The direct electrochemistry of glucose oxidase (GOD) immobilized on the designed titanium carbide‐Au nanoparticles‐fullerene C60 composite film modified glassy carbon electrode (TiC‐AuNPs‐C60/GCE) and its biosensing for glucose were investigated. UV‐visible and Fourier‐transform infrared spectra of the resulting GOD/TiC‐AuNPs‐C60 composite film suggested that the immobilized GOD retained its original structure. The direct electron transfer behaviors of immobilized GOD at the GOD/TiC‐AuNPs‐C60/GCE were investigated by cyclic voltammetry in which a pair of well‐defined, quasi‐reversible redox peaks with the formal potential (E0′) of ‐0.484 V (vs. SCE) in phosphate buffer solution (0.05 M, pH 7.0) at the scan rate of 100 mV·s?1 were obtained. The proposed GOD modified electrode exhibited an excellent electrocatalytic activity to the reduction of glucose, and the currents of glucose reduction peak were linearly related to glucose concentration in a wider linearity range from 5.0 × 10?6 to 1.6 × 10?4 M with a correlation coefficient of 0.9965 and a detection limit of 2.0 × 10?6 M (S/N = 3). The sensitivity and the apparent Michaelis‐Menten constant (KMapp) were determined to be 149.3 μA·mM?1·cm?2 and 6.2 × 10?5 M, respectively. Thus, the protocol will have potential application in studying the electron transfer of enzyme and the design of novel electrochemical biosensors.  相似文献

A mixed‐valence cluster of cobalt(II) hexacyanoferrate and fullerene C60‐enzyme‐based electrochemical glucose sensor was developed. A water insoluble fullerene C60‐glucose oxidase (C60‐GOD) was prepared and applied as an immobilized enzyme on a glassy carbon electrode with cobalt(II) hexacyanoferrate for analysis of glucose. The glucose in 0.1 M KCl/phosphate buffer solution at pH = 6 was measured with an applied electrode potential at 0.0 mV (vs Ag/AgCl reference electrode). The C60‐GOD‐based electrochemical glucose sensor exhibited efficient electro‐catalytic activity toward the liberated hydrogen peroxide and allowed cathodic detection of glucose. The C60‐GOD electrochemical glucose sensor also showed quite good selectivity to glucose with no interference from easily oxidizable biospecies, e.g. uric acid, ascorbic acid, cysteine, tyrosine, acetaminophen and galactose. The current of H2O2 reduced by cobalt(II) hexacyanoferrate was found to be proportional to the concentration of glucose in aqueous solutions. The immobilized C60‐GOD enzyme‐based glucose sensor exhibited a good linear response up to 8 mM glucose with a sensitivity of 5.60 × 102 nA/mM and a quite short response time of 5 sec. The C60‐GOD‐based glucose sensor also showed a good sensitivity with a detection limit of 1.6 × 10‐6 M and a high reproducibility with a relative standard deviation (RSD) of 4.26%. Effects of pH and temperature on the responses of the immobilized C60‐GOD/cobalt(II) hexacyanoferrate‐based electrochemical glucose sensor were also studied and discussed.  相似文献

A nanocylindrical wall structure was obtained by layer‐by‐layer (LbL) assembly of poly‐L ‐arginine (PLA) and human serum albumin (HSA) and characterized by scanning electron microscopy (SEM), scanning force microscopy (SFM), and cryogenic transmission electron microscopy (cryo‐TEM). SEM and SFM measurements of a lyophilized powder of (PLA/HSA)3 nanotubes yielded images of round, chimney‐like architectures with approximately 100 nm wall thickness. Cryo‐TEM images of the hydrated sample revealed that the tube walls are composed of densely packed HSA molecules. Moreover, when small‐angle X‐ray scattering was used to characterize the individual PLA and HSA components in aqueous solutions, maximum diameters of approximately 28 nm and 8 nm were obtained, respectively. These values indicate the minimum thickness of wall layers consisting of PLA and HSA. It can also be concluded from SEM as well as from cryo‐TEM images that the protein cylinders are considerably swollen in the presence of water. Furthermore, HSA retains esterase activity if assembled in nanotubes, as indicated by measurements of para‐nitrophenyl acetate hydrolysis under semi‐physiological conditions (pH 7.4, 22 °C). The enzyme activity parameters (Michaelis constant, Km, and catalytic constant, kcat) were comparable to those of free HSA.  相似文献

Metal–organic frameworks (MOFs) are suitable enzyme immobilization matrices. Reported here is the in situ biomineralization of glucose oxidase (GOD) into MOF crystals (ZIF‐8) by interfacial crystallization. This method is effective for the selective coating of porous polyethersulfone microfiltration hollow fibers on the shell side in a straightforward one‐step process. MOF layers with a thickness of 8 μm were synthesized, and fluorescence microscopy and a colorimetric protein assay revealed the successful inclusion of GOD into the ZIF‐8 layer with an enzyme concentration of 29±3 μg cm?2. Enzymatic activity tests revealed that 50 % of the enzyme activity is preserved. Continuous enzymatic reactions, by the permeation of β‐d ‐glucose through the GOD@ZIF‐8 membranes, showed a 50 % increased activity compared to batch experiments, emphasizing the importance of the convective transport of educts and products to and from the enzymatic active centers.  相似文献

A novel composite was fabricated through dispersing multiwalled carbon nanotubes (MWNTs) in gold nanoparticle (GPs) colloid stabilized by chitosan and ionic liquid (i.e., 1‐butyl‐3‐methylimidazolium tetrafluoroborate, BMIMBF4). Transmission electron microscopy (TEM) experiment showed that the GPs highly dispersed on the MWNTs probably due to the electrostatic interaction among GPs, MWNTs and the imidazolium cation of BMIMBF4. X‐ray photoelectron spectroscopy (XPS) indicated that thus‐formed gold nanostructure was mediated by BMIMBF4. When glucose oxidase (GOD) was immobilized on the composite (MWNTs‐GPs) its ultraviolet‐visible absorption spectrum kept almost unchanged. The immobilized GOD coated glassy carbon electrode (GOD/MWNTs‐GPs/GC) exhibited a pair of well‐defined peaks in 0.10 M pH 7.0 phosphate buffer solution (PBS), with a formal potential of ?0.463 V (vs. SCE). The electrochemical process involved two‐electron transfer. The electron transfer coefficient was ca.0.56 and the electron transfer rate constant was 9.36 s?1. Furthermore, the immobilized GOD presented good catalytic activity to the oxidation of glucose in air‐saturated PBS. The Km and Im values were estimated to be 13.7 μM and 0.619 μA. The GOD/MWNTs‐GPs/GC electrode displayed good stability and reproducibility.  相似文献

A new concept of enzyme inhibition‐based biosensor involving the appearance of an amperometric signal for an inhibition by mercury was developed. The bienzyme sensor was composed of two layers of clay materials. The inner layer was constituted of layered double hydroxides entrapping laccase wired by ABTS. The outer laponite layer contained glucose oxidase (GOD). GOD catalyzed the glucose oxidation with the reduction of O2 into H2O2. This induced a drastic decrease of the biosensor response to O2 by the electrically wired laccase. HgCl2 inhibited the O2 consumption by GOD leading to a signal increase of the electroenzymatic reduction of O2.  相似文献

The direct electrochemistry of glucose oxidase (GOD) was achieved based on the immobilization of GOD on a natural nano‐structural attapulgite (ATP) clay film modified glassy carbon (GC) electrode. The immobilized GOD displayed a pair of well‐defined quasi‐reversible redox peaks with a formal potential (E0′) of ?457.5 mV (vs. SCE) in 0.1 mol·L?1 pH 7.0 phosphate buffer solution. The peak current was linearly dependent on the scan rate, indicating that the direct electrochemistry of GOD in that case was a surface‐controlled process. The immobilized glucose oxidase could retain bioactivity and catalyze the oxidation of glucose in the presence of ferrocene monocarboxylic acid (FMCA) as a mediator with the apparent Michaelis‐Menten constant Kappm of 1.16 mmol·L?1. The electrocatalytic response showed a linear dependence on the glucose concentration ranging widely from 5.0×10?6 to 6.0×10?4 mol·L?1 (with correlation coefficient of 0.9960). This work demonstrated that the nano‐structural attapulgite clay was a good candidate material for the direct electrochemistry of the redox‐active enzyme and the construction of the related enzyme biosensors. The proposed biosensors were applied to determine the glucose in blood and urine samples with satisfactory results.  相似文献

Glucose oxidase (GOD) was encapsulated in chitosan matrix and immobilized on a glassy carbon electrode, achieving direct electron transfer (DET) reaction between GOD and electrode without any nano‐material. On basis of such DET, a novel glucose biosensor was fabricated for direct bioelectrochemical sensing without any electron‐mediator. GOD incorporated in chitosan films gave a pair of stable, well‐defined, and quasireversible cyclic voltammetric peaks at about ?0.284 (Epa) and ?0.338 V (Epc) vs. Ag/AgCl electrode in phosphate buffers. And the peak is located at the potentials characteristic of FAD redox couples of the proteins. The electrochemical parameters, such as midpoint potential (E1/2) and apparent heterogeneous electron‐transfer rate constants (ks) were estimated to ?0.311 V and 1.79 s?1 by voltammetry, respectively. Experimental results indicate that the encapsulated GOD retains its catalytic activity for the oxidation of glucose. Such a GOD encapsulated chitosan based biosensor revealed a relatively rapid response time of less than 2 min, and a sufficient linear detection range for glucose concentration, from 0.60 to 2.80 mmol L?1 with a detection limit of 0.10 mmol L?1 and electrode sensitivity of 0.233 μA mmol?1. The relative standard deviation (RSD) is under 3.2% (n=7) for the determination of practical serum samples. The biologic compounds probably existed in the sample, such as ascorbic acid, uric acid, dopamine, and epinephrine, do not affect the determination of glucose. The proposed method is satisfactory to the determination of human serum samples compared with the routine hexokinase method. Both the unique electrical property and biocompatibility of chitosan enable the construction of a good bio‐sensing platform for achieved DET of GOD and developed the third‐generation glucose biosensors.  相似文献

A facile strategy is presented to form 3D porous Cu@Cu2O aerogel networks by self‐assembling Cu@Cu2O nanoparticles with the diameters of ca. 40 nm for constructing catalytic interfaces. Unexpectedly, the prepared Cu@Cu2O aerogel networks display excellent electrocatalytic activity to glucose oxidation at a low onset potential of ca. 0.25 V. Moreover, the Cu@Cu2O aerogels also can act as mimicking‐enzymes including horseradish peroxidase and NADH peroxidase, and show obvious enzymatic catalytic activities to the oxidation of dopamine (DA), o‐phenyldiamine (OPD), 3,3,5,5‐tetramethylbenzidine (TMB), and dihydronicotinamide adenine dinucleotide (NADH) in the presence of H2O2. These 3D Cu@Cu2O aerogel networks are a new class of porous catalytic materials as mimic peroxidases and electrocatalysts and offer a novel platform to construct catalytic interfaces for promising applications in electrochemical sensors and artificial enzymatic catalytic systems.  相似文献

ZnO nanowire was tailored both physically and chemically to immobilize the enzyme glucose oxidase (GOD) for construction of a glucose sensor with high performance, which was ascribed to its high specific surface area and high isoelectric point value for efficient immobilization of high concentration of acidic enzymes and the mediating effect by the redox reaction of ZnO nanowires. The apparent Michaelis constants Jmax, and KM were adjusted in a large scope by tailoring the thickness of the GOD/ZnO nanowire layer and the enzyme load in the nanowired network. Thus, a variety of linear region, sensitivities and reaction rates of the sensor could be easily achieved. Moreover, the glucose sensor showed long term stability with the incorporation of the inorganic zinc oxide nanowire.  相似文献

Chaos and the natural evolution of tumor systems can lead to the failure of tumor therapies. Herein, we demonstrate that iridium oxide nanoparticles (IrOx) possess acid‐activated oxidase and peroxidase‐like functions and wide pH‐dependent catalase‐like properties. The integration of glucose oxidase (GOD) unlocked the oxidase and peroxidase activities of IrOx by the production of gluconic acid from glucose by GOD catalysis in cancer cells, and the produced H2O2 was converted into O2 to compensate its consumption in GOD catalysis owing to the catalase‐like function of the nanozyme, thus resulting in the continual consumption of glucose and the self‐supply of substrates to generate superoxide anion and hydroxyl radical. Moreover, IrOx can constantly consume glutathione (GSH) by self‐cyclic valence alternation of IrIV and IrIII. These cascade reactions lead to a “butterfly effect” of initial starvation therapy and the subsequent pressure of multiple reactive oxygen species (ROS) to completely break the self‐adaption of cancer cells.  相似文献

A gene of α‐L ‐arabinofuranosidase (Abf) from Trichoderma koningii G‐39 was successfully expressed in Pichia pastoris. The recombinant enzyme was purified to > 90% homogeneity by a cation‐exchanged chromatography. The purified enzyme exhibits both α‐L ‐arabinofuranosidase and β‐D ‐xylosidase (Xyl) activities with p‐nitrophenyl‐α‐L ‐arabionfuranoside (pNPAF) and 2,4‐dinitrophenyl‐β‐D ‐xylopyanoside (2,4‐DNPX) as substrate, respectively. The stability and the catalytic feature of the bifunctional enzyme were characterized. The enzyme was stable for at least 2 h at pH values between 2 and 8.3 at room temperature when assayed for Abf and Xyl activities. Enzyme activity decreased dramatically when the pH exceeded 9.5 or dropped below 1.5. The enzyme lost 35% of Abf activity after incubation at 55 °C for 2 h, but retained 95% of Xyl activity, with 2,4‐DNXP as substrate, under the same conditions. Further investigation of the active site topology of both enzymatic functions was performed with the inhibition study of enzyme activities. The results revealed that methyl‐α‐L ‐arabinofuranoside inhibition is noncompetitive towards 2,4‐DNPX as substrate but competitive towards pNPAF. Based on the thermal stability and the inhibition studies, we suggest that the enzymatic reactions of Abf and Xyl are performed at distinct catalytic sites. The recombinant enzyme possesses both the retaining transarabinofuranosyl and transxylopyranosyl activities, indicating both enzymatic reactions proceed through a two‐step, double displacement mechanism.  相似文献

A new anhydro disaccharide monomer, 1,6‐anhydro‐2,3‐di‐o‐benzyl‐4‐o‐(2′,3′,4′,6′‐tetra‐o‐benzyl‐β‐D ‐galactopyranosyl)‐β‐D ‐glucopyranose (benzylated 1,6‐anhydro lactose (LSHBE)), was synthesized from D ‐lactose to investigate the polymerizability and biological activities of the resulting branched polysaccharides. The ring‐opening polymerization of LSHBE was carried out with phosphorus pentafluoride as a catalyst under high vacuum to give a stereoregular benzylated (1 → 6)‐α‐D ‐lactopyranan. The molecular weights of poly(LSHBE)s increased with an increase in the amount of CH2Cl2 solvent, and polymerization temperatures were affected in both molecular weights and yields of the polymers. The copolymerization of LSHBE with benzylated 1,6‐anhydro‐β‐D ‐glucopyranose (LGTBE) gave the corresponding copolysacchrides having different proportions of lactose and glucose units in good yields. After debenzylation to recover hydroxyl groups and then sulfation, sulfated homopoly(lactose)s and copoly(lactose and glucose)s were obtained. Sulfated homopoly(lactose)s had moderate anti‐HIV (EC50 = 5.9 and 1.3 μg/mL) and blood anticoagulant activities (AA = 18 and 13 unit/mg), respectively. Sulfated copoly(lactose and glucose) having 15 mol % lactose units gave high anti‐HIV and blood anticoagulant activities of 0.3 μg/mL and 54 unit/mg, respectively. These biological results suggest that the distance between branched units on the main chain plays an important role in the anti‐HIV and blood anticoagulant activities. © 2008 Wiley Periodicals, Inc. This study demonstrated a novel nanographene platelets (NGPs)-based glucose/O2 biofuel cell (BFC) with the glucose oxidase (GOD) as the anodic biocatalysts and the laccase as the cathodic biocatalysts. The GOD/NGPs-modified electrode exhibited good catalytic activity towards glucose oxidation and the laccase/NGPs-modified electrode exhibited good catalytic activity towards O2 electroreduction. The maximum power density was ca. 57.8 μW cm? 2 for the assembled glucose/O2 NGPs-based BFC. These results indicated that the NGPs were very useful for the future development of novel carbon-based nanomaterials BFC device.  相似文献

本文选用生物相容性好的壳聚糖作为基体材料,使其与戊二醛交联成网状结构包埋葡萄糖氧化酶制成电化学传感器。这种壳聚糖膜不仅可以减小葡萄糖氧化酶的流失,而且能为酶提供了适宜的微环境。用红外光谱、紫外光谱及透射电镜对膜的形态和性质进行了表征。实验结果表明该传感器具有很快的响应速度,很好的稳定性和重现性,能选择性地催化葡萄糖并测定其浓度。该传感器的制备方法简单,成本低,于冰箱中放置两周信号保持在90%以上,对葡萄糖测量的线性范围为1×10-5 - 3.4×10-3mol•L-1,当信噪比为3:1时检测限为5×10-6mol•L-1。  相似文献

The glycosylation of cell‐penetrating poly(disulfide)s (CPDs) is introduced to increase the solubility of classical CPDs and to achieve multifunctional cellular uptake. With the recently developed sidechain engineering, CPDs decorated with α‐d ‐glucose (Glu), β‐d ‐galactose (Gal), d ‐trehalose (Tre), and triethyleneglycol (TEG) were readily accessible. Confocal laser scanning microscopy images of HeLa Kyoto cells incubated with the new CPDs at 2.5 μm revealed efficient uptake into cytosol and nucleoli of all glycosylated CPDs, whereas the original CPDs and TEGylated CPDs showed much precipitation into fluorescent aggregates at these high concentrations. Flow cytometry analysis identified Glu‐CPDs as most active, closely followed by Gal‐CPDs and Tre‐CPDs, and all clearly more active than non‐glycosylated CPDs. In the MTT assay, all glyco‐CPDs were non‐toxic at concentrations as high as 2.5 μm . Consistent with thiol‐mediated uptake, glycosylated CPDs remained dependent on thiols on the cell surface for dynamic covalent exchange, their removal with Ellman's reagent DTNB efficiently inhibited uptake. Multifunctionality was demonstrated by inhibition of Glu‐CPDs with d ‐glucose (IC50 ca. 20 mm ). Insensitivity toward l ‐glucose and d ‐galactose and insensitivity of conventional CPDs toward d ‐glucose supported that glucose‐mediated uptake of the multifunctional Glu‐CPDs involves selective recognition by glucose receptors at the cell surface. Weaker but significant sensitivity of Gal‐CPDs toward d ‐galactose but not d ‐glucose was noted (IC50 ca. 110 mm ). Biotinylation of Glu‐CPDs resulted in the efficient delivery of streptavidin together with a fluorescent model substrate. Protein delivery with Glu‐CPDs was more efficient than with conventional CPDs and remained sensitive to DTNB and d ‐glucose, i.e., multifunctional.  相似文献

The amperometric biosensing of aromatic amines using a composite glucose oxidase (GOD)‐peroxidase (HRP) biosensor in reversed micelles is reported. Rigid composite pellets of graphite and Teflon, in which GOD and HRP were coimmobilized by simple physical inclusion, were employed for the biosensor design. This design allows the in situ generation of the H2O2 needed for the enzyme reaction with the aromatic amines, thus preventing the negative effect that the presence of a high H2O2 concentration in solution has on HRP activity. The H2O2 in situ generation is performed by oxidation of glucose catalyzed by GOD. The effect of the composition of the reversed micelles, i.e., the nature of the organic solvent used as the continuous phase, the nature and concentration of the surfactant used as emulsifying agent, the aqueous 0.05 mol L?1 phosphate buffer percentage used as the dispersed phase, and the glucose concentration in the aqueous phase, on the biosensor response was evaluated. Reversed micelles formed with ethyl acetate, a 5% of phosphate buffer (pH 7.0) containing 3.0×10?3 mol L?1 glucose, and 0.1 mol L?1 AOT (sodium dioctylsulfosuccinate), were selected as working medium. Well‐defined and reproducible amperometric signals at 0.00 V were obtained for p‐phenylenediamine, 2‐aminophenol, o‐phenylenediamine, m‐phenylenediamine, 1‐naphthylamine, o‐toluidine and aniline. The useful lifetime of one single biosensor was of 60 days. The trend in sensitivity observed for the aromatic amines is discussed considering the effect of their structure on the stabilization of the radicals formed in the enzyme reaction which are electrochemically reduced. The behavior of the composite bienzyme electrode was also evaluated in a FI (flow injection) system using reversed micelles as the carrier. The suitability of the composite bienzyme electrode for the analysis of real samples was demonstrated by determining aniline in spiked carrots.  相似文献

本文以中性红为核,二氧化硅为壳,利用反相微乳液技术,通过正硅酸四乙酯的水解制备了掺杂有中性红的二氧化硅纳米粒子,并用TEM技术进行了表征。核中性红能够催化测定葡萄糖,乳酸和L-谷氨酸的反应,而壳二氧化硅不仅克服了电活性物质中性红易流失的缺点,且具有高的生物亲和性。分别与葡萄糖氧化酶、乳酸氧化酶以及L-谷氨酸氧化酶混合后,修饰在碳阵列电极表面。最后在该酶阵列电极表面滴加一层Nafion, 防止电活性物质抗坏血酸、尿酸等的干扰。该酶阵列传感器与流动注射分析技术(FIA)相结合,可应用于同时检测大鼠血样中的葡萄糖,乳酸和L-谷氨酸浓度。该方法无需通过传统的色谱柱的分离,大大简化了实验条件,为这一领域的研究提供了有效的分析方法。  相似文献

The highly porous Mn2O3‐Ag nanofibers were fabricated by a facile two‐step procedure (electrospinning and calcination). The structure and composition of the Mn2O3‐Ag nanofibers were characterized by SEM, TEM, XRD, EDX and SAED. The as‐prepared Mn2O3‐Ag nanofibers were then employed as the immobilization matrix for glucose oxidase (GOD) to construct an amperometric glucose biosensor. The biosensor shows fast response to glucose, high sensitivity (40.60 µA mM?1 cm?2), low detection limit (1.73 µM at S/N=3), low Km,app value and excellent selectivity. These results indicate that the novel Mn2O3‐Ag nanfibers‐GOD composite has great potential application in oxygen‐reduction based glucose biosensing.  相似文献

A simple and effective glucose biosensor based on immobilization of glucose oxidase (GOD) in graphene (GR)/Nafion film was constructed. The results indicated that the immobilized GOD can maintain its native structure and bioactivity, and the GR/Nafion film provides a favorable microenvironment for GOD immobilization and promotes the direct electron transfer between the electrode substrate and the redox center of GOD. The electrode reaction of the immobilized GOD shows a reversible and surface‐controlled process with the large electron transfer rate constant (ks) of 3.42±0.08 s?1. Based on the oxygen consumption during the oxidation process of glucose catalyzed by the immobilized GOD, the as‐prepared GOD/GR/Nafion/GCE electrode exhibits a linear range from 0.5 to 14 mmol·L?1 with a detection limit of 0.03 mmol·L?1. Moreover, it displays a good reproducibility and long‐term stability.  相似文献
