Four mononuclear metal complexes (Cu(II) ( 1 ), Ni(II) ( 2 ), Zn(II) ( 3 ) and V(IV) ( 4 )) were synthesized using the Schiff base ligand 2,2′‐{cyclohexane‐1,2‐diylbis[nitrilo(1E )eth‐1‐yl‐1‐ylidine]}bis[5‐(prop‐2‐yn‐1‐yloxy)phenol] and structurally characterized by various spectral techniques. The catecholase‐mimicking activities of 1 – 4 were investigated and the results reveal that all the complexes have ability to oxidize 3,5‐di‐tert ‐butylcatechol (3,5‐DTBC) to 3,5‐di‐tert ‐butylquinone in aerobic conditions. Electrospray ionization mass spectrometry studies were performed for 1 – 4 in the presence of 3,5‐DTBC to determine the possible complex–substrate intermediates. X‐band electron paramagnetic resonance spectroscopy results indicate that the metal centres are involved in the catecholase activity. Ligand‐centred radical generation was further confirmed by density functional theory calculation. The phosphatase‐like activity of 1 – 4 was investigated using 4‐nitrophenylphosphate as a model substrate. All the complexes exhibit excellent phosphatase activity in acetonitrile medium. The interactions of 1 – 4 with calf thymus DNA (CT‐DNA) and bovine serum albumin (BSA) protein were investigated using absorption and fluorescence titration methods. All the complexes strongly interact with CT‐DNA and BSA protein. The complexes exhibit significant hydrolytic cleavage of supercoiled pUC19 DNA. Complexes 1 – 4 exhibit significant in vitro cytotoxicity against MCF7 (human breast cancer) and MIA‐PA‐CA‐2 (human pancreatic cancer) cell lines. Moreover, the molecular docking technique was employed to determine the binding affinity with DNA and protein molecules.  相似文献

The antioxidant activity of two primary antioxidants, 2,2′‐methylenebis(4‐methyl‐6‐tert ‐butylphenol) (MMBPH2) and 2,2′‐methylenebis(4,6‐di‐tert ‐butylphenol) (MDBPH2), has been studied using the 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) method. The synthesized compounds have been successfully characterized systematically using elemental analyses, infrared, 1H NMR and 13C NMR spectra and GC–MS. Importantly, it has been found that the compound MMBPH2 in particular is more active in DPPH radical scavenging. In addition, density functional theory calculations (B3LYP) have been used to predict the antioxidant activity and predict structural geometries of the compounds in the gas phase.  相似文献

Schiff base derivatives have gained great importance due to revealing a great number of biological properties. Schiff bases were synthesized by treatment of 4-amino-1,5-dimethyl-2-phenyl-1H-pyrazol-3(2H)-one ( 1 ) with various aldehydes in methanol at reflux. In addition, diamine was reacted with an aldehyde to yield the corresponding Schiff bases. The structures of synthesized Schiff bases were elucidated by spectroscopic methods such as microanalysis, 1H-NMR, 13C-NMR, and FTIR. Antioxidant activities of synthesized Schiff bases were carried out using different antioxidant assays such as 1,1-diphenyl-2-picryl-hydrazyl free radical (DPPH) scavenging, 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging, and reducing power activity. (E)-4-((1H-indol-3-yl)methyleneamino)-1,5-dimethyl-2-phenyl-1H-pyrazol-3(2H)-one ( 3 ), (E)-1,5-dimethyl-4-((2-methyl-1H-indol-3-yl)methyleneamino)-2-phenyl-1H-pyrazol-3(2H)-one ( 5 ), (E)-1,5-dimethyl-2-phenyl-4-(thiophen-2-ylmethyleneamino)-1H-pyrazol-3(2H)-one ( 7 ), (E)-1,5-dimethyl-2-phenyl-4-(quinolin-2-ylmethyleneamino)-1H-pyrazol-3(2H)-one ( 9 ), (1S,2S,N1,N2)-N1,N2-bis((1H-indol-3-yl)methylene)cyclohexane-1,2-diamine ( 11 ), and (1S,2S,N1,N2)-N1,N2-bis((2-methyl-1H-indol-3-yl)methylene)cyclohexane-1,2-diamine ( 12 ) were synthesized in high yields. Compound 5 displayed a good ABTS•+ activity. Compound 3 revealed the outstanding activity in all assays. Compound 7 has the best-reducing power ability in comparison to other synthesized compounds. Although compounds 5, 11, 12 are new, compounds 3, 7, 9 are known. Due to revealing a good antioxidant activity, the synthesized compounds ( 3, 5, 7 ) have the potential to be used as synthetic antioxidant agents.  相似文献

Some new derivatives 7‐chloro‐2‐[2‐(2,6‐dichlorophenyl)amino]benzyl‐3‐[4‐(2‐substituted phenyl‐4‐oxo‐ thiazolidin‐3‐yl)phenyl]sulfonamido‐quinazolin‐4(3H)‐ones 5a – 5l were synthesized from 2‐[2‐(2,6‐dichloro‐phenyl)amino]phenyl acetic acid via acid chloride, benzoxazinone, amino quinazolin‐4(3H)‐one and Schiff base formation. The synthesized compounds were screened for in vitro antibacterial and antifungal activities by broth micro dilution method. Some of the Schiff base as well as 4‐thiazolidinone derivatives showed promising antibacterial activity while pronounced antifungal activity was observed against C. albicans.  相似文献

A series of homo‐ and hetero‐trinuclear cobalt(II) complexes [Co3(L)(OAc)2(CH3CH2OH)(H2O)] ( 1 ), [Co2Ba(L)(OAc)2] ( 2 ) and [Co2Ca(L)(OAc)2]·CHCl3 ( 3 ), containing an acyclic naphthalenediol‐based ligand H4L were synthesized. All the three complexes were characterized by elemental analyses, IR, UV – vis spectra and single crystal X‐ray diffraction analyses. Comparative studies of the structures and spectroscopic properties are carried out on these complexes. All of the complexes show catechol oxidase activities in MeCN. Using UV – vis spectroscopy, we monitored the aerial oxidation of 3,5‐di‐tert ‐butylcatechol (3,5‐DTBCH2) to 3,5‐di‐tert ‐butylquinone (3,5‐DTBQ), which confirms the essential role of these complexes in enhancing the catalytic reaction.  相似文献

1‐Amino‐2‐pyrid‐3‐yl‐5‐(2‐benzoylethylthio)‐s‐triazole ( 1 ) was condensed with 1‐amino‐3‐mercapto‐5‐ [(un)substituted phenyl]‐s‐triazoles and subsequently substituted with chloroacetic acid to afford bis‐s‐triazole sulfanylacetic acid mono‐Schiff bases ( 3a – 3e ), which were condensed with 9‐formylanthracene to produce asymmetric bis(s‐triazole Schiff base) sulfanylacetic acids ( 4a – 4e ). The structures of new synthesized compounds were characterized by elemental analysis and spectral data, and their in vitro antitumor activity against L1210, CHO and HL60 cell lines was evaluted via the respective IC50 values by methylthiazole trazolium (MTT) assay.  相似文献

The reaction of the β‐diketoiminate lithium complex (dipp)NacNacLi · OEt2 ((dipp)NacNac = 2‐((2,6‐diisopropylphenyl)amino)‐4‐((2,6‐diisopropylphenyl)imino)‐pent‐2‐enyl) with iPrMgCl and MgI2 yield the corresponding (dipp)NacNacMgiPr · OEt2 ( 1 ) and (dipp)NacNacMgI · OEt2 ( 2 ). The reaction of 2 with NaBH4 in diethylether gives (dipp)NacNacMg(μ‐H)3BH · OEt2 ( 3 ). The core element of compounds 1 – 3 is a six‐membered ring formed by N(1)–C(1)–C(2)–C(3)–N(2) and magnesium. The structures of 1 and 2 show the β‐diketoiminate backbone in a boat‐conformation with the tetrahedrally coordinated metal center at the prow and the opposing carbon atom at the stern. The magnesium atom in 3 is octahedrally coordinated and out of the β‐diketoiminate plane.  相似文献

Synthesis of ferrocenyl Schiff bases ( 1–6 ) was carried out by the condensation reaction of 4‐ferrocenyl aniline with different substituted aromatic aldehydes and acetyl acetone. Compounds were characterized by physical measurements, elemental analysis, FT‐IR, 1H‐NMR and 13C‐NMR spectroscopy. Single crystal X‐ray analysis of compound 2 showed the co‐planarity of both aromatic rings connected by a C–N double bond. Compounds demonstrated reversible one‐electron redox behavior and their peak currents were found to increase linearly with the square root of the sweep rate ν1/2. The overall electrode processes were found to be diffusion controlled. Compounds 1 and 4 showed low cytotoxicity and appreciable antifungal, antioxidant and DNA protection activities. 2-Amino substituted benzothiazole 4a--4I and p-acetamidobenzenesulfonyl chloride 2 were used to prepare 2-(p-aminophenylsulfonamido) substituted benzothiazole 6a--6I using mixture of pyridine and acetic anhydride which formed an electrophilic complex (N-acetyl pyridinium) to facilitate condensation to give desired product by removal of HC1. 2-{p-[(3-Carboxypyrid-2-y1)amino]phenylsulfonamido}benzothiazoles 8a--81 were synthesized from 2-chloropyridine-3-carboxylic acid 7 and 6a--6I in 2-ethoxy ethanol using Cu-powder and K2CO3. Acid chlorides 9a--91 were condensed with 2-hydroxyethyl piperazine 10 and 2,3-dichloropiperazine 11 for amide deriva- tives 2-(p-((3-(4-(2-hydroxyethy1)piperazin-1-ylcarbonyl)pyrid-2-y1)amino)phenylsulfonamido)benzothiazoes 12a -121 and 2-{p-[3-(2,3-dichloropiperazin-l-ylcarbonyl)pyrid-2-ylamino]phenylsulfonamido}benzothiazoles 13a- 131 respectively. The structures of the new compounds have been established on the basis of their chemical analysis and spectral data (IR, 1↑H NMR and mass). All the compounds have been screened for their antibacterial and antifungal activities.  相似文献

A unique hexanuclear zinc(II) ( 1 ) and two mononuclear copper(II) ( 2 and 3 ) complexes anchored with imino phenol ligand HL 1 and HL 2 were synthesized with good yield and purity (where HL 1  = 4‐tert‐butyl‐2,6‐bis((mesitylimino)methylphenol and HL 2   =  5‐tert‐butyl‐2‐hydroxy‐3‐((mesitylimino)methyl)benzaldehyde). These complexes were characterized by utilizing various spectroscopic protocols like NMR, FTIR, UV as well as ESI‐Mass spectrometry, elemental analysis and single crystal X‐ray diffraction studies. Their potential to bind calf thymus DNA (CT‐DNA) was tested utilizing different techniques such as UV–visible and fluorescence spectroscopy. The experiment implies that they interact with CT‐DNA via non‐intercalative mode with moderate capabilities (Kb ~ 104 M?1). On the other hand, these complexes have high capabilities to quench the fluorescence of bovine serum albumin (BSA) following the static pathway. In addition, they are active catalysts for the oxidation reaction of 3,5‐di‐tert‐butylcatechol (3,5‐DTBC) to 3,5‐di‐tert‐butylquinone (3,5‐DTBQ) under aerobic condition. From the recorded EPR signals of all complexes, it has been concluded that the oxidation reaction proceeds via ligand oriented radical pathway instead of metal based redox participation. Kinetic studies using 1 – 3 indicate that it follows Michaelis–Menten type of equation with moderate to high turnover number (kcat). Apart from these aspects, complexes 1 – 3 were screened for their cytotoxic behavior towards HeLa cells (human cervical carcinoma) and found quite active with comparable IC50 values to cisplatin.  相似文献

Six novel derivatives of 2‐ferrocenyl‐5‐aryl‐1,3,4‐oxadiazoles have been synthesized through catalytic oxidation of benzoylhydrazone with nitrophthalocyanine iron (II) as catalyst and tert‐butyl hydroperoxide as oxidant. All of the compounds have been characterized by high‐resolution mass spectrometry, 1H, 13C NMR, and infrared spectroscopy. Moreover, the antifungal activities of all new compounds have been evaluated and the results showed that the compounds exhibited excellent antifungal activities. In addition, the molecular structure of compound 1c could also be determined by X‐ray single‐crystal diffraction.  相似文献

Synthesis of the title compounds 4(a – i) was accomplished through a two‐step process. The synthetic route involves the cyclization of equimolar quantities of 2,2′‐methylene(methyl)bis(4,6‐di‐tert‐butyl‐phenol) ( 1 ) with tris‐(2‐chloro‐ethyl) phosphite ( 2a ), tris‐(2‐bromo‐ethyl) phosphine ( 2b ), and tris‐bromo methyl phosphine ( 2c ) in the presence of sodium hydride in dry tetrahydrofuran at 45–50°C. They were further converted to the corresponding oxides, sulfides, and selenides under N2 atmosphere by reacting them with hydrogen peroxide, sulfur, and selenium, respectively ( 4a – c , 4d – f, and 4g – i ). But the compounds 6a , b were prepared by the direct cyclocondensation of equimolar quantities of 1 with (2‐chloro‐ethyl)‐phosphonic acid dibromomethyl ester ( 5a ) and (2‐chloro‐ethyl)‐phosphonic acid bis(2‐bromo‐ethyl) ester ( 5b ) in the presence of sodium hydride in dry tetrahydrofuran at 45–50°C in moderate yields. All the newly synthesized compounds 4 ( a – i ) and 6 ( a – b ) exhibited moderate in vitro antibacterial and antifungal activities.  相似文献

A series of mono‐ and di‐Schiff bases formed between benzil dihydrazone {BDH; systematic name: (1Z)‐[(2E)‐2‐hydrazinylidene‐1,2‐diphenylethylidene]hydrazine} and three isomeric chlorobenzaldehydes were designed and synthesized to be used as model compounds to help to explain the reaction mechanisms for the formation of Schiff bases. These compounds are 1‐(2‐chlorobenzylidene)‐2‐{2‐[2‐(2‐chlorobenzylidene)hydrazin‐1‐ylidene]‐1,2‐diphenylethylidene}hydrazine (BDHOCB), and the 3‐chloro (BDHMCB) and 4‐chloro (BDHPCB) analogues, all having the formula C28H20Cl2N4. Surprisingly, only di‐Schiff bases were obtained; our attempts to push the reaction in favour of the mono‐Schiff bases all failed. Density functional theory (DFT) calculations were performed to explain the trend in the experimental results. In the case of the systems studied, the type of Schiff base produced exhibits a clear dependence on the HOMO–LUMO energy gaps (ΔEHOMO–LUMO), i.e. the product is mainly governed by its stability. The compounds were characterized by single‐crystal X‐ray diffractometry, elemental analysis, melting point, 1H NMR and 13C NMR spectroscopy. The structural features of the three new Schiff bases are similar. For instance, they have the same chemical formula, all the molecules have a symmetrical double helix structure, with each Ph—C=N—N=C—Ph arm exhibiting an anti conformation, and their supramolecular interactions include intermolecular π–π and weak C—H...π stacking interactions. The crystal systems are different, however, viz. triclinic (space group P) for BDHPCB, monoclinic (space group P21/n) for BDHOCB and orthorhombic (space group Pnna) for BDHMCB.  相似文献

A series of novel thiazolidinones, that is, 2‐(2‐hydroxy‐5‐((aryl)‐diazenyl)phenyl)‐3‐(4‐hydroxyphenyl)‐thiazolidin‐4‐one, have been synthesized by reaction of various Schiff bases 2‐(4‐hydroxyphenylimino)methyl)‐4‐(aryl)diazenyl)phenol with ethanolic thioglycolic acid. Schiff bases were obtained by the reactions of 4‐amino phenol with 2‐hydroxy‐5‐((aryl)diazenyl)benzaldehyde. The structures of the newly synthesized compounds were confirmed by IR, 1H NMR, mass spectra, and C, H, N elemental analysis. The thiazolidinone derivatives were evaluated for their antibacterial and antifungal activity.  相似文献

Three new metal complexes [Cu(L)2] (1), [Co(L)2] (2) and [Zn(L)2] (3) have been prepared by the reaction of hydrated salts of metal (II) acetate with new Schiff base ligand HL, [2‐((4‐(dimethylamino)phenylimino)methyl)‐4,6‐di‐t‐butylphenol] and characterized by different physico‐chemical analyses such as elemental analysis, single XRD, 1H NMR, FTIR and UV–Vis spectroscopic techniques. Their biomolecular docking, antimicrobial and cytotoxicity studies have also been demonstrated. The proposed structure of Schiff base ligand HL and complex 2 are confirmed by Single crystal X‐ray crystallography study. This analysis revealed that metal (II) complexes remain in distorted tetrahedral coordination environments. The electronic properties such as HOMO and LUMO energies are carried out by gaseous phase DFT/B3LYP calculations using Gaussian 09 program. Complex 1 showed a good binding propensity to the DNA and HSA, during the assessment of docking studies. Schiff base ligand HL and its metal (II) complexes, 1–3 screened for their in vitro antimicrobial activities using the disc diffusion method against selected microbes. Complex 1 shows higher antimicrobial activity than complexes 2, 3 and Schiff base ligand HL. According to the results obtained from the cytotoxic studies, Schiff base ligand HL and its metal (II) complexes 1–3 have better cytotoxicity against MCF‐7 cell lines with potency higher than the currently used chemotherapeutic agent cyclophosphamide.  相似文献

A new series of (E)‐1‐(4‐((1‐benzyl‐1H‐1,2,3‐triazol‐4‐yl)methoxy)phenyl)‐3‐phenylprop‐2‐en‐1‐one 1a (4‐((1‐benzyl‐1H‐1,2,3‐triazol‐4‐yl) methoxy)phenyl)‐1‐phenylprop‐2‐en‐1‐one 1b – 15b were designed, synthesized based on click chemistry, and biologically evaluated for their activity on tyrosinase. The result showed that most of prepared compounds 1a – 15a have potent activating effect on tyrosinase, especially for 3a , 8a – 10a and 14a – 15a . Among them, compounds 10a and 14a demonstrated the best activity with EC50=1.71 and 5.60 µmol·L?1 respectively, even better than the positive control 8‐MOP (EC50=14.8 µmol·L?1). Conversely, compounds 3b , 5b – 6b , 9b – 10b , and 15b induced enzymatic inhibition on tyrosinase.  相似文献
