Double or nothing : The title reaction converts a range of aromatic aldehydes and allenylmethyl/allyl silanes into 1,6‐dioxecane derivatives in good to excellent yields (see scheme; Ar=aryl, Tf=triflate, THF=tetrahydrofuran, TMS=trimethylsilyl). In addition, the bisdiene product has been transformed into the corresponding tricyclic compound through a Diels–Alder reaction.

Catalyzed by [CpRu(CH3CN)3]PF6, the hydrative cyclization of dipropargylic sulfone substrates provides an effective way to synthesize highly functionalized substituted 3‐sulfolenes. The amount of water is crucial for the reactivity of this cycloisomerization reaction. The scope and limitations of the Ru‐catalyzed cycloisomerization are discussed. A marked ketone‐directing effect was observed for the first time in ruthenium‐catalyzed cyclizations. A plausible mechanism for the ketone‐directed cycloisomerization is also rationalized. The utility of this method was demonstrated by both sulfur dioxide extrusion of the 3‐sulfolenes to afford 1,3‐dienes and subsequent inter‐ or intramolecular Diels–Alder reactions.  相似文献

Salvinorin A ( 1 ) is natural hallucinogen that binds the human κ‐opioid receptor. A total synthesis has been developed that parlays the stereochemistry of l ‐(+)‐tartaric acid into that of (?)‐ 1 via an unprecedented allylic dithiane intramolecular Diels–Alder reaction to obtain the trans‐decalin scaffold. Tsuji allylation set the C9 quaternary center and a late‐stage stereoselective chiral ligand‐assisted addition of a 3‐titanium furan upon a C12 aldehyde/C17 methyl ester established the furanyl lactone moiety. The tartrate diol was finally converted into the C1,C2 keto‐acetate.  相似文献

The construction of complex polycyclic terpenoid products in an efficient and step‐economical manner using multicomponent and tandem processes is highly valuable. Herein, we report a tandem cyclization sequence that initiates with a multicomponent double Diels–Alder reaction of cross‐conjugated diynones, followed by a Nazarov cyclization to efficiently produce [6‐5‐6] tricyclic products with excellent regio‐ and diastereoselectivity. This methodology generates five new carbon–carbon bonds, three rings, quaternary or vicinal quaternary carbons, and stereogenic centers in a one‐pot reaction.  相似文献

In this study, (5α,7α)‐4,5‐epoxy‐3,6‐dimethoxy‐17‐methyl‐6,14‐ethenomorphinan‐7‐carboxylic acid hydrazide ( 5 ) was synthesized by the condensation of methyl (5α,7α)‐4,5‐epoxy‐3,6‐dimethoxy‐17‐methyl‐6,14‐ethenomorphinan‐7‐carboxylate ( 4 ) with NH2NH2⋅H2O. The (5α,7α)‐4,5‐epoxy‐3,6‐dimethoxy‐17‐methyl‐6,14‐ethenomorphinan‐7‐carboxylic acid 2‐[(arylamino)carbonyl]hydrazides 6a – 6q were prepared by the reaction of 5 with corresponding substituted aryl isocyanates, and the N‐{5‐[(5α,7α)‐4,5‐epoxy‐3,6‐dimethoxy‐17‐methyl‐6,14‐ethenomorphinan‐7‐yl]‐1,3,4‐oxadiazol‐2‐yl}arenamines 7a – 7q were obtained via the cyclization reaction of 6a – 6q in the presence of POCl3. The synthesized compounds have a rigid morphine structure, including the 6,14‐endo‐etheno bridge and the 5‐(arylamino)‐1,3,4‐oxadiazol‐2‐yl residue at C(7) adopting the (S)‐configuration (7α). The structures of the compounds were confirmed by high‐resolution mass spectrometry (HR‐MS) and various spectroscopic methods such as FT‐IR, 1H‐NMR, 13C‐NMR, APT, and 2D‐NMR (HETCOR, COSY, INADEQUATE).  相似文献

A new pentacene‐type silaborin, in which three benzene rings are bridged by silicon and boron atoms, has been synthesized and characterized by using NMR spectroscopy and X‐ray crystallographic analysis. The precursor, 1,4‐bis(dimesitylboryl)‐2,5‐bis(phenylsilyl)benzene ( 4 ), was prepared by stepwise introduction of a silyl group and a boryl group to a benzene ring starting from 1,4‐dibromobenzene. Double cyclization of 4 proceeds by a H‐Mes exchange and a B‐H/C‐H dehydrogenative condensation to afford pentacene‐type silaborin 5 . X‐ray crystal structure analysis reveals that 5 adopts a bent structure rather than a planar one. UV/Vis spectra and DFT calculations for 5 reveal a lowering of the LUMO energy level compared with corresponding anthracene‐type 3 .  相似文献

(Acetoxymethyl)silanes 2 , 7 a – c , and 10 a – c with at least one alkoxy group, of the general formula (AcOCH2)Si(OR)3?n(CH3)n (R: Me, Et, iPr; n=0, 1, 2), were synthesized from the corresponding (chloromethyl)silanes 1 , 6 a – c , and 9 a – c by treatment with potassium acetate under phase‐transfer‐catalysis conditions. These compounds were found to provide 2,2,5,5‐organo‐substituted 1,4‐dioxa‐2,5‐disilacyclohexanes 3 , 8 a – c , and 11 a – c if treated with organotin(IV) catalysts such as dioctyltin oxide. The reaction proceeds through transesterification of the acetoxy and alkoxy units followed by ring‐closure to form a dimeric six‐membered ring. The corresponding alkyl acetates are formed as the reaction by‐products. With these mild conditions, the method overcomes the drawbacks of previously reported synthetic routes to furnish 2,2,5,5‐tetramethyl‐1,4‐dioxa‐2,5‐disilacyclohexane ( 3 ) and even allows the synthesis of 1,4‐dioxa‐2,5‐disilacyclohexanes bearing hydrolytically labile alkoxy substituents at the silicon atom in good yields and high purity. These new materials were fully characterized by NMR spectroscopy, elemental analysis, mass spectrometry, and X‐ray analysis (trans‐ 8 a ).  相似文献

A novel, efficient synthesis of a series of functionalized, benzo‐annelated decahydrofuro[3,2‐h][1,6]naphthyridine derivatives 3 has been achieved. The protocol is based on the intramolecular hetero‐Diels–Alder (IMHDA) reaction of in situ formed imines derived from an N‐prenylated sugar aldehyde 1 and different aromatic amines 2 in the presence of bismuth(III) chloride as catalyst. The reactions could be run under very mild conditions at room temperature, and were complete within 30 min, affording exclusively and stereoselectively the corresponding trans‐fused products 3 in good‐to‐excellent yields (Table).  相似文献

The high‐yielding synthesis of 5‐bromopenta‐2,4‐diynenitrile (BrC5N) was achieved for the first time. Its reactivity with triisopropylsilylacetylene and triisopropylsilylbutadiyne in the presence of copper and palladium as co‐catalysts and diisopropylamine was evaluated. It revealed an unprecedented cascade reaction leading to a diene in one case and to a benzofulvene in the other case, with a unique structure. Both of them were characterized by X‐ray crystallography, among other techniques. The mechanism of the reaction leading to the diene was investigated experimentally. Theoretical calculations at the DFT level suggest that the mechanism leading to the benzofulvene relies on a hexa‐dehydro Diels–Alder (HDDA)‐type of mechanism. This work constitutes an example of an unanticipated reactivity leading to an important increase of chemical complexity.  相似文献

The Photo‐Dehydro‐Diels–Alder (PDDA) reaction is shown to be a versatile method for the preparation of highly functionalized naphthalenes. Thus, ketones 1 could be cyclized to the 1H‐benzo[g]isochromen‐4‐(3H)‐ones 11 and 12 , mostly in good yields. The influence of various substituents on the regioselectivity of the reaction was investigated, and the mechanism is discussed based on theoretical calculations.  相似文献

A highly diastereoselective and practical biomimetic total synthesis of (±)‐basiliolide B has been achieved through the study of the two proposed biosynthetic pathways (O‐methylation and O‐acylation) for the unprecedented 7‐methoxy‐4,5‐dihydro‐3H‐oxepin‐2‐one (C ring). The synthesis featured a cyclopropanation/ring opening strategy for establishing the stereogenic centers at C8 and C9, a biomimetic 2‐pyrone Diels–Alder cycloaddition for the synthesis of the ABD ring system, and finally a highly efficient biomimetic intramolecular O‐acylation for the C ring formation. This result provides an important perspective on the biosynthetic origin of the unprecedented 7‐membered acyl ketene acetal moiety of the C ring.  相似文献
