Ramulus Cinnamomi (RC)–Radix Glycyrrhizae (RG) is a classic herb pair, which is commonly used as a fixed form to treat cardiovascular disease in the clinic. Our work aimed to compare the pharmacokinetic difference of cinnamic acid, liquiritin, isoliquiritigenin and glycyrrhetinic acid in rats after oral administration of the RC–RG herb pair extracts [Guizhigancao Decoction (GGD) and Lingguizhugan Decoction (LGZGD)] and the single RC or RG extract. A HPLC‐MS method was developed and validated to study comparative pharmacokinetics. The pharmacokinetic parameters (Cmax, AUC, MRT) of four compounds between the RC–RG herb pair group and the single herb (RC or RG) group showed significant differences (p < 0.05). Compared with the single herb (RC or RG) group, higher peak concentration, slower elimination and larger exposure could be observed after giving the RC–RG herb‐pair extracts. The pharmacokinetic differences might indicate the relativity of remedy in the RC–RG herb pair and provide scientific information for rational administration of the drug in the clinic. Cumulative evidence shows that eicosanoids such as prostaglandins, leukotrienes, thromboxanes and hydroxy eicosatetraenoic acids play an important role in associating inflammation with human colorectal cancer (CRC). In this study an ultra‐pressure liquid chromatography/tandem mass spectrometry (UPLC/MS/MS) method was developed and validated for the targeted profiling of eight relevant eicosanoids and the major metabolic precursor, arachidonic acid (AA), in human colon. Multiple reaction monitoring (MRM) experiments were performed in negative electrospray ionization mode. The metabolites were separated using a C18 column consisting of 1.7 µm ethylene‐bridged hybrid particles (100 × 2.1 mm i.d.) and gradient elution (50 to 95% of solvent B) with a mobile phase comprising water (0.1% formic acid) [solvent A] and acetonitrile (0.1% formic acid) [solvent B] at a flow rate of 0.4 mL/min. The analysis time for each sample was 5.5 min. Our UPLC/MS/MS method demonstrated satisfactory validation results in terms of selectivity, sensitivity, matrix effect, linearity, extraction efficiency, intra‐ and inter‐day precision, accuracy and autosampler stability. The method was applied for the clinical profiling of matched pairs of cancerous and normal colon mucosae obtained from eight colorectal cancer patients. Endogenous levels of AA and selected eicosanoids such as prostaglandin E2 (PGE2), prostacyclin (PGI2) [assayed as its stable hydrolytic product 6‐keto‐prostaglandin (6‐k PGF)] and 12‐hydroxy‐5Z,8Z,10E,14Z‐eicosatetraenoic acid (12‐HETE) were found to be significantly different (p <0.05; paired t‐test) between cancerous and normal mucosae. Eleven TLC solvent systems were evaluated for the separation of prostaglandins F, E2 and D2, thromboxane B2, 12-OH-5,8,10-heptadecatrienoic acid (HHT), 12-OH-5,8,10,14-eicosatetraenoic acid (HETE), and arachidonic acid (AA) from each other. A three-solvent development TLC procedure, which can conveniently be used for routine resolution of mixtures of these metabolites on a single TLC plate, is reported. Zengmian Yiliu (ZMYL), a traditional Chinese formula, is designed to improve clinical efficacy and reduce adverse effects in combination with cisplatin in ovarian cancer chemotherapy. In ZMYL, Radix Paeoniae Alba (RPA, made from root of Paeonia lactiflora Pall.) acts as an adjunctive drug in cancer treatment by ameliorating side effects induced by radio‐ and chemotherapy. The pharmacokinetics differences between isomer albiflorin and paeoniflorin, the main components of RPA, after oral administration decoction of single‐herb RPA and ZMYL were compared using a sensitive and accurate UPLC‐MS/MS. The results indicate that there are statistically significant differences between the pharmacokinetic parameters: decreasing area under the plasma concentration–time curve (AUC), maximum concentration (Cmax), elimination rate constant (Ke) and increasing apparent volume of distribution (Vd) and clearance (CL) for albiflorin, increasing distribution half‐life (T1/2d) and decreasing elimination half‐life (T1/2e), distribution rate constant (Kd) and absorption rate constant (Ka) for paeoniflorin in the ZMYL group compared with the single‐herb RPA group. In comparison with albiflorin, the pharmacokinetic parameters of paeoniflorin included significantly increasing mean residence time (MRT) and Vd, decreasing CL and Ke in the single‐herb RPA group and increasing MRT and T1/2d and decreasing CL, Ke and Kd in the ZMYL group. Both paeoniflorin and albiflorin are more likely, as the main active ingredients in RPA and ZMYL, to play a variety of pharmacological effects, and herb–herb interactions occur, resulting in different pharmacokinetics of albiflorin and paeoniflorin in RPA and ZMYL. The structures of [Cu(AA)6](ClO4)2, (I), and [Mn(AA)6](ClO4)2, (II) (AA is acrylamide, also known as prop‐2‐enamide; C3H5NO), display both intra‐ and intermolecular N—H...O hydrogen bonding. A three‐dimensional network is propagated via the perchlorate counter‐ions. There are two crystallographically independent molecules in the copper complex, with the most significant difference between them being the conformation of one symmetry‐related pair of AA ligands which are in the unusual syn conformation. The copper complex exhibits syn/anti disorder of the =CH2 group in one pair of symmetry‐related AA ligands. The CuII and MnII centres are both situated on centres of inversion. The copper complex cation has octahedral coordination geometry with typical Jahn–Teller distortions.  相似文献

In traditional Chinese medicine theory, Panax ginseng and Veratrum nigrum L. is an important incompatible herb pair. Studies on the content variation of main components and the influences on the metabolism in rat intestinal bacteria are useful to understand the mechanism of incompatibility of this herb pairs. In this study, the content variation of ginsenosides and their metaboltic profiles in the extracts of P. ginseng and compatibility of P. ginseng with V. nigrum L. (G‐V) were investigated using relative quantitative method of electrospray ionization mass spectrometry (ESI‐MS) and UPLC‐MSn, respectively. The relative contents of most ginsenosides were reduced in the extract of G‐V. Furthermore, ginsenosides Rb1, Rb2, Rc and Rd could be metabolized to Rd, F2 and C‐K in rat intestinal bacteria. The metabolic speeds of Rb1, Rb2 and Rc in the G‐V extracts at ratios of 10:5, 10:7 and 10:10 and the metabolic rates of ginsenosides Rb1, Rb2 and Rc to Rd, Rd to F2 in all compatibility extracts were lower than that in the P. ginseng extract. In conclusion, this study illustrated the mechanism of effect‐reducing by comparison of the relative contents and metabolic profiles of ginsenosides after compatibility of P. ginseng and V. nigrum L.  相似文献

A recent study showed that sarcosine may be potentially useful for the diagnosis and prognosis of prostate cancer (PCa). The aim of this study was to validate diagnostic value of sarcosine for PCa, to evaluate urine metabolomic profiles in patients with PCa in comparison of non-cancerous control, and to further explore the other potential metabolic biomarkers for PCa. Isotope dilution gas chromatography/mass spectrometry (ID GC/MS) metabolomic approach was applied to evaluate sarcosine using [methyl-D3]-sarcosine as an internal standard. Microwave-assisted derivatization (MAD) together with GC/MS was utilized to obtain the urinary metabolomic information in 20 PCa patients compared with eight patients with benign prostate hypertrophy and 20 healthy men. Acquired metabolomic data were analyzed using a two-sample t test. Diagnostic models for PCa were constructed using principal component analysis and were assessed with receiver–operating characteristic curves. Results showed that the urinary sarcosine level has no statistical difference between the PCa group and the control group. In addition, nine metabolomic markers between the PCa group and the healthy male group were selected, which constructed a diagnostic model with a high area under the curve value of 0.9425. We conclude that although urinary sarcosine value has limited potential in the diagnostic algorithm of PCa, urinary metabolomic panel based on GC/MS assay following MAD may potentially become a diagnostic tool for PCa.  相似文献

Copolymerization of acrylic acid and p‐chloromethylstyrene (p‐CMS) in dioxane initiated with α,α′‐azobisisobutyronitrile was carried out to produce macroinitiator P(AA‐co‐CMS) containing PhCH2Cl group at 65°C. Then methyl methacrylate was grafted onto P(AA‐co‐CMS) backbone using PhCH2Cl group as an initiation site and FeCl2/triphenyl phosphine complex as a catalyst. The resulted copolymer (AA‐co‐CMS)‐g‐PMMA with a comb‐like branched structure has a hydrophilic backbone (PAA) and hydrophobic side chains (PMMA). Compositions and structures of macroinitiator and the grafted product of P(AA‐co‐CMS)‐g‐PMMA were determined by 1H‐NMR, infrared (IR), and gel permeation chromatography (GPC). The average graft number, the average length of branch chains, the graft ratio, and the graft efficiency were investigated. The swelling behavior of the comb‐like branched polymer was also investigated. The gradual increase of swelling ratios was accompanied by an increase of pH and temperature. The kinetic exponents indicated that the swelling transport mechanisms transformed from Fickian diffusion to non‐Fickian transport as the decreasing pH. Alkaloids from Cortex Phellodendron amurense Rupr. were identified to determine the material basis for the bioactivity of this herb. HPLC–ESI‐MS with photodiode array detection coupled to XCharge C18 column was applied to analyze the alkaloids qualitatively and quantitatively. A total of 37 alkaloids were identified and tentatively characterized from the ethanol extract by online ESI‐MSn fragmentation and UV spectral analysis. A total of ten alkaloids, including four novel natural products, were tentatively identified for the first time in P. amurense. The fragmentation pathways for certain compounds were analyzed. The contents of a pair of isomers (columbamine and jatrorrhizine) and four main alkaloids (phellodendrine, magnoflorine, berberine, and palmatine) were simultaneously quantified using the aforementioned method. Results showed that the newly discovered and known components of P. amurense were helpful in determining the material basis for the bioactivity of the herb. The application of the XCharge C18 column is a suitable and practical method for the isolation of alkaloids in plants.  相似文献

Centella asiatica has been used as a culinary vegetable or medicinal herb. In this study, the hepatoprotective effect of the standardized extract of C. asiatica (ECa233) in rotenone‐treated rats was examined using a GC–MS‐based metabolomic approach. ECa233 contains >80% triterpenoids with a ratio of madecassoside to asiaticoside of 1.5(±0.5):1. Rats were randomly divided into three groups (with six rats/group): sham negative control, rotenone positive control and the ECa233 test group. Rats in the ECa233 group received 10 mg/kg ECa233 orally for 20 days, followed by 2.5 mg/kg intraperitoneal rotenone injection to induce toxicity before being sacrificed. Metabolomic analysis showed that supplementation of ECa233 protected rat liver against rotenone toxicity. Pipecolinic acid was one of the most important metabolites; its level was decreased in the rotenone group as compared with the control. Supplementation with ECa233 before administration of rotenone raised pipecolinic acid to levels intermediate between controls and rotenone alone. The metabolomics approach also helped discover a possible new genuine epimetabolite in the present work. Antioxidant tests revealed that ECa233 inhibited lipid peroxidation and increased catalase activities in liver tissue.  相似文献

Substance P (SP) belongs to the tachykinin family and plays an essential role in pain transmission and in neurogenic inflammation. It can be detected in the central and peripheral nervous systems. The objectives of this study were to establish SP metabolic stability in liver microsomes in three species (rat, mouse and human), and identify and characterize SP metabolites by LC‐MS/MS. Endogenous peptide metabolism is not well documented and this is particularly true for neuropeptides participating in neurogenic inflammation. In vitro, T1/2 results in pooled liver microsomes were 9.2, 5.6 and 18.6 min for rat, mouse and human liver microsomes, respectively. Five major SP metabolites were identified and quantified, including C‐terminal SP fragments SP3–11, SP5–11, SP6–11, SP8–11 as well as N‐terminal fragment SP1–7. The results suggest significant differences between species in SP metabolite concentrations. Consequently, the metabolic profile of each species is distinctive and may have a significant impact on biomolecular mechanisms involved in specific pathophysiological changes. In the present study, the metabolism of ritonavir was explored in the presence of rCYP3A4 using a well‐established strategy involving liquid chromatography–mass spectrometry (LC–MS) tools. A total of six metabolites were formed, of which two were new, not reported earlier as CYP3A4‐mediated metabolites. During LC–MS studies, ritonavir was found to fragment through six principal pathways, many of which involved neutral loss of CO2, as indicated through 44‐Da difference between masses of the precursors and the product ions. This was unusual as the drug and the precursors were devoid of a terminal carboxylic acid group. Apart from the neutral loss of CO2, marked differences were also observed among the fragmentation pathways of the drug and its metabolites having intact N‐methyl moiety as compared to those lacking N‐methyl moiety. These unusual fragmentation behaviours were successfully explained through energy distribution profiles by application of the density functional theory. A sensitive and high‐throughput inhibition screening liquid chromatography–mass spectrometry (LC‐MS/MS) method was developed and validated for the simultaneous quantification of five probe metabolites (7‐hydroxycoumarin, CYP2A6; 4‐hydroxytolbutamide, CYP2C9; 4′‐hydroxymephenytoin, CYP2C19; α‐hydroxymetoprolol, CYP2D6; and 1‐hydroxymidazolam, CYP3A4) for in vitro cytochrome P450 activity determination in human liver microsome and recombinant. All the metabolites and the internal standard, tramadol, were separated on a Waters 2695 series liquid chromatograph with a Phenomenex Luna C18 column (150 × 2.0 mm, 5 µm). Quality control samples and a positive control CYP inhibitor were included in the method. The IC50 values determined for typical CYP inhibitors were reproducible and in agreement with the literature. The method was selective and showed good accuracy (99.13–103.37%), and inter‐day (RSD < 6.20%) and intra‐day (RSD < 6.13%) precision. Also, the incubation extracts of the sample were stable at room temperature (20 °C) for 48 h and for 96 h in the autosampler (4 °C). The presented method is the first HPLC‐MS/MS method of this combination for simultaneous detection of the five metabolites 7‐hydroxycoumarin, 4‐hydroxytolbutamide, 4′‐hydroxymephenytoin, α‐hydroxymetoprolol and 1‐hydroxymidazolam in a single‐run process. The herb couple has special clinical significance in reducing the toxicity and increasing the efficacy of drugs. The combination of Radix Angelicae Dahuricae (Baizhi, BZ) and Rhizoma Chuanxiong (ChuanXiong, CX) is a traditional herb couple. The combination performs better than the CX extract alone in the treatment of migraine and has been used for thousands of years. However, the specific compatibility mechanisms are still unclear. Ligustilide, dl ‐3‐n‐butylphthalide and senkyunolide A are the major active ingredients in CX and BZ–CX decoction. However, a comprehensive study of the pharmacokinetics of CX has not been carried out. A gas chromatography–mass spectroscopy (GC–MS) method with high selectivity, sensitivity and accuracy was developed. An SH‐Rxi‐5Sil (30 m × 0.25 mm i.d., and 0.25 μm film thickness) column was employed in the GC separation. Selectivity, linearity, precision, accuracy, recovery, matrix effect and stability were used to validate the current GC–MS method. Using the validated method, this is the first time to study on the comparative pharmacokinetics of ligustilide, dl ‐3‐n‐butylphthalide and senkyunolide A from CX alone and BZ–CX decoction in rat plasma. The pharmacokinetic parameters (Cmax, Tmax, T1/2, AUC0–t, AUC0–∞ and CLz/F) of all of the detected ingredients showed significant differences between the two groups (P < 0.05). The results are helpful for further investigation of the compatibility mechanism of BZ–CX decoction.  相似文献

Methylation is one of the important posttranslational modifications of biological systems. At the metabolite level, the methylation process is expected to convert bioactive compounds such as amino acids, fatty acids, lipids, sugars, and other organic acids into their methylated forms. A few of the methylated amino acids are identified and have been proved as potential biomarkers for several metabolic disorders by using mass spectrometry–based metabolomics workstation. As it is possible to encounter all the N‐methyl forms of the proteinogenic amino acids in plant/biological systems, it is essential to have analytical data of all N‐methyl amino acids for their detection and identification. In earlier studies, we have reported the ESI‐MS/MS data of all methylated proteinogenic amino acids, except that of mono‐N‐methyl amino acids. In this study, the N‐methyl amino acids of all the amino acids ( 1 ‐ 21 ; including one isomeric pair) were synthesized and characterized by ESI‐MS/MS, LC/MS/MS, and HRMS. These data could be useful for detection and identification of N‐methyl amino acids in biological systems for future metabolomics studies. The MS/MS spectra of [M + H]+ ions of most N‐methyl amino acids showed respective immonium ions by the loss of (H2O, CO). The other most common product ions detected were [MH‐(NH2CH3]+, [MH‐(RH)]+ (where R = side chain group) ions, and the selective structure indicative product ions due to side chain and N‐methyl group. The isomeric/isobaric N‐methyl amino acids could easily be differentiated by their distinct MS/MS spectra. Further, the MS/MS of immonium ions inferred side chain structure and methyl group on α‐nitrogen of the N‐methyl amino acids.  相似文献

Hedyotis diffusa, a traditional Chinese herbal medicine, is widely used for oncotherapy and shows a positive effect in the clinical treatment. But its mechanism of anticancer activities is complicated and unclear. This study was undertaken to assess the therapeutic effects and reveal detailed mechanisms of Hdiffusa for oncotherapy. A Walker 256 tumor‐bearing rat model was established, and metabolomic profiles of plasma and urine were obtained from 1H NMR technique. Multivariate statistical analysis methods were used to characterize the discriminating metabolites between control (C), Walker 256 tumor‐bearing rats model (M), and Hdiffusa treatment (H) groups. Finally, 13 and 10 metabolomic biomarkers in urine and plasma samples were further identified as characteristic metabolites in M group, whereas H group showed a partial metabolic balance recovered, such as ornithine, N‐acetyl‐l ‐aspartate, l ‐aspartate, and creatinine in urine samples, and acetate, lactate, choline, l ‐glutamine, and 3‐hydroxybutyrate in plasma samples. On the basis of the methods above, we hypothesized Hdiffusa treatment reduced the injury caused by Walker 256 tumor and maintained a metabolic balance. Our study demonstrated that this method provided new insights into metabolic alterations in tumor‐bearing biosystems and researching on the effects of Hdiffusa on the endogenous metabolism in tumor‐bearing rats.  相似文献

The goal of this study is to investigate the biotransformation of ginsenoside Rg1 in vivo. A highly sensitive and specific LC‐MS/MS method was developed and used for metabolite identification in rat feces and urine after oral administration of ginsenoside Rg1. Four metabolites of Rg1 were detected in rat feces and three metabolites of Rg1 were detected in rat urine. Deglycosylation and oxygenation were found to be the major metabolic pathways of ginsenoside Rg1 after oral administration in rat. Except for the reported metabolites Rh1 and protopanaxatriol, mono‐oxygenated Rg1 and mono‐oxygenated protopanaxatriol were detected for the first time after oral administration of Rg1. The in vivo metabolite profiling of ginsenoside Rg1 in rat was proposed. Viewed collectively, Rg1 was metabolized to mono‐oxygenated Rg1, Rh1, protopanaxatriol and the secondary metabolite mono‐oxygenated protopanaxatriol in rat. In this study, tamoxifen metabolic profiles were investigated carefully. Tamoxifen was administered to two healthy male volunteers and one female patient suffering from breast cancer. Urinary extracts were analyzed by liquid chromatography quadruple time‐of‐flight mass spectrometry using full scan and targeted MS/MS techniques with accurate mass measurement. Chromatographic peaks for potential metabolites were selected by using the theoretical [M + H]+ as precursor ion in full‐scan experiment and m/z 72, 58 or 44 as characteristic product ions for N,N‐dimethyl, N‐desmethyl and N,N‐didesmethyl metabolites in targeted MS/MS experiment, respectively. Tamoxifen and 37 metabolites were detected in extraction study samples. Chemical structures of seven unreported metabolites were elucidated particularly on the basis of fragmentation patterns observed for these metabolites. Several metabolic pathways containing mono‐ and di‐hydroxylation, methoxylation, N‐desmethylation, N,N‐didesmethylation, oxidation and combinations were suggested. Sini decoction (SND) is an important traditional Chinese multiherbal formula, which is widely used to treat cardiovascular disease. Acontium carmichaeli (AC) is a leading herb in SND, whose main components are monoester‐diterpenoid alkaloids (MDAs). The aim of this study is to compare the pharmacokinetics of three MDAs in rat plasma after oral administration of AC extract and its compatibility with other herbal medicines in SND. A sensitive, accurate and specific LC‐MS/MS method was developed to determine the contents of three MDAs in rat plasma. Male Sprague–Dawley rats were randomly assigned to four groups: AC, AC + ZO, AC + GU and SND groups. There were significant differences in the pharmacokinetic parameters (Cmax, Tmax, t1/2, AUC(0–24), MRT and CL). Compared with the AC group, Cmax, AUC(0–24) and CL of three MDAs increased and t1/2 decreased in AC + ZO, AC + GU and SND groups. Little changed in the AC + GU group in comparison with AC + ZO group, which indicated that other ingredients in ZO may promote the absorption rate and accelerate excretion rate of MDAs. The results could be helpful for revealing the compatibility mechanism of Chinese multiherbal medicine and providing clinical medication guidance on AC and its compatibility with other herbal medicines in SND.
