring. Treatment of dihydropyridazine endoperoxides with water followed by phenyliodosyl bis(trifluoroacetate) oxidation provided acrylic acid derivatives and dimethyl pyridazine‐3,6‐dicarboxylate.  相似文献

Using 3‐(4‐cyano phenoxy)‐6‐methyl‐4‐(3‐trifluoromethylphenyl) pyridazine (compound A ) as a leading compound, a total of 24 novel 3‐(substituted phenoxy)‐6‐methyl‐4‐(3‐trifluoromethylphenyl) pyridazine derivatives containing two electron‐withdrawing groups on the benzene ring (acylamine and oxime ether) were synthesized. Their herbicidal, insecticidal activities were bioassayed, and the herbicidal activity of compound CD-2 against Brassica campestris was 97.6% at 300 g/ha, which was better than the commercial herbicide diflufenican at the this concentration and is equal to the activity of the leading compound A . Compound CD-4 , CD-5 , CJ-3 , and CJ-5 displayed excellent insecticidal activity against Aphis laburni Kaltenbach (>95%). The results show that the oxime ether substitutions exhibit better bleaching and herbicidal activity than the acylamine ones. The bleaching and herbicidal activity of para‐position substitutions is better than the meta‐position ones. It seems that the para‐position on the benzene ring of oxime ether pyridazine derivatives is one of the key active sites that affect their herbicidal activities.  相似文献

Diels-Alder adducts were formed in the lead tetraacetate oxidations of substituted cyclic hydrazides of furan, pyridazine and pyrrole dicarboxylic acids in the presence of 1,3-cyclo-hexadiene or 1,3-cyclopentadiene. The products resulting were furo[3,4-g]pyridazino[1,2-a]-pyridazine-6,10-diones, pyridazino[4,5-g]pyridazino[1,2-a]pyridazine-6,11-diones, and pyrrolo-[3,4-g]pyridazino[1,2-a]pyridazine-6,10-diones, respectively. Indoles are omnipresent in natural products, bioactive molecules, and organic materials. Consequently, their synthesis or functionalization are important fields of research in organic chemistry. Most works focus on installation or modification of the pyrrole ring. To access benzene‐ring‐functionalized indoles with an unsubstituted pyrrole ring remains more challenging. Reported herein is a platinum‐catalyzed cyclization/alkynylation domino process to selectively obtain C5‐ or C6‐functionalized indoles starting from easily available pyrroles. The work combines, for the first time, a platinum catalyst with ethynylbenziodoxole hypervalent iodine reagents in a domino process for the synthesis of polyfunctionalized arene rings and gives access to important building blocks for the synthesis of bioactive compounds and organic materials.  相似文献

New tricyclic 1,2,3‐triazolo‐1,2,4‐triazolo‐pyridazine derivatives, bearing a methyl substituent on the 1,2,3‐triazole ring, were prepared as potential biological agents. N‐Methylation of dimethyl 1,2,3‐triazole‐4,5‐dicarboxylate allowed synthesis of the isomeric 1‐methyl‐4,7‐dihydroxy and 2‐methyl‐4,7‐dihydroxy triazolo‐pyridazines 4a and 4b which, by a chlorination reaction, gave the corresponding 1‐methyl‐4‐chloro‐( 6a ), 1‐methyl‐7‐chloro‐ ( 6b ) and 2‐methyl‐4‐chloro‐ ( 9 ) substituted 1,2,3‐triazolo‐pyridazines. The nucle‐ophilic substitution with hydrazine hydrate and the suitable cyclization to form the 1,2,4‐triazole ring, provided the expected tricyclic isomeric derivatives 8a, 8b and 11 respectively. The p‐methoxybenzyl substituent, introduced as a leaving group to obtain either v‐triazolo‐pyridazine or v‐triazolo‐s‐triazolo‐pyri‐dazine derivatives unsubstituted on the 1,2,3‐triazole ring, appeared inadequate. Some compounds underwent binding assays toward the adenosine A1and A2A receptors.  相似文献

A new hydrogen terminated 2,2′‐bidipyrrin ligand was prepared from a bipyrrole dialdehyde and 3,4‐diethylpyrrole by a POCl3 induced condensation and isolated as the nickel(II) chelate. Unexpectedly a side reaction occured when base‐deficient and aerobic conditions were chosen in the metalation step. This side reaction led to a novel pentapyrrolic nickel(II) complex with one externally bound pyrrole ring. Further studies showed that the reactions of 2,2′‐bidipyrrins with 3,4‐diethyl‐ or 2,3,4‐trimethylpyrrole and an oxidant resulted in a stepwise exchange of the terminal pyrrole moieties and, in the former case, the introduction of one additional pyrrole ring into one of the two meso positions of the open‐chain tetrapyrrole.  相似文献

Triazolo[3,4‐a]phthalazine as well as their chloro and nitro derivatives were subjected to the reactions with the carbanions typical for the vicarious nucleophilic substitution (VNS) of hydrogen. The reactions were strongly dependent on the substituents present on the triazolo[3,4‐a]phthalazine ring and resulted not only in the substitution of hydrogen but also in exchange of chlorine atom and pyridazine ring scission; the latter process dominated for the unsubstituted triazolophthalazine. Two of the products showed promising stimulating activity towards the central nervous system with no significant toxic effects.  相似文献

Pyrrole and pyrrolidine derivatives of pyrimidine were prepared in which the nitrogen atom of the pyrrole or pyrrolidine ring is bonded directly to the 2‐ or 4‐carbon atom of the pyrimidine ring. Pyrrole derivatives were prepared by the dry distillation of an intimate mixture of an aminopyrimidine with mucic acid and by the reaction of a chloropyrimidine with potassium pyrrole. Pyrrolidine derivatives were prepared by the reaction of a chloropyrimidine with pyrrolidine and, in a single instance, by the catalytic hydrogenation of a pyrimidinylpyrrole. At a concentration of 200 mcg/mL, 1‐(2‐pyrimidinyl)pyrrole inhibited two plant pathogenic bacteria — Xanthomanus phaseoli (pathogenic on the bean plant) and Xanthomanus malvacearum (pathogenic on the cotton plant).  相似文献

Three new heterocyclic ring systems, isoxazolo[4,5‐d]‐1,2,4‐triazolopyridazines 12‐15 , tetrazolo‐[4,3‐b]pyridazine 18 and isoxazolo[4,5‐d]pyridazino[2,3‐c]2H‐triazines 16 , 17 along with isoxazolo[4,5‐d]pyridazines 2 , 5‐10 have been synthesized. Preliminary screening of these tricyclic heterocycles revealed that some of them possess significant antibacterial and antifungal activity.  相似文献

Dimedone adduct with 1,2-dibenzoylethene in reaction with p-phenylenediamine formed a bispyrrole derivative, and with benzidine and ethylenediamine afforded the corresponding monopyrroles. The reaction of the initial adduct with tryptamine results in a product containing a pyrrole and an indole fragments. Both versions of N-heterocyclization proceed in the reaction with urea yielding derivatives of tetrahydroindole and pyrrole. In the reaction with hydrazine hydrate a substituted pyridazine was obtained as a result of a dimedone molecule elimination from the intermediate cyclization product.  相似文献
