Graphene/Fe3O4 nanocomposite was prepared for the immobilization of hemoglobin (Hb) to improve the electron transfer between Hb and glass carbon electrode (GCE). The characterization of nanocomposites was described by transmission electron microscopy, Fourier transform infrared, Raman spectroscopy, and X-ray photoelectron spectroscopy, respectively. The electrochemistry of Hb on the graphene/Fe3O4-based GCE was investigated by cyclic voltammetry and amperometric measurement. The modified electrode showed a wide linear range from 0.25 μmol/L to 1.7 mmol/L with a correlation coefficient of 0.9967. The detection limit of the H2O2 biosensor was estimated at 6.0?×?10?6?mol/L at a signal-to-noise ratio of 3.  相似文献

A sensitive hydrogen peroxide (H2O2) biosensor was developed based on a reduced graphene oxide|carbon ceramic electrode (RGO|CCE) modified with cadmium sulfide‐hemoglobin (CdS‐Hb). The electron transfer kinetics of Hb were promoted due to the synergetic function of RGO and CdS nanoparticles. The transfer coefficient (α) and the heterogeneous electron transfer rate constant (ks) were calculated to be 0.54 and 2.6 s?1, respectively, indicating a great facilitation achieved in the electron transfer between Hb and the electrode surface. The biosensor showed a good linear response to the reduction of H2O2 over the concentration range of 2–240 µM with a detection limit of 0.24 µM (S/N=3) and a sensitivity of 1.056 µA µM?1 cm?2. The high surface coverage of the CdS‐Hb modified RGO|CCE (1.04×10?8 mol cm?2) and a smaller value of the apparent Michaelis? Menten constant (0.24 mM) confirmed excellent loading of Hb and high affinity of the biosensor for hydrogen peroxide.  相似文献

In this paper, a gold nanoparticle-modified indium tin oxide electrode (Au/ITO) was prepared without the use of any cross-linker or stabilizer reagent. The prepared Au/ITO was used as a new platform to achieve the direct electron transfer between Hb and the modified electrode. The proposed electrode exhibited a pair of well-defined redox peaks with a formal potential of ?0.073 V (vs. Ag/AgCl). The immobilized Hb showed excellent electrocatalytic activity toward H2O2 and the electrocatalytic current values were linear with the increasing concentration of H2O2 ranging from 1.0?×?10?6?M to 7.0?×?10?4?M. The detection limit was 2.0?×?10?7?M (S/N?=?3) and the Michaelis–Menten constant was calculated to be 0.2 mM. A new amperometric biosensor for hydrogen peroxide (H2O2) was developed by adsorbing hemoglobin (Hb) on an organic sol‐gel film. The organic sol‐gel was prepared using resorcinol and formaldehyde as monomers. This sol‐gel film shows a biocompatible microenvironment for retaining the native activity of the adsorbed Hb. The direct electron transfer between Hb and electrode is achieved. Hb adsorbed on the film shows an enzyme‐like catalytic activity for the reduction of H2O2. The reduction peak currents are proportional linearly to the concentration of hydrogen peroxide in the range of 6×10?8 to 3.6×10?6 M, with a detection limit of 2.4×10?8 M (S/N=3). This research enlarges the applications of organic sol‐gel materials in biosensor field.  相似文献

This paper reports a core-shell nanoparticle system coated on the carbon paste electrode (CPE) for determination of hydrogen peroxide. The amino-functionalized shell-magnetic core nanoparticles have been proven to be an effective material for Hb immobilization. The core-shell nanoparticle system was constructed by immobilizing hemoglobin (Hb) on amino-functionalized shell@magnetic core composite nanoparticles (NH2-SiO2-CoFe2O4) with the bridge of gold nanoparticles (AuNPs). Electrochemical impedance spectroscopy, cyclic voltammetry, and chronoamperometry were used to characterize the obtained biosensor. The Hb/AuNPs/NH2-SiO2-CoFe2O4/CPE showed a linear range from 1.9 × 10?6 to 4.6 × 10?3 M, with a detection limit of 6.3 × 10?7 M (S/N = 3) under the optimized experimental conditions. A good affinity was shown due to the small apparent Michaelis–Menten constant of 2.68 mM.  相似文献

A glassy carbon electrode was modified with PdO-NiO composite nanofibers (PdO-NiO-NFs) and applied to the electrocatalytic reduction of hydrogen peroxide (H2O2). The PdO-NiO-NFs were synthesized by electrospinning and subsequent thermal treatment, and then characterized by scanning electron microscopy, energy dispersive X-ray spectroscopy, X-ray diffraction, and X-ray photoelectron spectroscopy. Factors such as the composition and fraction of nanofibers, and of the applied potential were also studied. The sensor exhibits high sensitivity for H2O2 (583.43 μA?·?mM?1?·?cm?2), a wide linear range (from 5.0 μM to 19 mM), a low detection limit (2.94 μM at an SNR of 3), good long term stability, and is resistant to fouling.

Hemoglobin (Hb) was successfully immobilized on a gold electrode modified with gold nanoparticles (AuNPs) via a molecule bridge 1,6-hexanedithiol (HDT). The AFM images suggested that the HDT/gold electrode could adsorb more AuNPs. UV-vis spectra indicated that Hb on AuNPs/HDT film retained its near-native secondary structures. The electrochemical behaviors of the sensor were characterized with cyclic voltammetric techniques. The resultant electrode displayed an excellent electrocatalytical response to the reduction of hydrogen peroxide (H2O2). The linear relationship existed between the catalytic current and the H2O2 concentration ranging from 5.0 × 10?8 to 1.0 × 10?6 mol · L?1. The highly efficient H2O2 biosensor was fabricated on the basis of the complex films of hemoglobin (Hb), nano ZnO, chitosan (CHIT) dispersed solution and nano Au immobilized on glassy carbon electrode (GCE). Biocompatible ZnO‐CHIT composition provided a suitable microenvironment to keep Hb bioactivity (Michaelis‐Menten constant of 0.075 mmol L?1). The presence of nano Au in matrix could effectively enhance electron transfer between Hb and electrode. The electrochemical behaviors and effects of solution pH values were carefully examined in this paper. The (ZnO‐CHIT)‐Au‐Hb/GCE demonstrated excellently electrocatalytical ability for H2O2. This biosensor had a fast response to H2O2 less than 4 s and excellent linear relationships were obtained in the concentration range from1.94×10?7 to 1.73×10?3 mol L?1 with the detection limit of 9.7×10?8 mol L?1 (S/N=3) under the optimum conditions. Moreover, the stability and reproducibility of this biosensor were evaluated with satisfactory results.  相似文献

The direct electrochemistry of catalytically active cytochrome C (Cyt c) adsorbed together with a 3-dimensional network of chemically synthesized graphene on glassy carbon electrode has been readily obtained in aqueous phosphate buffer. Direct electrical communication between the redox center of Cyt c and the modified graphene-based electrode was established. The modified electrode was employed as a high-performance hydrogen peroxide (H2O2) biosensor. The Cyt c present in modified electrode exhibited a pair of quasi-reversible redox peaks with a midpoint potential of ?0.380 and ?0.2 V, cathodic and anodic, respectively. Investigations into the electrocatalytic activity of the modified electrode upon hydrogen peroxide exposure revealed a rapid amperometric response (5 s). Under optimized conditions, the linear range of response to H2O2 concentration ranged from 5 × 10?7 to 2 × 10?4 M with a detection limit of 2 × 10?7 M at a signal-to-noise ratio of 3. The stability, reproducibility, and selectivity of the proposed biosensor are discussed in relation to the morphology and composition of the modified electrode.  相似文献

A novel matrix, gold nanoparticles-bacterial cellulose nanofibers (Au-BC) nanocomposite was developed for enzyme immobilization and biosensor fabrication due to its unique properties such as satisfying biocompatibility, good conductivity and extensive surface area, which were inherited from both gold nanoparticles (AuNPs) and bacterial cellulose nanofibers (BC). Heme proteins such as horseradish peroxidase (HRP), hemoglobin (Hb) and myoglobin (Mb) were successfully immobilized on the surface of Au-BC nanocomposite modified glassy carbon electrode (GCE). The immobilized heme proteins showed electrocatalytic activities to the reduction of H2O2 in the presence of the mediator hydroquinone (HQ), which might be due to the fact that heme proteins retained the near-native secondary structures in the Au-BC nanocomposite which was proved by UV-vis and IR spectra. The response of the developed biosensor to H2O2 was related to the amount of AuNPs in Au-BC nanocomposite, indicating that the AuNPs in BC network played an important role in the biosensor performance. Under the optimum conditions, the biosensor based on HRP exhibited a fast amperometric response (within 1 s) to H2O2, a good linear response over a wide range of concentration from 0.3 μM to 1.00 mM, and a low detection limit of 0.1 μM based on S/N = 3. The high performance of the biosensor made Au-BC nanocomposite superior to other materials as immobilization matrix.  相似文献

A novel hydrogen peroxide (H2O2) biosensor was developed by immobilizing hemoglobin on the gold colloid modified electrochemical pretreated glassy carbon electrode (PGCE) via the bridging of an ethylenediamine monolayer. This biosensor was characterized by UV-vis reflection spectroscopy (UV-vis), electrochemical impendence spectroscopy (EIS) and cyclic voltammetry (CV). The immobilized Hb exhibited excellent electrocatalytic activity for hydrogen peroxide. The Michaelis–Menten constant (K m) was 3.6 mM. The currents were proportional to the H2O2 concentration from 2.6 × 10−7 to 7.0 × 10−3 M, and the detection limit was as low as 1.0 × 10−7 M (S/N = 3).  相似文献

A novel nanocomposite integrating the good biocompatibility of polyacrylic resin nanoparticles (PAR) and the good conductivity of colloidal gold nanoparticles was proposed to construct the matrix for the immobilization of hemoglobin (Hb) on the surface of a glassy carbon electrode (GCE). UV‐vis spectra demonstrated that Hb preserved its native structure after being entrapped into the composite film. The direct electrochemistry of hemoglobin (Hb) in this nanocomposite films showed a pair of well‐defined and quasi‐reversible cyclic voltammetric peaks with a formal potential of ?0.307 mV and a constant electron transfer rate of 2.51±0.2 s?1. The resultant amperometric biosensor showed fast responses to the analytes with excellent detection limits of 0.2 µM for H2O2 and 0.89 µM for TCA (S/N=3), and high sensitivity of 1108.6 for H2O2 and 77.14 mA cm?2 M?1 for TCA, respectively. The linear current response was found in the range from 0.59 to 7.3 µM (R2=0.9996) for H2O2 and from 5 to 85 µM (R2=0.9996) for TCA, while the superior apparent Michaelis–Menten constant was 0.012 mM for H2O2 and 0.536 mM for TCA, respectively. Therefore, the PAR‐Au‐Hb nanocomposite as a novel matrix opens up a possibility for further study on the direct electrochemistry of other proteins.  相似文献

A novel method for preparation of hydrogen peroxide biosensor was presented based on immobilization of hemoglobin (Hb) on carbon‐coated iron nanoparticles (CIN). CIN was firstly dispersed in a chitosan solution and cast onto a glassy carbon electrode to form a CIN/chitosan composite film modified electrode. Hb was then immobilized onto the composite film with the cross‐linking of glutaraldehyde. The immobilized Hb displayed a pair of stable and quasireversible redox peaks and excellent electrocatalytic reduction of hydrogen peroxide (H2O2), which leading to an unmediated biosensor for H2O2. The electrocatalytic response exhibited a linear dependence on H2O2 concentration in a wide range from 3.1 μM to 4.0 mM with a detection limit of 1.2 μM (S/N=3). The designed biosensor exhibited acceptable stability, long‐term life and good reproducibility.  相似文献

A novel nitrite biosensor was constructed by simultaneous immobilization of hemoglobin (Hb) and a room temperature ionic liquid, octylpyridinium chloride ([OcPy][Cl]), on multi-walled carbon ionic liquid electrode (MWILE). The direct electron transfer of Hb showed a pair of redox peaks with a formal potential of ?0.187 V vs. Ag/AgCl in pH 5.0 acetate buffer solution. Nitrite (NO2 ?) catalysis on the modified electrode was investigated by cyclic voltammetry and amperometry. The biosensor exhibited a wide linear range for NO2 ? detection from 0.01 to 15 mM, with a detection limit (3σ) of 1.46 μM. MWILE provided an excellent matrix for protein immobilization and biosensor fabrication which could be used for the determination of NO2 ? with a low detection limit, fast response, long linearity, and excellent sensitivity.  相似文献

We report on a novel electrochemical biosensor that was fabricated by immobilizing hemoglobin (Hb) onto the surface of a gold electrode modified with a chitosan@Fe3O4 nano-composite. The Fe3O4 nanoparticles were prepared by co-precipitation and have an average size of 25 nm. They were dispersed in chitosan solution to obtain the chitosan@Fe3O4 nano-composite particles with an average diameter of 35 nm as verified by transmission electron microscopy. X-ray diffraction patterns and Fourier transform IR spectroscopy confirmed that the crystallite structure of the Fe3O4 particles in the nano-composite has remained unchanged. At pH 7.0, Hb gives a pair of redox peaks with a potential of about ?0.21 V and ?0.36 V. The Hb on the film maintained its biological activity and displays good electrocatalytic reduction activity towards hydrogen peroxide. The linear range for the determination of H2O2 is from 2.3 μM to 9.6 mM, with a detection limit at 1.1 μM concentration (at S/N?=?3). The apparent Michaelis-Menten constant is 3.7 mM and indicates the high affinity of Hb for H2O2. This biosensor also exhibits good reproducibility and long-term stability. Thus, it is expected to possess potential applications in the development of the third-generation electrochemical biosensors.

Titanium dioxide nanorods (TNR) were grown on a titanium electrode by a hydrothermal route and further employed as a supporting matrix for the immobilization of nafion-coated horseradish peroxidase (HRP). The strong electrostatic interaction between HRP and TNR favors the adsorption of HRP and facilitates direct electron transfer on the electrode. The electrocatalytic activity towards hydrogen peroxide (H2O2) was investigated via cyclic voltammetry and amperometry. The biosensor exhibits fast response, a high sensitivity (416.9 μA·mM?1), a wide linear response range (2.5 nM to 0.46 mM), a detection limit as low as 12 nM, and a small apparent Michaelis-Menten constant (33.6 μM). The results indicate that this method is a promising technique for enzyme immobilization and for the fabrication of electrochemical biosensors.

A robust and effective composite film combined the benefits of Nafion, room temperature ionic liquid (RTIL) and multi‐wall carbon nanotubes (MWNTs) was prepared. Hemoglobin (Hb) was successfully immobilized on glassy carbon electrode surface by entrapping in the composite film. Direct electrochemistry and electrocatalysis of immobilized Hb were investigated in detail. A pair of well‐defined and quasi‐reversible redox peaks of Hb was obtained in 0.10 mol·L?1 pH 7.0 phosphate buffer solution (PBS), indicating that the Nafion‐RTIL‐MWNTs film showed an obvious promotion for the direct electron transfer between Hb and the underlying electrode. The immobilized Hb exhibited an excellent electrocatalytic activity towards the reduction of H2O2. The catalysis current was linear to H2O2 concentration in the range of 2.0×10?6 to 2.5×10?4 mol·L?1, with a detection limit of 8.0×10?7 mol·L?1 (S/N=3). The apparent Michaelis‐Menten constant (Kmapp) was calculated to be 0.34 mmol·L?1. Moreover, the modified electrode displayed a good stability and reproducibility. Chitosan (CS) was chosen for dispersing multi‐wall carbon nanotubes (MWNTs) to form a stable CS‐MWNTs composite, which was first coated on the surface of a glassy carbon electrode to provide a containing amino groups interface for assembling colloidal gold nanoparticles (GNPs), followed by the adsorption of hemoglobin (Hb). Repeating the assembly step of GNPs and Hb resulted in {Hb/GNPs}n multilayers. The assembly of GNPs onto CS‐MWNTs composites was confirmed by transmission electron microscopy. The consecutive growth of {Hb/GNPs}n multilayers was confirmed by cyclic voltammetry and UV‐vis absorption spectroscopy. The resulting system brings a new platform for electrochemical devices by using the synergistic action of the electrocatalytic activity of GNPs and MWNTs. The resulting biosensor displays an excellent electrocatalytic activity and rapid response for hydrogen peroxide. The linear range for the determination of H2O2 was from 5.0×10?7 to 2.0×10?3 M with a detection limit of 2.1×10?7 M at 3σ and a Michaelis–Menten constant KMapp value of 0.19 mM.  相似文献

A novel H2O2 amperometric biosensor based on the electrodeposition of gold nanoparticles (AuNPs) and CdS quantum dots (CdS QDs) onto a carbon paste electrode (CPE) and immobilizing hemoglobin (Hb) with ionic liquid (IL), is presented in this article. The modification process of the electrode was monitored by scanning electron microscopy (SEM), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Due to synergistic effects of AuNPs, CdS QDs and IL, the biosensor exhibited high stability and good bioelectrocatalytic ability to H2O2 with a linear concentration range from 10 to 750 µM and a detection limit of 4.35 µM (S/N=3).  相似文献
