The one‐pot four‐component reaction of benzohydrazide, acetylenedicarboxylate, aromatic aldehydes and malononitrile in ethanol with triethylamine as base catalyst afforded functionalized 1‐benzamido‐1,4‐dihydropyridines in satisfactory yields. Under similar conditions, picolinohydrazide or nicotinohydrazide can also be successfully utilized in the reactions to give corresponding functionalized 1,4‐dihydropyridines. 1H NMR data indicated that an equilibrium of cis/trans‐conformations exist in 1‐benzamido‐1,4‐dihydropyridines.  相似文献

A four‐component reaction for the synthesis of 1,2‐dihydroisoquinoline derivatives is described. The Huisgen 1,4‐dipolar intermediate, which is produced from isoquinoline and an electron‐deficient acetylene compound 1 , reacts with H2O in the presence of diketene to produce 1,2‐dihydroisoquinoline derivatives 2 (Scheme 1). In addition, reaction of isoquinoline, dibenzoylacetylene (=1,4‐diphenylbut‐2‐yne‐1,4‐dione), and diketene in the presence of H2O leads to pyrroloisoquinoline derivative 7 . The structures of the compounds 2a – f and 7 were corroborated spectroscopically (IR, 1H‐ and 13C‐NMR, EI‐MS) and by elemental analyses. A plausible mechanism for the reaction is proposed (Schemes 2 and 3).  相似文献

Synthesis of substituted 2‐isoxazolines derivatives with sulfone functional groups was accomplished by 1,3‐dipolar cycloaddition reaction. The 1,3‐dipolar nitrile oxides generated in situ by reacting α‐formaldoximes with N‐bromosuccinimide in the presence of triethylamine on reaction with activated alkenes in toluene at room temperature afford the corresponding 2‐isoxazolines in high yields. All the 2‐isoxazoline derivatives were assayed for their antibacterial activities against Staphylococcus aureus, Bacillus cereus, Escherichia coli, and Pseudomonas aeruginosa and antifungal activities against Aspergillus niger and Candida albicans. Most of the compounds showed good antibacterial and antifungal activities.  相似文献

The one‐pot multicomponent reaction of alkyl isocyanide, alkylidene‐substituted Meldrum's acid, and arylcarboxylic acids affords new derivatives of iminofuranone in fair yields. The structure of the products was deduced from their spectroscopic data. Two equivalents of the respective isocyanides participate in this reaction.  相似文献

A scalable, one‐pot, enantioselective catalytic synthesis of 2‐pyrazolines from beta‐substituted enones and hydrazines is described. Pivoting on a two‐stage catalytic Michael addition/condensation strategy, the use of an aldehyde to generate a suitable hydrazone derivative of the hydrazine was found to be key for curtailing background reactivity and tuning the catalyst‐controlled enantioselectivity. The new synthetic method is easy to perform, uses a new and readily prepared cinchona‐derived bifunctional catalyst, is broad in scope, and tolerates a range of functionalities with high enantioselectivity (up to >99:1 e.r.). The significant scalability of this methodology was demonstrated with the synthesis of more than 80 grams of a pyrazoline product with 89 % catalyst recovery.  相似文献

A simple, convenient, and eco‐friendly synthetic protocol has been developed via a one‐pot three‐component reaction between 2‐chloroquinoline‐4‐amines, different substituted aromatic aldehydes, and malononitrile using ethanol as reaction medium. Employing this protocol, a series of 5‐chloro‐4‐phenyl benzo[f ][1,6] naphthyridine‐2‐amino‐3‐carbonitrile derivatives were synthesized in an environmentally friendly approach under operational simplicity, short time reactions, easy work‐up procedure, and comparatively high yields. This chemistry provides a convenient and promising synthetic strategy for the construction of the napthyridine skeleton. All synthesized compounds were identified on the basis of their spectral data.  相似文献

A series of novel 1,3,5‐triarylpyrazoles 3a , 3b , 3c , 3d , 3e , 3f , 3g , 3h , 3i , 3j , 3k , 3l , 3m , 3n , 3o , 3p , 3q , 3r , 3s , 3t , 3u , 3v , 3w , 3x were synthesized from flavanones, arylhydrazines, and trimethyl phosphate in an one‐pot procedure. Facile reaction process, easy after‐reaction workshop, and good yields are the distinct characteristics of the developed protocol. The target compounds were characterized by element analysis, infrared ray (IR), 1H NMR spectra, and electrospray ionization‐mass spectrometry. The structure of representative compound 3h (C23H20N2O3, Mr = 372.42) was further confirmed by X‐ray diffraction. It crystallizes in monoclinic, space group P 21/c, a = 8.9720(5), b = 24.5523(13), c = 8.9687(6) Å, α = 90.0000, β = 102.6417(17), γ = 90.0000°, V = 1927.76(20) Å3, Z = 4, μ(MoKα) = 0.086, F(000) = 784, Dc = 1.283 g/cm3, the final R = 0.0349 and wR = 0.0844 for 1668 observed reflections (I > 2σ(I))  相似文献

A versatile synthesis of 3‐cyano‐2‐pyridones via a one‐pot, four‐component condensation of ethyl cyanoacetate, ketones, aldehydes, and ammonium acetate under very mild conditions has been developed. A true one‐step one‐pot aldol‐reaction procedure has been developed for the synthesis of β‐hydroxy ketones and esters. The reaction can be run at room temperature by simply mixing four components in CH2Cl2, with medium‐to‐high yields of aldol products obtained after regular workup. Mechanistically, the process probably proceeds via Mg‐enolate formation of the ketone or ester component, followed by addition to the electrophilic aldehyde.  相似文献

An efficient method for the one‐pot synthesis of 2‐oxazolines from ethyl α‐cyanocinnamate derivatives with N‐bromoacetamide in the presence of K3PO4 has been developed. The reaction performed smoothly and cleanly to give 2‐oxazolines in good to excellent yields (up to 98%) within 4.5 h in acetone at room temperature without protection of inert gases. A total of 13 examples have been investigated. A possible nucleophilic addition reaction mechanism is proposed.  相似文献

An efficient method for the synthesis of polysubstituted indolizines has been developed based on formal [4+2] annulation of 1‐acetylaryl 2‐formylpyrroles with enals, followed by oxidative aromatization. Pyridine‐type six‐membered rings were constructed in this transformation. This transition metal‐free reaction features mild reaction conditions, a broad substrate scope, and excellent functional group tolerance. Notably, the formyl group is well tolerated under reaction conditions.  相似文献

Multicomponent reactions are excellent tools for rapid generation of small molecules with broad chemical diversity and molecular complexity. Herein, a novel one‐pot multicomponent synthesis of β‐amino amides from aldehydes, anilines, carboxylic acids and ynamides has been successfully developed. This process is practical and efficient to unravel synthetic utility and scalability. Moreover, an isotope labeling reaction was conducted to elucidate a plausible reaction mechanism.  相似文献

A one‐pot synthesis of 3,4,5‐ and 1,3,5‐pyrazoles from tertiary propargylic alcohols and para‐tolylsulfonohydrazide has been accomplished. The pyrazoles are formed through a four‐step cascade sequence, including FeCl3‐catalyzed propargylic substitution, aza‐Meyer–Schuster rearrangement, base‐mediated 6π electrocyclization, and thermal [1,5] sigmatropic shift. In this reaction, the 3,4,5‐ and 1,3,5‐pyrazoles are produced selectively according to different substituents in the starting alcohols.  相似文献
