The knowledge of the spatial distribution and the local concentration of trace elements in tissues are of great importance since trace elements are involved in a number of metabolic and physiological processes in the human body, and their deficiency and excess may lead to different metabolic disorders. In this way, the main goal of this work is to compare the elemental concentration in different brain structures, namely temporal cortex, entorhinal cortex, visual cortex and hippocampus, from Wistar female rats (n = 15) with different ages: 2, 8 and 48 weeks. The measurements were performed at the Synchrotron Light Brazilian Laboratory, Campinas, São Paulo, Brazil. In the entorhinal cortex, the following elements decreased with age: Zn, S, Cl, K, Ca and Br. In the temporal cortex, Ca, Fe and Br levels increased with aging and on the other hand, P, S, Cl, K and Rb levels decreased with aging. In the visual cortex almost all the elements decreased with aging: Cl, Ca, Fe, Ni and Zn. In the hippocampus, in turn, most of the elements identified, increased with aging: Al, P, S, K, Fe, Cu, Zn and Rb. The increase of Fe with aging in the hippocampus is an important fact that will be studied, since it is involved in oxidative stress. It is believed that oxidative stress is the one of the main causes responsible for neuronal death in Parkinson's disease.  相似文献

Synchrotron radiation X-ray fluorescence (SRXRF) spectroscopy is an advanced method of quantitative multielemental analysis with space resolution of several microm and sensitivities in the microg g(-1) range. It can be used for keeping track of trace elements after an electrophoretic separation of biological samples. In this paper, proteins in human liver cytosol were separated with gel filtration chromatography and thin layer isoelectric focusing (IEF). The contents of metal ions in protein bands were determined by SRXRF. The results showed that in the molecular weight (MW) range of 10 approximately 25 kDa, there were at least 2 Zn-containing bands with isoelectric point (pI) of 5 approximately 6 and 6.2 approximately 7, respectively and about 11 Fe-containing proteins with pI of 4.4, 4.6, 4.8, 5.0, 5.2, 5.3, 5.5, 5.6, 6.6, 6.8, and 7.2, respectively, present in human liver cytosol. The Zn-containing band with pI of 5-6 is the dominant species of zinc in this MW range. In addition, the Cu-containing bands with pI of 5.0 and below 4.8 were also detected. It is demonstrated that the procedure could be widely used in further investigations of the chemical species of trace elements in biological samples.  相似文献

Measurements of the refractive index (RI) and elemental analysis using synchrotron radiation X-ray fluorescence spectrometry (SR-XRF) were applied to the forensic discrimination of sheet-glass samples from different origins. The refractive index was calculated from the matching temperature at which the glass fragments became invisible in silicone oil. Fragments smaller than 1 mm in maximum diameter were taken from each of 11 sheet glasses and subjected to analysis by SR-XRF. The XRF spectrum of these samples indicated that a comparison of 6 elements (Ca, Fe, Sr, Zr, Ba and Ce) was useful for the discrimination of sheet glasses. Cluster analysis was performed using 33 sets of SR-XRF data obtained by triplicate measurements for the 11 glasses. Comparing 528 pairs among 33 samples, 515 pairs could be correctly discriminated. The number of indistinguishable pairs could be reduced from 36 to 4 by comparing the SR-XRF data. Elemental analysis by SR-XRF could provide small glass fragments with a more evidential value than the solely measurement of only RI, through a significant improvement of the discrimination capability.  相似文献

This paper aims to study the environmental pollution in the tree development, in order to evaluate its use as bioindicator in urban and country sides. The sample collection was carried out in Piracicaba city, São Paulo State, which presents high level of environmental contamination in water, soil and air, due to industrial activities, vehicles combustion, sugar-cane leaves burning in the harvesting, etc. The species Caesalpinia peltophoroides (“Sibipiruna”) was selected because it is widely used in urban forestation. Synchrotron Radiation Total Reflection X-ray Fluorescence technique (SR-TXRF) was employed to identify and quantify the elements and metals of nutritional and toxicological importance in the wood samples. The analysis was performed in the Brazilian Synchrotron Light Source Laboratory, using a white beam for excitation and a Si(Li) detector for X-ray detection. In several samples, P, K, Ca, Ti, Fe, Sr, Ba and Pb were quantified. The K/Ca, K/P and Pb/Ca ratios were found to decrease towards the bark.  相似文献

The concentration of copper in traditional homemade alcoholic distillates produced in Venezuela (Cocuy de Penca) were determined by total reflection X-ray fluorescence (TXRF) using vanadium as internal standard. The results were compared to those obtained by flame atomic absorption spectrometry (FAAS). Three preparative methods of addition of vanadium were compared: classical internal standard addition, ‘layer on layer’ internal standard addition and in situ addition of internal standard. The TXRF procedures were accurate and the precision was comparable to that obtained by the FAAS technique. Copper levels were above the maximum allowed limits for similar beverages. Zinc and iron in commercial and homemade distilled beverages were also analyzed by TXRF with in situ addition of internal standard demonstrating the usefulness of this technique for trace metal determination in distillates.  相似文献

Summary The study of trace element levels is of great importance due to their relevance in agingand several neurodegenerative diseases. This work compares the elemental concentrations in different postnatal ages and between the temporal cortex, entorhinal cortex and hippocampus from Wistar rats, using X-ray total reflection fluorescence with synchrotron radiation. Ten elements were determined in brain samples: Ti, Cr, Mn, Fe, Cu, Zn, (at trace level) and P, S, Cl and K (at major levels). The elements that increased with aging in cortical areas were: S, K, Fe, Cu and Zn. Ca and Zn levels decreased with advancing age in the hippocampus. In addition to this, Ti, Mn and Fe levels were more conspicuous in the entorhinal cortex.  相似文献

Knowledge about the spatial distribution and the local concentration of trace elements in tissues is of great importance, since trace elements are involved in many biological functions of living organisms. However, there are few methods available to measure the spatial (two (three)-dimensional) elemental distribution in animal brain. X-ray microfluorescence with synchrotron radiation is a multielemental mapping technique, which was used in this work to determine the topographic of iron, zinc and copper in coronal sections of female Wistar rats of different ages. Young (14 days old) and middle-aged (20 months old) rats (n = 8) were analyzed. The measurements were carried out at the XRF beam line at the Synchrotron Light National Laboratory (Campinas, Brazil). Two-dimensional scanning was performed in order to study the tendency of elemental concentration variation. The acquisition time for each pixel was 10 s/step and the step size was 300 microm/step in both directions. It was observed that the iron distribution was more conspicuous in the cortical area, thalamus and bellow the thalamus. On the other hand, the zinc distribution was more pronounced in the hippocampus. The iron, copper and zinc levels increased with advancing age. Therefore, this study reinforces the idea that these elements are involved in the chemical mechanisms of the brain that induce some neurological diseases, since they are also present in high levels in specific areas of the brain, such as the hippocampus and the substantia nigra of patients with these disorders.  相似文献

The method of proton-induced X-ray emission (or PIXE) analysis has been investigated with reference to the study of minor and trace elements in biological tissue. Methods of specimen preparation have been investigated, and optimum instrumental conditions arrived at. The lower limits of detection for lead and zinc in blood have been investigated and have been found to be approximately 0.3 and 0.1 ppm for lead and zinc respectively, for a proton energy of 2.5 MeV. The concentrations of copper, zinc and iron have been studied in normal and diseased human liver tissue. In cirrhotic tissue the copper levels are substantially raised. We note also a diminution in liver iron in a case of alcoholic cirrhosis associated with clinical anaemia; and a blood zinc deficiency in primary biliary cirrhosis. The results are discussed in relation to existing work.  相似文献

This paper presents an alternative analytical method employing energy dispersive X-ray fluorescence (EDXRF) to determine copper, iron, nickel and zinc ions in ethanol fuel samples after a pre-concentration procedure. Our pre-concentration strategy utilizes analyte retention on cation exchange chromatography paper, a convenient substrate for direct EDXRF measurements. The repeatability, expressed in terms of RSD of standard solutions containing 0.25 μg mL−1 of Cu, Fe, Ni and Zn, and calculated from fifteen consecutive measurements, was 2.5, 2.8, 3.0, and 2.7%, respectively. The limits of detection (LOD), defined as the analyte concentration that gives a response equivalent to three times the standard deviation of the blank (n = 10), were found to be 13, 15, 15 and 12 μg L−1 for Cu, Fe, Ni and Zn, respectively. The proposed method was applied to Cu, Fe, Ni and Zn determination in hydrated ethanol fuel samples collected from different gas stations.  相似文献

Bromine (20-40 ppm), chlorine (200-500 ppm), sulphur (0.2-3%) and phosphorus (300-1000 ppm) in peat have been determined by X-ray fluorescence spectrometry (XRFS) combined with the standard-addition method. Chlorine, sulphur and phosphorus have also been determined by other methods and agreement between the results is good. Theoretical calculations based on the Sherman equation were made to validate the linearity of the standard-addition curves. A multi-element standard-addition technique has been tested with addition of all elements at the same time. The results for chlorine were high but after correction for the difference in attenuation coefficient between the sample and added compound the results agreed with those from single-element standard addition.  相似文献

The presented work provides baseline data on the existing airborne conditions in the Miyagi Museum of Art in Sendai, Japan, during the summer of 2000. The chemical composition, size and indoor and outdoor origin of the suspended particulate matter were identified using a number of advanced X-ray techniques, such as Electron Probe X-Ray Microanalysis (EPXMA) and Energy Dispersive X-Ray Fluorescence Analysis (EDXRF). Our results, to the best of our knowledge, represent the first detailed study of the chemical nature of the indoor particulate matter in a Japanese museum and, as such, may contribute to future improvements of the air quality inside museums and to the lasting conservation of works of art.  相似文献

Trace amounts of zinc, copper, lead and manganese (10–75 μg) are coprecipitated on iron(III) hydroxide at pH 8.5 after masking of phosphate by calcium. Reliable results are obtained for mixtures with widely different concentrations. The limit of detection is about 1 μg.  相似文献

Total reflection X-ray fluorescence spectrometry and chemical pre-concentration procedures have been applied for the analysis of trace concentrations of copper, mercury, and lead in drinking water samples. A simple total reflection module has been used in X-ray measurements. The elements under investigation were pre-concentrated by complexation using a mixture of carbamates followed by solvent extraction with methyl isobutyl ketone. The preconcentration procedure was tested with the use of twice-distilled water samples and samples of mineral and tap water spiked with known additions of copper, mercury, and lead. The obtained recovery and precision values are presented. The minimum detection limits for the determination of these elements in mineral and tap water samples were found to be 40 ng l−1, 60 ng l−1, and 60 ng l−1, respectively.  相似文献

Archaeometallurgical materials from “La Bastida de Moixent”, a site in Valencia (Spain), from the second Iberian iron age (4th Century B.C.) have been studied using metallographic techniques, microanalysis, chemometrics and image analysis. The materials come from various phases of iron production and cupellation of argentiferous lead to obtain silver. Scanning electron microscopy (SEM) is used to determine the morphological, microstructural and topographic characteristics of the samples. Image analysis was used to obtain a numeric estimate of the main components in these materials. X-ray microanalysis (SEM/EDX) provides qualitative and quantitative information about the elements in the sample. The semiquantitative results have been treated by chemometric tools such as Principal Component Analysis (PCA) to group the different archaeological materials. These analyses provide information on the lead and iron metallurgical processes, such as silver production by cupellation used in the period. Macroscopic and microscopic analyses indicate a high quality, acceptably uniform manufactured product, indicating broad technical skill in the later metallurgical process of transformation and refinement of these materials to obtain ingots and manufactured products to trade.  相似文献

Chromatin-bound metal ions interact with enzymatic reactions involved in DNA repair processes. Content of Cu, Zn and Mn was studied in the chromatin of lymphocytes derived from patients with some light sensitive skin diseases, i.e. 26 with polymorphic light eruption and 10 with porphyria cultanea tarda. After isolating the lymphocytes by ficoll-urographin method and separating the chromatin by lysis of the cells, the trace elements studied were determined by neutron activation analysis. The Zn and Mn content in the chromatin of the patients did not differ from that of the controls. The copper concentration was significantly higher in the chromatin of the cells derived from patients with polymorphic light eruption, but not in cases with porphyria cutanea tarda. The results are discussed in connection with the UV-light induced excision repair.  相似文献
