The photocycloaddition of 6-azauracil (AU) with olefins have been carried out. Four new compounds,1,2,4-triazabicyclo [4,2,0] octane and 1,2,4-triazatricyclo [4,2,01.6,07.12]dodecane derivatives,were obtained. Based on structure identificationof productsand photosensitization behavior of this syste,the regioselection and the multiplicity of thisreaction were discussed.  相似文献

4-Phenyl-1,2,4-triazoline-3,5-dione (PTAD) acts as an efficient Michael acceptor in reactions with 1-unsubstituted 2-pyrazolines, giving substituted (3,5-dioxo-4-phenyl-1,2,4-triazol-idin-1-yl)-1-pyrazolines in quantitative yields. Through elimination of molecular nitrogen, the latter are easily transformed into the corresponding cyclopropanes. A reaction of methyl 5-methyl-2-pyrazoline-5-carboxylate with a twofold excess of PTAD leads to an intermediate bisadduct, which undergoes dediazotization to form a 1,3,5-triazabicyclo[3.3.0]octane-2,4-dione derivative. 1-Substituted 2-pyrazolines also add to PTAD with the exception that the 2-pyrazoline structure is retained in the product because of the migration of the C(3)H proton to PTAD.  相似文献

Three new bis-triazolinediones, 3,3′-dimethyl-4,4′-bis-[3,5-dioxo-1,2,4-triazoline-4-yl]biphenyl, t-1,4-bis-[3,5-dioxo-1,2,4-triazoline-4-yl]methyl cyclohexane, and 4,4′-bis-[3,5-dioxo-1,2,4-triazoline-4-yl] phenyl ether, were synthesized from their corresponding bis-amines or bis-isocyanates. The compounds were identified by their quantitative ene reaction with 2,3-dimethyl-2-butene. The high degree of reactivity of the triazoline moiety makes solvent selection for reaction media rather difficult. This fact prompted a study of rates of reaction with a variety of polar and nonpolar solvents, including halogenated aliphatics, aromatics, tetrahydrofuran (THF), and N,N-dimethylformamide (DMF). The compounds exhibited reasonable stability in the halogenated solvents, as well as in the aliphatic and aromatic hydrocarbons, but they underwent reaction with THF and DMF. The structure of the reaction product of N-phenyl-1,2,4-triazoline-3,5-dione in DMF solution was determined, and a mechanism for product formation was proposed. Two of the bis- triazolinediones were polymerized via a base-catalyzed condensation mechanism which eliminates N2 from the triazolinedione ring.  相似文献

8-Acetyl-7-aryl-2-arylmethylene-8,9-diaza- and 4,8,9-triazabicyclo[4.3.0]non-9-enes react with4-phenyl-4,5-dihydro-3H-1,2,4-triazole-3,5-dione, following the ene addition pattern. Under similar conditons 7-aryl-2-arylmethylene-8-methyl-8,9-diazabicyclo[4.3.0]non-9-enes give rise to both mono- and polyadditionproducts. The product structures were studied by 1H and 13C NMR, IR, and UV spectroscopy and singlecrystal X-ray diffraction.  相似文献

Acid-catalyzed reaction of 6,10a-dihydroxy-3,4a,7,9-tetra(tert-butyl)-1,2,4a,10a-tetrahydrodibenzo-[b,e][1,4]dioxine-1,2-dione with 2-methylquinoline derivatives led to the formation of a previously unknown system 6-[(Z)-2-(quinolin-2-yl)-1-hydroxyethen-1-yl]pyran-2-one. The molecular structure of 3,5-di(tert-butyl)-6-[(Z)-2-(7,8-dimethyl-4-chloroquinolin-2-yl)-1-hydroxyvinyl]pyran-2-one was established by XRD method; the energy and structural characteristics of its isomers in the gas phase and in a polar solvent were calculated by quantumchemical methods (B3LYP/6-31G**).  相似文献

Thermolysis of 3-aroylpyrrolo[1,2-d][1,4]benzoxazine-1,2,4(4H)-triones generates aroyl(2-oxo-1,4-benzoxazin-3-yl)ketenes which react as dienes at the aroylketene fragment in [4 + 2]-cycloaddition at the C=N bond of N,N??-dicyclohexylcarbodiimide with formation of 3-[6-aryl-4-oxo-3-cyclohexyl-2-cyclohexylimino-3,4-dihydro-2H-1,3-oxazin-5-yl]-2H-1,4-benzoxazin-2-ones. Thermolysis of the latter is accompanied by elimination of N,N??-dicyclohexylcarbodiimide, and aroyl(2-oxo-1,4-benzoxazin-3-yl)ketenes thus generated undergo cyclodimerization to produce 7-aroyl-6,10-dioxo-9-(2-oxo-2H-1,4-benzoxazin-3-yl)-6,10-dihydrobenzo[b]pyrido[1,2-d][1,4]oxazin-8-yl benzoates.  相似文献

Highly reactive 4-substituted-1,2,4-triazoline-3,5-diones (TDs) have been studied extensively as dienophiles, but little work has been done on their role as enophiles and particularly on their use as propagating species in polymerization studies. The copolymerization between bis-4-substituted-1,2,4-triazoline-3,5-diones (bis-TDs) and styrene has been reported. The purpose of the present work was to synthesize new copolymers derived from a variety of substituted styrenes and bis-TDs and to study the mechanism and kinetics of this novel polymerization. Three bis-TDs were prepared: 3,3′-dimethyl-4,4′-bis[3,5-dioxo-1,2,4-triazoline-4-yl] biphenyl (8), t-1,4-bis[3,5-dioxo-1,2,4-triazoline-4-yl] methyl cyclohexane (9), and 4,4′-bis[3,5-dioxo-1,2,4-triazoline-4-yl] phenyl ether (10). Their structures were fully established by spectroscopic studies, elemental analyses, and indirectly, their quantitative ene reactions with 2,3-dimethyl-2-butene. Copolymerization between bis-TDs and substituted styrenes was carried out in dimethylformamide (DMF), tetrahydrofuran (THF), or dichloroethane (DCE). Polymers formed were characterized by infrared (IR) and nuclear magnetic resonance (NMR) spectroscopy, differential scanning calorimetry (DSC), gel permeation chromatography (GPC), and viscometry. Molecular weights of polymers range from 5000 to 16,000 in most cases. They were stable up to 250°C and higher. Polymers derived from bis-TDs and p-t-butylstyrene, α-methylstyrene, p-nitrostyrene, and p-acetoxystyrene contained only Diels-Alder-ene (DAe) repeating units, whereas those derived from styrene, p-chlorostyrene, p-bromostyrene, p-methylstyrene, p-methoxystyrene, and 4-vinylbiphenyl contained both DAe and double Diels-Alder (dDA) repeating units. A kinetic study of the copolymerization of 4,4′-bis-(3,5-dioxo-1,2,4-triazoline-4-yl) phenyl ether with α-methylstyrene, p-t-butylstyrene, styrene, p-chlorostyrene, and p-nitrostyrene in DCE was carried out; the copolymerization rate constants were 60.9, 49.8, 8.4, 5.5, and 0.8 (1 mol?1s), respectively.  相似文献

This paper reports our recent efforts to develop novel tricycles based on 4H-benzo[1,4]thiazin-3-one ( 2) and 1,1-dioxo-1,4-dihydro-2H-1lambda(6)-benzo[1,4]thiazin-3-one (3) using 1,5-difluoro-2,4-dinitrobenzene (1). All of these tricycles integrate two privileged structures into one skeleton, including 3,8-dihydro-5-thia-1,3,8-triaza-cyclopenta[b]naphthalene-7-one (4, 10, 12), 5,5-dioxo-3,5,6,8-tetrahydro-5lambda(6)-thia-1,3,8-triaza-cyclopenta[b]naphthalene-7-one (5, 11), 3,8-dihydro-5-thia-1,2,3,8-tetraaza-cyclopenta[b]naphthalene-7-one (6), 5,5-dioxo-3,5,6,8-tetrahydro-5lambda(6)-thia-1,2,3,8-tetraaza-cyclopenta[b]naphthalene-7-one (7), 3,8-dihydro-1H-5-thia-1,3,8-triaza-cyclopenta[b]naphthalene-2,7-dione (8), and 5,5-dioxo-3,5,6,8-tetrahydro-1H-5lambda(6)-thia-1,3,8-triaza-cyclopenta[b]naphthalene-2,7-dione (9). A typical library of scaffold 5 was synthesized in a parallel solution-phase manner and analyzed by HPLC-UV-MS or HPLC-UV-ELSD method.  相似文献

1,2-Cycloaddition reactions of several enamines with N-substituted maleimides and 4-phenyl-1,2,4-triazoline-3,5-dione have been carried out. The adducts were transformed into 7-(substituted-amino)-3-azabicyclo[3.2.0]heptanes and a 1,2,4-triazolidine-1-acetaldehyde, respectively. These are the first reported examples of 3-azabicyclo[3.2.0]heptanes substituted in any position other than the 3-position. The reaction of an ynamine with N-benzylmaleimide afforded a bicyclic intermediate which upon hydrolysis gave a 2,5-dioxo-3-pyrrolidineacetic acid.  相似文献

The reaction of 3,4-diamino-5-oxo-4,5-dihydro-l,2,4-triazine or its 6-methyl or 6-phenyl substituted derivatives and ethyl acetoacetate gave three compounds: 4,7-dioxo-9-methyl-1,4,6,7-tetrahydro-as-triazino[4,3-b]-1,2,4-triazepine in poor yield, isomeric 4,9-dioxo-7-methyl-1,4,8,9-tetrahydro-as -triazino[4,3-b]-1,2,4-triazepine and by competitive cyclisation, 2-methyl-7-oxo-3,7-dihydro-s-triazolo[3,2-c]-1,2,4-triazine. By condensation of 3-methylamino-4-amino-5-oxo-4,5-dihydro-1,2,4-triazine with ethyl acetoacetate, the formation of 4,9-dioxo-7,10-dimethyl-4,8,9,10-tetrahydro-as-triazino[4,3-b]-1,2,4-triazepine was strongly favored.  相似文献

Treatment of 6-amino-5-arylazo-1,3-dimethyluracils with urea or N,N′-carbonyldiimidazole gave the respective 6-aryl-1,3-dimethyl-6,7-dihydro-6-azalumazin-7-(6H)ones, which were hydrolyzed with alkali to afford 2-aryl-2,3,4,5-tetrahydro-3,5-dioxo-1,2,4-triazine-6-carboxylic acids (1-aryl-6-azauracil-5-carboxylic acids). Thermal decomposition of these carboxylic acids gave the corresponding 2-aryl-1,2,4-triazine-3,5-(2H,4H)diones (1-aryl-6-azauracils). A group of 5-(2-chlorophenyl)-10-(substituted)-7H-pyrido[4,3-f][1,2,4]triazolo[4,3-a][1,4]diazepines 7a-c were synthesized by the acid catalyzed reaction of 5-(2-chlorophenyl)-2-hydrazino-3H-pyrido[3,4-e]-[1,4]diazepine ( 6 ) with either trimethyl orthoformate, triethyl orthoacetate or triethyl orthobenzoate, respectively. 5-(2-Chlorophenyl)-7H-pyrido[4,3-f][1,2,4]triazolo[4,3-a][1,4]diazepine ( 7a ) and 5-(2-chlo-rophenyl)-10-methyl-7H-pyrido[4,3-f][1,2,4]triazolo[4,3-a][1,4]diazepine ( 7b ) exhibited good anticonvulsant activity in the subcutaneous metrazol anticonvulsant screen which serves as a model for absence (petit mal) epilepsy.  相似文献

Seventeen of the newly synthesized compounds were selected by the NCI, Maryland, USA, and were tested for their anticancer activity in an initial single high dose in the full NCI 60 cell line panel among which [1,4]thiazino[4,3,2‐gh ]purine‐7,8‐dione 2 , 7‐benzyl‐[1,4]thiazino[4,3,2‐gh ]purine 10b , and 3‐(2,4‐dimethoxyphenyl)‐7H ‐[1,2,4]triazolo[4,3‐i]purine 23 were found to possess very potent anticancer activity against most of the cancer cell lines.  相似文献
