The time-dependent adsorption behavior of β-Lactoglobulin (β-Lg) on ATR crystal (ZnSe) surface was studied by two-dimensional (2D) correlation ATR/FTIR spectroscopy. More bands were resolved by 2D correlation spectroscopy compared to the results from second derivative (SD) and Fourier self-deconvolution (FSD) analyses, but some of the new bands resolved may originate from bandwidth changes, wavenumber shifts, etc. The integrated/overall sequential order of the intensity changes of the four sub-bands in amide I region obtained from 2D correlation spectroscopy was not consistent with the experimental observation. Adsorption-induced conformational changes did not occur until 10 min of adsorption of β-Lg molecules on the ZnSe crystal surface. The relative contents of the low-wavenumber component of the antiparallel β-strands (1627cm(-1)) and random segments with α-helix (1651cm(-1)) changed prior to β-turns (1666cm(-1)) and the high-wavenumber component of the antiparallel β-strands (1684cm(-1)). More specifically, from about 10 to 15min of adsorption, the loss content of the low-wavenumber component of the antiparallel β-strands (1627cm(-1)) was simultaneously transformed into random segments (1651cm(-1)). After 20 min of adsorption, the content of β-turns (1666cm(-1)) started to decrease, and the loss of β-turns (1666cm(-1)) was also transformed into antiparallel β-strands (high-wavenumber component at 1684cm(-1)) in a cooperative way as the β-Lg molecules become more extended.  相似文献

The adsorption behavior of butyl xanthate on the surface of lead oxide was investigated using continuous online in situ attenuated total reflectance Fourier transform infrared(ATR-FTIR) spectroscopy technique and two dimensional(2D) correlation analysis.The adsorbed layer studied was prepared by coating α-PbO particles onto the surfaces of the ZnSe crystal.The appearance of spectral peaks at 1203 cm-1,1033 cm-1 and their red shift indicated the formation and aggregation of xanthate at the surface of α-PbO.According to 1R intensity changes after rinsing with deionized water and a NaOH solution,the adsorption was proved to be a chemisorption type.The competition between xanthate and OH for the surfaces leads to desorption of xanthate at higher pH.The technique of 2D correlation ATR-FTIR spectroscopy was used to evaluate the changing order of spectral intensities in the adsorption process,and the results indicated that xanthate micelles were formed at the surfaces.The adsorption kinetics of butyl xanthate was found to be a pseudo-second-order reaction model and the adsorption capacity of butyl xanthate at α-PbO was as high as 281 mg g-1 after 150 min.  相似文献

Infrared spectra of hen egg white lysozyme and bovine serum albumin (BSA) adsorbed on a solid poly tris(trimethylsiloxy)silylstyrene (pTSS) surface in D2O solution were measured using attenuated total reflection (ATR) Fourier transform infrared spectroscopy. From the area and shape of the amide I' band of each spectrum, the adsorption amount and the secondary structure were determined simultaneously, as a function of adsorption time. We could show that the average conformation for all the adsorbed lysozyme molecules was solely determined by the adsorption time, and independent of the bulk concentration, while the adsorption amount increased with the bulk concentration as well as the adsorption time. These results suggest that lysozyme molecules form discrete assemblies on the surface, and that the surface assemblies grow over several hours to have a definite architecture independent of the adsorption amount. As for BSA, the extent of the conformational change was solely determined by the adsorption amount, regardless of the bulk concentration and the adsorption time. These differences in the adsorption properties of lysozyme and BSA may reflect differences in their conformational stabilities.  相似文献

The conformation of antifreeze glycoprotein (AFGP) molecules adsorbed at the ice/water interface was studied by attenuated total reflection (ATR)-FTIR spectroscopy. Measurements were carried out for AFGP/D2O solution films formed on the surface of an ATR prism as a function of temperature. Using the FTIR spectrum from the O-D stretching band of D2O molecules, we monitored the supercooled and frozen states of the film and measured the thickness of the quasi-liquid layer (QLL) at the ice/prism interfaces. The AFGP structure was determined for the liquid, supercooled, and frozen states of the solution film using the amide I band spectra. No noticeable differences in conformation were observed in the solution conformation from room temperature down to the 15 K supercooling studied, whereas the alpha-helical content of AFGP suddenly increased when the supercooled solution film froze at -15 degrees C. This change in conformation can increase the overall interaction between the AFGP molecules and ice surface and allow a stronger adsorption. In contrast, the alpha-helical content of AFGP in the frozen film gradually decreased with increasing temperature and finally returned to its solution-state level at the melting point of D2O ice. This gradual decrease in the alpha-helix content directly correlates with the measured increase in QLL thickness. Finally, we conclude that the differences in the alpha-helix signals between the frozen and supercooled states indicate the conformational change of AFGP molecules upon adsorption at the ice/water interface, emphasizing the importance of the structure-function relationship, even for this highly flexible antifreeze.  相似文献

Conformational analyses of PRP1, a proline-rich acidic salivary protein and major component of the acquired enamel pellicle, have been carried out in solution and upon binding to two enamel prototypes, hydroxyapatite (HA) and carbonated hydroxyapatite (CHA), using Fourier transform infrared spectroscopy (FTIR) in attenuated total reflection (ATR) mode. We have shown for the first time that, in solution, large portions of PRP1 adopt the hydrated polyproline type II (PPII) helical structure in addition to the random coil structure, with the maximum absorbance of the amide I band around 1620 cm(-1). Upon binding to HA or CHA, the protein undergoes significant conformational changes, loosing a considerable portion of hydrated PPII and random coil domains with a shift in the maximum absorbance to 1666 cm(-1), indicating that a large fraction of the protein is composed of beta turns. A small fraction of PPII in a calcium-bound or anhydrous form (approximately 1642 cm(-1)) was also observed in the HA- and CHA-bound proteins, which could play a role in protein-mineral interactions. The conformational changes in PRP1 adsorbed on CHA and HA were similar in nature; however, these changes were greater in the protein bound to HA. Interestingly, these results are in agreement with protein adsorption data that show that less protein is adsorbed onto CHA than onto HA. Our results demonstrate that binding to apatitic mineral surfaces leads to major conformational changes in PRP1, which might reflect the expulsion of water and the formation of protein-mineral and/or protein-protein interactions in the adsorbed layer.  相似文献

Stabilization of arsenic contaminated soils by iron oxides has been proposed as a remediation technique to prevent leaching of arsenate into the environment. Fundamental studies are needed to establish under which conditions the complexes formed are stable. In the present work, a powerful technique, viz. ATR-FTIR spectroscopy, is adapted to the studies of adsorption of arsenate species on iron oxides. This technique facilitates acquisition of both quantitative and qualitative in situ adsorption data.In the present work, about 800 nm thick films of 6-lineferrihydrite were deposited on ZnSe ATR crystals. Arsenate adsorption on the ferrihydrite film was studied at pD values ranging from 4 to 12 and at an arsenate concentration of 0.03 mM in D2O solution. The amount of adsorbed arsenate decreased with increasing pD as a result of the more negatively charged iron oxide surface at higher pD values. The adsorption and desorption kinetics were also studied. Arsenate showed a higher adsorption rate within the first 70 min and a much lower adsorption rate from 70 to 300 min. The low adsorption rate at longer reaction times was partly due to a low desorption rate of already adsorbed carbonate species adsorbed at the surface. The desorption of carbonate species was evidenced by the appearance of negative absorption bands. The desorption of adsorbed arsenate complexes was examined by flushing with D2O at pD 4 and 8.5 and it was found that the complexes were very stable at pD 4 suggesting formation of mostly inner-sphere complexes whereas a fraction of the complexes at pD 8.5 were less stable than at pD 4, possibly due to the formation of outer-sphere complexes.In summary, the ATR technique was shown to provide in situ information about the adsorption rate, desorption rate and the speciation of the complexes formed within a single experiment, which is very difficult to obtain using other techniques.  相似文献

The adsorption of two dextrin-based polymers, a regular wheat dextrin (TY) and a carboxymethyl-substituted (CM) dextrin, onto an anatase TiO(2) particle film has been studied using in situ attenuated total reflection (ATR) FTIR spectroscopy. Infrared spectra of the polymer solutions and the polymer adsorbed at the anatase surface were acquired for two solution conditions: pH 3 and pH 9; below and above the isoelectric point (IEP) of anatase, respectively. Comparison of the polymer solution spectra and the adsorbed layer spectra highlighted a number of spectral differences that were attributed to involvement of the carboxyl group of CM Dextrin interacting with the anatase surface directly and the adsorption of oxidized dextrin chains in the case of regular dextrin (TY) at high pH. The adsorption/desorption kinetics were determined by monitoring spectral peaks of the pyranose ring of both polymers. Adsorption equilibrium was not established for Dextrin TY for many hours, whereas CM Dextrin reached equilibrium in its adsorption within 60 min. The extent of desorption of Dextrin TY (observed by flowing a background electrolyte dextrin-free solution) was extensive at both pH values, which reflects the poor affinity and binding of the polymer on anatase. In contrast, CM Dextrin underwent almost no desorption, indicating a high affinity between the carboxyl groups of the polymer and the anatase surface.  相似文献

Poly-l-lysine can form either of three different conformers as α-helix, anti-parallel β-sheet and random coil stably under appropriate conditions. In buffer solution poly-l-lysine exists in a random coil at about pH 4, an α-helix above pH 12, and transforms from α-helix to β-sheet when the sample is heated to 46 °C for 30 min. The effects of elevated hydrostatic pressure on three different initial conformers of poly-l-lysine are investigated with Fourier transform infrared spectroscopy and two-dimensional correlation analysis. Changes observed in the amide I′ band indicate that the α-helix conformer undergo hydration enhancement at low pressure (<400 MPa), then gradually transition into an α′-helix. Two initial conformers, the β-sheet and random coiled polypeptide, undergo conformational changes to an α-helix at low pressure and to an α′-helix at high pressure. Moreover, the conversion occurred at a lower pressure for the β-sheet (∼250 MPa) than for the α-helix (∼300 MPa) and the random coil (∼850 MPa).  相似文献

In-situ attenuated total reflection (ATR)-FTIR spectroscopy was used to monitor the consecutively alternating adsorption of polyethylenimine (PEI) and poly(acrylic acid) (PAC) onto both Si crystals (SiO) and CO2 plasma-treated polypropylene (PP) films. The vibration band vas(COO) and v(CO) of PAC are diagnostic for the polyelectrolyte layer build-up and sensitive to protonation changes. Human serum albumine (HSA) adsorption experiments revealed a strong decrease of fouling for the PP films, which were modified with polyelectrolyte multilayers, in comparison to the unmodified ones.  相似文献

The adsorption of N-acetyl-L-cysteine from ethanol solution on gold has been studied by in situ attenuated total reflection infrared (ATR-IR) spectroscopy, polarization modulation infrared reflection absorption spectroscopy, and a quartz crystal microbalance. After an initial fast adsorption, in situ ATR-IR revealed two considerably slower processes, besides further adsorption. The appearance of carboxylate bands and the partial disappearance of the carboxylic acid bands demonstrated that part of the molecules on the surface underwent deprotonation. In addition, the C=O stretching vibration of the carboxylic acid group shifted to lower and the amide II band to higher wavenumbers, indicating hydrogen-bonding interactions within the adsorbate layer. Based on the initial ATR-IR spectrum, which did not reveal deprotonation, the orientation of the molecule within the adsorbate layer was determined. For this, density functional theory was used to calculate the transition dipole moment vectors of the vibrational modes of N-acetyl-l-cysteine. The projections of the latter onto the z-axis of the fixed surface coordinate system were used to determine relative band intensities for different orientations of the molecule. The analysis revealed that the amide group is tilted with respect to and points away from the surface, whereas the carboxylic acid is in proximity to the surface, which is also supported by a shift of the C-O-H bending mode. This position of the acid group favors its deprotonation assisted by the gold surface and easily enables intermolecular interactions. Periodic acid stimuli revealed reversible protonation/deprotonation of part of the adsorbed molecules. However, only non-hydrogen-bonded carboxylic acid groups showed a response toward the acid stimuli.  相似文献

Rich and complex Raman scattering and Raman optical activity (ROA) spectra have been measured monitoring the pH induced alpha-helix-to-disordered conformational transition in poly(L-glutamic acid). Two-dimensional (2D) correlation techniques have been applied to facilitate a comprehensive analysis of these two complementary spectral sets. Synchronous contour plots have identified band assignments of alpha-helical and disordered conformations, and have revealed bands characteristic of changes in the protonation state of the polypeptide. Asynchronous plots, on the other hand, have probed the relative sequential orders of intensity changes indicating a decrease in intensity of alpha-helical bands in the backbone skeletal stretch region, followed by a subsequent decrease in intensity in the extended amide III and amide I regions, underlying the appearance of disordered structure, including poly(L-proline) II (PPII) helix. The application of a 2D correlation 'moving' window has also disclosed two distinct phases during helix unfolding in the alpha-helix-to-disordered transition, occurring at approximately pH 4.9 and approximately pH 5.2, possibly a result of the difference in helical stability between the end and central regions of the alpha-helix. This paper demonstrates the potential value of combining 2D Raman, 2D ROA and moving window correlation techniques for the detailed investigation of complex and subtle changes of secondary structure during the unfolding mechanisms of polypeptides and proteins.  相似文献

The NMR coupling constants ((3)J(H(N), H(alpha))) of dipeptides indicate that the backbone conformational preferences vary strikingly among dipeptides. These preferences are similar to those of residues in small peptides, denatured proteins, and the coil regions of native proteins. Detailed characterization of the conformational preferences of dipeptides is therefore of fundamental importance for understanding protein structure and folding. Here, we studied the conformational preferences of 13 dipeptides using infrared and Raman spectroscopy. The main advantage of vibrational spectroscopy over NMR spectroscopy is in its much shorter time scale, which enables the determination of the conformational preferences of short-lived states. Accuracy of structure determination using vibrational spectroscopy depends critically on identification of the vibrational parameters that are sensitive to changes in conformation. We show that the frequencies of the amide I band and the A12 ratio of the amide I components of dipeptides correlate with the (3)J(H(N), H(alpha)). These two infrared vibrational parameters are thus analogous to (3)J(H(N), H(alpha)), indicators for the preference for the dihedral angle phi. We also show that the intensities of the components of the amide III bands in infrared spectra and the intensities of the skeletal vibrations in Raman spectra are indicators of populations of the P(II), beta, and alpha(R) conformations. The results show that alanine dipeptide adopts predominantly a PII conformation. The population of the beta conformation increases in valine dipeptides. The populations of the alpha(R) conformation are generally small. These data are in accord with the electrostatic screening model of conformational preferences.  相似文献

Undesired adsorption of proteins brings big troubles to marine structures.The settled proteins change the physical and chemical properties of the surfaces,which allow marine fouling organisms to settle down on the structures.Therefore,to understand the adsorption mechanism of proteins is very helpful to find an environment-friendly solution against biofouling.Many approaches have been developed to study protein adsorption,but most of them are insufficient to give the chemical interaction information between proteins and surfaces.Fourier transform infrared spectroscopy with attenuated total reflection(FTIR-ATR)is an efficient,fast and non-destructive method for in situ surface measurement,which greatly minimizes the interference of water to infra red spectra,because of the very small depth of penetration of the evanescent wave.In this paper,an in situ FTIR-ATR technology was used to investigate the adsorption process of trypsin on a bare ZnSe surface and on a TiO2 coated ZnSe surface,and the effect of calcium cation strength and ultraviolet light irradiation on the secondary structure of trypsin were also evaluated.FTIR spectra of trypsin showed that Amide I band red shift and AmideⅡband blue shift in aqueous environment on both surfaces compared with the dry trypsin powder,and the addition of calcium cations further changed the Amide bands position,which indicated that the change of the secondary structure could be interfered by the environment.The hydrogen bond formation between water and trypsin,the interaction between surface and trypsin,the interaction between hydrated calcium cations and trypsin,are major facto rs to change the secondary structure of trypsin,and UV light irradiation also showed its influence for the secondary structure.  相似文献

采用傅里叶变换红外(FTIR)光谱法和二维相关分析(2D Correlation analysis)技术研究了卵粘蛋白(Ovomucin)的构象转变与温度之间的关系. 结果表明, 当温度为55~65℃时, 卵粘蛋白的红外谱峰的位置和强度发生较大改变. 二维相关分析表明, 在升温过程中, 与肽链相比卵粘蛋白分子中的糖链对温度变化更为敏感, 且优先发生构象改变, 糖链分子的存在利于维持卵粘蛋白构象的热稳定性. 在25~95℃升温过程中, 卵粘蛋白分子二级结构的变化次序依次为α-螺旋、 β-折叠、 β-转角和无规卷曲. 由温度变化引起的卵粘蛋白分子结构动态变化的微观信息, 为揭示变温微扰引起的蛋白构象变化机理提供了初步的理论依据.  相似文献

To study the surface‐adsorbate properties of ZnO nanowires, a hydrothermal method was modified to grow ZnO nanowires directly on ZnSe, which were then characterized by attenuated total reflection infrared (ATR‐IR) spectroscopy. To prepare ZnO nanowires directly on ATR sensing element of ZnSe, ZnO seed layers were first formed by annealing of ZnO seeds on ZnSe surfaces. The ZnO seed layers then were exposed to growth solution, forming ZnO nanowires directly on the ATR crystals. The interaction properties of the resulting surfaces were studied by an ATR‐IR method. The diameter, length and distribution of the ZnO nanowires can be tuned by adjusting the growth conditions, particularly the growing time and the concentrations of reagents. Two surfaces, namely Zn‐rich and Zn‐O ion‐pair surfaces were studied in detail for their adsorption properties toward compounds bearing different functional groups. By examination of several volatile organic compounds (VOCs), it was found that the Zn‐rich surface is less selective and interacts with compounds bearing the functional groups of amino and hydroxyl. The Zn‐O ion‐pair surface is more selective and a much stronger interaction was observed with non‐aromatic amino compounds. These results indicate that the improving of the selectivity of a ZnO‐based sensing device can be achieved by tuning the surface structure of the ZnO nanomaterials.  相似文献

The potential of an optical sensor based on mid-infrared spectroscopy, utilising a zinc selenide (ZnSe) attenuated total reflectance (ATR) element coated with an amorphous Teflon polymer, to determine chlorinated hydrocarbon species (CHC) in an aqueous environment is examined. The polymer coating concentrates the analytes within the penetration depth of the Fourier transform infrared (FTIR) evanescent wave and excludes water from the region. Teflon AF (Amorphous Fluoropolymer) is a family of amorphous copolymers based on polytetrafluoroethylene (PTFE), and is commercially available in two polymeric grades. Teflon AF is highly amorphous in nature with a large 'void volume', exhibits excellent chemical resistance and low water absorption. Such properties identify it as an excellent candidate for enrichment coating on an ATR/FTIR sensor. The potential of both polymeric grades of Teflon AF as enrichment membranes for ATR/FTIR analysis of CHC species was examined and contrasted. A rapid, repeatable, reversible response was observed with both grades to a range of CHC species. Linear responses in the mg/L region, with detection limits in the low mg/L region were achieved with the system used.  相似文献

Temperature‐induced phase separation of poly(N‐isopropylacrylamide) in aqueous solutions was studied by attenuated total reflectance (ATR)/Fourier transform infrared spectroscopy. The main objectives of the study were to understand, on a molecular level, the role of hydrogen bonding and hydrophobic effects below and above the phase‐separation temperature and to derive the scenario leading to this process. Understanding the behavior of this particular system could be quite relevant to many biological phenomena, such as protein denaturation. The temperature‐induced phase transition was easily detected by the ATR method. A sharp increase in the peaks of both hydrophobic and hydrophilic groups of the polymer and a decrease in the water‐related signals could be explained in terms of the formation of a polymer‐enriched film near the ATR crystal. Deconvolution of the amide I and amide II peaks and the O? H stretch envelope of water revealed that the phase‐separation scenario could be divided, below the phase‐separation temperature, into two steps. The first step consisted of the breaking of intermolecular hydrogen bonds between the amide groups of the polymer and the solvent and the formation of free amide groups, and the second step consisted of an increase in intramolecular hydrogen bonding, which induced a coil–globule transition. No changes in the hydrophobic signals below the separation temperature could be observed, suggesting that hydrophobic interactions played a dominant role during the aggregation of the collapsed chains but not before. © 2001 John Wiley & Sons, Inc. A 3D attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy is applied to the in situ monitoring of the growth of breast cancer cell (MCF-7) on a ZnSe surface. The good biocompatibility of the ZnSe crystal results in a tight adhesion of the cells on the surface, with corresponding remarkable IR signals. The series spectrum curves are corresponding to each stage of the cell growth, elucidating the distinguishing phase of the cell cycle. The characteristic absorbance bands of MCF-7 breast cancer cells shift in comparison to the spectra of breast normal cells. However, no changes are observed as compared to the spectra of dead cells with those of the live cells.  相似文献

The interaction between bacterial cells of Pseudomonas fluorescens (ATCC 17552) and gold electrodes was analyzed by cyclic voltammetry (CV) and attenuated total reflection-surface-enhanced infrared absorption spectroscopy (ATR-SEIRAS). The voltammetric evaluation of cell adsorption showed a decrease in the double-layer capacitance of polyoriented single-crystal gold electrodes with cell adhesion. As followed by IR spectroscopy in the ATR configuration, the adsorption of bacterial cells onto thin-film gold electrodes was mainly indicated by the increase in intensity with time of amide I and amide II protein-related bands at 1664 and 1549 cm(-1), respectively. Bands at 1448 and 2900 cm(-1) corresponding to the scissoring and the stretching bands of CH2 were also detected, together with a minor peak at 1407 cm(-1) due to the vs COO- stretching. Weak signals at 1237 cm(-1) were due to amide III, and a broad band between 1100 and 1200 cm(-1) indicated the presence of alcohol groups. Bacteria were found to displace water molecules and anions coadsorbed on the surface in order to interact with the electrode intimately. This fact was evidenced in the SEIRAS spectra by the negative features appearing at 3450 and 3575 cm-1, corresponding to interfacial water directly interacting with the electrode and water associated with chloride ions adsorbed on the electrode, respectively. Experiments in deuterated water confirmed these assignments and allowed a better estimation of amide absorption bands. In CV experiments, an oxidation process was observed at potentials higher than 0.4 V that was dependent on the exposure time of electrodes in concentrated bacterial suspensions. Adsorbed bacterial cells were found to get closer to the gold surface during oxidation, as indicated by the concomitant increment in the main IR bacterial signals including amide I, a sharp band at 1240 cm(-1), and a broad one at 1120 cm(-1) related to phosphate groups in the bacterial membranes. It is proposed to be due to the oxidation of lipopolysaccharides on the outermost bacterial surface.  相似文献
