Superoxide is involved in the pathogenesis of various diseases, such as inflammation, ischemia-reperfusion injury and carcinogenesis. Superoxide dismutases (SODs) catalyze the disproportionation reaction of superoxide to produce oxygen and hydrogen peroxide, and can protect living cells against the toxicity of free radicals derived from oxygen. Thus, SODs and their functional mimics have potential value as pharmaceuticals. We have previously reported that Fe(II)tetrakis-N,N,N',N'-(2-pyridylmethyl)ethylenediamine (Fe(II)TPEN) has an excellent SOD activity (IC50 = 0.5 microM) among many iron complexes examined (J. Biol. Chem., 264, 9243-9249 (1989)). Fe(II)TPEN can act like native SOD in living cells, and protect Escherichia coli cells from free radical toxicity caused by paraquat. In order to develop more effective SOD functional mimics, we synthesized Fe(II)TPEN derivatives with electron-donating or electron-withdrawing groups at the 4-position of all pyridines of TPEN, and measured the SOD activities and the redox potentials of these complexes. Fe(II) tetrakis-N,N,N',N'-(4-methoxy-2-pyridylmethyl)ethylenediamine (Fe(II)(4MeO)4TPEN) had the highest SOD activity (IC50 = 0.1 microM) among these iron-based SOD mimics. In addition, a good correlation was found between the redox potential and the SOD activity of 15 Fe(II) complexes, including iron-based SOD mimics reported in the previous paper (J. Organometal. Chem., in press). Iron-based SOD mimics may be clinically applicable, because these complexes are generally tissue-permeable and show low toxicity. Therefore our findings should be significant for the development of clinically useful SOD mimics.  相似文献

To gain insight into the mechanism of INH activation by KatG and to understand how resistance is conferred by the single active-site point mutation of KatG(S315T), we have employed pulse radiolysis as the means to initiate a catalytic pathway capable of mimicking the in vivo oxidation of isoniazid (INH). Radiolysis of a solution containing WT KatG revealed two intermediates: compound III (oxyferrous KatG) [415 (Soret), 545, 580 nm] formed [k1 = (4.47 +/- 0.91) x 105 M-1 s-1] in the absence of INH and compound II (410 (Soret), 540, 575 nm) formed [k1 = (4.43 +/- 0.69) x 105 M-1 s-1] in the presence of INH, with a comparison of the rates suggesting that compound III (rate-limiting) precedes compound II formation. By contrast, radiolysis of KatG(S315T) only led to compound III formation, whether INH was present [k1 = (4.72 +/- 0.99) x 105 M-1 s-1] or not [k1 = (4.51 +/- 1.38) x 105 M-1 s-1]. HPLC studies to determine the rates of INH-NADH adduct formation (an inhibitor of InhA) as catalyzed by KatG were also performed employing various oxidants: air [WT: (7.18 +/- 1.25) x 10-4, S315T: (0.74 +/- 0.39) x 10-4], superoxide (SOTS-1) [WT: (9.22 +/- 1.10) x 10-4, S315T: not detected], and tert-butylhydroperoxide [WT: (20.5 +/- 1.13) x 10-4, S315T: (10.15 +/- 0.19) x 10-4]. Taken together, the results from the pulse radiolysis work as well as the InhA inhibitor studies allow us to propose a mechanism capable of correlating the inability for the oxyferrous intermediate of KatG(S315T) to oxidize ("activate") INH to the suppressed formation of the INH-NADH adduct, thereby leading to INH resistance in Mycobacterium tuberculosis.  相似文献

The water-soluble, non-mu-oxo dimer-forming porphyrin, [5,10,15,20-tetrakis-4'-t-butylphenyl-2',6'-bis-(N-methylene-(4'-t-butylpyridinium))porphyrinato]iron(III) octabromide, (P(8+))Fe(III), with eight positively charged substituents in the ortho positions of the phenyl rings, was characterized by UV-vis and 1H NMR spectroscopy and 17O NMR water-exchange studies in aqueous solution. Spectrophotometric titrations of (P(8+))Fe(III) indicated a pKa1 value of 5.0 for coordinated water in (P(8+))Fe(III)(H2O)2. The monohydroxo-ligated (P(8+))Fe(III)(OH)(H2O) formed at 5 < pH < 12 has a weakly bound water molecule that undergoes an exchange reaction, k(ex) = 2.4 x 10(6) s(-1), significantly faster than water exchange on (P(8+))Fe(III)(H2O)2, viz. k(ex) = 5.5 x 10(4) s(-1) at 25 degrees C. The porphyrin complex reacts with nitric oxide to yield the nitrosyl adduct, (P(8+))Fe(II)(NO+)(L) (L = H2O or OH-). The diaqua-ligated (P(8+))Fe(III)(H2O)2 binds and releases NO according to a dissociatively activated mechanism, analogous to that reported earlier for other (P)Fe(III)(H2O)2 complexes. Coordination of NO to (P(8+))Fe(III)(OH)(H2O) at high pH follows an associative mode, as evidenced by negative deltaS(double dagger)(on) and deltaV(double dagger)(on) values measured for this reaction. The observed ca. 10-fold decrease in the NO binding rate on going from six-coordinate (P(8+))Fe(III)(H2O)2 (k(on) = 15.1 x 10(3) M(-1) s(-1)) to (P(8+))Fe(III)(OH)(H2O) (k(on) = 1.56 x 10(3) M(-1) s(-1) at 25 degrees C) is ascribed to the different nature of the rate-limiting step for NO binding at low and high pH, respectively. The results are compared with data reported for other water-soluble iron(III) porphyrins with positively and negatively charged meso substituents. Influence of the porphyrin periphery on the dynamics of reversible NO binding to these (P)Fe(III) complexes as a function of pH is discussed on the basis of available experimental data.  相似文献

A new type of thiourea cross-linked chitosan with Fe(III) as template (TCCTS template) was synthesized. The adsorption of Fe(II) and Fe(III) on this TCCTS template was studied. The factors affecting adsorption such as pH and contact time were considered. The results showed that the optimum pH value for adsorption was pH = 5.0 and the adsorption equilibrium time was about 60 min. The adsorption isotherms and kinetics were investigated, and the equilibrium data agreed very well with the Langmuir model and the pseudo second-order model could describe adsorption process better than the pseudo first-order model. Results also showed that TCCTS template was a favourable adsorbent for Fe(II) and Fe(III) in aqueous solution.  相似文献

The new heterodinuclear mixed valence complex [Fe(III)Mn(II)(BPBPMP)(OAc)(2)]ClO(4) (1) with the unsymmetrical N(5)O(2) donor ligand 2-bis[((2-pyridylmethyl)-aminomethyl)-6-((2-hydroxybenzyl)(2-pyridylmethyl))-aminomethyl]-4-methylphenol (H(2)BPBPMP) has been synthesized and characterized. Compound 1 crystallizes in the monoclinic system, space group P2(1)/c, and has an Fe(III)Mn(II)(mu-phenoxo)-bis(mu-carboxylato) core. Two quasireversible electron transfers at -870 and +440 mV versus Fc/Fc(+) corresponding to the Fe(II)Mn(II)/Fe(III)Mn(II) and Fe(III)Mn(II)/Fe(III)Mn(III) couples, respectively, appear in the cyclic voltammogram. The dinuclear Fe(III)Mn(II) center has weakly antiferromagnetic coupling with J = -6.8 cm(-1) and g = 1.93. The (57)Fe M?ssbauer spectrum exhibits a single doublet, delta = 0.48 mm s(-1) and DeltaE(Q) = 1.04 mm s(-1) for the high spin Fe(III) ion. Phosphatase-like activity at pH 6.7 with the substrate 2,4-bis(dinitrophenyl)phosphate reveals saturation kinetics with the following Michaelis-Menten constants: K(m) = 2.103 mM, V(max) = 1.803 x 10(-5) mM s(-1), and k(cat) = 4.51 x 10(-4) s(-1).  相似文献

Electronic absorption and resonance Raman (RR) spectra are reported for hydroxide and aqua complexes of iron(II)-protoporphyrin IX (Fe(II)PP) respectively formed in alkaline and neutral aqueous solutions. These compounds with weak axial ligand(s) represent a biomimetic approach of the unusual coordination of the atypical heme c(i) of membrane cytochrome b6f complexes. Absorption spectra and spectrophotometric titrations show that Fe(II)PP in alkaline aqueous cetyltrimethylammonium bromide (CTABr) binds one hydroxide ion, forming a five-coordinated high-spin (HS) complex. In alkaline aqueous ethanol, we confirm the formation of a dihydroxy complex of Fe(II)PP. In the RR spectra of Fe(II)PP dissolved in neutral aqueous CTABr, a mixture of a four-coordinated intermediate spin form with an HS monoaqua complex (Fe(II)PP(H2O)) was observed. The spectroscopic information obtained for Fe(II)PP(OH-), Fe(II)PP(H2O), and Fe(II)PP(OH-)2 was compared with that previously reported for the 2-methylimidazole and 2-methylimidazolate complexes of Fe(II)PP, representative of the most common axial ligation in HS heme proteins. This investigation reveals a very remarkable analogy in the spectral properties of, in one hand, the Fe(II)PP(H2O) and mono-2-methylimidazole complexes and, in the other hand, the Fe(II)PP(OH-) and mono-2-methylimidazolate complexes. The comparisons of the absorption and RR spectra of Fe(II)PP(OH-) and Fe(II)PP(OH-)2 clearly establish that both a redshift of the pi-pi electronic transitions and an upshift of the v8 RR frequency are spectral parameters indicative of porphyrin doming in HS ferrous complexes. Based upon isotopic substitutions (16OH-,16OD-, and 18OH-), stretching modes of the Fe-OH bond(s) of a ferrous porphyrin were assigned for the first time, i.e., at 435 cm(-1) for Fe(II)PP(OH-) (nu(Fe(II)-OH-)) and at 421 cm(-1) for Fe(II)PP(OH-)2 (nu(s)(Fe(II)-(OH-)2). The spectroscopic and redox properties of Fe(II)PP(H2O), Fe(II)PP(OH-), and heme c(i) were discussed and favor a water coordination for the heme c(i) iron.  相似文献

The H-point standard addition method (HPSAM) was applied to kinetic data for simultaneous determination of Fe(III) and Fe(II) or selective determination of Fe(III) in the presence of Fe(II). The method is based on the difference in the rate of two processes; reduction of Fe(III) with Co(II) and subsequent complex formation of resulted Fe(II) with 1,10-phenanthroline, and direct complex formation between Fe(II) and 1,10-phenanthroline in pH 3 and cetyl trimethyl ammonium bromide, CTAB, micellar media. Fe(III) can be determined in the range of 0.75-5.13 mug ml(-1)with satisfactory accuracy and precision in the presence of excess Fe(II) under working conditions. The proposed method was successfully applied to the simultaneous determination of Fe(III) and Fe(II) and also to the selective determination of Fe(III) in the presence of Fe(II) in several synthetic mixtures containing different concentration ratios of Fe(III) to Fe(II).  相似文献

Formation of free radical intermediates in a NaOH/H(2)O(2)/Fe(III) system has been studied by ESR spectroscopy in the presence of the spin trap 5,5-dimethy-1-pyrroline N-oxide (DMPO). DMPO/O(2(*) ) (-) and DMPO/(*)OH signals were simultaneously detected in this system, but only the DMPO/(*)OH signal could be observed in the absence of Fe(III). Effects of pH values and Fe(III) concentrations on the ESR signal intensities were investigated in detail. Formation of DMPO/O(2(*) ) (-) adduct was inhibited by the addition of superoxide dismutase (SOD), catalase or nitro blue tetrazolium (NBT), and by chelating the Fe(III) with some chelators, including ethylenediaminetetraacetic acid (EDTA), diethylenetriaminepentaacetic acid (DTPA), and desferrioxamine (DFO). Deoxygenation from the NaOH/H(2)O(2)/Fe(III) mixture had a slight effect on the formation of DMPO/O(2(*) ) (-). DMPO/O(2(*) ) (-) signal was also detected from the NaOH/H(2)O(2)/Fe(II) mixture, but it can be totally suppressed under anaerobic conditions. Considering the hydrolysis of Fe(III) into polymerization iron species with oxide phases in the alkaline medium, Fe(2)O(3) was directly suspended into a mixture of NaOH/H(2)O(2) for comparison. Fortunately, the presence of Fe(2)O(3) suspension was found to be of benefit to the production of DMPO/O(2(*) ) (-). Influence of aging time of hydrolytic iron species on the superoxide anion radical generation was also studied. These results suggest that the generation of O(2(*) ) (-) from the NaOH/H(2)O(2)/Fe(III) system was probably caused by the heterogeneous surface catalysis initiated by hydrolytic iron species.  相似文献

A series of ternary copper(II) complexes containing same coordination sphere but difference in the counter ions, viz., [Cu(PMDT)(OAc)]PF(6)(1); [Cu(PMDT)(OAc)]ClO(4)(2); [Cu(PMDT)(OAc)]BF(4)(3) and [Cu(PMDT)(OAc)]BPh(4)(4) where PMDT=N,N,N',N',N'-pentamethyldiethylenetriamine, OAc=Acetate ion were synthesized and characterized by means of spectroscopic, magnetic and cyclic voltammetric measurements. In frozen solution e.p.r. spectra, an interesting relation g|| >g(perpendicular) has been observed which is atypical of the axially symmetric d(9) Cu(II) (S(Cu)=1/2) having an unpaired electron in a d (x2-y2) orbital. Single crystal X-ray analysis of (1) has revealed the presence of distorted square planar geometry. The influence of the counter ion on the complexes has been examined by performing some biological experiments like superoxide dismutase and anti-microbial activity.  相似文献

The one-electron reduced iron(II)-dioxygen adduct, {Fe(II)-O(2)}(-), is known to be an important intermediate in the catalytic cycle of heme (mono)oxygenases. The same type of species, considered as Fe(III)-peroxo, can be formed in a direct reaction between a Fe(II) center and superoxide. In a unique high-pressure study of the reaction between superoxide and the Fe(II) complex of a crown ether porphyrin conjugate in dimethylsulfoxide (DMSO), the overall Fe(II)-superoxide interaction mechanism could be visualized and the nature of all species that occur along the reaction coordinate could be clarified. The equilibrium between the low-spin and high-spin forms of the starting Fe(II) complex was quantified, which turns out to be the actual activation step toward substitution and subsequent inner-sphere electron transfer reactions. The constructed reaction volume profile demonstrates that the reaction product consists of Fe(III)-peroxo and Fe(II)-superoxo species that exist in equilibrium, which can better account for the versatile reactivity of {Fe(II)-O(2)}(-) adducts toward different substrates.  相似文献

A spectrophotometric method for the determination of Fe(II) in the presence of large amounts (up to 800 mg/l.) of Fe(III) is suggested. The Fe(III) is effectively masked by complexing with fluoride at pH 2.0-2.4 before development of the violet Fe(II) complex with 2,4,6-tri(2'-pyridyl)-1,3,5-triazine. The absorbance is measured at 595 nm. Various commonly occurring ions which complex with Fe(II) and/or Fe(III) do not interfere.  相似文献

Chelation studies of cobalt(III) with 1,2 bis(beta-aminoethoxy)ethane N,N,N',N' sodium sulfonate triacetic acid (ASTA) were performed. The results showed the effectiveness of ASTA as a chelating agent by using: molar ratio, continuous variation and slope ratio methods. Stable complex 1∶1 was formed at pH from 6.0 to 10.5. Solutions of Co-ASTA chelate of different molar ratios at pH 6.5 and 8.0 were irradiated by different gamma-radiation doses. The results showed a linear decrease of absorbance with gamma-radiation dose which can be utilized as a dosimeter for low dose rate measurement in the range studied. A proposed radiolytic mechanism is discussed. The degradation of the ASTA ligand has been related to hydroxyl radical attack.  相似文献

The light-induced high-spin-->low-spin relaxation for the Fe(II) spin-crossover compounds [Fe(btpa)](PF6)2 and [Fe(b(bdpa))](PF6)2 in solution, where btpa is the potentially octadentate ligand N,N,N',N'-tetrakis(2-pyridylmethyl)-6,6'-bis(aminomethyl)-2,2'-bipyridine and b(bdpa) is the analogous hexadentate ligand N,N'-bis(benzyl)-N,N'-bis(2-pyridylmethyl)-6,6'-bis(aminomethyl)-2,2'- bipyridine, respectively, has been studied by temperature-dependent laser flash photolysis. [Fe(b(bdpa))](PF6)2 shows single-exponential 5T2-->1A1 relaxation kinetics, whereas [Fe(btpa)](PF6)2 exhibits solvent-independent biphasic relaxation kinetics. The fast process of [Fe(btpa)](PF6)2 with a rate constant, kHL, of 2.5 x 10(7) s-1 at 295 K and an activation energy, Ea, of 1294(26) cm-1 in methanol can be assigned to the 5T2-->1A1 relaxation as well. The slow process with a kHL(295 K) of 3.7 x 10(5) s-1 and a Ea of 2297(32) cm-1 in methanol--which is the slowest light-induced relaxation process observed so far for an Fe(II) spin-crossover complex in solution--is assigned to a coupling of the 5T2-->1A1 relaxation process to a geometrical rearrangement within the pendent pyridyl arms.  相似文献

The radiolysis of NTA complexes with U(VI), Fe(III) and Ni(II) has been studied spectrophotometrically at different pH values, at different doses of60Co gamma-radiation. The absorbed doses were measured using the Fricke dosimeter. A clear relation between the radiolytic degradation of the complexes and the ligand concentration was observed. A per-O-methylated beta-cyclodextrin dimer, Py2CD, was conveniently prepared via two steps: the Williamson reaction of 3,5-bis(bromomethyl)pyridine and beta-cyclodextrin (beta-CD) yielding 2A,2'A-O-[3,5-pyridinediylbis(methylene)bis-beta-cyclodextrin (bisCD) followed by the O-methylation of all the hydroxy groups of the bisCD. Py2CD formed a very stable 1:1 complex (Fe(III)PCD) with [5,10,15,20-tetrakis(p-sulfonatophenyl)porphinato]iron(III) (Fe(III)TPPS) in aqueous solution. Fe(III)PCD was reduced with Na2S2O4 to afford the Fe (II)TPPS/Py2CD complex (Fe(II)PCD). Dioxygen was bound to Fe(II)PCD, the P(1/2)(O2) values being 42.4 +/- 1.6 and 176 +/- 3 Torr at 3 and 25 degrees C, respectively. The k(on)(O2) and k(off)(O2) values for the dioxygen binding were determined to be 1.3 x 10(7) M(-1) s(-1) and 3.8 x 10(3) s(-1), respectively, at 25 degrees C. Although the dioxygen adduct was not very stable (K(O2) = k(on)(O2)/k(off)(O2) = 3.4 x 10(3) M(-1)), no autoxidation of the dioxygen adduct of Fe(II)PCD to Fe(III)PCD was observed. These results suggest that the encapsulation of Fe (II)TPPS by Py2CD strictly inhibits not only the extrusion of dioxygen from the cyclodextrin cage but also the penetration of a water molecule into the cage. The carbon monoxide affinity of Fe(II)PCD was much higher than the dioxygen affinity; the P(1/2)(CO), k(on)(CO), k(off)(CO), and K(CO) values being (1.6 +/- 0.2) x 10(-2) Torr, 2.4 x 10(6) M(-1) s(-1), 4.8 x 10(-2) s(-1), and 5.0 x 10(7) M(-1), respectively, at 25 degrees C. Fe(II)PCD also bound nitric oxide. The rate of the dissociation of NO from (NO)Fe(II)PCD ((5.58 +/- 0.42) x 10(-5) s(-1)) was in good agreement with the maximum rate ((5.12 +/- 0.18) x 10(-5) s(-1)) of the oxidation of (NO)Fe(II)PCD to Fe(III)PCD and NO3(-), suggesting that the autoxidation of (NO)Fe(II)PCD proceeds through the ligand exchange between NO and O2 followed by the rapid reaction of (O2)Fe(II)PCD with released NO, affording Fe(II)PCD and the NO3(-) anion inside the cyclodextrin cage.  相似文献

The 1:1 inclusion complex of 5,10,15,20-tetrakis(4-sulfonatophenyl)porphinato iron(II) (Fe(II)TPPS) and an O-methylated beta-cyclodextrin dimer having a pyridine linker (1) binds dioxygen reversibly in aqueous solution. The O2 adduct was very stable (t(1/2) = 30.1 h) at pH 7.0 and 25 degrees C. ESI-MS and NMR spectroscopic measurements and molecular mechanics (MM) calculations indicated the inclusion of the sulfonatophenyl groups at the 5- and 15-positions of Fe(III)TPPS or Fe(II)TPPS into two cyclodextrin moieties of 1 to form a supramolecular 1:1 complex (hemoCD1 for the Fe(II)TPPS complex), whose iron center is completely covered by two cyclodextrin moieties. Equilibrium measurements and laser flash photolysis provided the affinities ( and ) and rate constants for O2 and CO binding of hemoCD1 (k(O2)(on), k(O2)(off), k(CO)(on), and k(CO)(off)). The CO affinity relative to the O2 affinity of hemoCD1 was abnormally high. Although resonance Raman spectra suggested weak back-bonding of d(pi)(Fe) --> pi(CO) and hence a weak CO-Fe bond, the CO adduct of hemoCD1 was very stable. The hydrophobic CO molecule dissociated from CO-hemoCD1 hardly breaks free from a shallow cleft in hemoCD1 surrounded by an aqueous bulk phase leading to fast rebinding of CO to hemoCD1. Isothermal titration calorimetry furnished the association constant (K(O2)), DeltaH degrees , and DeltaS degrees for O2 association to be (2.71 +/- 0.51) x 10(4) M(-1), -65.2 +/- 4.4 kJ mol(-1), and -133.9 +/- 16.1 J mol(-1) K(-1), respectively. The autoxidation of oxy-hemoCD1 was accelerated by H+ and OH-. The inorganic anions also accelerated the autoxidation of oxy-hemoCD1. The O2-Fe(II) bond is equivalent to the O2.--Fe(III) bond, which is attacked by the inorganic anions or the water molecule to produce met-hemoCD1 and a superoxide anion.  相似文献

A mixed-valence complex, [Fe(III)Fe(II)L1(μ-OAc)(2)]BF(4)·H(2)O, where the ligand H(2)L1 = 2-{[[3-[((bis(pyridin-2-ylmethyl)amino)methyl)-2-hydroxy-5-methylbenzyl](pyridin-2-ylmethyl)amino]methyl]phenol}, has been studied with a range of techniques, and, where possible, its properties have been compared to those of the corresponding enzyme system purple acid phosphatase. The Fe(III)Fe(II) and Fe(III)(2) oxidized species were studied spectroelectrochemically. The temperature-dependent population of the S = 3/2 spin states of the heterovalent system, observed using magnetic circular dichroism, confirmed that the dinuclear center is weakly antiferromagnetically coupled (H = -2JS(1)·S(2), where J = -5.6 cm(-1)) in a frozen solution. The ligand-to-metal charge-transfer transitions are correlated with density functional theory calculations. The Fe(III)Fe(II) complex is electron paramagnetic resonance (EPR)-silent, except at very low temperatures (<2 K), because of the broadening caused by the exchange coupling and zero-field-splitting parameters being of comparable magnitude and rapid spin-lattice relaxation. However, a phosphate-bound Fe(III)(2) complex showed an EPR spectrum due to population of the S(tot) = 3 state (J= -3.5 cm(-1)). The phosphatase activity of the Fe(III)Fe(II) complex in hydrolysis of bis(2,4-dinitrophenyl)phosphate (k(cat.) = 1.88 × 10(-3) s(-1); K(m) = 4.63 × 10(-3) mol L(-1)) is similar to that of other bimetallic heterovalent complexes with the same ligand. Analysis of the kinetic data supports a mechanism where the initiating nucleophile in the phosphatase reaction is a hydroxide, terminally bound to Fe(III). It is interesting to note that aqueous solutions of [Fe(III)Fe(II)L1(μ-OAc)(2)](+) are also capable of protein cleavage, at mild temperature and pH conditions, thus further expanding the scope of this complex's catalytic promiscuity.  相似文献

Three immobilized Cu(II) complexes were generated by the following: (a) homopolymerization of the N,N',N' '-chelate ligand tris[2-(1-vinylimidazolyl)]phosphine (1) and subsequent metalation with CuCl2; (b) copolymerization of 1 with ethyleneglycol dimethacrylate (EGDMA) und subsequent metalation with CuCl2; or (c) molecular imprinting with the organometallic Mo-complex [Mo(eta3-C4H7)(CO)2(1)](TsO) (5) and EGDMA and subsequent replacement of Mo(II) by Cu(II). All three polymeric Cu complexes were found to efficiently promote the hydrolysis of activated phosphoesters with the relative activity being dependent on the nature of the polymer and the substrate.  相似文献
