A rapid, specific, and sensitive fluorescence polarization immunoassay (FPIA) method was developed to detect residues of furazolidone, a forbidden drug in feed. An immuno-hapten was designed for monoclonal antibody preparation. Furthermore, seven novel tracers were synthesized and the “heterology tracer” could improve the sensitivity of FPIA significantly. An optimized FPIA method was established with a cross-reactivity of less than 0.1%, IC50 of 5.5 ng/mL. The limits of detection (LOD) calculated from feed samples were 0.5–0.9 ng/mL. Recoveries of fortified samples at levels of 5, 20, and 50 ng/mL ranged from 79.0% to 85.0%. The coefficients of variation were less than 12%.  相似文献

A chemiluminescent competitive indirect enzyme-linked immunosorbent assay, based on a mutant single-chain variable fragment (scFv), was developed to detect a broad range of fluoroquinolones (FQs) in fish and shrimp matrices. In this study, the best scFvC4A9H1_mut2 was adopted, which showed 10-fold improved affinity to sarafloxacin (SAR), difloxacin (DIF), and trovafloxacin (TRO), while the affinity to other FQs was fully inherited from wild-type scFvC4A9H1. In the optimized generic test, scFvC4A9H1_mut2 in combination with norfloxacin–ovalbumin conjugate and horseradish peroxidase-labeled anti-c-myc 9E10 antibody showed 50 % binding inhibition (IC50) at 0.12 μg kg?1 for norfloxacin in buffer. Screening for the class of FQ antibiotics is accomplished using a simple, rapid extraction carried out with ethanol/acetic acid (99:1, v/v). This common extraction was able to detect 20 FQ residues such as s ciprofloxacin (CIP), danofloxacin, DIF, enoxacin, enrofloxacin (ENR), fleroxacin, amifloxacin, flumequine, levofloxacin, lomefloxacin hydrochloride, marbofloxacin, norfloxacin (NOR), ofloxacin, orbifloxacin, pazufloxacin, pefloxacin-d5 (PEF), prulifloxacin, SAR, sparfloxacin, and TRO in fish and shrimp. The limit of detection (LOD) for NOR was 0.2 μg kg?1 and the LODs for CIP and ENR were all <0.2 μg kg?1. Values of LODs inferred from the cross-reactivity data will range from approximately 0.23 μg kg?1 for PEF to 2.1 μg kg?1 for TRO. Field fish and shrimp samples were analyzed and compared to the results obtained from liquid chromatography tandem mass spectrometric method. All five instances (from 0.25 to 15.6 μg kg?1) in which FQs were present at concentrations near or above the assay LOD were identified as positive by the newly developed assay, demonstrating the usefulness of this assay as a screening tool.

An indirect competitive enzyme-linked immunosorbent assay (icELISA) based on polyclonal antibody for the estrogen diethylstilbestrol (DES) was developed. With this aim, two different haptens mono-O-3-carboxypropyldiethylstilbestrol (DES-CP) and mono-O-carboxymethyldiethylstilbestrol (DES-CM) with carboxylic group that preserve the molecular structure character of diethylstilbestrol were synthesized. The haptens were conjugated with the carder proteins bovine serum albumin (BSA) by mixed-anhydride method for immunogen and conjugated with ovalbumin (OVA) by active ester method for coating antigen. Polyclonal antibodies for diethylstilbestrol were raised by immunizing mice with immune antigen DES-CP-BSA. Under optimized system, the lowest limit of detection (LLD) of diethylstilbestrol was 0.01 ng/mL, and IC50= 1.02 ng/mL. Its analogs were tested and no obvious cross-reactivity was found to anti-diethylstilbestrol antibody. DES-fortified water samples were determined by simple dilution to diminish the matrix effect. The comparison between the amount of DES estimated by ELISA and the amount added indicates good agreement for all water samples tested, with mean recovery values ranging from 86% to 120.2%.  相似文献

A time-resolved fluoroimmunoassay for the specific determination of methyl-3-quinoxaline-2-carboxylic acid in animal tissues, a marker residue of olaquindox, was developed. The IC50 of the assay was found to be 1.46 ± 0.19 ng/mL of methyl-3-quinoxaline-2-carboxylic acid in phosphate-buffered saline samples and the detection limit was 0.16 ± 0.03 ng/mL. For porcine liver and muscle samples spiked with 5, 10, and 15 ng/g, the recovery ranges were 95.7–112.3% and 98.5–116.2% and the coefficients of variation were 9.3–11.5% and 8.9–14.2%, respectively. The time-resolved fluoroimmunoassay results correlated well with high performance liquid chromatography results (correlation coefficients of 0.991 for liver and 0.988 for muscle). This study suggests that this method is simple, fast, and sensitive for the high-throughput determination of methyl-3-quinoxaline-2-carboxylic acid in animal tissues.  相似文献

This article describes an up-conversion immunoassay (UCIA) method for the determination of diethylstilbestrol with homogenous and rapid properties. Biotinylated diethylstilbestrol was used as a bridge for the formation of a streptavidinated beads-biotin-diethylstilbestrol‐antibody-acceptor beads complex in the immunoassay. The complex gave out imitation fluorescence at a wavelength of 680 nm when its excitation wavelength was at 615 nm as a result of the generation of singlet oxygen. The optimal test conditions and analytical performance of the method were studied. The optimized reaction time of ≥ 80 min was determined. The best buffer of phosphate buffer containing the surfactant of Tween 20 was established. Additionally, the sound analysis result can be obtained with highest antibody concentration, which was set in the experiments. Calibration curve, fit by 4-parameters formulation, resulted in R2 values of 0.9989, a linear range from 0.025 to 12.5 ng/mL and the limit of detection (LOD) of 0.01 ng/mL. The results of trace diethylstilbestrol concentrations in water samples showed recoveries ranging from 92.0% to 95.0%, and coefficients of variation between 3–8%. The data suggest that UCIA method is a good method with sensitivity for the determination of diethylstilbestrol in water.  相似文献

A novel, simple and rapid method, termed dispersive liquid–liquid microextraction with solidification of floating organic drop coupled to high performance liquid chromatography, was developed for analysis of three phenolic oestrogens including diethylstilbestrol, dienestrol and hexestrol in human urine and water samples. The parameters of dispersive liquid–liquid microextraction with solidification of floating organic drop procedure including sample pH, type and volume of disperser solvent, and type and volume of extraction solvent were optimised. High performance liquid chromatography was applied for the phenolic oestrogens’ analysis. Under the optimum extraction and detection conditions, excellent analytical performances were attained. Good linear relationships (r ≥ 0.998) between peak area and concentration for diethylstilbestrol and dienestrol were optimised from 0.1 to 20 µg/mL, for hexestrol from 2 to 50 µg/mL. Method detection limits of 28.6–666.7 ng/mL were achieved. Satisfactory relative recoveries ranging from 72% to 122% were determined for urine, lake and tap water samples, with relative standard deviations (RSDs, n = 6) of 1.5–9.8%. The developed dispersive liquid–liquid microextraction with solidification of floating organic drop-high performance liquid chromatography method has a great potential in routine residual analysis of trace phenolic oestrogens in biological and water samples.  相似文献

A simple synthetic method for haptens of organophosphorus (OP) pesticides with a spacer arm (aminocarboxylic acid) attached at the pesticide thiophosphate group was developed. While the previous synthetic approach for this type of haptens requires seven steps, the present method involves only two steps. Using this method, five haptens of fenitrothion were synthesized and two of them were conjugated to proteins to be used as immunogens for production of polyclonal antibodies. Using the antibodies and a coating antigen, an indirect competitive enzyme-linked immunosorbent assay (ELISA) for fenitrothion was developed, which showed an IC50 of 3.7 ng/mL with a detection limit of 0.5 ng/mL. A direct competitive ELISA using an enzyme tracer was also developed, which showed an IC50 of 5.6 ng/mL with a detection limit of 0.4 ng/mL. The antibodies in both assays showed negligible cross-reactivity with other OP pesticides except with the insecticide parathion-methyl only in the direct ELISA. Recoveries of fenitrothion from fortified lettuce and rice samples ranged 84-116 and 100-121%, respectively.  相似文献

The steroid 19-Nortestosterone (NT) has been illegally used in horse racing, dog games to boost physical performance, and also in animal husbandry to accelerate weight gain. This paper reports an indirect heterologous competitive enzyme-linked immunosorbent assay (icELISA) using polyclonal antibody for rapid, sensitive analysis of NT in animal urines. After derivation, NT haptens were conjugated to bovine serum albumin (BSA) and ovalbumin (OVA) through 1-Ethyl-3-(3-dimethylaminopropy) carbodiimide (EDC) method and mixed-anhydride technology respectively. Two antisera raised in rabbits and two coating antigens synthesized were screened and optimized using homologous and heterologous ELISA formats. The most sensitive heterologous icELISA (antisera generated from NT-17-BSA, with NT-3-OVA as coating antigen) gave an IC50 value of 27 ng/mL, with a dynamic range from 4 to 198 ng/mL, and the limit of detection (LOD) of 1.9 ng/mL. Except for cross-reactivity (CR) toward 17α-Nortestosterone (67%) and β-Boldenone (16.4%), no significant CR was observed for other chemical analogues tested. When applied to the authentic animal urines, the intra- and inter-assay precision values were below 8.4%, while the recoveries were in the range of 95–110%. The correlation coefficient between the established icELISA and LC-MS/MS method was greater than 0.98.  相似文献

Diethylstilbestrol (DES) is a synthetic estrogen that has ever been used worldwide. Polyclonal antibodies (PAbs) were used in immunoassay for detection of DES residues in environmental and agricultural samples in previous paper. In this paper, an indirect competitive enzyme-linked immunosorbent assay (icELISA) was developed based on monoclonal antibody (MAb) for the determination of diethylstilbestrol. Mono-o-carboxypropyldiethylstilbestrol (DES-CP) and mono-o-carboxymethyldiethylstilbestrol (DES-CME) were synthesized to be haptens. DES-CP was coupled to bovine serum albumin (BSA) to be an immunogen in BALB/c female mouse for MAb production. The MAb was characterized for specificity and affinity to DES in icELISA. Under the optimum condition, the icELISA showed an ICs0 of 9.8 ng/mL, the limit of detection (IC20) of 2.3 ng/mL and a working range of 2-42 ng/mL. Hexestrol and dienestrol exhibited cross-reactivity values were 44% and 27%, respectively. Cross-reactivity of natural estrogen 17β-estradiol was less than 0.1%. The influences of some factors such as salt concentration, pH and organic solvent concentration on the assay were evaluated. The concentrations of DES in the fortified water samples determined by the assay were correlated well with the fortification levels. The results were conf'm'ned with analysis by HPLC.  相似文献

A rapid easy-to-use trace level direct competitive enzyme-linked immunosorbent assay (dc-ELISA) detection of total residual malachite green (MG), crystal violet (CV) and their corresponding primary metabolites leucomalachite green (LMG) and leucocrystal violet (LCV) in fishery products in a single assay was developed. The monoclonal antibodies, anti-MG and anti-CV mAbs, were prepared using carboxyl-malachite green (CMG) and cationized bovine serum albumin (cBSA) conjugates as immunogen. The linear range for the quantitative detection of total MG, CV and their primary metabolites LMG and LCV was between 0.15 to 4.5?ng?mL?1 with a half maximal inhibitory concentration (IC50) at 0.56?±?0.04?ng?mL?1 (n?=?5). The anti-MG mAbs exhibited 98% cross-reactivity to CV, less than 0.1% cross-reactivity with LMG and LCV, and no cross-reactivity with chloramphenicol, enrofloxacin, sulfadiazine, and tetracycline. Application of the dc-ELISA in fish tissue samples gave a limit of detection (LOD) of 0.37?ng?g?1. The improved total detection lead to a recovery of 74.60?±?8.38% at 0.5?ng?g?1 and 87.47?±?12.83% at 2.0?ng?g?1 that was better than existing techniques. The dc-ELISA showed total MG in 7 out of 44 field fish samples that were confirmed with LC-MS/MS. The easy-to-use, inexpensive, and rapid dc-ELISA for the detection of total MG, CV and their corresponding primary metabolites holds promise for field applications.  相似文献

A novel nanocomposite electrode material constituted of gold nanoparticles (AuNPs), multi-walled carbon nanotubes (MWCNTs) and n-octylpyridinium hexafluorophosphate (OPPF6) ionic liquid was prepared and checked for the development of electrochemical (bio)sensing devices. AuNPs/MWCNTs/OPPF6 paste electrodes with micrometer dimensions (500 μm, i.d.) were constructed and applied to the determination of cortisol and androsterone hormones. Regarding cortisol determination, the microsized paste electrode was used to detect 1-naphtol generated upon addition of 1-naphthyl phosphate as enzyme substrate in the competitive immunoassay between alkaline phosphatase-labelled cortisol and cortisol. Squarewave voltammetry allowed determining the hormone within the 0.1- to 10-ng/mL linear range (r?=?0.990) with a detection limit of 15 pg/mL and a EC50 value of 0.46?±?0.06 ng/mL cortisol. The method was applied to the determination of cortisol in urine and serum samples containing a certified cortisol content. Moreover, a microsized enzyme biosensor prepared by bulk modification of the AuNPs/MWCNTs/OPPF6 electrode with the enzyme 3α-hydroxysteroid dehydrogenase was used for the determination of androsterone through the amperometric detection of reduced nicotinamide adenine dinucleotide. A calibration plot with a linear range between 0.1 and 120 μg/mL (r?=?0.993) and a limit of detection of 89 ng/mL were obtained. The biosensor was applied to the analysis of human serum spiked with androsterone at the 250 ng/mL concentration level.  相似文献

Ecgonine is suggested to be a promising marker of cocaine (COC) ingestion. A combined mass spectrometry (MS) and tandem MS (MS/MS) method was developed to simultaneously determine ecgonine and seven other metabolites of cocaine in human urine and whole blood with ultra-high-pressure liquid chromatography coupled with quadrupole time-of-flight mass spectrometry. The compounds were extracted from as little as 100 μL of sample by solid-phase extraction with a 96-well μElution solid-phase extraction plate. The protonated molecules or fragment ions at accurate mass acquired in MS mode were used to quantify specific analytes, following by dedicated MS/MS identification. The assay was linear in the range from 5 to 50-100 ng/mL for urine samples, except for ecgonine methyl ester (10-200 ng/mL) and ecgonine (40-400 ng/mL), and was linear from 1-2 to 50 ng/mL for whole blood samples, except for ecgonine methyl ester (20-1,000 ng/mL) and ecgonine (40-2,000 ng/mL). The correlation coefficients were all greater than 0.99. The limits of detection ranged from 0.2 to 16 ng/mL, and the lower limits of quantification ranged from 1 to 40 ng/mL. The repeatability and intermediate precision were 18.1 % or less. The accuracy was in the range from 80.0 to 122.9 %, process efficiencies were in the range from 8.6 to 177.4 %, matrix effects were in the range from 28.7 to 171.0 %, and extraction recoveries were in the range from 41.0 to 114.3 %, except for ecgonine (12.8 % and 9.3 % at low and high concentrations, respectively). This method was highly sensitive in comparison with previously published methods. The validated method was successfully applied to the analysis of real samples derived from forensic cases, and the results verified that, on the basis of data from four positive samples, ecgonine is a promising marker of cocaine ingestion.

A novel, sensitive, and robust method has been developed to detect 9 β2-agonists in porcine urine to monitor illegal use of β2-agonists in swine rearing. The method based on the molecular imprinted polymer (MIP) rapid extraction followed ultra-performance liquid chromatography coupled tandem mass spectrometry (UPLC-MS/MS) detection. The cleaning efficiency of MIP cartridges was demonstrated by comparing with common ion exchange solid phase extraction. The presented method was validated in accordance with the European Commission Decision 2002/657/EC. The linearity, decision limit (CCα), detection capability (CCβ), recovery, precision, robustness, and stability were studied in detail. CCα and CCβ values were from 0.006 ng/mL to 0.03 ng/mL and from 0.02 ng/mL to 0.08 ng/mL, respectively. The mean recoveries and repeatability varied from 68.8% to 94.2% and from 2.8% to 10.1%. The proposed method was applied to test 170 porcine urine samples from the Shaanxi province in China and two urine samples were confirmed as clenbuterol positive and the concentrations of clenbuterol in positive urine samples were about 0.08 ng/mL and 0.1 ng/mL, respectively. The developed method was demonstrated to be more sensitive and robust for the determination of 9 β2-agonists in porcine urine. The method was proven to be simple and easy in operation with high selectivity and good reproducibility.  相似文献

Four haptens of the organophosphorus (OP) insecticide diazinon were synthesized to develop enzyme-linked immunosorbent assays (ELISAs) for this pesticide. One of them was conjugated to KLH to be used as the immunogen for production of monoclonal antibodies. By using the antibodies and a coating antigen, an indirect competitive ELISA was developed, which showed an IC50 of 4.0 ng/mL with a detection limit of 0.7 ng/mL. A direct competitive ELISA using an enzyme tracer was also developed, which showed an IC50 of 6.0 ng/mL with a detection limit of 0.9 ng/mL. The antibodies in both assays showed negligible cross-reactivity with metabolites of diazinon and other OP pesticides. Recovery of diazinon from fortified lettuce and rice samples was satisfactory except at the fortified concentration of 100 ppb.  相似文献

Abstract

This paper reports on the preparation of a monoclonal antibody (Mab) against maduramicin (MD) and the development of a simple and sensitive ELISA for MD in chicken tissues. MD was conjugated to bovine serum albumin for immunogens and ovalbumin for coating antigens by mixed anhydride (MA) and active ester (AE) methods. Hybridoma cells were generated using spleen cells from a mouse immunized with MD-BSA conjugate. After screening with ELISA, the Mab with high anti-interference ability and high affinity was selected, and it exhibited negligible cross-reactivity with other usual-used polyether antibiotics. After optimization, the developed ELISA showed an IC50 value of 2.12 ± 0.46 ng/ml (n = 20). Chicken muscle and liver samples were extracted with methanol-8% NaCl solution (4:1) and then directly diluted with PBS-10% methanol for analysis. The recoveries of MD from spiked chicken tissues at levels of 40–480 µg/kg ranged from 81.3% to 91.3% with variation coefficients of 5.2–12.1%, and the detection limits were 6.5 µg/kg in muscle and 9.2 µg/kg in liver, respectively (n = 20).  相似文献

This study was designed to evaluate the isolated flavonoids (chrysin 1, and umbelliferone 2) from Potentilla evestita for cytotoxic, antitumour-promoting and inhibition of protein denaturation activities. The results showed marked cytotoxic effect of compounds 1 and 2 in brine shrimp cytotoxic assay at various concentrations with LD50 of 34.5 and 31.8 mg/mL, respectively. In Epstein–Barr-virus early antigen activation assay, both compounds 1 and 2 illustrated significant antitumour-promoting effect with IC50 values of 462 and 308 mol ratio/32 pmol TPA, respectively. The cytotoxic and antitumour-promoting effects of compounds were strongly supported by inhibition of protein denaturation activity with IC50 of 119 and 112 μg/mL, respectively. In conclusion, both compounds possess strong cytotoxic, antitumour-promoting and inhibition of protein denaturation activities.  相似文献

Artesunate (AS), a hemisuccinate derivative of artemisinin, is readily soluble in water and can easily be used in formulations for parenteral treatment of severe malaria. AS is rapidly hydrolyzed to the active metabolite dihydroartemisinin (DHA) and primarily eliminated by biliary excretion after glucuronidation. To investigate systematically the AS metabolism and pharmacokinetics, a novel liquid chromatography–tandem mass spectrometry (LC-MS/MS) method for the simultaneous quantification of AS and its metabolites DHA and DHA glucuronide (DHAG) in human plasma samples was developed. Compared to previous methods, our method includes for the first time the quantification of the glucuronide metabolite using a newly synthesized stable isotope-labeled analogue as internal standard. Sample preparation was performed with only 50 μL plasma by high-throughput solid-phase extraction in the 96-well plate format. Separation of the analytes was achieved on a Poroshell 120 EC-C18 column (50*2.1 mm, 2.7 μm, Agilent Technologies, Waldbronn, Germany). The method was validated according to FDA guidelines. Calibration curves were linear over the entire range from 1 to 2,500 nM (0.4–961.1 ng/mL), 165 to 16,500 nM (46.9–4,691.8 ng/mL), and 4 to 10,000 nM (1.8–4,604.7 ng/mL) for AS, DHA, and DHAG, respectively. Intra- and interbatch accuracy, determined as a deviation between nominal and measured values, ranged from ?5.7 to 3.5 % and from 2.7 to 5.8 %, respectively. The assay variability ranged from 1.5 to 10.9 % for intra- and interbatch approaches. All analytes showed extraction recoveries above 85 %. The method was successfully applied to plasma samples from patients under AS treatment.

Snake venom contains bioactive materials for drug development, diagnosis, and treatment. After separating and purifying the kallikrein-like enzyme (AHP-Ka) from Agkistrodon halys pallas venom for the first time, a monoclonal antibody against AHP-Ka was prepared and characterized. An indirect sandwich enzyme-linked immunosorbent assay (ELISA) based on the monoclonal antibody was developed and validated for the pharmacokinetic analysis of AHP-Ka in rat plasma. The method was calibrated using rat plasma and 1:100 dilution of plasma was selected to prepare a calibration curve to validate the precision, accuracy, and stability of the ELISA method. A good linear relationship was obtained in a working range from 3.9 ng/mL to 62.5 ng/mL with a limit of detection of 2.94 ng/mL. Intra- and inter-batch precision were less than 10%. The average recovery ranged from 94.6% to 104.4% in rat plasma at the concentrations of 5 ng/mL, 15 ng/mL, and 45 ng/mL, respectively. The ELISA method was successfully used for the pharmacokinetic study of AHP-Ka in Sprague-Dawley rat plasma after intravenous administration. The work is expected to contribute to future preclinical development of AHP-Ka.  相似文献

This study was designed to investigate antioxidant and anticholinesterase potential of Iris germanica var; florentina. Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitory potential of plant samples were investigated by Ellman’s assay. Antioxidant activity was performed using DPPH, H2O2 and ABTS free radical scavenging assays. Total phenolics and flavonoids contents were expressed in mg GAE/g dry weight and mg RTE/g, respectively. In AChE inhibition assay, Ig.Fl, Ig.Sp and Ig.Cf fractions exhibited highest activity with IC50 values of < 0.1, 5.64 and 19 μg/mL, respectively. In BChE inhibitory assay, Ig.Fl, Ig.Sp, Ig.Cf and Ig.Cr were most active with IC50 of < 0.1, < 0.1, 31 and 78 μg/mL, respectively. In DPPH assay, Ig.Fl and Ig.Cf exhibited highest inhibition of free radicals, 80.52% (IC50 = 9 μg/mL) and 78.30% (IC50 = 8 μg/mL), respectively. In ABTS assay Ig.Cr, Ig.Cf, Ig.Fl and Ig.Sp exhibited IC50 values of < 0.1, 2, 2 and 3 μg/mL, respectively.  相似文献

Cyphostemma digitatum stem and roots extracts were investigated for antioxidant, antimicrobial, urease inhibition potential and phytochemical analysis. Phytochemical screening of the roots and stem extract revealed the presence of secondary metabolites including flavonoids, alkaloids, coumarins, saponins, terpenoids, tannins, carbohydrates/reducing sugars and phenolic compounds. The methanolic extracts of the roots displayed highest antioxidant activity (93.518%) against DPPH while the crude methanolic extract of the stem showed highest antioxidant activity (66.163%) at 100 μg/mL concentration. The methanolic extracts of both stem and roots were moderately active or even found to be less active against the selected bacterial and fungal strains (Tables S2 and S3). The roots extract (methanol) showed significant urease enzyme inhibition activity (IC50 = 41.2 ± 0.66; 0.2 mg/mL) while the stem extract was found moderately active (IC50 = 401.1 ± 0.58; 0.2 mg/mL) against thiourea (IC50 = 21.011; 0.2 mg/mL).  相似文献
