Neferine and isoliensinine are the two major compositions of the Chinese medicine lotus plumule. They have extensive cardiovascular activity, such as antiarrhythmic, antithrombic and antihypertensive. In order to confirm the structures reported in the literature by explicit 1H and 13C assignments, we used a series of NMR experiments including 1H, 1H-COSY, HSQC and HMBC. The absolute configuration of neferine (C-1 and C-1') was determined as R and S and that of isoliensinine (C-1 and C-1') was determined as R and R.  相似文献

In the (1)H and (13)C NMR spectra of selenophene-2-carbaldehyde azine, the (1)H-5, (13)C-3 and (13)C-5 signals of the selenophene ring are shifted to higher frequencies, whereas those of the (1)H-1, (13)C-1, (13)C-2 and (13)C-4 are shifted to lower frequencies on going from the EE to ZZ isomer or from the E moiety to the Z moiety of EZ isomer. The (15)N chemical shift is significantly larger in the EE isomer relative to the ZZ isomer and in the E moiety relative to the Z moiety of EZ isomer. A very pronounced difference (60-65 mg/g) between the (77)Se resonance positions is revealed in the studied azine isomers, the (77)Se peak being shifted to higher frequencies in the ZZ isomer and in the Z moiety of EZ isomer. The trends in the changes of the measured chemical shifts are reasonably reproduced by the GIAO calculations at the MP2 level of the (1)H, (13)C, (15)N and (77)Se shielding constants in the energy-favorable conformation with the syn orientation of both selenophene rings relative to the C = N groups. The NBO analysis suggests that such an arrangement of the selenophene rings may take place because of a higher energy of some intramolecular interactions.  相似文献

The synthesis and NMR elucidation of five novel penta-cycloundecane amine derivatives are reported. These compounds are potential antituberculosis agents. The (1)H and (13)C spectra showed major overlapping of methine signals of the cage skeleton making it extremely difficult to elucidate these compounds. The overlapping occurs as a result of the additions made to the carbonyl carbon (C-8/C-11) of the cage. The two-dimensional NMR technique proved to be a useful tool in overcoming this problem. All compounds reported are meso compounds thereby not only simplifying the NMR structure elucidation, but also making it indeed possible.  相似文献

(1)H and (13)C NMR spectroscopic data for 5alpha-androstanes and halo-5alpha-androstanes with different substituents at positions C-3, C-9, C-11 and C-17 were examined and assigned by a combination of 1D and 2D NMR experiments. The substituent effects on the (13)C chemical shifts were compared with those of epi-androsterone, used as a reference compound. The coupling constants (n)J((19)F,(13)C) were measured for compounds 6, 8, 11 and 14.  相似文献

Gladiolin is a macrolide antibiotic isolated from Burkholderia gladioli BCC0238 with promising activity against Mycobacterium tuberculosis, including several multidrug resistant strains. The configuration of all but one of the stereogenic centers of gladiolin has previously been elucidated using a combination of NOESY NMR experiments and predictive sequence analysis of the polyketide synthase responsible for its assembly. However, it was not possible to assign the configuration of the C-35 methyl group using such methods. Here we report the synthesis of C-33/C-35-syn and C-33/C-35-anti mimics of the C-30 to C-38 fragment of gladiolin from (R) and (S)-citronellol, respectively. Comparison of HSQC NMR data for the mimics and the natural product showed that the C-35 methyl is anti to the C-33 hydroxyl group, indicating that gladiolin has the 35S configuration.  相似文献

The 1H and 19F NMR spectra of the α- and β-pyranose anomers of 4-deoxy-4-fluoro-D -glucose (4FG) and 6-deoxy-6-fluoro-D -galactose (6FGA) in methanol-d4, DMSO-d6, acetone-d6 and D2O solution are reported. Computer analysis of the ABMX spectra of the CH CH2F fragments gives accurate vicinal HH and HF coupling constants. An iterative computational analysis of the observed vicinal couplings in this fragment for 6FG, 6FGA and other molecules allows the determination of both the individual rotamer couplings and the rotamer populations. Consideration of the derived rotamer couplings strongly suggests that the correct assignment for the prochiral C-6 methylene protons in 6FG is that with the 6S proton having the larger coupling to H-5. This is the reverse of the assignment of these protons in D -glucose. In contrast, the assignment of these protons in 6FGA follows that given previously for D -galactose. The relative energies for the conformations about the C-5 C-6 bond for 4FG, 6FG and 6FGA are given from the derived rotamer populations. For 6FGA the rotamer in which the fluorine is antiperiplanar to C-4 is particularly favoured. For 4FG the rotamer with OH anti-periplanar to the ring O is highly unfavoured, but the other two rotamers are of almost equal energy. Consideration of the effect of replacing hydroxyl by fluorine in these molecules indicates that any hydrogen bonding involving the C-4 or C-6 hydroxyls plays little part in determining the conformer energies of glucose or galactose in polar solutions.  相似文献

The (1)H and (13)C NMR spectra of 2,4,6,8-tetraaryl-3,7-diazabicyclo[3.3.1]nonan-9-ones (1-2), oximes (3-8) and O-benzyl oximes (9-12) were recorded. The chemical shifts were unambiguously assigned using 1D and 2D NMR spectral data. The results clearly indicate that the compounds exist in chair-boat conformation with equatorial and axial orientation of the aryl groups in the chair and boat forms, respectively. Since the molecules are flexible and dynamic in solution, the chair and boat forms are mutually interconvertible. In 3-12, because of the effect of oximation/oximination, all the protons in the heterobicyclic systems gave distinct signals except the benzylic protons of the boat form. In all synthesized compounds, the aryl group protons at C-6,8 are shielded by the aryl groups at C-2,4 and therefore appear in the lower frequency region than the aryl groups at C-2,4.  相似文献

The replacement of hydroxyl groups by fluorine atoms on hexopyranoside scaffolds may allow access to invaluable tools for studying various biochemical processes. As part of ongoing activities toward the preparation of fluorinated carbohydrates, a systematic investigation involving the synthesis and biological evaluation of a series of mono- and polyfluorinated galactopyranosides is described. Various monofluorogalactopyranosides, a trifluorinated, and a tetrafluorinated galactopyranoside have been prepared using a Chiron approach. Given the scarcity of these compounds in the literature, in addition to their synthesis, their biological profiles were evaluated. Firstly, the fluorinated compounds were investigated as antiproliferative agents using normal human and mouse cells in comparison with cancerous cells. Most of the fluorinated compounds showed no antiproliferative activity. Secondly, these carbohydrate probes were used as potential inhibitors of galactophilic lectins. The first transverse relaxation-optimized spectroscopy (TROSY) NMR experiments were performed on these interactions, examining chemical shift perturbations of the backbone resonances of LecA, a virulence factor from Pseudomonas aeruginosa. Moreover, taking advantage of the fluorine atom, the 19F NMR resonances of the monofluorogalactopyranosides were directly monitored in the presence and absence of LecA to assess ligand binding. Lastly, these results were corroborated with the binding potencies of the monofluorinated galactopyranoside derivatives by isothermal titration calorimetry experiments. Analogues with fluorine atoms at C-3 and C-4 showed weaker affinities with LecA as compared to those with the fluorine atom at C-2 or C-6. This research has focused on the chemical synthesis of “drug-like” low-molecular-weight inhibitors that circumvent drawbacks typically associated with natural oligosaccharides.  相似文献

13C-NMR chemical shifts were measured for C-4 and C-6 in a collection of eight crystalline glucoses and glucosides. The influence of the hydroxymethyl conformation was greater at C-4 than at C-6, with mean chemical shifts for gauche–trans molecules displaced 3.1 ppm (C-4) and 2.5 ppm (C-6) relative to gauche–gauche molecules. This information was used to interpret 13C-NMR spectra of crystalline celluloses. Chemical shifts for C-4 in the crystallite cores of celluloses I and II differed by just 0.2 ppm, but the corresponding chemical shifts for well-ordered crystallite surfaces differed by 3.0 ppm. The separation between crystallite-surface signals was attributed to different hydroxymethyl conformations at the cellulose–water interface, i.e., gauche–gauche and gauche–trans on crystallites of cellulose I and cellulose II, respectively. A broad C-4 signal in the spectrum of cellulose II indicated gauche–gauche conformations in disordered cellulose. Chemical shifts for C-6 were consistent with these conformations.  相似文献

The present paper covers the structure of a new sesterterpene, phyllofenone A(1) , isolated from the South China Sea sponge Phyllospongia foliascens. Phyllofenone A is a member of the scalarane class of sesterterpenes with a (C-20, C-24)-bishomo-(C-25)-nortetra-cyclic skeleton. The structure charcterization was based on one- and two-dimensional NMR and mass spectroscopy with the assistance of molecular modeling. Phyllofenone A shows weak antifungal activity against Candida pseudotropicalis.  相似文献

An unexpected six-membered silacycle was obtained using ring-closing metathesis of diallylsilane 3. The ring size of the compound formed was derived using a 2D 13C-13C inadequate experiment. All proton and carbon-13 resonances were assigned from classical 2D NMR experiments and the proton coupling pattern was clarified. Molecular modeling calculation was then applied using NMR data as constraints, giving the 3D structure of the synthesized compound.  相似文献

Leucanthoside A, a new allose-containing triterpenoid saponin (1), was isolated from the aerial parts of Cephalaria leucantha L. Its structure was determined by electrospray ionization mass spectrometry and NMR spectroscopy. Complete assignments of the 1H and 13C NMR chemical shifts were achieved by two-dimensional NMR experiments: DQF-COSY, NOESY, TOCSY, HSQC, DINE-HSQC, HMBC, 13C-1H 2D-J-resolved spectroscopy, and 1,1-ADEQUATE.  相似文献

(13)C NMR chemical shift assignments of five alpha- and beta- epimeric pairs of cholestanes functionalized at C-3 are presented. Empirical increment estimations proved to be a valuable tool for the unequivocal structural elucidation when compared with the chemical shift values of cholestanes derivatized by introduction of N- and S-containing groups at C-3 in equatorial and axial positions. The feasibility of Carbon-13 Radiofrequency (RF) Amplification by Stimulated Emission of Radiation (C-13 RASER) is demonstrated on a bolus of liquid hyperpolarized ethyl [1-13C]acetate. Hyperpolarized ethyl [1-13C]acetate was prepared via pairwise addition of parahydrogen to vinyl [1-13C]acetate and polarization transfer from nascent parahydrogen-derived protons to the carbon-13 nucleus via magnetic field cycling yielding C-13 nuclear spin polarization of approximately 6 %. RASER signals were detected from samples with concentration ranging from 0.12 to 1 M concentration using a non-cryogenic 1.4T NMR spectrometer equipped with a radio-frequency detection coil with a quality factor (Q) of 32 without any modifications. C-13 RASER signals were observed for several minutes on a single bolus of hyperpolarized substrate to achieve 21 mHz NMR linewidths. The feasibility of creating long-lasting C-13 RASER on biomolecular carriers opens a wide range of new opportunities for the rapidly expanding field of C-13 magnetic resonance hyperpolarization.  相似文献

By employing concerted 1 and 2D NMR techniques, exact NMR spectral assignments have been made of the acyl (2-7) and methyl (8 and 9) derivatives of mangiferin (1) isolated from the leaves of Bombax ceiba. Derivatives 2, 8 and 9 have been reported in literature, while 3-7 represent new compounds. The acetates 2 and 3 were found to be unstable and were converted into the same penta-acetate 4 at room temperature. Extensive NMR studies on mangiferin (1) and its derivatives showed that H-4 exchanges with deuterium of the solvent molecule more easily. This exchange under acidic conditions occurred at that position (C-4) where electrophilic substitution reactions can easily take place. This is the first report describing the exchange of C-4 proton of mangiferin (1), or any other xanthone, with deuterium of solvent molecules.  相似文献
