Abstract

A highly hydrophilic, nontoxic, and conductive effect of colloidal gold nanoparticles (GNP) and multi-walled carbon nanotubes (MWCNT) on pyrolytic graphite electrode has been demonstrated. The direct electron transfer of catalase (CAT) was achieved based on the immobilization of MWCNT/CAT-GNP on a pyrolytic graphite electrode by a Nafion film. The immobilized catalase displayed a pair of well-defined and nearly reversible redox peaks in 0.1 M phosphate buffer solution (PBS) (pH 6.98). The dependence of E°′on solution pH indicated that the direct electron transfer reaction of catalase was a single-electron-transfer coupled with single-proton-transfer reaction process. The immobilized catalase maintained its biological activity, showing a surface controlled electrode process with an apparent heterogeneous electron transfer rate constant (k s) of 1.387±0.1 s?1 and charge-transfer coefficient (α) of 0.49, and displayed electrocatalytic activity in the electrocatalytic reduction of hydrogen peroxide. Therefore, the resulting modified electrode can be used as a biosensor for detecting hydrogen peroxide.  相似文献

A sensitive hydrogen peroxide (H2O2) biosensor was developed based on a reduced graphene oxide|carbon ceramic electrode (RGO|CCE) modified with cadmium sulfide‐hemoglobin (CdS‐Hb). The electron transfer kinetics of Hb were promoted due to the synergetic function of RGO and CdS nanoparticles. The transfer coefficient (α) and the heterogeneous electron transfer rate constant (ks) were calculated to be 0.54 and 2.6 s?1, respectively, indicating a great facilitation achieved in the electron transfer between Hb and the electrode surface. The biosensor showed a good linear response to the reduction of H2O2 over the concentration range of 2–240 µM with a detection limit of 0.24 µM (S/N=3) and a sensitivity of 1.056 µA µM?1 cm?2. The high surface coverage of the CdS‐Hb modified RGO|CCE (1.04×10?8 mol cm?2) and a smaller value of the apparent Michaelis? Menten constant (0.24 mM) confirmed excellent loading of Hb and high affinity of the biosensor for hydrogen peroxide.  相似文献

We report on the redox behaviour of the microperoxidase‐11 (MP‐11) which has been electrostatically immobilized in a matrix of chitosan‐embedded gold nanoparticles on the surface of a glassy carbon electrode. MP‐11 contains a covalently bound heme c as the redox active group that exchanges electrons with the electrode via the gold nanoparticles. Electroactive surface concentration of MP‐11 at high scan rate is between 350±50 pmol cm?2, which reflects a multilayer process. The formal potential (E°′) of MP‐11 in the gold nanoparticles‐chitosan film was estimated to be ?(267.7±2.9) mV at pH 7.0. The heterogeneous electron transfer rate constant (ks) starts at 1.21 s?1 and levels off at 6.45 s?1 in the scan rate range from 0.1 to 2.0 V s?1. Oxidation and reduction of MP‐11 by hydrogen peroxide and superoxide, respectively have been coupled to the direct electron transfer of MP‐11.  相似文献

Stable magnetic nanocomposite of gold nanoparticles (Au‐NPs) decorating Fe3O4 core was successfully synthesized by the linker of Boc‐L‐cysteine. Transmission electron microscope (TEM), energy dispersive X‐ray spectroscopy (EDX) and cyclic voltammograms (CV) were performed to characterize the as‐prepared Fe3O4@Au‐Nps. The results indicated that Au‐Nps dispersed homogeneously around Fe3O4 with the ratio of Au to Fe3O4 nanoparticles as 5–10/1 and the apparent electrochemical area as 0.121 cm2. After self‐assembly of hemoglobin (Hb) on Fe3O4@Au‐Nps by electrostatic interaction, a hydrogen peroxide biosensor was developed. The Fe3O4@Au‐Nps/Hb modified GCE exhibited fast direct electron transfer between heme center and electrode surface with the heterogeneous electron transfer rate constant (Ks ) of 3.35 s−1. Importantly, it showed excellent electrocatalytic activity towards hydrogen peroxide reduction with low detection limit of 0.133 μM (S /D =3) and high sensitivity of 0.163 μA μM−1, respectively. At the concentration evaluated, the interfering species of glucose, dopamine, uric acid and ascorbic acid did not affect the determination of hydrogen peroxide. These results demonstrated that the introduction of Au‐Nps on Fe3O4 not only stabilized the immobilized enzyme but also provided large surface area, fast electron transfer and excellent biocompatibility. This facile nanoassembly protocol can be extended to immobilize various enzymes, proteins and biomolecules to develop robust biosensors.  相似文献

The electrochemical behavior of cytochrome c (cyt‐c) that was electrostatically immobilized onto a self‐assembled monolayer (SAM) of captopril (capt) on a gold electrode has been investigated. Cyclic voltammetry, scanning electrochemical microscopy (SECM) and electrochemical impedance spectroscopy were employed to evaluate the blocking property of the capt SAM. SECM was used to measure the bimolecular electron transfer (ET) kinetics (kBI) between a solution‐based redox probe and the immobilized protein. In addition, the tunneling ET between the immobilized protein and the underlying gold electrode was calculated. A kBI value of (5.0±0.6)×108 mol?1 cm3 s?1 for the bimolecular ET and a standard tunneling rate constant (k0) of 46.4±0.2 s?1 for the tunneling ET have been obtained.  相似文献

A novel biopolymer/room‐temperature ionic liquid composite film based on carrageenan, room temperature ionic liquid (IL) [1‐butyl‐3‐methylimidazolium tetra?uoroborate ([BMIM]BF4)] was explored for immobilization of hemoglobin (Hb) and construction of biosensor. Direct electrochemistry and electrocatalytic behaviors of Hb entrapped in the IL‐carrageenan composite ?lm on the surface of glassy carbon electrode (GCE) were investigated. UV‐vis spectroscopy demonstrated that Hb in the IL‐carrageenan composite ?lm could retain its native secondary structure. A pair of well‐de?ned redox peaks of Hb was obtained at the Hb‐IL‐carrageenan composite ?lm modi?ed electrode through direct electron transfer between the protein and the underlying electrode. The heterogeneous electron transfer rate constant (ks) was 2.02 s?1, indicating great facilitation of the electron transfer between Hb and IL‐carrageenan composite film modi?ed electrode. The modi?ed electrode showed excellent electrocatalytic activity toward reduction of hydrogen peroxide with a linear range of 5.0×10?6 to 1.5×10?4 mol/L and the detection limit was 2.12×10?7 mol/L (S/N=3). The apparent Michaelis‐Menten constant KMapp for hydrogen peroxide was estimated to be 0.02 mmol/L, indicating that the biosensor possessed high af?nity to hydrogen peroxide. The direct electron transfer between immobilized myoglobin (Mb) and colloidal gold modified carbon paste electrode was studied. The Mb immobilized on the colloidal gold nanoparticles displayed a pair of redox peaks in 0.1 M pH 7.0 PBS with a formal potential of –(0.108 ± 0.002) V (vs. NHE). The response showed a surface‐controlled electrode process with an electron transfer rate constant of (26.7 ± 3.7) s ?1 at scan rates from 10 to 100 mV s?1 and a diffusion‐controlled process involving the diffusion of proton at scan rates more than 100 mV s?1. The immobilized Mb maintained its activity and could electrocatalyze the reduction of both hydrogen peroxide and nitrite. Thus, the novel renewable reagentless sensors for hydrogen peroxide and nitrite were developed, respectively. The activity of Mb with respect to the pseudo peroxidase with a KMapp value of 0.65 mM could respond linearly to hydrogen peroxide concentration from 4.6 to 28 μM. The sensor exhibited a fast amperometric response to NO2? reduction and reached 93% of steady‐state current within 5 s. The linear range for NO2? determination was from 8.0 to 112 μM with a detection limit of 0.7 μM at 3σ.  相似文献

A new composite electrode has been fabricated based on coating multi‐walled carbon nanotubes (MWCNTs) and n‐octylpyridinum hexafluorophosphate (OPPF6) ionic liquid composite on a glassy carbon (GC) electrode (OPPF6‐MWCNTs/GCE). This electrode shows very attractive electrochemical performances for electrooxidation of risperidone (RIS) compared to conventional electrodes using carbon and mineral oil, notably improved sensitivity and stability. The oxidation peak potentials in cyclic voltammogram of RIS on the OPPF6‐MWCNTs/GCE was occurred around 230 mV vs. SCE at Britton–Robinson (B–R) buffer (pH 4.0) at scan rate of 100 mV s?1. The electrochemical parameters such as diffusion coefficient (D), charge transfer coefficient (α) and the electron transfer rate constant (k/s) were determined using cyclic voltammetry. Under the optimized conditions, the peak current was linear to risperidone concentration over the concentration range of 10–200 nM with sensitivity of 0.016 μA/nM?1 using differential pulse voltammetry. The detection limit was 6.54 nM (S/N = 3). The electrode also displayed good selectivity and repeatability. In the presence of clozapine (CLZ) the response of RIS kept almost unchanged. Thus this electrode could find application in the determination of RIS in some real samples. The analytical performance of the OPPF6‐MWCNTs/GCE was demonstrated for the determination of RIS in human serum and pharmaceutical samples.  相似文献

Direct electron transfer of immobilized copper, zinc‐superoxide dismutase (SOD) onto electrodeposited nickel‐oxide (NiOx) nanoparticle modified glassy carbon (GC) electrode displays a well defined redox process with formal potential of ?0.03 V in pH 7.4. Cyclic voltammetry was used for deposition of (NiOx) nanoparticles and immobilization of SOD onto GC electrode. The surface coverage (Γ) and heterogeneous electron transfer rate constant (ks) of immobilized SOD are 1.75×10?11 mol cm?2 and 7.5±0.5 s?1, respectively. The biosensor shows a fast amperometric response (3 s) toward superoxide at a wide concentration range from 10 µM to 0.25 mM with sensitivity of 13.40 nA µM?1 cm?2 and 12.40 nA µM?1 cm?2, detection limit of 2.66 and 3.1 µM based on anodically and cathodically detection. The electrode mechanism of Mo(VI) reduction was studied under catalytic adsorptive stripping mode by means of square‐wave voltammetry (SWV). Mo(VI) creates a stable surface active complex with mandelic acid. The electrode reaction of Mo(VI)‐mandelic acid system undergoes as one‐electron reduction, exhibiting properties of a surface electrode process. In the presence of chlorate, bromate, and hydrogen peroxide, the electrode reaction is transposed into a catalytic mechanism. The experimental results are compared with the recent theory for surface catalytic reaction, enabling qualitative characterization of the electrode mechanism in the presence of different catalytic agents. Utilizing both the method of “split SW peaks” and “quasireversible maximum” the standard redox rate constant of Mo(VI)‐mandelic acid system was estimates as ks=150±5 s?1. By fitting the experimental and theoretical results, the following catalytic rate constants have been estimated: (8.0±0.5)×104 mol?1 dm3 s?1, (1.0±0.1)×105 mol?1 dm3 s?1, and (3.2±0.1)×106 mol?1 dm3 s?1, for hydrogen peroxide, chlorate, and bromate, respectively.  相似文献

The present study was aimed at investigating the use of a mixture multiwall carbon nanotube (MWCNT) and thionine (Th) dye in designing of a thionine‐based electrochemical biosensor containing catalase (Ct) enzyme (MWCNT‐Nafion‐Th/Ct) onto a glassy carbon electrode (GCE). The effects of pH, MWCNT concentration and thionine concentration on electrochemical response were explored for optimum analytical performance. The modified electrode exhibited a pair of well‐defined, quasi‐reversible peaks at formal potential (Eo′) = ‐0.218 ± 0.017 V vs. Ag/AgCl corresponding to the Thox/Thred redox couples in the presence of MWCNT, Nafion, and Ct. The electrochemical parameters, including charge‐transfer coefficient (0.36), and apparent heterogeneous electron transfer rate constant (4.28 ± 0.26 s?1) were determined. Using differential pulse voltammetry, the prepared enzyme electrode exhibited a linear response to hydrogen peroxide (H2O2) in the range of 10.0‐100.0 μM with a detection limit 8.7 μM and a sensitivity of 6051.0 μA mM?1 cm?2.  相似文献

A new electrochemical method was proposed for the determination of adenosine‐5′‐triphosphate (ATP) based on the electrooxidation at a molecular wire (MW) modified carbon paste electrode (CPE), which was fabricated with diphenylacetylene (DPA) as the binder. A single well‐defined irreversible oxidation peak of ATP appeared on MW‐CPE with adsorption‐controlled process and enhanced electrochemical response in a pH 3.0 Britton‐Robinson buffer solution, which was due to the presence of high conductive DPA in the electrode. The electrochemical parameters of ATP were calculated with the electron transfer coefficient (α) as 0.54, the electron transfer number (n) as 1.9, the apparent heterogeneous electron transfer rate constant (ks) as 2.67 × 10?5 s?1 and the surface coverage (ΓT) as 4.15 × 10?10 mol cm?2. Under the selected conditions the oxidation peak current was proportional to ATP concentration in the range from 1.0 × 10?7 mol L?1 to 2.0 × 10?3 mol L?1 with the detection limit as 1.28 × 10?8 mol L?1 (3σ) by sensitive differential pulse voltammetry. The proposed method showed good selectivity without the interferences of coexisting substances and was successful applied to the ATP injection samples detection.  相似文献

In this paper a molecular wire modified carbon paste electrode (MW‐CPE) was firstly prepared by mixing graphite powder with diphenylacetylene (DPA). Then a graphene (GR) and chitosan (CTS) composite film was further modified on the surface of MW‐CPE to receive the graphene functionalized electrode (CTS‐GR/MW‐CPE), which was used for the sensitive electrochemical detection of adenosine‐5′‐triphosphate (ATP). The CTS‐GR/MW‐CPE exhibited excellent electrochemical performance and the electrochemical behavior of ATP on the CTS‐GR/MW‐CPE was carefully studied by cyclic voltammetry with an irreversible oxidation peak appearing at 1.369 V (vs. SCE). The electrochemical parameters such as charge transfer coefficient (α) and electrode reaction standard rate constant (ks) were calculated with the results of 0.53 and 5.28×10?6 s?1, respectively. By using differential pulse voltammetry (DPV) as detection technique, the oxidation peak current showed good linear relationship with ATP concentration in the range from 1.0 nM to 700.0 µM with a detection limit of 0.342 nM (3σ). The common coexisting substances, such as uric acid, ascorbic acid and guanosine‐5′‐triphosphate (GTP), showed no interferences and the modified electrode was successfully applied to injection sample detection.  相似文献

An amperometric biosensor for hydrogen peroxide (H2O2) has been constructed by immobilizing cytochrome c on an indium/tin oxide (ITO) electrode modified with a macroporous material. Cyclic voltammetry showed that the direct and quasi-reversible electron transfer of cytochrome c proceeds without the need for an electron mediator. A surface-controlled electron transfer process can be observed with an apparent heterogeneous electron-transfer rate constant (ks) of 29.2?s?1. The biosensor displays excellent electrocatalytic responses to the reduction of H2O2 to give amperometric responses that increase steadily with the concentration of H2O2 in the range from 5???M to 2?mM. The detection limit is 0.61???M at pH?7.4. The apparent Michaelis-Menten constant (Km) of the biosensor is 1.06?mM. This investigation not only provided a method for the direct electron transfer of cytochrome c on macroporous materials, but also established a feasible approach for durable and reliable detection of H2O2.

The reduction process of molybdenum in the presence of fulvic acids and phenanthroline was investigated by square-wave voltammetry (SWV). The mixed-ligand complex of molybdenum exhibits a pronounced tendency to adsorb onto the mercury electrode surface. The electrode reaction proceeds as a surface process in which both components of the redox couple are firmly confined to the electrode surface. The kinetics of the electrode reaction is studied utilizing the properties of “split SW peaks” and “quasireversible maximum”. The kinetic parameters obtained with two different square-wave voltammetric methods are in good agreement. In 0.5 mol/L NaCl solution with pH 2.5 the kinetic parameters are: standard rate constant ks=8±2 s−1, cathodic electron transfer coefficient α=0.41±0.05, and number of exchanged electrons n=2. The SW kinetic measurements are confirmed by cyclic voltammetric method.  相似文献

The electrochemical behavior of oxadiargyl at a graphene‐paste electrode modified with an azo dye, 2‐(4‐((4‐acetylphenyl)diazenyl)phenylamino)ethanol (ADPE), ADPE/MGRPE was investigated. The modified electrode showed high electrocatalytic activity toward oxadiargyl. The apparent electron transfer rate constant (ks) and charge transfer coefficient (α) between electrode and ADPE were 1.16 s?1 and 0.41, respectively. The differential pulse voltammetry response of the modified graphene‐paste electrode was linear against the concentration of oxadiargyl in the range from 0.03 to 1.4 mg L?1. The limit of detection was found to be 1.3 µg L?1 (S/N=3). The practical analytical utility of this electrode was demonstrated by measurement of oxadiargyl in river water, soil and rice samples.  相似文献

利用瞬态吸收光谱技术研究了不同条件下C6H5Cl与H2O2水溶液的激光闪光光解情况, 初步考察了其瞬态物种的生长和衰减等行为. 研究表明, •OH自由基和C6H5Cl反应生成C6H5Cl-OH adduct, 其反应速率常数在近中性、酸性条件下约为(5.89±0.65)×109和(7.07±0.61)×109 L•mol-1•s-1; 其衰减则符合双分子二级反应, 速率常数2k/εl=1.1×106 s-1, 而在碱性时则为(4.34±0.51)×109 L•mol-1•s-1, 衰减呈准一级反应, 速率常数为2.11×105 s-1. 在有氧条件下, O2与C6H5Cl-OH adduct反应生成C6H5Cl-OHO2 adduct, 其反应速率常数为6.8×108 L•mol-1•s-1.  相似文献

The direct electron transfer and electrocatalysis of hemoglobin (Hb) entrapped in polyvinyl alcohol (PVA)–room temperature ionic liquid (i.e., 1-octyl-3-methylimidazolium hexafluorophosphate [OMIM]PF6) composition has been investigated by using cyclic voltammetry and chronocoulometry. It is found that the composition can promote the direct electron transfer of Hb and the heterogeneous electron transfer rate constant (ks) of immobilized Hb is enhanced to 19.9 s−1. The immobilized Hb also shows high electro-catalytic activity towards the redox of oxygen, hydrogen peroxide and nitrite. The Michaelis constants (Km) decrease to 1.2 × 10−4 M (for hydrogen peroxide) and 9.4 × 10−3 M (for nitrite). The surface concentration of electroactive Hb is estimated and it is ca. 1.4 × 10−10 mol cm−2, meaning that several layers of immobilized Hb take part in the electrochemical reaction. When gold nanoparticles (GNP) is introduced into the composition, the resulting PVA–GNP–[OMIM]PF6 composition presents better performance. The electrochemical characteristic of immobilized Hb is improved further. Thus PVA–GNP–[OMIM]PF6 composition is more suitable for the immobilization of Hb. Therefore, it is a good strategy to prepare novel composition for protein immobilization by using several materials with different function.  相似文献

An ionic liquid (IL) 1‐(3‐chloro‐2‐hydroxy‐propyl)‐3‐methylimidazolium trifluoroacetate was used as the modifier for the preparation of the modified carbon paste electrode (CPE). The IL‐CPE showed excellent electrocatalytic activity towards the oxidation of guanosine‐5′‐triphosphate (5′‐GTP) in a pH 5.0 Britton‐Robinson buffer solution. Due to the presence of high conductive IL on the electrode surface, the electrooxidation of 5′‐GTP was greatly promoted with a single well‐defined irreversible oxidation peak appeared. The electrode reaction was an adsorption‐controlled process and the electrochemical parameters of 5′‐GTP on IL‐CPE were calculated with the electron transfer coefficient (α) as 0.44, the electron transfer number (n) as 1.99, the apparent heterogeneous electron transfer rate constant (ks) as 2.21 × 10?9 s?1 and the surface coverage (ΓT) as 1.53 × 10?10 mol cm?2. Under the selected conditions a linear calibration curve between the oxidation peak currents and 5′‐GTP concentration was obtained in the range from 2.0 to 1000.0 μmol L?1 with the detection limit as 0.049 μmol L?1 (3σ) by differential pulse voltammetry. The proposed method showed good selectivity to the 5‘‐GTP detection without the interferences of coexisting substances and the practical application was checked by measurements of the artificial samples.  相似文献

This work demonstrated for the first time that myoglobin cross‐linked in polylysine films is electrochemically active at 6 °C. At 6 °C, these protein films exhibited reversible reduction/oxidation peaks which are characteristic of FeIII/FeII redox couple. The estimated current function densities (J=1.6×10?4 C/V cm2), surface concentrations (ΓT=0.10 nmol/cm2) and standard electron transfer constant (ks=13.86 s?1) at 6 °C for the data taken at a scan rate of 0.1 V/s were similar to those which were obtained at 10, 15 and 23 °C. Basically, this study shows a possible electrocatalytic application of these myoglobin/polylysine films, for example in low temperature sensing applications.  相似文献
