Two simple and sensitive extractive spectrophotometric methods have been described for the analysis of clarithromycin in pure form and in pharmaceutical formulations. The methods involved formation of yellow colored chloroform extractable ion‐association complexes of clarithromycin with bromothymol blue (BTB) and cresol red (CR) in buffered aqueous solution at pH 4. The extracted complexes showed maximum absorbance at 410 and 415 nm for BTB and CR, respectively. Beer's law is obeyed in the concentration ranges 0.1–20 μg mL?1 and 2.0–20 μg mL?1 of clarithromycin with molar absorptivity of 2.01 × 104 and 4.378 × 103 for BTB and CR, respectively. The composition ratio of the ion‐association complex was clarithromycin: BTB and CR = 1:1 as established by Job's method. The methods have been applied to the determination of drug in commercial formulations. The results of analysis were validated statistically and through recovery studies.  相似文献

The present study describes three simple and sensitive spectrophotometric methods developed for the determination of linezolid (LZD) in pure and tablet forms. These methods are based on the oxidation of LZD by ferric chloride in the presence of 1,10-phenanthroline (method A), or 2,2′-bipyridyl (method B), or potassium ferricyanide (method C). The colored complexes were measured at 510, 522 and 758 nm for methods A, B and C, respectively. In all the methods, the absorbance is found to increase linearly with increasing LZD concentration. Beer’s law is obeyed over the concentration ranges of 0.5–6.0, 0.5–9.0 and 1.0–9.0 μg/mL for methods A, B and C, respectively. The calculated molar absorptivity values are 5.8 × 104, 3.6 × 104 and 4.8 × 104 L/mol cm for methods A, B and C, respectively, and the corresponding Sandell’s sensitivities are 5.8 × 10?3, 9.8 × 10?3 and 7 × 10?3 μg/cm2, respectively. The developed methods are applied successfully to the determination of LZD in the pharmaceutical formulations and the results tallied well with label claims.  相似文献

A simple, sensitive and accurate spectrophotometric method has been described for the assay of diphenhydramine hydrochloride (DPH) in raw material and in biological samples. The method is based on extraction of DPH into dichloromethane as ion-pair complexes with patent blue (PB), eriochrome black T (EBT), methyl orange (MO) and bromocresol purple (BCP) in acidic medium. The coloured species exhibited absorption maxima at 632, 514, 428 and 414 nm for PB, EBT, MO and BCP, with molar absorptivity values of 1.32 × 105, 2.36 × 104, 3.68 × 104 and 3.07 × 104 l mol?1 cm?1, respectively. The reaction conditions were optimized to obtain the maximum colour intensity. Beer’s law was obeyed with a good correlation coefficient (0.9982–0.9993) in the concentration ranges 0.5–3, 2.0–16, 2.0–10 and 1.0–10 μg ml?1 for PB, EBT, MO and BCP methods, respectively. The composition ratio of the ion-association complexes was found to be 1:1 in all cases as established by Job’s method. The conditional stability constant (Kf) and the free energy changes (ΔG°) were determined for all complexes formed. The proposed method was successfully applied for the determination of DPH in tablets and human urine with good accuracy and precision. Statistical comparison of the results with those obtained by the official method showed good agreement and indicated no significant difference in accuracy and precision.  相似文献

Two simple, rapid and sensitive extractive spectrophotometric methods have been developed for the assay of nortriptyline hydrochloride (NTPH) in pure and pharmaceutical formulations. These methods are based on the formation of chloroform soluble ion-association complexes of NTPH with bromocresol green (BCG) and with methyl orange (MO) in KCl-HCl buffer of pH 2. The colored species exhibited absorption maxima at 416 and 422 nm for BCG and MO with molar absorptivity values of 2.88 × 104 and 2.29 × 104 L/mol cm, respectively. Reaction conditions were optimized to obtain the maximum color intensity. Various analytical parameters have been evaluated and the results have been validated by statistical data. The methods were successfully applied to the analysis of NTPH in pharmaceutical formulations. The article is published in the original.  相似文献

Three sensitive and accurate spectrophotometric procedures were developed for the analysis of cephapirine sodium in pure form and in its pharmaceutical formulation. Method A: A kinetic method based on the observation that in acidic medium cephapirine reduces sodium molybdate to molybdenum blue, the absorbance of which is proportional to the amount of antibiotic present at a fixed time of 40 minutes; the formed product was spectrophotometrically measured at 780 nm. The concentration of drug calculated using its calibration by fixed concentration and rate constant methods is feasible with the calibration equations obtained, but the fixed time method proved to be more applicable. Method B is based on chetale formation with palladium(II) chloride in buffered medium as the interaction between metal ions and ligand anions or moleules capable of the formation of complexes which results in the development of colors suitable for the characterization of quantitative determination of metal or ligand. Metals containing easily excited d or f electrons were suitable for the formation of colored complexes. Method C, is based on the formation of colored complex between palladium(II), eosin and cephapirine Na. Sodium lauryl sulphate is used as surfactant to increase the solubility and intensity of the formed complex. Under optimum conditions, the complexes showed maximum absorption at Δ370 and Δ550 for methods B and C, respectively. Apparent molar absorpitivities were 5.2 × 103, 5.5 × 103, 1.4 × 104; Sandell's sensitivities were 1.17 × 10?3, 1.24 × 10?3, 3.1 × 10?3, for methods A, B and C, respectively. The solution of the products obeyed Beer's Law in the concentration ranges 10–70, 20–70, 2–48, μg mL?1 for methods A, B, and C. The proposed methods were applied to the determination of the drug in pure or pharmaceutical preparations. The results obtained were compared statistically with those given by the official method.  相似文献

Differential pulse cathodic adsorptive stripping (DPCAdSV) and square wave cathodic adsorptive stripping (SWCAdSV) voltammetric methods were developed for the determination of antimony and lead in gunshot residues. Linear working ranges for DPCAdSV and SWCAdSV methods were (2.0×10?9–5.0×10?7) M and (2.0×10?9–7.0×10?7) M for antimony and 2.0×10?9–3.0×10?7 M (both methods) for lead. The detection of antimony limits were found to be 1.3×10?9 M for DPCAdSV and 7.3×10?10 M for SWCAdSV while the corresponding values for lead were 3.0×10?9 M and 5.8×10?10 M. Antimony and lead contents obtained by these methods in gunshot residues are in good agreement with those obtained by graphite furnace atomic absorption spectrometric method within a confidence limit of 95%.  相似文献

Vitamin B1‐selective electrodes with PVC membrane were developed that contain ion associates of vitamin B1 with an inorganic anion, BiI4?, and an organic anion, brilliant yellow, as electrode‐active substances. The linearity ranges of the electrode function are 1.0×10?5–1.0×10?2 and 1.0×10?4–1.0×10?2 M, the electrode function slopes are 33.0±1.0 and 33.1±1.1 mV decade?1, the detection limits are 5.5×10?6 and 8.3×10?5 M for BiI4? and brilliant yellow respectively. The working range of pH is 5–12. The efficiency of the use of electrodes for the vitamin B1 content control in multivitamin pharmaceutical preparations was shown by direct potentiometry and potentiometric titration methods.  相似文献

Simple, selective and accurate sensors were developed for the determination of melatonin and oxomemazine in biological samples (urine) and in pharmaceutical preparations. Potentiometric measurements were based on bismus tetraiodate‐drug ion‐pair as novel electroactive materials incorporating a plasticized PVC membrane with o‐nitrophenyl octyl ether or dioctyl phthalate. Each sensor was conditioned for at least two days in 0.1 M drug solution before use. It exhibited fast and stable Nernstian response for melatonin and oxomemazine over the concentration range of 1.0×10?6–1.0×10?2 M and 1.0×10?5–1.0×10?2 M, pH range of 3.0–6.5 and 3.5–6.0 for melatonin and oxomemazine sensors, respectively. Results with an average recovery not more than 101 % and a mean standard deviation less than 1.0 % of the nominal were obtained for the four sensors. The sensors showed reasonable selectivity towards investigated drugs in presence of many cations.  相似文献

A polymerized film of Adizol Black B (ABB) on the surface of glassy carbon (GC) electrode was prepared for the simultaneous determination of ascorbic acid (AA), epinephrine (EP), and uric acid (UA). This new electrode presented an excellent electrocatalytic activity towards the oxidation of AA, EP, and UA by differential pulse voltammetry method. The oxidation peaks of the three compounds were well defined and had the enhanced peak currents. The separation of the oxidation peak potentials for AA–EP and EP–UA were about 180 and 130 mV, respectively. The calibration curves obtained for AA, EP, and UA were in the ranges of 2.0–1,970.0, 0.1–64.0, and 0.1–1,700.0 μmol L–1, respectively. The detection limits (S/N?=?3) were 0.01, 0.007, and 0.02 μmol L–1 for AA, EP, and UA, respectively. The diffusion coefficient and the catalytic rate constant for the oxidation reaction of EP at poly(ABB) film-coated GC electrode were calculated as 1.54(±0.10)?×?10?4 cm2 s?1 and 4.5?×?103 mol?1 L s?1, respectively. The present method was applied to the determination of EP in pharmaceutical, AA in commercially available vitamin C tablet, and UA in urine samples.  相似文献

Liquid chromatography with electrochemical detection (LC-ED), coupled with in vivo microdialysis sampling, has been used for analysis of thiols. An acetylene black–dihexadecyl hydrogen phosphate (AB–DHP) composite film-modified electrode was used as working electrode. The AB–DHP-modified electrode enabled efficient electrocatalytic oxidation of l-cysteine (l-Cys) and glutathione (GSH) with relatively high sensitivity, stability, and longevity. The peak currents of l-Cys and GSH were linear in the concentrations ranges 2.0 × 10?7–2.0 × 10?4 and 3.0 × 10?7–5.0 × 10?4 mol L?1, respectively, with calculated detection limits (S/N = 3) of 1.0 × 10?7 and 2.0 × 10?7 mol L?1, respectively. The method has been successfully used to measure the amounts of l-Cys and GSH in striatal microdialysate of freely moving rats.  相似文献

A reliable and simple electrochemical method has been proposed for the simultaneous determination of paracetamol (PAR) and p‐aminophenol (PAP) in pharmaceutical formulations. The oxidation and reduction peak potentials in cyclic voltammetry (CV) for PAR on carbon ionic liquid electrode (CILE) were occurred at 370 and 225 mV vs. Ag/AgCl, respectively at pH 7.0, while those for PAP on CILE appeared at 128 mV and 68 mV, respectively at the scan rate of 0.05 V s?1. In comparison to the conventional carbon paste electrode, the apparent reversibility and kinetics of the electrochemical reactions of PAR and PAP were significantly improved on CILE. In differential pulse voltammetric technique, the peak potentials for PAR and PAP appeared at 345 and 130 mV, respectively, with the peak separation of 215 mV, sufficient for their simultaneous determination in samples containing these two species. The proposed method was used for simultaneous determination of PAR and PAP in tablets. PAR and PAP can be determined in the ranges of 2.0×10?6–2.2×10?3 M and 3.0×10?7–1.0×10?3 M, with the detection limits of 5.0×10?7 and 1.0×10?7 M (calculated by 3σ), respectively. The relative standard deviations for the determination of PAR and PAP were less than 2%.  相似文献

A novel chemiluminescence (CL) method for the determination of hydrogen peroxide is described. Method is based on the transition metals in highest oxidation state complex, which include diperiodatoargentate (DPA) and diperiodatonickelate (DPN) and show excellent sensitisation on the luminol-H2O2 CL reaction with low luminol concentration in alkaline medium. In particular, the sensitiser which was previously reported (such as Co2+, Cu2+, Ni2+, Mn2+, Fe3+, Cr3+, KIO4, K3Fe(CN)6 etc.) to be unobserved CL due to poor sensitisation with such low concentration of luminol which makes the method hold high selectivity. Based on this observation, the detection limits were 6.5?×?10?9?mol?L?1 and 1.1?×?10?8?mol?L?1 hydrogen peroxide for the DPN- and DPA-luminol CL systems, respectively. The relative CL intensity was linear with the hydrogen peroxide concentration in the range of 2.0?×?10?8–6.0?×?10?6?mol?L?1 and 4.0?×?10?8–4.0?×?10?6?mol?L?1 for the DPN- and DPA-luminol CL systems, respectively. The proposed method had good reproducibility with a relative standard deviation of 3.4% (8.0?×?10?7?mol?L?1, n?=?7) and 1.0% (2.0?×?10?6?mol?L?1, n?=?7) for the DPN- and DPA-luminol CL systems, respectively. A satisfactory result has been gained for the determination of H2O2 in rainwater and artificial lake water by use of the proposed method.  相似文献

A cobalt oxide nanoparticles (Co3O4NPs) and multi walled carbon nanotubes (MWCNTs) modified carbon paste electrodes were used to study the electrochemical behavior of linagliptin and empagliflozin in Britton Robinson buffer solution of pH 8.0 using cyclic and square wave voltammetry. The above mentioned modified electrodes showed highly sensitive sensing and gave an excellent anodic response for both drugs. The peak current varied linearly over the concentration ranges: 3.98×10?5–1.53×10?3 mol L?1 (18.82–723.00 μg/mL) and 7.94×10?6–1.07×10?4 mol L?1 (3.65–48.25 μg/mL) with determination coefficients of 0.9999 and 0.9998 for linagliptin and empagliflozin, respectively. The recoveries and relative standard deviations were found in the following ranges: 98.80 %–102.00 % and 0.23 %–1.90 % for linagliptin and 98.30 %–101.80 % and 0.11 %–1.86 % for empagliflozin. The detection and quantification limits were 1.13×10?5 and 3.76×10?5 mol L?1 (5.34and17.77 μg/mL) for linagliptin, 1.71×10?6and 5.68×10?6 mol L?1 (0.77 and 2.56 μg/mL) for empagliflozin. The proposed sensors have been successfully applied for the determination of the drugs in bulk, pharmaceutical formulations and biological fluids.  相似文献

Abstract

An amperometric biosensor for the determination of phenols is proposed using a crude extract of sweet potato (Ipomoea batatas (L.) Lam.) as an enzymatic source of polyphenol oxidase (PPO; tyrosinase; catechol oxidase; EC 1.14.18.1). The biosensor is constructed by the immobilization of sweet potato crude extract with glutaraldehyde and bovine serum albumin onto an oxygen membrane. This biosensor provides a linear response for catechol, pyrogallol, phenol and p-cresol in the concentration ranges of 2.0×10?5-4.3×10?4mol L?1, 2.0×10?5-4.3×10?4 mol L?1, 2.0×10?5-4.5×10?4 mol L?1 and 2.0×10?5-4.5×10?4mol L?1, respectively. The response time was about 3–5 min for the useful response range, and the lifetime of this electrode was excellent for fifteen days (over 220 determinations for each enzymatic membrane). Application of this biosensor for the determination of phenols in industrial wastewaters is presented.  相似文献

The electrooxidation of dextromethorphan on a composite constructed with carbon nanotube–ionic liquid–carbon microparticles was investigated by cyclic voltammetry in a 100 mM phosphate buffer solution, pH 7.40. In the voltammograms, an irreversible diffusion-controlled anodic peak appeared. The diffusion coefficient of dextromethorphan, the electron-transfer coefficient, and the standard rate constant of the electrooxidation process were found to be 3.45?×?10?6 cm2 s?1, 0.65, and 1.67?×?10?3 cm s?1, respectively. A sensitive and timesaving determination procedure was developed for the analysis of dextromethorphan, and the corresponding analytical parameters were reported. Using this method, dextromethorphan was determined with an LOD and LOQ of 8.81 and 29.36 μM in a linear range of 2.5?×?10?4 to 3.3?×?10?3 M, respectively. The proposed amperometric method was successfully applied to the analysis of commercial pharmaceutical products (syrup and oral drop), and the results were in good agreement with the declared values.  相似文献

A simple, rapid and a highly selective method for direct electrochemical determination of acebutolol hydrochloride (AC) was developed. The developed method was based on the construction of three types of sensors conventional polymer (I), carbon paste (II) and modified carbon nanotubes (MCNTs) carbon paste (III). The fabricated sensors depend mainly on the incorporation of acebutolol hydrochloride with phosphotungstic acid (PTA) forming ion exchange acebutolol‐phosphotungstate (AC‐PT). The performance characteristics of the proposed sensors were studied. The sensors exhibited Nernstian responses (55.6 ± 0.5, 57.14 ± 0.2 and 58.6 ± 0.4 mV mol L?1) at 25 °C over drug concentration ranges (1.0 × 10?6‐1.0 × 10?2, 1.0 × 10?7‐1.0 × 10?2 and 5.0 × 10?8‐1.0 × 10?2 mol L?1 with lower detection limits of (5.0 × 10?7, 5.0 × 10?8 and 2.5 × 10?8 mol L?1 for sensors (I), (II) and (III), respectively. The influence of common and possible interfering species, pharmaceutical additives and some related pharmacological action drugs was investigated using separate solution method and no interference was found. The stability indicating using forced degradation of acebutolol hydrochloride was studied. The standard addition method was used for determination of the investigated drug in its pharmaceutical dosage forms and biological fluids. The results were validated and statistically analysed and compared with those from previously reported methods.  相似文献
