Abstract

A flow‐injection spectrophotometric procedure was developed for determining N‐acetylcysteine in pharmaceutical formulations. The sample was dissolved in deionized water and 400 µl of the solution was injected into a carrier stream of 1.0×10?2 mol l?1 sodium borate solution. The sample flowed through a column (70 mm length×2.0 mm i.d.) packed with Zn3(PO4)2 immobilized in a polymeric matrix of polyester resin and Zn(II) ions were released from the solid‐phase reactor because of the formation of the Zn(II) (N‐acetylcysteine)2 complex. The mixture merged with a stream of borate buffer solution (pH 9.0) containing 5.0×10?4 mol l?1 Alizarin red S and the Zn(II)Alizarin red complex formed was measured spectrophotometrically at 540 nm. The analytical curve was linear in the N‐acetylcysteine concentration range from 3.0×10?5 to 1.5×10?4 mol l?1 (4.9 to 24.5 µg ml?1) with a detections limit of 8.0×10?6 mol l?1 (1.3 µg ml?1). The relative standard deviations (RSDs) were smaller than 0.5% (n=10) for solutions containing 5.0×10?5 mol l?1 (8.0 µg ml?1) and 8.0×10?5 mol l?1 (13.0 µg ml?1) of N‐acetylcysteine, and the analytical frequency was 60 determinations per hour. A paired t‐test showed that all results obtained for N‐acetylcysteine in commercial formulations using the proposed flow‐injection procedure and a comparative procedure agreed at the 95% confidence level.  相似文献

Abstract

A simple, rapid, and validated method for separation and determination of terbutaline enantiomers was developed. Terbutaline was separated and determined on a Vancomycin Chirobiotic V column (250 × 4.6 mm), using a mixture of methanol, acetic acid, and triethylamine (100:0.1:0.1% v/v/v) as a mobile phase at 20°C and at a flow rate of 1 ml/min. The UV detector was set to 276 nm. Acetyl salicylic acid (aspirin) was used as an internal standard. The applied high-performance liquid chromatography (HPLC) method allowed separation and quantification of terbutaline enantiomers with good linearity (r > 0.999) in the studied range. The relative standard deviations (RSD) were 1.10 and 1.32% for the terbutaline enantiomers with accuracy of 99.80 and 99.55. The limit of detection and limit of quantification of terbutaline enantiomers were found to be 0.05 and 0.10 µg · ml?1, respectively. The method was validated through the parameters of linearity, accuracy, precision, and robustness. The HPLC method was applied for the quantitative determination of terbutaline in pharmaceutical formulations.  相似文献

An SPE-HPTLC method for simultaneous identification and quantification of seven pharmaceuticals in production wastewater was optimized and validated. The studied compounds were enrofloxacine, oxytetracycline, trimethoprim, sulfamethazine, sulfadiazine, sulfaguanidine and penicillin G/procaine. The method involves solid-phase extraction on hydrophilic-lipophilic balance cartridges with methanol and HPTLC analysis of extracts on CN modified chromatographic plates followed by videodensitometry at 254 and 366 nm. Optimization of chromatographic separation was performed by systematic variation of the mobile phase composition using genetic algorithm approach and the optimum mobile phase composition for TLC separation was 0.05 M H2C2O4:methanol = 0.81:0.19 (v/v). Linearity of the method was demonstrated in the ranges from 1.5 to 15.0 μg L−1 for enrofloxacine, 100–500 μg L−1 for oxytetracycline, 150–600 μg L−1 for trimethoprim, 300–1100 μg L−1 for sulfaguanidine and 100–400 μg L−1 for sulfamethazine, sulfadiazine and penicillin G/procaine with coefficients of determination higher than 0.991. Mean recoveries ranged from 74.6 to 117.1% and 55.1 to 108.0% for wellspring water and production wastewater, respectively. Only sulfaguanidine showed lower results. The described method has been applied to the determination of pharmaceuticals in wastewater samples from pharmaceutical industry.  相似文献

An HPLC method was developed and validated for the determination of butamyrate citrate. The HPLC separation was achieved on a diol column (300 × 4.6 mm) packed with 5.0 μm particle size using a mobile phase of ammonium acetate buffer (pH = 6.5) and methanol (750:250, v/v) at a flow rate of 1.4 ml min?1. The UV detector was operated at 225 nm. The method was validated for specificity, linearity, precision, accuracy and robustness. The retention time was 5.9 min. The proposed method provided linear responses within the concentration range 75–225 μg ml?1 with LOD and LOQ values of 0.69 and 2.29 μg ml?1, respectively. Correlation coefficient (r) of the regression equation was 0.9999. The method was found to be precise, accurate, and reproducible.  相似文献

Abiraterone acetate is a prodrug of Abiraterone widely used for the treatment of metastatic castration resistant prostate cancer. In this study, a simple, sensitive, and rapid stability-indicating reverse phase HPLC method was developed and validated for the determination of Abiraterone acetate in bulk and its pharmaceutical formulation. The method was developed by HPLC using a Hypersil ODS C-18 (150 mm × 4.6 mm, 5 µm) column in a isocratic mode with mobile phase constituted by potassium phosphate buffer and acetonitrile (40:60, v/v%) flow rate was 1.0 mL min?1, column temperature of 30°C, UV detection wavelength 235 nm, and injection volume of 20 µL. The validated parameters were in accordance with FDA and ICH specifications, assay exhibited a linear range of 25–250 µg mL?1 with regression (r2) coefficient 0.9998. The limits of detection and quantification were 0.23 and 0.70 µg mL. Accuracy was between 99.34 and 100.07%. The drug was subjected to various stress conditions like acidic, base hydrolysis, oxidation, thermal, and photolytic degradation. Stress study Abiraterone acetate was found susceptible to degrade under hydrolytic (acid and base) conditions. The proposed method has stability indicating the resolution of the main peak from their degradation peaks. The validated method is suitable for quality control application and reduced analysis time.  相似文献

Abstract

A simple and reliable high-performance liquid chromatographic (HPLC) method was developed for the determination of belotecan in the plasma, urine, and bile samples of rats. Belotecan was analyzed with HPLC using a C18 column with fluorescence detector. A mixture of acetonitrile–0.1 M potassium phosphate buffer at pH 2.4 (25:75, v/v) and 0.2% trifluoroacetic acid was used as the mobile phase. The lower limits of quantitation (LOQ) were 5 ng mL?1 for the plasma and 5 µg mL?1 for the urine and bile samples. The method has been readily applied for the routine pharmacokinetic study of belotecan in small laboratory animals.  相似文献

Abstract

Sunset Yellow (SY), Carmoisine (C), Ponceau 4R (P), and Patent Blue V (PB) are synthetic organic dyes which are under governmental regulations all over the world because of their toxicity and carcinogenicity.

In this study, a simple and fast analytical procedure was proposed for the simultaneous determination of food dyes (SY, C, P, and PB) in powder drinks by means the partial least‐square treatment of spectrophotometric absorbance between 450 –730 nm, taken at 10 nm intervals. The experimental calibration matrix was constructed with 27 samples. The concentration ranges considered were 2, 3, 4 µg · ml?1 for SY, 7, 8, 9 µg · ml?1 for C, 9, 10, 11 µg · ml?1 for P, and 0.3, 0.4, 0.5 µg · ml?1 for PB. The method was applied to the determination of dyes in different commercially available powder drinks. The results obtained by the application of the PLS‐2 method were statistically compared with those obtained by an HPLC method using the F and t tests. Very similar values were found by two methods. No time consuming pretreatment was needed and this method also provides rapid, accurate and economical analysis of these colors.  相似文献

Abstract

A simple and selective spectrophotometric method has been developed for the determination of trace amounts of arsenic using azure B as a chromogenic reagent. The proposed method is based on the reaction of arsenic(III) with potassium iodate in acid medium to liberate iodine. The liberated iodine bleaches the violet color of azure B and is measured at 644 nm. This decrease in absorbance is directly proportional to the As(III) concentration, and Beer's law is obeyed in the range 0.2–10 µg ml?1 of As(III). The molar absorptivity, Sandell's sensitivity, detection limit, and quantitation limit of the method were found to be 1.12×104 l mol?1cm?1, 6.71×10?3 µg cm?2, 0.02 µg ml?1 and 0.08 µg ml?1, respectively. The optimum reaction conditions and other analytical parameters were evaluated. The proposed method has been successfully applied for the determination of arsenic in various environmental and biological samples.  相似文献

Two simple, sensitive, and specific high-performance liquid chromatography and thin-layer chromatography methods were developed for the simultaneous estimation of Amlodipine besilate (AM) and Valsartan (VL). Separation by HPLC was achieved using a xTerra C18 column and methanol /acetonitrile /water/ 0.05% triethylamine in a ratio 40:20:30:10 by volume as mobile phase, pH was adjusted to 3 ± 0.1 with o-phosphoric acid. The flow rate was 1.2 mL min?1. The linearity range was 0.2 to 2 µg mL?1 for amlodipine besilate and 0.4 to 4 µg mL?1 for Valsartan with a mean percentage recovery of 99.59 ± 0.523% and 100.61 ± 0.400% for amlodipine besilate and valsartan, respectively. The TLC method used silica gel 60 F254 plates; the optimized mobile phase was ethyl acetate/ methanol / ammonium hydroxide (55:45:5 by volume). Quantitatively, the spots were scanned densitometrically at 237 nm. The range was 0.5–4.0 µg spot?1 for amlodipine besilate and 2.0–12.0 µg spot?1 for valsartan. The mean percentages recovery was 99.80 ± 0.451% and 100.61 ± 0.363% for amlodipine besilate and valsartan, respectively. The HPLC method was found to be simple, selective, precise, and reproducible for the estimation of both drugs from spiked human plasma.  相似文献

A stability-indicating HPLC method has been developed and subsequently validated for the simultaneous determination of domperidone and pantoprazole in commercial tablets. The proposed HPLC method utilizes Phenomenex® Gemini C18 column (150 mm × 4.6 mm i.d., 5 μm) and mobile phase consisting of methanol-acetonitrile-20 mM dipotassium hydrogen phosphate and phosphoric acid buffer pH 7.0 (20:33:47, v/v/v) at a flow rate of 1.19 mL min?1. Quantitation was achieved with UV detection at 285 nm based on peak area with linear calibration curves at concentration ranges 0.5–5.0 μg mL?1 for domperidone and 1.0–10 μg mL?1 for pantoprazole (R 2 > 0.999 for both drugs). The method was validated in terms of accuracy, precision, linearity, limits of detection, limits of quantitation and robustness. This method has been successively applied to pharmaceutical formulation and no interference from the tablet excipients was found. Domperidone, pantoprazole and their combination drug product were exposed to acid, base and neutral hydrolysis, oxidation, dry heat and photolytic stress conditions and the stressed samples were analyzed by the proposed method. As the proposed method could effectively separate the drug from its degradation products, it can be employed as stability-indicating method for the determination of instability of these drugs in bulk and commercial products.  相似文献

A simple and rapid HPLC method using phenacetin (PHN) as internal standard has been developed for simultaneous determination of acetaminophen, caffeine, and chlorphenamine maleate in the product compound paracetamol and chlorphenamine maleate granules. Separation and quantitation were achieved on a 250 mm × 4.6 mm, 5 μm particle, C18 column. The mobile phase was methanol 0.05 mol L?1 aqueous KH2PO4 solution, 45:55 (v/v), containing 0.1% triethylamine and adjusted to pH 3.6 by addition of phosphoric acid; the flow rate was 1.0 mL min?1. Detection of all compounds was by UV absorbance at 260 nm and elution of the analytes was achieved in less than 12 min. The linearity, accuracy, and precision of the method were acceptable to good over the concentration ranges 6.4–153.6 μg mL?1 for acetaminophen, 5.0–120.0 μg mL?1 for caffeine, and 9.6–230.4 μg mL?1 for chlorphenamine maleate.  相似文献

Abstract

A simple, rapid, sensitive, and reproducible procedure for assaying norfloxacin (NOR), ciprofloxacin (CIP), and ofloxacin (OFL) was investigated. The procedure is based on the reaction of selected drugs with Sudan II (I), Congo red (II), and Gentian violet (III) in universal buffer to give soluble ion‐pair complexes. The effects of various parameters have been studied. Beer's law plots were obeyed in the concentration ranges 0.5–11 µg ml?1, whereas Ringbom optimum ranges were 0.7–9.5 µg ml?1. The apparent molar absorptivity (6.4×104 L mol?1 cm?1), Sandell sensitivity (4.99 ng cm?2), detection (0.13 µg ml?1), and quantification (0.44 µg ml?1) limits were calculated. The relative standard deviation for ten determinations, for samples containing 4.0 µg ml?1, was found to be 1.40%. The influence of commonly employed excipients in the determination of the studied drugs was examined. There was no interference from degradate product results from thermal and hydrolytic treatments. The results obtained by the proposed procedure were statistically validated. The developed procedure was successfully applied to the determination of the studied drugs in dosage forms and biological fluids.  相似文献

Two sensitive and selective stability-indicating methods were developed for the determination of the antibiotic cefpirome sulfate in bulk powder, pharmaceutical formulation and in presence of its acid, alkaline, photo- and oxidative degradation products. Method A was based on HPLC separation of cefpirome sulfate in the presence of its degradation products on a reversed phase column C18, 250 × 4.6 mm, 5-μm particle size and mobile phase consisting of 0.1 M disodium hydrogen phosphate dihydrate pH 3.9 adjusted with phosphoric acid–acetonitrile (85:15, v/v). Quantitation was achieved with UV detection at 270 nm. The linear calibration curve was in the range 5.0–50.0 μg mL?1. Method B was based on reversed phase TLC separation of the cited drug in the presence of its degradation products followed by densitometric measurement of the intact drug at 270 nm. The separation was carried out using disodium hydrogen phosphate dihydrate 2.0 g %w/v, at pH 3.5 adjusted with phosphoric acid–acetone (15:10, v/v) as a developing system. The calibration curve was in the range of 1.0–10.0 μg/spot. The HPLC method was used to study the kinetic of cefpirome sulfate acid degradation. The results obtained were statistically analyzed and compared with those obtained by applying the official Japanese method.  相似文献

Mirabegron is a novel β3-adrenoceptor agonist containing an amide group. It was subjected to stress conditions of acidic and alkaline hydrolyses. The hydrolytic degradation product was isolated and its structure was confirmed using mass and IR spectrometry. Two stability-indicating chromatographic methods have been proposed for the determination of mirabegron. TLC method was applied using silica gel as stationary phase and chloroform–methanol–ammonia (9.0:1.0:0.1 by volume) as the mobile phase, and chromatograms were scanned at 250 nm. Accurate determination of the drug was achieved over the concentration range of 2–12 μg per band. In addition, an isocratic HPLC method was developed on Agilent C18 column (150 mm × 4.5 mm I.D., particle size 5 µm) using ethanol-phosphate buffer pH 2.5 (30:70, by volume) as a mobile phase with flow rate of 1 mL min?1.The intact drug was detected at 250 nm with running time less than 5 min. Mirabegron was determined accurately in a concentration range of 1–25 µg mL?1. The proposed chromatographic methods were applied successfully for the assay of mirabegron in pharmaceutical dosage form and both methods were validated as per the International Conference on Harmonization guidelines and statistically compared with a reported gradient HPLC method.  相似文献

Abstract

Brazilian sugarcane spirits were analyzed to elucidate similarities and dissimilarities by principal component analysis. Nine aldehydes, six alcohols, and six metal cations were identified and quantified. Isobutanol (LD 202.9 µg L?1), butiraldehyde (0.08–0.5 µg L?1), ethanol (39–47% v/v), and copper (371–6068 µg L?1) showed marked similarities, but the concentration levels of n-butanol (1.6–7.3 µg L?1), sec-butanol (LD 89 µg L?1), formaldehyde (0.1–0.74 µg L?1), valeraldehyde (0.04–0.31 µg L?1), iron (8.6–139.1 µg L?1), and magnesium (LD 1149 µg L?1) exhibited differences from samples.  相似文献

A stability-indicating ultra-high-performance liquid chromatography (UHPLC) method with a diode array detector was developed and validated for the determination of cis/trans isomers of perindopril l-arginine in bulk substance and pharmaceutical dosage form. The separation was achieved on a Poroshell 120 Hilic (4.6 × 150 mm, 2.7 µm) column using a mobile phase composed of acetonitrile–0.1 % formic acid (20:80 v/v) at a flow rate of 1 mL min?1. The injection volume was 5.0 µL and the wavelength of detection was controlled at 230 nm. The selectivity of the UHPLC-DAD method was confirmed by determining perindopril l-arginine in the presence of degradation products formed during acid–base hydrolysis and oxidation as well as degradation in the solid state, at an increased relative air humidity and in dry air. The method’s linearity was investigated in the ranges 0.40–1.40 µg mL?1 for isomer I and 0.40–2.40 µg mL?1 for isomer II of perindopril l-arginine. The UHPLC-DAD method met the precision and accuracy criteria for the determination of the isomers of perindopril l-arginine. The limits of detection and quantitation were 0.1503 and 0.4555 µg mL?1 for isomer I and 0.0356 and 0.1078 µg mL?1 for isomer II, respectively.  相似文献

Speciation of mercury was accomplished by using a simple interface with photo-induced chemical vapour generation in a high performance liquid chromatography—atomic fluorescence spectrometry (HPLC-AFS) hyphenated system. Acetic acid and 2-mercaptoethanol in the mobile phase were used as photochemical reagent. The operating parameters were optimized to give limits of detection of 0.53 µg L?1, 0.22 µg L?1, 0.18 µg L?1 and 0.25 µg L?1 for inorganic mercury, methylmercury, ethylmercury and phenylmercury, respectively. The method was validated with the certified reference material DORM-2 and applied to the analysis of seafood samples. The HPLC-AFS hyphenated system is simple, environmentally friendly, and represents an attractive alternative to the conventional peroxothiosulfate-borohydride method.  相似文献

The possibility of using ionic liquid based chitosan sorbent for the separation and preconcentration of fluoroquinolone antibiotics (marbofloxacin, enoxacin, ofloxacin, ciprofloxacin, and enrofloxacin) has been studied. For this reason, different ionic liquids were prepared and coated on the chitosan sorbent. The conditions of the preconcentration of fluoroquinolones on a microcolumn have been optimized and the extraction efficiencies of the prepared sorbents have been compared. The compounds were eluted with 5 mL of 20% NH3 (v/v, MeOH) solution and determined by HPLC with diode array and fluorescence detector. The limits of detection were found as 4.23 µ g L?1 for marbofloxacin, and 1.09 µg L?1 for enoxacin; 3.23 × 10?3 µg L?1 for ofloxacin; 8.39 × 10?3 µg L?1 for ciprofloxacin; and 19.50 × 10?3 µg L?1 for enrofloxacin. The developed method was applied for the analysis of fluoroquinolone in milk, egg, fish, bovine, and chicken samples and the recoveries were obtained in the range 70–100%.  相似文献

A simple, sensitive, precise and accurate reversed phase liquid chromatographic method has been developed for the simultaneous estimation of atorvastatin (AT) calcium, ramipril (RA) and aspirin (AS) from capsule dosage form. The method was developed using a Phenomenex Luna C18 (250 mm, 4.6 mm i.d., 5 µm) column with a mobile phase consisting of 0.1%, orthophosphoric acid buffer:acetonitrile:methanol (45:50:5 v/v/v), pH 3.3, at a flow rate of 1 mL min?1. Detection was carried out with ultra-violet detection at 210 nm. The retention times were about 12.19, 2.35, and 3.95 min for AT calcium, RA and AS, respectively. The developed method was validated for linearity, accuracy, precision, limit of detection, limit of quantitation and robustness. The linearity ranges were 1–6 µg mL?1 for AT calcium, 0.5–3 µg mL?1 for RA and 7.5–45 µg mL?1 for AS with mean recoveries of 100.59 ± 0.68, 100.62 ± 0.83 and 100.49 ± 0.73% for AT calcium, RA and AS, respectively. Limit of detection obtained were 29.85 ng mL?1 for AT calcium, 4.71 ng mL?1 for RA and 85.13 ng mL?1 for AS. Impurity of salicylic acid was found in capsule dosage form at the retention time of about 4.84 min. The proposed method can be used for the estimation of these drugs in combined dosage forms.  相似文献
