An analytical method for the determination of three polyamines (putrescine, spermidine, and spermine) and five acetylpolyamines [N1-acetylspermidine (N1AcSpd), N8-acetylspermidine (N8AcSpd), N1-acetylspermine, N1,N8-diacetylspermidine, and N1,N12-diacetylspermine] involved in the polyamine catabolic pathway has been developed using a hybrid tandem mass spectrometer. Heptafluorobutyryl (HFB) derivatives of these compounds and respective internal standards labeled with stable isotopes were analyzed simultaneously by TOF MS, based on peak areas appearing at appropriate m/z values. The isomers, N1AcSpd and N8AcSpd were determined from their fragment ions, the acetylamidopropyl and acetylamidobutyl groups, respectively, using MS/MS with 13C2-N1AcSpd and 13C2-N8AcSpd which have the 13C2-acetyl group as an internal standard. The TOF MS method was successfully applied to measure the activity of enzymes involved in polyamine catabolic pathways, namely N1-acetylpolyamine oxidase (APAO), spermine oxidase (SMO), and spermidine/spermine N1-acetyltransferase (SSAT). The following natural substrates and products labeled with stable isotopes considering the application to biological samples were identified; for APAO, [4,9,12-15N3]-N1-acetylspermine and [1,4,8-15N3]spermidine (15N3-Spd), respectively; for SMO, [1,4,8,12-15N4]spermine and 15N3-Spd, respectively; and for SSAT, 15N3-Spd and [1,4,8-15N3]-N1-acetylspermidine, respectively.  相似文献

The active ingredient of ecstasy, N-methyl-3,4-methyldioxyphenylisopropylamine (MDMA) can be manufactured by a number of easy routes from simple precursors. We have synthesised 45 samples of MDMA following the five most common routes using N-precursors from 12 different origins and three different precursors for the aromatic moiety. The 13C and 15N contents of both the precursors and the MDMA samples derived therefrom were measured by isotope ratio mass spectrometry coupled to an elemental analyser (EA-IRMS). We show that within-pathway correlation between the 15N content of the precursor and that of the derived MDMA can be strong but that no general pattern of correlation can be defined. Rather, it is evident that the δ15N values of MDMA are strongly influenced by a combination of the δ15N values of the source of nitrogen used, the route by which the MDMA is synthesised, and the experimental conditions employed. Multivariate analysis (PCA) based on the δ15N values of the synthetic MDMA and of the δ15N and δ13C values of the N-precursors leads to good discrimination between the majority of the reaction conditions tested.  相似文献

Mercury cyanide complexes of alkyldiamines (16), [Hg(L)(CN)2] (where L?=?en (1,2-diaminoethane), pn (1,3-diaminopropane), N-Me-en, N, N′-Me2-en, N, N′-Et2-en, and N, N′-ipr2-en), have been synthesized and characterized by elemental analysis, IR, 13C, and 15N solution NMR in DMSO-d6, as well as 13C, 15N, and 199Hg solid-state NMR spectroscopy. Complexes 1 and 2 have been studied computationally, built and optimized by GAUSSIAN03 using DFT at B3LYP level with LanL2DZ basis set. Binding modes of en and bn (where bn?=?1,4-diaminobutane) toward Hg(CN)2 are completely different. Complexes with en and pn show chelating binding to Hg(II), while bn behaves as a bridging ligand to form a polymeric structure, [Hg(CN)2-bn] [B.A. Al-Maythalony, M. Fettouhi, M.I.M. Wazeer, A.A. Isab. Inorg. Chem. Commun., 12, 540 (2009).]. The solution 13C NMR of the complexes demonstrates a slight shift of the ?C≡N (0.9 to 2?ppm) and ?C–NH2 (0.25 to 6?ppm) carbon resonances, while the other resonances are relatively unaffected. 15N labeling studies have shown involvement of alkyldiamine ligands in coordination to the metal. The principal components of the 13C, 15N, and 199Hg shielding tensors have been determined from solid-state NMR data. Antimicrobial activity studies show that the complexes exhibit higher antibacterial activities toward various microorganisms than Hg(CN)2.  相似文献

Two new complexes, [Cu(L1){N(CN)2}]·ClO4 (1) (L1 is 1,8-dimethyl-1,3,6,8,10,13-hexa-azacyclotetradecane) and [Co(L2)(N3)2]·ClO4 (2) (L2 is 5,7,7,12,14,14-hexamethyl-1,4,8,11-tetra-azacyclotetradecane) have been synthesized and characterized. The compounds crystallize in the monoclinic system P21 space group for 1 and P21/n for 2. Single crystal X-ray analysis reveals that the compound 1 assumes a one-dimensional structure via hydrogen-bonding interactions, in which each Cu(II) ion is coordinated by four nitrogen atoms from ligand L1 and one nitrogen atom from [N(CN)2] anion. For compound 2, each Co(III) ion is coordinated by four nitrogen atoms of ligand L2 and two nitrogen atoms from N3 anion.  相似文献

Abstract

A new analytical Spectrophotometric method has been developed for the determination of dissolved O2 in the range 4.6 × 10?6?4.1 × 10?5M, based on the CS2N? 3/HSO? 3/O2 reaction. In one single experiment 2.3 × 10?5 M of oxygen can be determined with an uncertainty of ± 7 × 10?7M for a confidence level of 95%.  相似文献

Complexes of fullerenes C60 and C70 with cobalt(II) and manganese(II) tetraphenylporphyrinates of compositions Mn(TPP)·(C60)2(CS2)1.5 (1), Mn(TPP)·C70(CS2) x , wherex<=1.25 (2), Co(TPP)·C60(CS2)0.5 (3), and Co(TPP)·C70(CS2) x , wherex<0.25 (4), were synthesized and studied by ESR spectroscopy. At 77 K, complexes1 and2 have singlet ESR spectra characteristic of the low-spin (S=1/2) state of MnII, withg=2.002 and linewidths of 250 G and 300 G, respectively, and differing significantly from that of the initial MnII(TPP) (g 1=5.9 andg=2.0,S=5/2). The spectra of complexes1 and2 exposed to oxygen exhibit hyperfine structure due to interaction with55Mn and14N nuclei. The ESR spectra of complexes3 and4 are asymmetric (<g>=2.4, ΔH pp=(500–600) G), which is due to the overlap of parallel and perpendicular spectral components. The absence of ESR signals from C60 .− and C70 .− radical anions makes it possible to conclude that the formation of complexes1–4 is not accompanied by electron transfer from Co(TPP) and Mn(TPP) to fullerences C60 and C70. The new cyclic phosph(V)azane ligand [(C6H5N)P(O)H]2 (2) is obtained from the reaction between PCl3 and PhNH2 in toluene followed by controlled hydrolysis of the product in an H2O–CHCl3 solution. Compound 2 is the first example of P(V) dimer [(µ-NC6H5)P(H)=O]2, a P2N2 ring with two P(O)H moieties. The reaction of 2 with ZnCl2 in a molar ratio of 1?:?1 in tetrahydrofuran yields the cyclophosph(V)azane complex Cl2Zn[(C6H5N)P(O)H]2 (3) in which Zn–O bonds form directly between a cyclic phosph(V)azane ligand and Zn(II). The products have been characterized by infrared, multinuclear (1H, 31P, 13C) NMR, mass spectrometry, and elemental analysis.  相似文献

Treatment of 2- and 4-amino-3-(tert-butyl-NNO-azoxy)pyridines with nitrating agents (N2O5or NO2BF4) afforded the first representatives of pyridoannelated 1,2,3,4-tetrazine di-N-oxides, viz., pyrido[2,3-e][1,2,3,4]tetrazine 1,3-dioxide (9), 7-nitropyrido[2,3-e][1,2,3,4]tetrazine 1,3-dioxide (10), and pyrido[3,4-e][1,2,3,4]tetrazine 2,4-dioxide (11). These compounds were studied by 1H, 13C, and 14N NMR spectroscopy. Thyroid hormones undergo extensive metabolism to regulate hormone activity. A labeled thyroid hormone would be useful to track hormone metabolism through various pathways. While radiolabeled thyroid hormones have been synthesized and used for in vivo studies, a stable isotope labeled form of thyroid hormone is required for studying thyroid hormone metabolism by LC-MS/MS, an analytical technique that has certain advantages without the complications of radioactivity. Here we report the synthesis of 13C9-15N-T2 and 13C9-15N-T4, two labeled thyroid hormone derivatives suitable for in vivo LC-MS/MS studies.

ABSTRACT

Compound independent calibration (CIC) can be used to measure stable isotopically labeled molecules and their metabolites. This study was designed to demonstrate that gas chromatography coupled to atomic emission detection and CIC are useful analytical tools for the specific and quantitative determination of 13C and 15N labeled caffeine and its metabolites. These labeled compounds can be measured by monitoring either 13C or 15N atomic emission lines and using only one calibration function from one of the analytes. This method can be helpful when it is difficult or impossible to obtain an authentic reference material, which is often the case in metabolic studies.  相似文献

New cadmium(II) complexes with phosphine telluride ligands of the type CdX2(R3PTe)n [X?=?ClO4?, n?=?4: R?=?n-Bu (1), Me2?N (2), C5H10?N (3), C4H8?N (4) or OC4H8?N (5); X?=?Cl, n?=?2: R?=?n-Bu (6), Me2?N (7), C5H10?N (8), C4H8?N (9) or OC4H8?N (10)] have been synthesized and characterized by elemental analyses, IR and multinuclear (31P, 125Te, and 113Cd) NMR spectroscopy. In particular, the solution structures of these complexes were confirmed by 113Cd NMR at low temperature, which displays a quintuplet for each of the perchlorate complexes and a triplet for each of the chloride complexes due to coupling with four and two equivalent phosphorus atoms, respectively, indicating a four-coordinate tetrahedral geometry for the metal center. These multiplet features were further accompanied by one bond Te–Cd couplings, clearly showing that the ligand is coordinated to the metal through tellurium. The results are discussed and compared with those obtained for closely related phosphine chalcogenide analogs.  相似文献

Two rare-earth metal coordination compounds, (NH4)4[SmIII2(Httha)2]·16H2O (1) (H6ttha?=?triethylenetetramine-N,N,N,N′′,N′′′,N′′′-hexaacetic acid) and (NH4)4[SmIII2(dtpa)2]·10H2O (2) (H5dtpa?=?diethylenetriamine-N,N,N,N′′,N′′-pentaacetic acid), have been synthesized through reflux and characterized by FT-IR spectroscopy, thermal analysis, and single-crystal X-ray diffraction techniques. SmIII of (NH4)4[SmIII2(Httha)2]·16H2O (1) is nine-coordinate, forming tricapped trigonal prismatic coordination with three amine nitrogens and six oxygens, in which four oxygens are from one ttha and two from the other ttha. (NH4)4[SmIII2(Httha)2]·16H2O (1) crystallizes in the monoclinic crystal system with P2(1)/c space group. The crystal data are: a?=?13.9340(13) Å, b?=?22.890(3) Å, c?=?20.708(2) (14) Å, β?=?99.521(2)°, and V?=?6513.7(13) Å3. There are two –NH+– groups in the [SmIII2(Httha)2]4?. The polymeric (NH4)4[SmIII2(dtpa)2]·10H2O (2) also is nine-coordinate with tricapped trigonal prismatic conformation and crystallizes in the triclinic crystal system with P–1 space group. The cell dimensions are: a?=?9.8240(8) Å, b?=?10.0329(9) Å, c?=?13.0941(11) Å, β?=?77.1640(10)°, and V?=?1227.30(18) Å3. In (NH4)4[SmIII2(dtpa)2]·10H2O, there are two types of ammonium cations, which connect [SmIII2(dtpa)2]4? and lattice water through hydrogen bonds, leading to a 2-D ladder-like layer structure.  相似文献

The methylation reactions at ten nucleophilic sites in four DNA base molecules with methanediazonium ions (CH3N2 +) have been theoretically investigated including solvent effects at the B3LYP/6-31G** and MP2/6-31G** levels. The results show that all the methylation reactions have relatively small activation energy (<33.5 kJ/mol), and the methylation process is exothermic reaction and easy to occur. This study shows that the ultimate carcinogen CH3N2 + by NDMA can easily methylate DNA base molecules and form carcinogenic products. Host lattice Ba3Si5O13−δNδ oxonitridosilicates have been synthesized by the traditional solid state reaction method. The lattice structure is based on layers of vertex-linked SiO4 tetrahedrons and Ba2+ ions, where each Ba2+ ion is coordinated by eight oxygen atoms forming distorted square antiprisms. Under an excitation wavelength of 365 nm, Ba3Si5O13−δNδ:Eu2+ and Ba3Si5O13−δNδ:Eu2+,Ce3+ show broad emission bands from about 400-620 nm, with maxima at about 480 nm and half-peak width of around 130 nm. The emission intensity is strongly enhanced by co-doping Ce3+ ions into the Ba3Si5O13−δNδ:Eu2+ phosphor, which could be explained by energy transfer. The excitation band from the near UV to the blue light region confirms the possibility that Ba3Si5O13−δNδ:Eu2+, Ce3+ could be used as a phosphor for white LEDs.  相似文献

Lithocholic acid N-(2-aminoethyl)amide (1) and deoxycholic acid N-(2-aminoethyl)amide(2) have been prepared and characterized by1H, 13C and 15N NMR. The accurate molecular masses of 1 and 2 have been determined by ESI MS. The formation of the Cd2+-complexes (1+Cd and 2+Cd) in CD3OD solution have been detected by 1H,13C, 15N and 113Cd NMR. The 13C NMR chemical shift assignments of 1 and 2 and their Cd2+-complexes are based on DEPT-135 and z-GS 1H,13C HMQC experiments as well as comparison with the assignments of the related structures. The 15N NMR chemical shiftassignments of the ligands and theirCd2+-complexes are based on z-GS1H,15N HMBC experiments. 13C NMR chemical shift differences between 1and its 1:1 Cd2+-complex based on ab initiocalculations at Hartree-Fock SCI-PCM level using3-21G(d) basis set are in agreement with theexperimental shift changes observed onCd2+-complexation.  相似文献

In this paper, we have illustrated the utilisation of a second-sphere coordination approach to construct supramolecular inclusion solids with varieties of guest molecules. A flexible molecule N,N,N′,N′-tetra-p-methylbenzyl-ethylenediamine (L1) bearing doubly protonated H-bond donors was designed, capable of forming N–H…Cl hydrogen bonds with a crystallographically unique chloride anion, to construct an anion-directed ligand. The pillared double-layered host framework was constructed by an anion-directed ligand and primary coordination sphere [CoCl4]2 ?  through weak C–H…Cl hydrogen-bonding interactions. A variety of guest molecules, such as p-anisaldehyde, 1,4-dimethoxy-2,5-bis(methoxymethyl)benzene, can be included, leading to the formation of novel supramolecular inclusion solids: [L1]·4[H]+·[CoCl4]2 ? ·2Cl·1.5[C8H8O2]·0.25[CH3OH] (1) and [L1]·4[H]+·[CoCl4]2 ? ·2Cl·1.5[C12H20O4]·0.5[CH3OH] (2).

ABSTRACT

Electroanalytical methods have been widely used for determination of Se(IV), but the nature of the reduction processes involved is not well understood. Polarographic reduction occurs in three waves (i1, i2, and i3) the height of which changes with pH. We proved that in wave i1, H3SeO3 + is reduced, in i2 H2SeO3, and in i3 HSeO3 -. SeO3 2? is not reducible. All reductions involve a transfer of six electrons and yield selenides. Limiting currents are controlled by the rate of protonation. As proton donors, in addition to H3O+, the acid forms of the buffer present also act. Limiting currents increase markedly with increasing concentration of the buffer. Tenfold increase in buffer concentration can result in up to 200% increase in limiting current.  相似文献

Dimethyl dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate can exist either in 1,2- or 1,4-dihydro tautomeric forms. The 15N NMR spectra of dimethyl dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate were measured at the 15N natural abundance level as well as in 15N doubly labelled selectively and in 15N completely labelled compounds (20% 15N). The J(15N,15N) value was determined in 15N completely labelled compounds (20% 15N) using 1D 15N INADEQUATE and was found to be 12.2 ± 0.2 Hz in deuteriochloroform, acetonitrile-d3, DMSO-d6 and CD3OH. Very similar 15N chemical shifts and 1J(15N,1H) values were also observed in all the solvents. This indicates that compound 1 exists completely in the 1,4-dihydro tautomeric form (i.e., as dimethyl 1,4-dihydro-1,2,4,5-tetrazine-3,6-dicarboxylate) in all the solvents tested.  相似文献
