Electron‐Deficient Alkynes as Cleavable Reagents for the Modification of Cysteine‐Containing Peptides in Aqueous Medium

Enriched cleavage : An efficient chemoselective cysteine modification of unprotected peptides and proteins has been developed by using electron‐deficient alkynes in aqueous media. Interestingly, terminal alkynone‐modified peptides could be converted back into the unmodified peptides by cleavage by adding thiols under mild conditions (see scheme).

     

Stabilization of an α Helix by β‐Sheet‐Mediated Self‐Assembly of a Macrocyclic Peptide

Protein roll call : Peptide‐based building blocks, in which both an α‐helix‐forming segment and a β‐sheet segment are located within a single macrocyclic structure, self‐assemble into α‐helix‐decorated artificial proteins. This approach provides a starting point for developing artificial proteins that can modulate α‐helix‐mediated interactions occurring in a multivalent fashion.

     

Direct Catalytic Asymmetric Synthesis of Highly Functionalized 2‐Methylchroman‐2,4‐diols via Barbas–List Aldol Reaction

A practical and sustainable chemical process for the synthesis of highly substituted aldol?lactol products was achieved for the first time through the asymmetric Barbas–List aldol (BLA) reaction of 2‐hydroxybenzaldehydes with acetone in the presence of a catalytic amount of trans‐4‐OH‐L ‐proline (see scheme).

     

Synthesis,Electronic Properties,and Reactivity of Phospholes and 1,1′‐Biphospholes Bearing 2‐ or 3‐Thienyl C‐Substituents

PS, I love you! Novel mixed phosphole/thiophene π‐conjugated systems were synthesized and their electronic properties have been studied both experimentally by UV/Vis spectroscopy and electrochemistry and by theoretical calculations. Exploiting the chemistry of both P‐ and S‐heteroles allows the generation of a diverse range of novel ring‐fused benzophosphole–thiophene derivatives.

     

Design,Synthesis, and Biological Evaluation of Artificial Macrosphelides in the Search for New Apoptosis‐Inducing Agents

A drug of two halves : New artificial compounds composed of a macrosphelide core skeleton and an epothilone side chain were designed and synthesized. These compounds were more potent inducers of apoptosis than the parent natural‐type macrosphelides.

     

Enhanced Electron‐Transfer Properties of Cofacial Porphyrin Dimers through π–π Interactions

π–π assisted : Photoinduced electron transfer from cofacial porphyrin dimers to electron acceptors is prominently accelerated, whereas the back electron transfer is decelerated, relative to the corresponding porphyrin monomer (see figure).

     

A C‐linked Glycomimetic in the Gas Phase and in Solution: Synthesis and Conformation of the Disaccharide Manα(1,6)‐C‐ManαOPh

The effect of carbon is subtle but sweet : The flexible C‐linkage in the newly synthesised C‐glycosyl mimetic, Manα(1,6)‐C‐ManαOPh allows OH? π bonding, both in the gas phase and in aqueous solution. This interaction is absent in the O‐linked disaccharide (see figure).

     

Oxidation of Ethers,Alcohols, and Unfunctionalized Hydrocarbons by the Methyltrioxorhenium/H2O2 System: A Computational Study on Catalytic C?H Bond Activation

A concerted mechanism that does not involve an ionic intermediate was revealed by a DFT study on oxidation of ethers, alcohols, and unfunctionalized hydrocarbons by methyltrioxorhenium/H₂O₂. Instead, C? H insertion occurs through hydride transfer and then turns into a hydroxide transfer/rebound in a concerted fashion. Light in Aminocatalysis: The Asymmetric Intermolecular α‐Alkylation of Aldehydes

Following the light: Photoredox catalysis along with aminocatalysis have proved to be the right combination for one of the most challenging asymmetric transformation in organic synthesis: the direct intermolecular α‐alkylation of aldehydes.

     

Investigation of sp2–sp Coupling for Electron‐Enriched Aryl Dihalides under Oxygen‐Free Sonogashira Coupling Reaction Conditions Using a Two‐Chamber Reaction System

Triethylamine hydroiodide crystals were formed during Sonogashira reactions; after complete reaction the solution retains a characteristic light color (see picture). Very sluggish Sonogashira reactions of electron‐enriched aryl diiodides have been carried out in high yield in an oxygen‐free, two‐chamber reaction system. The formation of triethylamine hydroiodide crystals was monitored to determine the completion of reaction.

     

Density Functional Theory Investigation of the Alkyl–Alkyl Negishi Cross‐Coupling Reaction Catalyzed by N‐Heterocyclic Carbene (NHC)–Pd Complexes

Why bigger is better : A “steric wall” created by the N‐(2,6‐diisopropylphenyl) substituent on the bulky NHC ligand IPr (1,3‐bis(2,6‐diisopropylphenyl)imidazol‐2‐ylidene) guides the reactants to and from the Pd center through weak, fleeting (IPr)H–Pd interactions that help the oxidative addition intermediate escape “the anti‐trap”. The alternative “side” approach leads to transmetalation (the rate‐limiting step) for which a novel Pd–Zn interaction was identified.

     

Heteroaromatic Tosylates as Electrophiles in Regioselective Mizoroki–Heck‐Coupling Reactions with Electron‐Rich Olefins

Easy and direct : Regioselective Mizoroki–Heck‐coupling reactions using heteroaromatic tosylates as electrophiles were achieved, thus providing direct and easy access to highly functionalized α‐heteroarylvinyl amides and ethers.

     

Iron(III)‐Catalyzed and Air‐Mediated Tandem Reaction of Aldehydes,Alkynes and Amines: An Efficient Approach to Substituted Quinolines

Economic and practical advantages are offered by the iron(III)‐catalyzed and air‐mediated tandem coupling/hydroarylation/dehydrogenation of simple readily available aldehydes, alkynes, and amines for the synthesis of 2, 4‐disubstituted quinolines (see scheme).

     

Isoprene‐Assisted Radical Coupling of (Co)polymers Prepared by Cobalt‐Mediated Radical Polymerization

Tackling blocks : The isoprene‐assisted radical coupling (I‐ARC) of polymers prepared by cobalt‐mediated radical polymerization (see picture) is the first efficient radical coupling method that is not restricted to short chains. When applied to AB diblock copolymers, I‐ARC constitutes a straightforward approach to the preparation of novel symmetrical ABA triblock copolymers.

     

Synthesis of some novel 2,6‐disubstituted pyridazin‐3‐ones as TMC120 analogues

Different analogues of TMC120 derived from pyridazin‐3(2H)‐one rings were synthesized by coupling of 3,6‐dichloropyridazine with arylacetonitriles, phenols and/or aniline derivative followed by hydrolysis and alkylation with different benzyl bromide derivatives.
