Biosynthesis and thermal properties of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) with different HV (hydrovalerate) content produced by a Bacillus cereus strain were investigated. A large variety of HV contents (up to about 90 mol%) of PHBV could be produced by this strain. Combined nitrogen sources containing both yeast extract and ammonium sulphate were better for cell growth and polyhydroxyalkanoates (PHA) production than either yeast extract or ammonium sulphate alone. Propionic acid is more favorable for the production of HV content than that of valeric acid. Finally, thermal properties of PHBV produced by this strain are found close to the results of other groups.  相似文献

This study investigates the biosynthesis and characterization of P(3HB-co-3HV-co-3HHx) terpolymer from mixtures of palm kernel oil and 3HV-precursors by using recombinant Cupriavidus necator PHB4/pBBREE32d13. Sodium valerate and propionate have been evaluated for the generation of 3HV monomers. The feeding time of these precursors was a crucial factor that significantly affected the 3HV molar fractions, which ranged from 2 to 60 mol%. Sodium valerate was generally the better precursor in initiating the accumulation of 3HV monomers while maintaining high cell dry weight (7.9 g/L) and good PHA accumulation (79 wt%). However, the 3HHx molar fractions in the terpolymers at 72 h were consistent at about 2-7 mol%. P(3HB-co-3HV) copolymers have been known to exhibit approximately the same degree of crystallinity throughout a wide range of 3HV composition. Interestingly, in this study, terpolymers containing 58 mol% 3HB, 39 mol% 3HV and 3 mol% 3HHx showed elastomeric behavior. This study demonstrates the suitability of palm kernel oil as the main carbon source and both sodium propionate and sodium valerate as 3HV-precursors for the synthesis of novel compositions of P(3HB-co-3HV-co-3HHx) terpolymers with interesting properties.  相似文献

Poly(3-hydroxybutyrate) [P(3HB)] homopolymer and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] copolymer was produced by Comamonas sp. EB172 using single and mixture of carbon sources. Poly(3-hydroxyvalerate) P(3HV) incorporation in the copolymer was obtained when propionic and valeric acid was used as precursors. Incorporation of 3HV fractions in the copolymer varied from 45 to 86 mol% when initial pH of the medium was regulated. In fed-batch cultivation, organic acids derived from anaerobically treated palm oil mill effluent (POME) were shown to be suitable carbon sources for polyhydroxyalkanoate (PHA) production by Comamonas sp. EB172. Number average molecular weight (Mn) produced by the strain was in the range of 153-412 kDa with polydispersity index (Mw/Mn) in the range of 2.2-2.6, respectively. Incorporation of higher 3HV units improved the thermal stability of P(3HB-co-3HV) copolymer. Thus the newly isolated bacterium Comamonas sp. EB172 is a suitable candidate for PHA production using POME as renewable and alternative cheap raw materials.  相似文献

In this study, activated sludge bacteria from a conventional wastewater treatment process were induced to accumulate polyhydroxyalkanoates (PHAs) under different carbon-nitrogen (C:N) ratios. As the C:N ratio increased from 20 to 140, specific polymer yield increased to a maximum of 0.38 g of polymer/g of dry cell mass while specific growth yield decreased. The highest overall polymer production yield of 0.11 g of polymer/g of carbonaceous substrate consumed was achieved using a C:N ratio of 100. Moreover, the composition of polymer accumulated was dependent on the valeric acid content in the feed. Copolymer poly (3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] was produced in the presence of valeric acid. The 3-hydroxyvalerate (3HV) mole fraction in the copolymer was linearly related tovaleric content in the feed, which reached a maximum of 54% when valeric acid was used as sole carbon source. When the 3HV U in the polymer increased from 0–54 mol%, the melting temperature decreased from 178° to 99°C. Thus, the composition, and hence the mechanical properties, of the copolymer produced from activated sludge can be controlled by adjusting the mole fraction of valeric acid in the feed medium.  相似文献

Unlike polyhydroxyalkanoates (PHAs) copolymers, the controlled and efficient synthesis of PHA terpolymers from triglycerides and fatty acids are yet to be established. This study demonstrates the production of P(3HB-co-3HV-co-3HHx) terpolymer with a wide range of 3HV monomer compositions from mixtures of crude palm kernel oil and 3HV precursors using a mutant Cupriavidus necator PHB4 transformant harboring the PHA synthase gene (phaC) of a locally isolated Chromobacterium sp. USM2. The PHA synthase of Chromobacterium has an unusually high affinity towards 3HV monomer. P(3HB-co-3HV-co-3HHx) terpolymers with 3HV monomer composition ranging from 2 to 91 mol% were produced. Generation of 3HHx monomers was affected by the concentration and feeding time of 3HV precursor. P(3HB-co-24 mol% 3HV-co-7 mol% 3HHx) exhibited mechanical properties similar to that of common low-density polyethylene. P(3HB-co-3HV-co-3HHx) terpolymers with a wide range of 3HV molar fraction had been successfully synthesized by adding lower concentrations of 3HV precursors and using a PHA synthase with high affinity towards 3HV monomer.  相似文献

The first objective of this study was the measurement of physical properties of P(3HB-co-3HV) copolymers with different (hydroxybutyrate) HB to (hydroxyvalerate) HV ratios produced by Bacillus cereus (TRY2) isolated from activated sludge. The 3HV PHBV copolymers were 0.05, 22.6, 39.2, 54.1, and 69.1 mol%, respectively. The second objective was to study possible wastewater treatment and production of PHAs at the same time by B. cereus (TRY2) and Pseudomonas spp. (TOB17) (both were isolated from activated sludge), recombinant Bacillus DH5α, and a combination of the above three bacteria. The results were satisfactory; the maximum COD and TOC of the sewage sludge reduced were 53.5% and 67.5%, respectively.  相似文献

Cupriavidus necator is well known for its ability to accumulate polyhydroxybutyrate (PHB). When supplemented with propionic acid (or sodium propionate) in the growth medium, the bacterium is also able to synthesize polyhydroxybutyrate-co-hydroxyvalerate (PHBV). In order to increase the fraction of 3-hydroxyvalerate (3HV) in PHBV, we cloned the propionate permease gene prpP from C. necator and the propionyl-CoA synthase gene prpE from Cupriavidus taiwanensis and transformed into an Escherichia coli containing phaCAB operon of C. necator. The effects on PHBV accumulation in cells co-expressed with phaCAB and prpE or prpP in the media contained mixed carbon sources (glucose and sodium propionate) were evaluated. The HV fraction in PHBV increased when prpE or prpP was overexpressed in the cells. Concentrations of yeast extracts could also affect the fraction of HV. In addition, when glucose was replaced by sodium pyruvate, sodium succinate, or sodium gluconate, only PHB were detected in the recombinant strains.  相似文献

Polyhydroxyalkanoate (PHA) granules with core-shell layered microstructure were synthesized in Ralstonia eutropha using periodic feeding of valeric acid into a growth medium containing excess fructose. The O2 consumption and CO2 evolution rates, determined by off-gas mass spectrometry, have been used as sensitive measures to indicate the type of nutrients utilized by R. eutropha during PHA synthesis. Domains of poly-3-hydroxybutyrate (PHB) were formed during polymer storage conditions when only fructose was present. Feeding of valeric acid (pentanoic acid) resulted in the synthesis of hydroxyvalerate (HV) monomers, forming a poly-3-hydroxybutyrate-co-valerate (PHBV) copolymer. The synthesis of desired polymer microstructures was monitored and controlled using online mass spectrometry (MS). The respiratory quotient (RQ) was unique to the type of polymer being synthesized due to increased O2 consumption during PHBV synthesis. MS data was used as the control signal for nutrient feeding strategies in the bioreactor. The core-shell structures synthesized were verified in cells using transmission electron microscopy after thin sectioning and staining with RuO4. It was demonstrated that the synthesis of core-shell microstructures can be precisely controlled utilizing a MS feedback control system.  相似文献

Vibrio alginolyticus is a halophilic organism usually found in marine environments. It has attracted attention as an opportunistic pathogen of aquatic animals and humans, but there are very few reports on polyhydroxyalkanoate (PHA) production using V. alginolyticus as the host. In this study, two V. alginolyticus strains, LHF01 and LHF02, isolated from water samples collected from salt fields were found to produce poly(3-hydroxybutyrate) (PHB) from a variety of sugars and organic acids. Glycerol was the best carbon source and yielded the highest PHB titer in both strains. Further optimization of the NaCl concentration and culture temperature improved the PHB titer from 1.87 to 5.08 g/L in V. alginolyticus LHF01. In addition, the use of propionate as a secondary carbon source resulted in the production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV). V. alginolyticus LHF01 may be a promising host for PHA production using cheap waste glycerol from biodiesel refining.  相似文献

Streptomyces omiyaensis SSM 5670 was characterized by its ability to use compression moulded samples of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) as its sole carbon source. Biodegradation of PHBV in liquid mineral salts medium was investigated using scanning electron microscopy, gravimetric measurements, capillary viscometry, tensile testing and wide angle X-ray spectroscopy. The biodegradation of PHBV proceeds via surface erosion mechanism, resulting in the formation of pits by microbial attack. PHBV specimens lost about 45% of their original weight after 45 days of exposure. During the degradation process the elastic modulus reduces less than 10%. The formation of pores and microcracks initiated at the degraded pits determines the reduction of the elongation and stress at break. However, the true stress at break is practically independent of the degradation time. No significant changes of PHBV molecular weight or crystallinity were observed during biodegradation. The polymer chain cleavage occurred only at the specimen surface and does not discriminate between crystalline and amorphous states.  相似文献

Ammonium surfactants are commonly used as clay organomodifiers in nanocomposites. Their effect on the thermal- and thermo-mechanical degradation of polyhydroxyalkanoates (PHAs) is reported. Two poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) grades were tested and compared to polyhydroxybutyrate (PHB). Thermal stabilities were determined from thermogravimetric data and determination of molecular weight changes after processing was performed. The data revealed that all surfactants enhance the PHBV degradation. The results also highlight the preponderant effect of the initial Mw rather than the HV content on the thermal stability. The thermo-mechanical study confirmed the role of surfactants and their different behaviour towards PHBV degradation. This study demonstrates that all surfactants enhance the PHA degradation since their decomposition products most likely act as catalytic agents.  相似文献

In this study, the ability of Cupriavidus sp. USMAA2-4 to synthesize polyhydroxyalkanoates (PHA) containing 4-hydroxyvalerate monomer (4HV) was studied through one-stage cultivation using γ-valerolactone as the carbon precursor. The presence of 4HV monomer unit in the polymer was detected through gas chromatography analysis, proving the capability of this wild strain bacterium to produce poly(3-hydrxybutyrate-co-3-hydroxyvalerate-co-4-hydroxyvalerate) [P(3HB-co-3HV-co-4HV)] terpolymer. Existence of a 4HV monomer unit in the PHA produced was further confirmed through 13C and 1H NMR analysis. P(3HB-co-88 % 3HV-co-1 % 4HV) terpolymer with the highest PHA content of 63 wt% was obtained through combination of 0.14 wt% C of γ-valerolactone with 0.42 wt% C of oleic acid. Various compositions of P(3HB-co-3HV-co-4HV) terpolymer with 3HV and 4HV compositions ranging from 11 to 94 mol% and from 1 to 4 mol%, respectively, were acquired by manipulating γ-valerolactone and oleic acid concentrations. The molecular weight and the thermal and mechanical properties of four different compositions of terpolymers—P(3HB-co-91 % 3HV-co-1 % 4HV), P(3HB-co-55 % 3HV-co-2 % 4HV), P(3HB-co-27 % 3HV-co-2 % 4HV), and P(3HB-co-9 % 3HV-co-1 % 4HV)—were characterized. Among these terpolymers, P(3HB-co-27 % 3HV-co-2 % 4HV) terpolymer with a molecular weight of 5.7 (105 Da) exhibited the highest elongation to break (264 %). The monomer unit compositional distributions of these terpolymers were investigated through acetone–water fractionation analysis. The results suggested that these produced terpolymers had broad 3HV compositional distribution and narrow 4HV compositional distribution.  相似文献

Summary : Haloferax mediterranei was investigated for the production of two different high-performance polyhydroxyalkanoates (PHAs). A copolyester containing 6 mol-% 3-hydroxyvalerate (3HV) was produced from whey sugars as sole carbon source. The maximum specific growth rate (µmax.) and the maximum specific PHA production rate (qp max.) were determined with 0.10 1/h and 0.15 1/h, respectively. The cells contained 72.8 wt.-% of P-(3HB-co-6%-3HV) which featured low melting points between 150 and 160 °C and narrow molecular mass distribution (polydispersity PDI = 1.5). Further, a PHA terpolyester with an increased 3HV fraction as well as 4-hydroxybutyrate (4HB) building blocks was accumulated by feeding of whey sugars plus 3HV - and 4HB precursors. Kinetic analysis of the process reveals a µmax. of 0.14 1/h and a qp max. of 0.23 1/h, respectively. The final percentage of P-(3HB-co-21.8%-3HV-co-5.1%-4HB) in biomass amounted to 87.5 wt.-%. Also this material showed a narrow molecular mass distribution (PDI = 1.5) and a high difference between the two melting endotherms of the material (between 140 and 150 °C) and the onset of decomposition at 236 °C. The accomplished work provides viable strategies to obtain different high-quality PHAs which might be potential candidates for application in the medical and pharmaceutical field.  相似文献

Pseudo-dendritic polyethyleniminated poly(oxypropylene)diamine (D400(EI)(20)) was used as a stabilizer and promoter to prepare Pt nanoparticles in aqueous solution, which was then deposited on carbon surface followed by calcination. After being deposited on carbon surface, no Pt(0) could be detected in the solution phase. In all steps, the increasing molar ratio of the amino groups of D400(EI)(20) to H(2)PtCl(6) ([N]/[Pt]) drastically reduced the size and the polydispersity and kept a constant low value after [N]/[Pt] = 20. Under a [N]/[Pt] ratio of 20, the particle sizes obtained from transmission electron microscopy (TEM) were very small in solution (2.7-2.4 nm) and remained the same after being deposited on carbon surface (2.7-2.4 nm), and were only slightly increased to 3.6-3.0 nm after calcination. The stabilizing ability of D400(EI)(20) to Pt on carbon surface before and after calcination can be interpreted by the existence of binding energy between Pt and amine nitrogen. The X-ray diffraction (XRD) pattern together with the TEM image reveals that the obtained Pt nanoparticles exist in single-crystal form. The results of photoelectron spectroscopy (XPS) evidence that the metallic Pt(0) rather than the oxidized Pt is the predominant species in the Pt/C catalysts. The electrochemical active surface (EAS) area of the Pt/C catalyst is only slightly higher than that of the E-TEK Pt/C catalyst, but the utilization factor (93.4%) is remarkably higher than the latter (62.8%). The increasing time of thermal treatment increases the crystallinity of Pt(0) on carbon, accompanied by the increasing EAS areas, which corresponds to its enhanced electrocatalytic performance to methanol oxidation.  相似文献

Poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-4-hydroxybutyrate) [P(3HB-co-3HV-co-4HB)] terpolymer was produced using Cupriavidus sp. USMAA2-4 via one-step cultivation process through combination of various carbon sources such as 1,4-butanediol or γ-butyrolactone with either 1-pentanol, valeric acid, or 1-propanol. Oleic acid was added to increase the biomass production. The composition of 3HV and 4HB monomers were greatly affected by the concentration of 1,4-butanediol and 1-pentanol. Terpolymers with 3HV and 4HB molar fractions ranging from 2 to 41 mol.% and 5 to 31 mol.%, respectively, were produced by varying the concentration of carbon precursors. The thermal and mechanical properties of the terpolymers containing different proportions of the constituent monomers were characterized using gel permeation chromatography (GPC), DSC, and tensile machine. GPC analysis showed that the molecular weights (M w) of the terpolymer produced were within the range of 346 to 1,710 kDa. The monomer compositions of 3HV and 4HB were also found to have great influences on the thermal and mechanical properties of the terpolymer P(3HB-co-3HV-co-4HB) produced.  相似文献

Crystallization behavior of blends of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) and poly(1,2-propandiolcarbonate) (PR(CO2)) has been investigated by polarized light microscopy (PLM). The spherulite growth rates (SGR) of all blends were faster than that of pure PHBV, and the spherulite growth rates of PHBV in the PHBV/PR(CO2) blends reduced with increasing PR(CO2) weight fraction. There are two melting peaks in both the pure PHBV and the PHBV/PR(CO2) blends. The melting peak of PHBV/PR(CO2) blends was reduced by lower temperature about 20K as compared to PHBV and the higher temperature melting peak was increased by about 10K in the blends.  相似文献

We describe the characterization of polyhydroxyalkanoate (PHA)-producing bacteria isolated from an ammunition-polluted soil in Kitakyushu City, Japan. Over 270 strains were evaluated for PHA accumulation based on a colony staining method using Nile red. Of these, nine strains were selected based on the intensity of Nile red fluorescence and the cells were quantitatively analyzed for PHA by gas chromatography. PHA accumulation was observed in five strains, all of which are inferred to be close to the Bacillus cereus group according to 16S rDNA sequence analysis. Interestingly, these strains produced a PHA copolymer, poly(3-hydroxybutyrae-co-3-hydroxyvalerate) [P(3HB-co-3HV)], with a 3HV fraction up to 2 mol% with glucose as a carbon source. Further characterization was performed on one isolate, B. cereus YB-4. Gel permeation chromatography analysis revealed that the number of average molecular weights of PHA accumulated in B. cereus YB-4 drastically changed from 722,000 to 85,000 over a 72-h cultivation period. Furthermore, the PHA synthase genes were cloned and the deduced amino acid sequences were determined. This study provides new insights into PHA biosynthesis by members of the B. cereus group.  相似文献

The characterization of polyhydroxyalkanoates (PHA) produced by mixed cultures is fundamental for foreseeing the possible final applications of the polymer. In this study PHA produced under aerobic dynamic feeding (ADF) conditions are characterized. The PHA produced shows a stable average molecular weight ([symbol: see text]) in the range (1.0-3.0) x 10(6), along three years of reactor operation. Attempts to improve the amount of PHA produced did not introduce significant variations on the values [symbol: see text]. Along this period, the polydispersity indices (PDI) were between 1.3 and 2.2. The use of different carbon sources allowed the tailoring of polymer composition: homopolymers of poly(3-hydroxybutyrate), P(3HB), were obtained with acetate and butyrate, whereas a mixture of acetate and propionate, and propionate and valerate, gave terpolymers of 3-hydroxybutyrate (3HB), 3-hydroxyvalerate (3HV), and 2-methyl-3-hydroxyvalerate (2M3HV). All of these PHA had [symbol: see text] between 2.0 x 10(6) and 3.0 x 10(6). Thermal characterization of the produced polymers showed values of glass transition temperature, melting temperature, melting enthalpy, and crystallinity slightly lower than those obtained for PHA from pure cultures. The introduction of a purification step during the polymer extraction process allowed the elimination of possible contaminants but did not significantly improve the polymer quality.  相似文献

Poly(3-hydroxybutyrate) [P(3HB)], a polymer belonging to the polyhydroxyalkanoate (PHA) family, is accumulated by numerous bacteria as carbon and energy storage material. The mobilization of accumulated P(3HB) is associated with increased stress and starvation tolerance. However, the potential function of accumulated copolymer such as poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] remained unknown. In this study, Delftia acidovorans DS 17 was used to evaluate the contributions of P(3HB) and P(3HB-co-3HV) granules during simulated exogenous carbon deprivation on cell survival by transferring cells with PHAs to carbon-free mineral salt medium supplemented with 1 % (w/v) nitrogen source. By mobilizing the intracellular P(3HB) and P(3HB-co-3HV) at 11 and 40 mol% 3HV compositions, the cells survived starvation. Surprisingly, D. acidovorans containing P(3HB-co-94 mol% 3HV) also survived although the mobilization was not as effective. Similarly, recombinant Escherichia coli pGEM-T::phbCAB Cn (harboring the PHA biosynthesis genes of Cupriavidus necator) containing P(3HB) granules had a higher viable cell counts compared to those without P(3HB) granules but without any P(3HB) mobilization when exposed to oxidative stress by photoactivated titanium dioxide. This study provided strong evidence that enhancement of stress tolerance in PHA producers can be achieved without mobilization of the previously accumulated granules. Instead, PHA biosynthesis may improve bacterial survival via multiple mechanisms.  相似文献

The influence of nanofiller, plasticizer and hydroxyvalerate (HV) contents on the properties of poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV)/clay nano-biocomposites is studied.Analyses by wide-angle X-ray diffraction and differential scanning calorimetry showed that both the organo-modified montmorillonite clay and acetyl tributyl citrate plasticizer facilitate the crystallization process of PHBV. Despite its nucleating effect, the nanoclay hampers the spherulite growth and leads to a reduction in the copolymer crystallinity, while the addition of plasticizer induces the formation of thicker spherulites.For all studied systems, it was shown that the variation of mechanical properties of these nano-biocomposites is strongly influenced by their changes in crystallinity.  相似文献
