A zinc-mediated addition of methyl and terminal alkynes to chiral N-tert-butanesulfinyl ketimines for the preparation of optical quaternary 3-amino oxindoles was reported. In general, the operationally simple reaction affords the desired products in high yields and good to excellent diastereoselectivities. Subsequent convenient cleavage of sulfinyl protecting group under mild conditions was presented without racemization. 相似文献

Reduction of (RS)-N-tert-butanesulfinyl alpha-halo imines afforded chiral aziridines in good to excellent yields. Upon reduction of (RS)-N-tert-butanesulfinyl alpha-halo imines with NaBH4 in THF, in the presence of 10 equiv of MeOH, (RS,S)-beta-halo sulfinamides were formed in excellent yield (up to 98%) with very good stereoselectivity (>98:2). Simple treatment of the latter (RS,S)-beta-halo-tert-butanesulfinamides with KOH afforded the corresponding (RS,S)-N-(tert-butylsulfinyl)aziridines in quantitative yields. On the contrary, its epimer, (RS,R)-N-(tert-butylsulfinyl)aziridine was synthesized by switchover of the reducing agent from NaBH4 to LiBHEt3. (RS,R)-N-(tert-Butylsulfinyl)aziridines were synthesized in good yields (up to 85%) and diastereoselectivity (up to 92:8) by reduction of (RS)-N-tert-butanesulfinyl alpha-halo imines with LiBHEt3 in dry THF and subsequent treatment with KOH. A three-component stereoselective reaction between an aldehyde, an amine and phenylacetylene to afford optically active propargyl amines in good yields was developed. The reaction is catalysed by copper complexes of enantiomerically pure bis-imines. The best results were obtained with imines readily prepared in very high yields from the commercially available binaphtyl diamine.
A very simple experimental procedure at room temperature allowed to obtain optically active propargyl amines in very good yields and enantioselectivity up to 75%. The extremely simple methodology and the mild reaction conditions, as well as the possibility of a modular approach for developing new and more efficient bis-imine-based chiral ligands make the present methodology very attractive. 相似文献

A general and efficient new method for the asymmetric synthesis of alpha-amino boronate esters has been developed. The key step is the Cu(I)-catalyzed addition of bis(pinacolato)diboron to N-tert-butanesulfinyl aldimines, which proceeds in good yields (52-88%) and with very high diastereoselectivities (>96:2) for a variety of aldimine substrates. This method was applied to an efficient synthesis of bortezomib, a potent alpha-amino boronic acid inhibitor of the proteasome that is in clinical use for the treatment of multiple myeloma and mantle cell lymphoma. 相似文献

A new method for the Rh(I)-catalyzed addition of arylboronic acids to N-tert-butanesulfinyl imino esters has been developed for the asymmetric synthesis of arylglycine derivatives. This method provides high yields (61-90%) and diastereoselectivities (>98:2) for a variety of functionalized arylboronic acids. The N-sulfinyl arylglycine ester products are versatile intermediates for further transformations, including selective protecting group removal, conversion to beta-amino alcohols, and direct incorporation into peptides. 相似文献

Treatment of novel chiral N-sulfinyl alpha-chloro ketimines with Grignard reagents resulted in the synthesis of chiral N-(1-substituted cyclopropyl)-tert-butanesulfinamides in acceptable to good yields and diastereoselectivity via 1,3-dehydrohalogenation and subsequent addition of the Grignard reagent to the intermediate cyclopropylideneamine. Only in the case of allylmagnesium chloride did the reaction lead to aziridines in high yield. An efficient method for the asymmetric synthesis of the trifluoromethylated propargylamines was described. Addition of lithium trifluoromethylacetylide, in situ prepared from lithium diisopropylamide and the 2-bromo-3,3,3-trifluoropropene, to various N-tert-butanesulfinyl imines provided a range of trifluoromethylated propargyl sulfinamides. Besides high yields and excellent diastereoselectivities, the additions featured that the diastereoselectivities could be reversed when polar or nonpolar solvent was used.
