Five new biflavonoids, 1 – 5 , were isolated from the stem bark of Daphne aurantiaca. The structures were elucidated as 2,2″‐bisteppogenin ( 1 ), 2,2″‐bisteppogenin 7‐Oβ‐glucopyranoside ( 2 ), 2′′′‐dehydroxy‐2,2″‐bisteppogenin ( 3 ), 2′′′‐dehydroxy‐2,2″‐bisteppogenin 7‐Oβ‐glucopyranoside ( 4 ), and 7‐methoxyneochamaejasmin B ( 5 ) on the basis of spectral analyses.  相似文献

There are abundant natural diterpenoids in the plants of the genus Daphne from the Thymelaeaceae family, featuring a 5/7/6-tricyclic ring system and usually with an orthoester group. So far, a total of 135 diterpenoids has been isolated from the species of the genus Daphne, which could be further classified into three main types according to the substitution pattern of ring A and oxygen-containing functions at ring B. A variety of studies have demonstrated that these compounds exert a wide range of bioactivities both in vitro and in vivo including anticancer, anti-inflammatory, anti-HIV, antifertility, neurotrophic, and cholesterol-lowering effects, which is reviewed herein. Meanwhile, the fascinating structure–activity relationship is also concluded in this review in the hope of providing an easy access to available information for the synthesis and optimization of efficient drugs.  相似文献

Two new secolignans and one new neolignan, named feddeiphenols A-C (1-3), together with eight known compounds (4-11), were isolated from the leaves and stems of Daphne feddei. Their structures were established on the base of spectroscopic methods, mainly extensive NMR, UV spectroscopy, and MS spectrometry. Compounds 1-11 were tested for their anti-human immunodeficiency virus (HIV)-1 activity and cytotoxicity. The results revealed that compounds 1, 2, 3, 7, and 9 showed therapeutic index (TI) values above 30, respectively, and the other compounds also showed weak anti-HIV-1 activity. Compound 1 showed modest cytotoxic activity. The other compounds also showed weak cytotoxic activity.  相似文献

Genus Daphne belongs to the Thymelaeaceae family and consists of 70 species. Its various species exist in Europe, Philippine Islands, temperate and subtropical Asia, North Africa, Australia and Pacific. In Pakistan, Daphne is represented by three species. Our focused Daphne oleoides is widely found in diverse climatic conditions from northern cold to central hot regions which creates a rich diversity and novelty in biosynthetic levels of its chemical constituents and hence is a great opportunity. Daphne oeloides is a proven rich source of a variety of unique and interesting nature-made skeletons with a wide range of therapeutic properties. D. oleoides possesses effective therapeutic properties, therefore, has been used in herbal medicines and is still being used to treat various diseases. The modern research by various groups, including ourselves, has resulted in the isolation of a number of natural molecules including some novel tris- and bis- coumarins, daphnane diterpenoids and lignoids. Therefore, due to novelty and richness of the nature-made molecules, and their therapeutic potential combined with our significant work on D. oleoides, this report covers chemical constituents isolated from D. oleoides. The pharmacological activities of the isolated compounds and use of this species in folk medicine have also been reviewed.  相似文献

The phytochemical investigation of the flower buds of Daphne genkwa yielded eight highly oxygenated daphnane-type diterpene esters, including a new one, genkwanine N 20-palmitate (1) and seven known ones (28). The molecular structures of the isolated compounds were elucidated on the basis of extensive spectroscopic analysis, including UV, IR, NMR, and MS, and comparison with literature data. Furthermore, compound 1 showed potential cytotoxic activity in vitro, against A549 and Hep G2 tumour cell lines.  相似文献

Screening for new natural anti-neuroinflammatory compounds was performed with the traditional folk medicine Genkwa Flos, which potently inhibited nitric oxide (NO) production by LPS-activated microglial BV-2 cells. Two new lathyrane-type diterpenes, genkwalathins A (1) and B (2), and 14 known daphnane-type diterpenes (316) were isolated. The lathyrane-type diterpenes were isolated for the first time from the Thymelaeaceae family in this study. Compounds 1 and 2 moderately inhibited LPS-induced NO production in BV-2 cells without affecting cell viability, while six daphnane-type diterpenes (3, 4, 6, 7, 9 and 10) potently reduced NO production with IC50 values less than 1 μM, although they did display weak cytotoxicity. A structure–activity relationship study on the daphnane-type diterpenes indicated that the stereochemistry at C-19, the benzoate group at C-20, and the epoxide moiety could be important for their anti-neuroinflammatory effects.  相似文献

Two new bis‐coumarins, namely daphnogitin ( 1 ), daphnogirin ( 2 ), and a flavan‐3‐ol derivative, 5‐O‐methylafzelechin ( 3 ), together with eleven known compounds, daphnetoxin, 1,2‐dihydrodaphnetoxin, daphnetin, daphnoretin, daphneticin, isodaphneticin, trans‐2,3‐dihydro‐3‐(4‐hydroxy‐3‐methoxyphenyl)‐2‐(hydroxymethyl)‐9H‐pyrano[2,3‐f]‐1,4‐benzodioxin‐9‐one (= demethoxyisodaphneticin; 4b ), (+)‐pinoresinol, acuminatin, (−)‐dihydrosesamin, and isosalicifoline, were isolated from the leaves and stems of Daphne giraldii. Compound 4b was isolated for the first time as a natural product, and 1,2‐dihydrodaphnetoxin, daphneticin, isodaphneticin, (+)‐pinoresinol, acuminatin, (−)‐dihydrosesamin, and isosalicifoline were isolated for the first time from Daphne giraldii. The structures of the three new compounds were elucidated by spectroscopic analysis.  相似文献

Three new phenolic glucosides named orcinosides A, B, and C ( 1, 2 , and 3 , resp.) were isolated in low yields (4.0×10−6, 11.5×10−6, 4.5×10−6%, resp.) from the rhizomes of Curculigo orchioides. Their structures were elucidated by comprehensive spectroscopic analyses including FAB‐MS, HR‐ESI‐MS, IR, and 1D‐ and 2D‐NMR (HSQC, HMBC) data. An ethyl acetate extract from the barks of the ethnic Chinese medicine Daphne tangutica Maxim. exhibited antihepatocellular carcinoma activity against HepG2 and Hep3B cell lines. By using high‐performance liquid chromatography based activity profiling in combination with offline liquid chromatography with mass spectrometry and NMR analysis, we rapidly identified ten major components of the extract, including seven active principles, coumarins ( 1–4 ) and biscoumarins ( 7, 8, 10 ), along with three inactive flavonoids ( 5, 6, 9 ). This study demonstrated that our combined protocol can be used as an important strategy for chemical profiling, dereplication as well as the identification of bioactive compounds from herbal medicines.  相似文献

Three new daphnane‐type diterpenoids, genkwanines M–O ( 1 – 3 , resp.), together with seven known daphnane‐type diterpenoids, genkwanines D and H, genkwaine F, genkwadaphnine, yuanhuatine, yuanhuafine, and yuanhuapine ( 4 – 10 , resp.), were isolated from the flower buds of Daphne genkwa during a phytochemical investigation.
