Enantioselective open tubular capillary electrochromatography with carboxymethyl‐β‐cyclodextrin conjugated gold nanoparticles as stationary phase was developed. This novel open tubular column was fabricated through layer‐by‐layer self‐assembly of gold nanoparticles on a 3‐mercaptopropyl‐trimethoxysilane‐modified fused‐silica capillary and subsequent surface functionalization of the gold nanoparticles through self‐assembly of 6‐mercapto‐β‐cyclodextrin. The 6‐mercapto‐β‐cyclodextrin was firstly synthesized and determined by extensive spectroscopic data. Scanning electron microscopy, energy dispersive X‐ray analysis spectroscopy, and electroosmotic flow experiments were carried out to characterize the prepared open tubular column. Then, the separation effectiveness of the open tubular column was verified by two pairs of ɑ‐tetralones derivatives enantiomers and two pairs of basic drug enantiomers (tramadol hydrochloride and zopiclone) as mode analytes. Factors that influence the enantioseparation were optimized, and under the optimized conditions, satisfactory separation results were obtained for the four enantiomers: compound A, compound B, tramadol hydrochloride, and zopiclone with resolutions of 3.79, 1.56, 1.03, 1.60, respectively. For the combination of gold nanoparticles and negatively charged carboxymethyl‐β‐cyclodextrin, the open tubular column exhibited wider separation range for neutral and basic drugs. Moreover, the repeatability and stability of the column were studied through the run‐to‐run and day‐to‐day investigations.  相似文献

Inspired by the distinct chemical and physical properties of nanoparticles, here a novel open‐tubular capillary electrochromatography column was prepared by electrostatic assembly of poly(diallydimethylammonium chloride) onto the inner surface of a fused‐silica capillary, followed by self‐adsorption of negatively charged SH‐β‐cyclodextrin/gold nanoparticles. The formation of the SH‐β‐cyclodextrin/gold nanoparticles coated capillary was confirmed and characterized by scanning electron microscopy and energy dispersive spectrometry. The results of scanning electron microscopy and energy dispersive spectrometry studies indicated that SH‐β‐cyclodextrin/gold nanoparticles were successfully coated on the inner wall of the capillary column. The performance of the SH‐β‐cyclodextrin/gold nanoparticles coated capillary was validated by the analysis of six pairs of chiral drugs, namely zopiclone, carvedilol, salbutamol, terbutaline sulfate, phenoxybenzamine hydrochloride, and ibuprofen. Satisfactory enantioseparation results were achieved, confirming the use of gold nanoparticles as the support could enhance the phase ratio of the open‐tubular capillary column. Additionally, the stability and reproducibility of the SH‐β‐cyclodextrin/gold nanoparticles coated capillary column were also investigated. Then, this proposed method was well validated with good linearity (≥0.999), recovery (90.0–93.5%) and repeatability, and was successfully used for enantioseparation of ibuprofen in spiked plasma samples, which indicated the new column's potential usage in biological analysis.  相似文献

A novel open‐tubular capillary electrochromatography column coated with β‐cyclodextrin was prepared using the sol‐gel technique. In the sol‐gel approach, owing to the three‐dimensional network of sol‐gel and the strong chemical bond between the stationary phase and the surface of capillary columns, good chromatographic characteristics and unique selectivity in separating enantiomers were shown. The influences of capillary inner diameter, coating time, organic modifier, buffer pH, and buffer concentration on separation were investigated. The sol‐gel‐coated β‐cyclodextrin column has shown improved enantioseparation efficiency of chlorphenamine, brompheniramine, pheniramine, zopiclone in comparison with the sol‐gel matrix capillary column. The migration time relative standard deviation of the separation of the enantiomers was less than 0.89% over five runs and 2.9% from column to column. This work confirmed that gold nanoparticles are promising electrochromatographic support to enhance the phase ratio of open‐tubular capillary electrochromatography column in capillary electrochromatography.  相似文献

Inspired by the chiral recognition ability of β‐cyclodextrin and the natural adhesive properties of polydopamine under alkaline conditions, in this study, a rapid and in situ modification strategy was developed to fabricate β‐cyclodextrin/polydopamine composite material coated‐capillary columns for open tubular capillary electrochromatography. The results of scanning electron microscopy, FTIR spectroscopy, streaming potential, and electro‐osmotic flow studies indicated that β‐cyclodextrin/polydopamine was successfully fixed on the inner wall of the capillary column. This coating can be achieved within 1 h affording a greatly reduced capillary preparation time. The performance of the β‐cyclodextrin/polydopamine‐coated capillary was validated by the analysis of seven pairs of chiral analytes, namely epinephrine, norepinephrine, isoprenaline, terbutaline, verapamil, tryptophane, carvedilol. Good enantioseparation efficiencies were achieved for all. For three consecutive runs, the relative standard deviations for the migration times of the analytes for intraday, interday, and column‐to‐column repeatability were in the range of 0.41–1.74, 1.03–4.18, and 1.66–8.24%, respectively. Moreover, the separation efficiency of the β‐cyclodextrin/polydopamine‐coated capillary column did not decrease obviously over 90 runs. The strategy should also be feasible to introduce and immobilize other chiral selectors on the inner walls surface of capillary columns.  相似文献

Derivatized β‐cyclodextrin (β‐CD) functionalized monolithic columns were prepared by a “one‐step” strategy using click chemistry. First, the intended derivatized β‐CD monomers were synthesized by a click reaction between propargyl methacrylate and mono‐6‐azido‐β‐CD and then sulfonation or methylation was carried out. Finally, monolithic columns were prepared through a one‐step in situ copolymerization of the derivatized β‐CD monomer and ethylene glycol dimethacrylate. The sulfated β‐CD‐based monolith was successfully applied to the hydrophilic interaction liquid chromatography separation of nucleosides and small peptides, while the methylated β‐CD‐functionalized monolith was useful for the separation of nonpolar compounds and drug enantiomers in capillary reversed‐phase liquid chromatography. The structures of the monomers were characterized by Fourier transform infrared spectroscopy and mass spectrometry. The physicochemical properties and column performance of monoliths were evaluated by scanning electron microscopy and micro high performance liquid chromatography. This strategy has considerable prospects for the preparation of other derivatized CD‐functionalized methacrylate monoliths.  相似文献

The separation and migration behavior of six isomeric dichlorophenols (DCPs) in cyclodextrin‐modified capillary zone electrophoresis (CD‐CZE) using a phosphate‐borate buffer at alkaline pH with β‐CD and hydroxypropyl‐β‐CD (HP‐β‐CD) as electrolyte modifiers were investigated. The influence of buffer pH and the concentration of β‐cyclodextrins were examined. The results indicate that baseline separation of six isomeric DCPs can be achieved with addition of β‐CD concentration in the range of 2.0‐10 mM or HP‐β‐CD concentration in the range of 4.0‐10 mM at pH 10.0. Binding constants of DCPs to β‐CDs were evaluated for a better understanding of the interaction of DCPs with β‐CDs.  相似文献

In this work, an open‐tubular capillary liquid‐phase column was prepared by modifying chain polymer on the inner surface of capillary and chemical bonding of metal organic frameworks, NH2‐UiO‐66, to the brushes of chain polymer (poly(glycidyl methacrylate)). Besides advantages of facial preparation and good permeability, the chain polymer effectively increases the modification amount of NH2‐UiO‐66 nanoparticles to increase the phase ratio of open‐tubular capillary column and enhance the interactions with analytes. The results of scanning electron microscope energy‐dispersive X‐ray spectra indicated that NH2‐UiO‐66 nanoparticles were successfully bonded to the chain polymer. Because of the hydrophobic interaction and hydrogen bonding interaction between the analytes and the ligand of NH2‐UiO‐66, different analytes were well separated on the NH2‐UiO‐66‐modified poly(glycidyl methacrylate) capillary (1.12 m × 25 μm id × 365 μm od) with the high absolute column efficiency reaching 121 477 plates, benefiting from an open‐tubular column and low mass transfer resistance provided by polymer brush and metal–organic framework crystal. The relative standard deviations of the retention time for run‐to‐run, day‐to‐day, and column‐to‐column (= 3) runs are below 4.28%, exhibiting good repeatability. Finally, the column was successfully applied to separation of flavonoids in licorice.  相似文献

To further evaluate the feasibility and applicability of the one‐pot strategy in monolithic column preparation, two novel β‐cyclodextrin‐functionalized organic polymeric monoliths were prepared using two β‐cyclodextrin derivatives, i.e. mono(6‐amino‐6‐deoxy)‐β‐cyclodextrin and heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin. In this improved method, mono(6‐amino‐6‐deoxy)‐β‐cyclodextrin or heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin reacted with glycidyl methacrylate to generate the corresponding functional monomers and were subsequently copolymerized with ethylene dimethacrylate. The polymerization conditions for both monoliths were carefully optimized to obtain satisfactory column performance with respect to column efficiency, reproducibility, permeability, and stability. The obtained poly(glycidyl methacrylate‐mono(6‐amino‐6‐deoxy)‐β‐cyclodextrin‐co‐ethylene dimethacrylate) and poly(glycidyl methacrylate‐heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin‐co‐ethylene dimethacrylate) monoliths exhibited a uniform structure, good permeability, and mechanical stability as indicated by scanning electron microscopy and micro‐high‐performance liquid chromatography experimental results. Because of the probable existence of multi‐glycidyl methacrylate linking spacers on the poly(glycidyl methacrylate‐heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin‐co‐ethylene dimethacrylate) monolith, the effect of the ratio of glycidyl methacrylate/heptakis(6‐amino‐6‐deoxy)‐β‐cyclodextrin was especially studied, and satisfactory reproducibility could still be achieved by strictly controlling the composition of the polymerization mixture. To investigate the effect of the degree of amino substitution of β‐cyclodextrin on column performance, a detailed comparison of the two monoliths was also carried out using series of analytes including small peptides and chiral acids. It was found that the β‐cyclodextrin‐functionalized monolith with mono‐glycidyl methacrylate linking spacers demonstrated better chiral separation performance than that with multi‐glycidyl methacrylate linking spacers.  相似文献

Tetracyclines are a group of broad spectrum antibiotics widely used in animal husbandry to prevent and treat diseases. However, the improper use of tetracyclines may result in the presence of their residues in animal tissues or waste. Recently, great attention has been drawn towards the green solvents ionic liquids. Ionic liquids have been employed as a coating material to modify the electroosmotic flow in capillary electrophoresis. In this study, a functionalized ionic liquid, mono‐6‐deoxy‐6‐(3‐methylimidazolium)‐β‐cyclodextrin tosylate, was synthesized and used for the simultaneous separation and quantification of tetracyclines by capillary electrophoresis. Good separation efficiency could be achieved due to the multiple functions of β‐cyclodextrin derived ionic liquid, including the electrostatic interaction, the hydrogen bonding, and the cavity structure in β‐cyclodextrin ionic liquid which can entrap the tetracyclines to form inclusion complex. After optimization, baseline separation achieved in 25 min with the running buffer consisted of 10 mmol/L, pH 7.2 phosphate buffer and 20 mmol/L β‐cyclodextrin ionic liquid. The satisfied result demonstrated that the β‐cyclodextrin ionic liquid is an ideal background electrolyte modifier in the separation of tetracyclines with high stability and good reproducibility. And it is an effective strategy to design and synthesize specific ILs as additive applied in separation.  相似文献

Recombinant human erythropoietin is an important therapeutic protein with high economic interest due to the benefits provided by its clinical use for the treatment of anemias associated with chronic renal failure and chemotherapy. In this work, a poly(norepinephrine)‐coated open tubular column was successfully prepared based on the self‐polymerization of norepinephrine under mild alkaline condition, the favorable film forming and easy adhesive properties of poly(norepinephrine). The poly(norepinephrine) coating was characterized by scanning electron microscopy and measurement of the electro‐osmotic flow. The thickness of the coating was about 431 nm. The electrochromatographic performance of the poly(norepinephrine)‐coated open tubular column was evaluated by separation of proteins. Some basic and acidic proteins including two variants of bovine serum albumin and two variants of β‐lactoglobulin achieved separation in the poly(norepinephrine)‐coated open tubular column. More importantly, the column demonstrated separation ability for the glycoforms of recombinant human erythropoietin. In addition, the column demonstrated good repeatability with the run‐to‐run, day‐to‐day, and column‐to‐column relative standard deviations of migration times of proteins less than 3.40%.  相似文献

Chiral separation of dausyl amino acids by capillary electrophoresis using mixed selectors of Mn(ll)-L-alanine complex and β-cyclodextrin (β-CD) was studied. Resolution was considerably superior to that obtained by using either Mn (Ⅱ)-L-alanine complex or β-CD alone. The effects of separation parameters, such as pH value of buffer solution, capillary temperature, the concentration of Mn (Ⅱ)-L-alanine complex, the types of CD and ligand on the migration times and resolutions were investigated. Six different transition metal complexes,Cu(Ⅱ), Zn(Ⅱ), Co(Ⅱ), Ni(Ⅱ), Hg(Ⅱ) and Cd(Ⅱ)-L-alanine complexes have been employed and compared with Mn(Ⅱ)complex. Differences in retention and selectivity were found.The substitution of Cu(Ⅱ), Zn(Ⅱ), Co(Ⅱ) and Ni(Ⅱ) for Mn(Ⅱ) resulted in a better chiral resolution while Hg(Ⅱ) and Cd(Ⅱ) showed poorer resolution abilities. The chiral separation mechanism was also discussed briefly.  相似文献

Submicron, non‐porous, chiral silica stationary phase has been prepared by the immobilization of functionalized β‐CD derivatives to isocyanate‐modified silica via chemical reaction and applied to the pressurized capillary electrochromatography (pCEC) enantio‐separation of various chiral compounds. The submicron, non‐porous, cyclodextrin‐based chiral stationary phases (sub_μm‐CSP2) exhibited excellent chiral recognition of a wide range of analytes including clenbuterol hydrochloride, mexiletine hydrochloride, chlorpheniramine maleate, esmolol hydrochloride, and metoprolol tartrate. The synthesized submicron particles were regularly spherical and uniformly non‐porous with an average diameter of around 800 nm and a mean pore size of less than 2 nm. The synthesized chiral stationary phase was packed into 10 cm × 100 μm id capillary columns. The sub_μm‐CSP2 column used in the pCEC system showed better separation of the racemates and at a higher rate compared to those used in the capillary liquid chromatography mode (cLC) system. The sub_μm‐CSP2 possessed high mechanical strength, high stereoselectivity, and long lifespan, demonstrating rapid enantio‐separation and good resolution of samples. The column provided an efficiency of up to 170 000 plates/m for n‐propylbenzene.  相似文献

Polystyrene (PS) nanoparticles coated by BSA, hereafter denoted as PS/BSA, were prepared and chemically immobilized for the first time onto a capillary inner wall for open‐tubular CEC (OTCEC). EOF and scanning electron micrography were used to characterize the prepared nanoparticle‐coated capillaries. To investigate the performance of the prepared columns in OTCEC, chiral separation of d ,l ‐tryptophan (dl ‐Trp) was performed in monolayer BSA‐modified capillary and PS/BSA nanoparticle‐coated columns. The results indicated that the nanoparticle‐modified column afforded a higher resolution compared with the monolayer type. Rapid enantioseparation of dl ‐Trp (within 3 min) was achieved with the PS/BSA‐immobilized column using an electroosmotic pump‐assisted CEC. Enantiomer separations of other compounds like dl ‐tyrosine and warfarin were also achieved with the column. Besides, run‐to‐run and column‐to‐column repeatabilities of the PS/BSA‐coated column in the chiral separation were systematically introduced.  相似文献

This work documents the influence of the position of single carboxymethyl group on the β‐cyclodextrin skeleton on the enantioselectivity. These synthesized monosubstituted carboxymethyl cyclodextrin (CD) derivatives, native β‐cyclodextrin, and commercially available carboxymethyl‐β‐cyclodextrin with degree of substitution approximately 3 were used as additives into the BGE consisting of phosphate buffer at 20 mmol/L concentration, pH 2.5, and several biologically significant low‐molecular‐mass chiral compounds were enantioseparated by CE. The results indicate that different substituent location on β‐cyclodextrin skeleton has a significant influence on the enantioseparation of the investigated enantiomers. The enantioselectivity of 2IO‐regioisomer was better than with native β‐cyclodextrin. Comparable results to native β‐cyclodextrin were obtained for 6IO‐ regioisomer and the enantioselectivity of 3IO‐regioisomer was even worse than with native β‐cyclodextrin. Commercially available derivative of CD provides better resolutions than the monosubstituted carboxymethyl CD derivatives for most of the investigated analytes.  相似文献

A novel open‐tubular CEC column coated with chitosan‐graft‐(β‐CD) (CDCS) was prepared using sol‐gel technique. In the sol‐gel approach, owing to the 3D network of sol‐gel and the strong chemical bond between the stationary phase and the surface of capillary columns, good chromatographic characteristics and unique selectivity in separating isomers were shown. The column efficiencies of 55 000~163 000 plates/m for the isomeric xanthopterin and phenoxy acid herbicides using the sol‐gel‐derived CDCS columns were achieved. Good stabilities were demonstrated that the RSD values for the retention time of thiourea and isoxanthopterin were 1.3 and 1.4% (run to run, n = 5), 1.6 and 2.0% (day to day, n = 3), 2.9 and 3.1% (column to column, n = 3), respectively. The sol‐gel‐coated CDCS columns have shown improved separations of isomeric xanthopterin in comparison with CDCS‐bonded capillary column.  相似文献

The nanocomposite (denoted as GR‐AuNPs‐CD‐CS) of graphene (GR), gold nanoparticles (AuNPs), chitosan (CS) and β‐cyclodextrin (β‐CD) was prepared to modify a glassy carbon electrode. The as‐modified electrode was explored for the ultrasensitive detection of dopamine (DA) and uric acid (UA). The modified electrode demonstrated linearly increased current response in the concentration range of 0.1–120 µm for DA and 0.05–70 µm for UA, with so far the best detection limit for DA and UA. Good stability and repeatability were further demonstrated for the as‐made sensor.  相似文献

The chiral organic‐inorganic hybrid materials can exhibit a high loading, and the chiral selector nanoparticles can create efficient stationary phases for open‐tubular capillary electrochromatography (OT‐CEC). Hence, a novel protocol for the preparation of an OT column coated with nano‐amylose‐2,3‐bis(3,5‐dimethylphenylcarbamate) (nano‐ABDMPC)‐silica hybrid sol through in situ layer‐by‐layer self‐assembly method was developed for CEC enantioseparation. By controlling the assembly cycle number of nano‐ABDMPC‐silica hybrid sol, a homogeneous, dense and stable coating was successfully prepared, which was confirmed by SEM and elemental analysis. As the main parameter influencing the chiral separating effect, the nano‐ABDMPC bearing 3‐(triethoxysilyl)propyl residues concentration was investigated. The experimental results showed that 10.0 mg/mL nano‐ABDMPC bearing 3‐(triethoxysilyl)propyl residues coated OT capillary column possessed chiral recognition ability toward the six enantiomers (phenylalanine, tyrosine, tryptophan, phenethyl alcohol, 1‐phenyl‐2‐propanol, and Tröger's base) at some of the different conditions tested. Additionally, the coated OT column revealed adequate repeatability concerning run‐to‐run, day‐to‐day and column‐to‐column. These results demonstrated the promising applicability of nano‐ABDMPC‐silica hybrid sol coated OT column in CEC enantioseparations.  相似文献

Chiral separation of 12 pairs of basic analyte enantiomers including oxybutynin, bambuterol, tradinterol, clenbuterol, clorprenaline, terbutaline, tulobuterol, citalopram, phencynonate, fexofenadine, salbutamol, and penehyclidine was conducted by capillary electrophoresis using a single‐isomer anionic β‐cyclodextrin derivative, heptakis‐(2,3‐diacetyl‐6‐sulfato)‐β‐cyclodextrin as the chiral selector. Parameters influencing separation were studied, including background electrolyte pH, heptakis‐(2,3‐diacetyl‐6‐sulfato)‐β‐cyclodextrin concentration, buffer concentration, and separation voltage. A background electrolyte consisting of 50 mM Tris‐H3PO4 and 6 mM heptakis‐(2,3‐diacetyl‐6‐sulfato)‐β‐cyclodextrin at pH 2.5 was found to be highly efficient for the separation of most enantiomers, with other conditions of normal polarity mode at 10 kV, detection wavelength of 210 nm using hydrodynamic injection for 3 s. Under the optimal conditions, baseline resolution (>1.50) for 11 pairs of enantiomers and somewhat lower resolution for penehyclidine enantiomers (1.17) were generated. Moreover, the possible mechanism of separation of clenbuterol, oxybutynin, salbutamol, and penehyclidine was investigated using a computational modeling method.  相似文献

The separation and determination of proteins in food is an important aspect in food industry. Inspired by the self‐polymerization of dopamine under alkaline conditions and the natural adhesive properties of polydopamine, in this paper, a simple and economical method was developed for the preparation of polydopamine‐coated open tubular column, in which ammonium persulfate was used as the source of oxygen to induce and facilitate the polymerization of dopamine to form polydopamine. In comparison with a naked fused‐silica capillary, the direction and magnitude of the electro‐osmotic flow of the as‐prepared polydopamine‐coated open tubular column could be manipulated by varying the pH values of background solutions due to the existence of amine and phenolic hydroxyl groups on polydopamine coating. The surface morphology of the polydopamine‐coated open tubular column was studied by scanning electron microscopy, and the thickness of polydopamine coating was 106 nm. The performance of the polydopamine‐coated open tubular column was validated by analysis of proteins. The relative standard deviations of migration times of proteins representing run‐to‐run, day‐to‐day, and column‐to‐column were less than 3.5%. In addition, the feasibility of the polydopamine‐coated open tubular column for real samples was verified by the separation of proteins in chicken egg white and pure milk.  相似文献

A supramolecular recognition functionalized electrode (βCD‐nanoAu/Fc‐ITO) which exhibits redox‐activity was prepared through supramolecular assembly of β‐cyclodextrin (βCD) capped gold nanoparticles (βCD‐nanoAu) on the ITO previously coated with a monolayer of ferrocene residues (Fc‐ITO). The immobilization of βCD‐nanoAu on Fc‐ITO was confirmed by atomic force microscopy (AFM), and the supramolecular nature of the immobilization approach was also confirmed by cyclic voltammetry. On the other hand, the electrocatalytic activity of βCD‐nanoAu/Fc‐ITO electrode was also studied. The electrocatalytic activity toward ascorbic acid (AA) was enhanced compared with that at the Fc‐ITO electrode, and a linear relationship existed between the anodic peak and the concentration of AA in the range of 5.3×10?5 to 3.0×10?3 M with a detection limit (S/N=3) of 4.1×10?6 M.  相似文献
