Abstract

Two important drugs griseofulvin and dexamethasone have been quantitatively estimated through complexation with fluoranil (tetrafluoro-2,5-cyclohexadiene - 1,4- dione). The optimal experimental conditions, applicability of the suggested method to assay the cited drugs in their tablet forms, and statistical analysis of the results were studied. The results obtained were compared with those of the corresponding official method of assay for each drug, showing good agreement with the official methods. Furthermore, the reaction products have been isolated and subjected to structure elucidation by means of IR-Spectroscopic technique.  相似文献

The photooxidative removal of analgesic pharmaceutical compound phenazopyridine (PhP) from aqueous solutions by UV/H2O2 system with a re-circulated photoreactor was investigated. Response surface methodology (RSM) was employed to optimize the effect of operational parameters on the photooxidative removal efficiency. The investigated variables were: the initial PhP and H2O2 concentrations, irradiation time, volume of solution and pH. The analysis of variance (ANOVA) of quadratic model demonstrated that the described model was highly significant. The predicted values of the photooxidative removal efficiency were found to be in a fair agreement with experimental values (R2 = 0.9832, adjusted R2 = 0.9716). The model predicted that the optimal reaction conditions for a maximum removal of PhP (>98%) were: initial PhP concentration less than 23 mg L–1, initial concentration of H2O2 higher than 470 mg L–1, solution volume less than 500 mL, pH close to 2 and irradiation time longer than 6 min.  相似文献

Abstract

Two simple and sensitive spectrophotometric procedures are suggested for analysis of triamterene. The first procedure is based on the reaction of triamterene with p-chloranilic acid (p-CA) in methylene chloride to form a highly stable coloured product, exhibiting maximum absorbance at λ 530 nm. Beer's law is obeyed in the range of 40–220 μg.ml?1 with a mean percentage accuracy of 99.98 ± 0.446. Limit of determination is 20 μg.ml?1. In the second procedure, the drug is determined via charge transfer complex formation with 2,3 dichloro-5,6-dicyano p-benzoquinone (DDQ) using methylene chloride as a solvent. Here the reaction product has two well defined maxima at 460 nm and 530 nm where each has been utilized for quantitative determination. Beer's law is obeyed in concentration ranges of 25–125 μg.ml?1 and 25–150 μg.ml?1 with mean percentage accuracies of 99.92 ± 0.449 and 100.00 ± 0.511 for both maxima. 460 and 530 nm. respectively. Limit of determination is 12.5 μg.ml?1 at both maxima. Optimum conditions for each procedure have been studied and the stoichiometry of both reactions was ascertained using Job's method of continuous variation. The validity of the suggested procedures was assessed by applying the standard addition technique using the drug capsules. Both procedures are statistically analyzed as compared with BP method for analysis of triamterene (non aqueous titration) revealing good accuracy and precision as indicated by t and F tests.  相似文献

Abstract

A colorimetric method for the determination of certain 8-hydroxyquinolines has been developed. The method is based on the coupling of 8-quinolinols with diazotriazole carboxylic acid in the presence of sodium carbonate (0.5% w/v) at ambient temperature (around 30°C). The resulting azo dyes are stable and give intense absorption in the range of 486–540 nm. Beer's law is valid in the concentration ranges of 1 – 8, 2.5 – 17.5, 5 – 35, 2 – 20, and 2 – 20 mcg/ml for 8-hydroxyquinoline (HQ), 5,7-diiodo-8-hydroxyquinoline (DIQ), 5,7-dibromo-8-hydroxyquinoline (DBQ), 5-chloro-7-iodo-8-hydroxyquinoline (CIQ) and 8-hydroxy-7-iodo-5-quinoline sulfonic acid (SIQ), respectively. The proposed method has been successfully applied for the determination of the studied compounds in pure form and in commercial formulations. The obtained results are in good agreement with those obtained from reported methods.  相似文献

ABSTRACT

The present study is based on the interaction of diazotized 4-amino-6-chloro-1,3-benzene disulphonamide (DACBS) with 8-hydroxyquinolines in slightly alkaline medium and with dibenzazepines in strongly acidic medium to form red coloured products in the range of 500- 555 nm. Under optimum reaction condition, Beer's law is obeyed in the concentration range 0.5-25 μg/ml for 8-hydroxyquinolines and 1.0-25 μg/ml for dibenzazepines. The stoichiometry of the reaction was investigated and was found to be 1:1 for all the studied drugs. Interference due to other co-formulated drugs, common additives and excepients was studied. The proposed method was applied successfully for the determination of the cited drugs in their pharmaceutical preparations and results were comparable with that of the official, reported method.  相似文献

ABSTRACT

The use of multivariate spectrophotometric calibration is reported for the analysis of tablets containing the antibiotics sulfamethoxazole and trimethoprim, and a combination of the former two drugs with the analgesic phenazopyridine. The resolution of these mixtures has been accomplished without prior separation, derivatisation or use of nonaqueous solvents, with the aid of partial least-squares (PLS-1) regression analysis of electronic absorption spectral data. Abstract

Direct spectrophotometric methods for the determination of carbochromen hydrochloride and dipyridamole, each in the presence of its oxidative degradation products, are presented. the methods are based on the first derivative (D1) and second derivative (D2) spectrophotometric measurement (absolute trough, U) at 336 nm and (Peak-trough, Y) at 309–342 nm for carbochromen hydrochloride and at 240–260 nm(U) and 246–268 nm(Y) for dipyridamole. Plots of D1 or D2 versus concentration were linear over the concentration range of 8.00–16.00 μg/ml for carbochromen hydrochloride and 4.00–12.00 μg/ml for dipyridamole. Oxidative degradation of these drugs has been optimized with respect to hydrogen peroxide concentration. Determining the intact in coexistence with its oxidative degradation product, the proposed derivative spectrophotometric methods proved to be of high potential in correcting the systematic error appearing in the results of the Amax method due to the latter. Assaying the commercial tablets, the proposed method gave results of high accuracy and reproducibility.  相似文献

Two simple and sensitive spectrophotometric methods are described for the determination of six penicillin derivatives. The methods are based on the reaction of these drugs as n-electron donors with either 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) or 7,7,8,8-tetracyanoquinodimethane (TCNQ) as pi-acceptors, to give a highly coloured radical anion. The coloured products are quantified spectrophotometrically at 460 and 842 nm for DDQ and TCNQ, respectively. The optimization of the different experimental conditions is described. The interference from streptomycin sulphate and common degradation products was also studied. The proposed methods were applied successfully to the determination of the different penicillins investigated, either in pure or dosage forms, with good accuracy and precision. The results were compared with those given by the official United States Pharmacopeial XXI method.  相似文献

采用循环伏安法和差分脉冲伏安法考察了临床药物盐酸非那吡啶在功能化复合介孔膜修饰的水/1,6-二氯己烷(W/DCH)界面上的转移行为. 研究结果表明, 盐酸非那吡啶中的非那吡啶(PAP) 可在酸性条件下发生质子化形成非那吡啶阳离子(HPAP+)并在该界面上发生转移. 根据循环伏安曲线峰电流与扫描速率的关系以及Randles-Sev$\check{c}$ik 方程, 计算得到HPAP+在复合介孔膜内水相中的扩散系数为5.14×10-8 cm2/s. 研究发现, 相对于空白膜修饰的W/DCH界面, 该复合介孔膜修饰的W/DCH界面可增强非那吡啶阳离子转移时的电流响应, 并提升非那吡啶阳离子的检测性能. 另外, 利用液/液界面电化学方法绘制了非那吡啶在功能化复合介孔膜修饰的W/DCH界面上的离子分布图, 从而得到非那吡啶在该界面上不同条件下的分布情况及其分配系数和吉布斯转移自由能.  相似文献

The lithio-anion of isoxazole 2 was found to ring open propylene oxide in good yields with complete regioselectivity. Vinylic and benzylic epoxides were utilized as key examples of electrophiles and found to produce a mixture of regioisomeric adducts. Additionally, the use of chiral epoxides was explored, and absolute configuration was determined by X-ray crystallography to prove that nucleophilic attack at the benzylic carbon of (R)-styrene oxide proceeds with 100% inversion at the benzylic carbon to afford the (S)-alcohol (4b).  相似文献

The combination of theoretical models of macromolecules that exist at different spatial and temporal scales has become increasingly important for addressing complex biochemical problems. This work describes the extension of concurrent multiscale approaches, introduces a general framework for carrying out calculations, and describes its implementation into the CHARMM macromolecular modeling package. This functionality, termed MSCALE, generalizes both the additive and subtractive multiscale scheme (e.g. QM/MM ONIOM-type), and extends its support to classical force fields, coarse grained modeling (e.g. ENM, GNM, etc.), and a mixture of them all. The MSCALE scheme is completely parallelized with each subsystem running as an independent, but connected calculation. One of the most attractive features of MSCALE is the relative ease of implementation using the standard MPI communication protocol. This allows external access to the framework and facilitates the combination of functionality previously isolated in separate programs. This new facility is fully integrated with free energy perturbation methods, Hessian based methods, and the use of periodicity and symmetry, which allows the calculation of accurate pressures. We demonstrate the utility of this new technique with four examples; (1) subtractive QM/MM and QM/QM calculations; (2) multi-force field alchemical free energy perturbation; (3) integration with the SANDER module of AMBER and the TINKER package to gain access to potentials not available in CHARMM; and (4) mixed resolution (i.e. coarse grain / all-atom) normal mode analysis. The potential of this new tool is clearly established and in conclusion an interesting mathematical problem is highlighted and future improvements are proposed.  相似文献

Simple and sensitive spectrophotometric methods for the assay of terfenadine are described. The first is based on the reaction of terfenadine with iodine to give a molecular charge-transfer complex, the terfenadine acting as an n-electron donor and iodine as a sigma-electron acceptor. The second depends on the formation of a highly coloured stable radical anion between terfenadine and 7,7,8,8-tetracyanoquinodimethane (TCNQ) as a pi-electron acceptor. Beer's law is obeyed over the terfenadine concentration range 0.2-1.2 mg/100 ml. The proposed methods have been successfully applied to the analysis of commercial terfenadine tablets.  相似文献

Background

Pregabalin (PG) is an anticonvulsant, analgesic and anxiolytic drug. A survey of the literature reveals that all the reported spectrophotometric methods are either don't offer high sensitivity, need tedious extraction procedures, recommend the measurement of absorbance in the near UV region where interference most probably occurs and/or use non specific reagent that don't offer suitable linearity range.

Results

Two new sensitive and simple spectrophotometric methods were developed for determination of pregabalin (PG) in capsules. Method (I) is based on the reaction of PG with 1,2-naphthoquinone-4-sulphonate sodium (NQS), yielding an orange colored product that was measured at 473 nm. Method (II) is based on the reaction of the drug with 2,4-dinitrofluorobenzene (DNFB) producing a yellow product measured at 373 nm. The different experimental parameters affecting the development and stability of the reaction product in methods (I) and (II) were carefully studied and optimized. The absorbance-concentration plots were rectilinear over the concentration ranges of 2-25 and 0.5-8 μg mL-1 for methods (I) and (II) respectively. The lower detection limits (LOD) were 0.15 and 0.13 μg mL-1 and the lower quantitation limits (LOQ) were 0.46 and 0.4 μg mL-1 for methods (I) and (II) respectively.

Conclusion

The developed methods were successfully applied to the analysis of the drug in its commercial capsules. The mean percentage recoveries of PG in its capsule were 99.11 ± 0.98 and 100.11 ± 1.2 (n = 3). Statistical analysis of the results revealed good agreement with those given by the comparison method. Proposals of the reaction pathways were postulated.  相似文献

Common oxidants used in chemical synthesis, including newly developed perruthenates, were evaluated in the context of understanding (and better appreciating) the sensitiveness and associated potential hazards of these reagents. Analysis using sealed cell differential scanning calorimetry (scDSC) facilitated Yoshida correlations, which were compared to impact sensitiveness and electrostatic discharge experiments (ESD), that enabled sensitiveness ranking. Methyltriphenylphoshonium perruthenate (MTP3, 8 ), isoamyltriphenylphosphonium perruthenate (ATP3, 7 ) and tetraphenylphosphonium perruthenate (TP3, 9 ) were found to be the most sensitive followed by 2-iodoxybenzoic acid (IBX, 2 ) and benzoyl peroxide (BPO, 10 ), whereas the most benign were observed to be Oxone ( 12 ), manganese dioxide (MnO2, 13 ), and N-bromosuccinimide (NBS, 17 ).  相似文献

A variety of IR‐active moieties with absorptions that are distinct from those of proteins have been developed as probes of local protein environments, including carbon‐deuterium bonds (C?D), cyano groups (CN), and azides (N3); however, no systematic analysis of their utility in a protein has been published. Previously, we characterized the N‐terminal Src homology 3 domain of the murine adapter protein Crk‐II (nSH3) with C?D bonds site‐selectively incorporated throughout, and showed that it is relatively rigid and electrostatically heterogeneous and that it thermally unfolds under equilibrium conditions via a simple two‐state mechanism. We now report the synthesis and characterization of eight variants of nSH3 with CN and/or N3 probes at five of the same positions. In agreement with previous studies, the position‐dependent spectra suggest that both probes are predominantly sensitive to hydration, and not to their local electrostatic environments. Importantly, both probes also tend to significantly perturb the protein if they are not incorporated at surface‐exposed positions. Thus, unlike C?D labels, which are both sensitive to their environment and non‐perturbative, CN and N3 probes should be used with caution.  相似文献

A simple, accurate, precise and sensitive colorimetric method for the determination of some skeletal muscle relaxant drugs, namely orphenadrine citrate (I), baclofen (II), antihistaminic drugs as acrivastine (III) and fexofenadine hydrochloride (IV) is described. This method is based on the formation of charge transfer complex with 4-chloro-7-nitro-2,1,3-benzoxadiazole (NBD-Cl) in non-aqueous medium. The orange color products were measured at 472, 465, 475 and 469 nm for drugs I, II, III and IV, respectively. The optimization of various experimental conditions was described. Beer's Law was obeyed in the range (2.5-17.5), (5-70), (2.5-25) and (10-50)microg/ml for drugs I, II, III and IV, respectively. The molar absorptivity (epsilon), sandell sensitivity, detection((LOD)) and quantitation limits((LOQ)) are calculated. The procedure was favorably applied for determination of certain pharmaceutical dosage forms containing the studied drugs. The obtained results were compared with the official and reported methods. There were no significant differences between proposed, reported and the official methods.  相似文献

On-chip electrophoresis can provide size separations of nucleic acids and proteins similar to more traditional slab gel electrophoresis. Lab-on-a-chip (LoaC) systems utilize on-chip electrophoresis in conjunction with sizing calibration, sensitive detection schemes, and sophisticated data analysis to achieve rapid analysis times (<120 s). This work describes the utility of LoaC systems to enable and augment systems biology investigations. RNA quality, as assessed by an RNA integrity number score, is compared to existing quality control (QC) measurements. High-throughput DNA analysis of multiplex PCR samples is used to stratify gene sets for disease discovery. Finally, the applicability of a high-throughput LoaC system for assessing protein purification is demonstrated. The improvements in workflow processes, speed of analysis, data accuracy and reproducibility, and automated data analysis are illustrated.  相似文献

The scope of the tandem Pauson-Khand reaction has been explored for the regiospecific construction of [5.5.5.5]- and [5.6.6.5]tetracyclic systems via the photolytic method of Livinghouse. The rapid regiospecific entry into the two dicyclopentapentanoid systems 17 and 29 was accomplished from the key diene-diynes 11 and 19b. A photochemically mediated catalytic tandem Pauson-Khand cyclization was employed to prepare the parent ring systems of dicyclopenta[a,e]pentalene (from 19b) and dicyclopenta[a,f]pentalene (from 11) in regiospecific fashion in a one-pot process. Under these conditions, conversion of acyclic diene-diyne 16 into tetracyclic system 17 was achieved in 74% yield, while a similar process was employed to convert 28 into tetracycle 29 in 90% yield. This is much improved over the previous conditions that employed NMO. Six carbon-carbon bonds were generated in this process constituting up to 98% yield for each carbon-carbon bond so formed. Furthermore, tetracyclic [5.6.6.5] systems such as dicyclopenta[b,g]decalins 37, 38, and 40 were prepared from similar diene-diyne precursors via the tandem Pauson-Khand cyclization. Importantly, acetal 36 provided the desired cis-fused [5.6.6.5] system 38a (via 40a/b) in stereospecific fashion. This reaction is unique in that it provides a cis-decalin ring system; moreover, the yield of each of the six carbon-carbon bonds formed in this process was at least 89%. The structure of cis diol 38a was confirmed by X-ray crystallography.  相似文献
