Abstract

The electrochemical behavior of 6-MP was studied by cyclic voltammetry at a hanging copper amalgam dropping electrode (HCADE). It was found that 6-MP could form a complex with the Cu(II) stripped from the HCADE, showing a new peak at ?0.19V in the medium of 0.1mol/L LiClO4-0.5mol/L HClO4 solution. The mechanism of the reaction was proposed. This new peak was sensitive and could be used for the determination of trace 6-MP by differential pulse adsorption cathodic stripping voltammetry (DPAdCSV). The linear range was from 3.6×10?10 to 5.3×10?6 mol/L, and the detection limit was about 1.2×10?10 mol/L (S/N=3). The method was also successfully applied to the determination of 6-MP in pharmaceutical tablets.  相似文献

The molecular structures of theendo (1a) andexo (1b) isomers of B4H8CO have been optimized at the ab initio MP2(Full)/6-31G* level of theory. The agreement of the computed geometrical parameters with the recently published electron-diffraction (GED) data is very good, even though a number of geometrical constraints were applied in the experimental determination. The IGLO (individual gauge for localized orbitals)11B NMR chemical shifts, calculated at the II//MP2/6-31G* level, are also in accord with experiment. The formation of1a and1b by association of B4H8 and CO is computed to be exothermic by 22.8 and 22.2 kcal/mol, respectively, at the MP2(Full)/6-31G*//MP2(Full)/6-31G* + ZPE(6-31G*) level of theory. The Lewis acid strength of B4H8 toward CO is comparable to that of BH3.  相似文献

The scavening of OH· radicals by 2- and 6-mercaptopurines (2-MP and 6-MP) and purine was studied spectrophotometrically using the competition method. The air-saturated aqueous solutions of these compounds in the presence of p-nitroso-N,N-dimethylaniline (NDMA) were -irradiated (-source—60Co). The absolute rate constants for the scavening of OH· radicals by 2-MP and 6-MP were calculated. Main product as produced by irradiation of 2-MP was identified.  相似文献

This paper developed optical fiber sensor based on molecular imprinted polymer as artificial recognition element for the determination of 6-mercaptopurine (6-MP) in human serum. This approach displayed high sensitivity by oxidizing 6-MP to a strong fluorescent compound with H2O2 in the alkaline media. It offered a relatively nice selectivity for 6-MP detection by molecular imprinted polymer's recognition. The relative standard deviation (R.S.D.) was 5% for a same sensor (n = 5) when 6-MP concentration was 1.0 × 10−7 g mL−1 in serum. The developed method was satisfactorily applied to the determination of 6-MP in human serum without any necessity for sample treatment or time-consuming extraction steps prior to the analysis.  相似文献

A method is presented for the measurement of 6-mercaptopurine (6-MP) in serum. Serum samples containing the drug were treated with dithioerythritol and saturated with ammonium carbonate. 6-MP was then extracted from the serum with a mixture of isopropanol-ethyl acetate (1:1, v/v) containing 0.01% ethanethiol. The solvent was evaporated, the residue dissolved in dilute hydrocholoric acid, washed with chloroform, and extracted with ethyl acetate. 6-MP was derivatized with trimethylanilinium hydroxide and measured gas chromatographically. The sulfhydryl-protecting reagents, dithioerythritol and ethanethiol, were added to prevent the decomposition of 6-MP. The extraction and clean-up method recovered 78 +/- 2% (S.E.) of the 6-MP present. Tracer amounts of [8-14C]6-MP served as the internal standard during the extraction part of the method. Theophyline was used as the internal standard during the gas chromatographic analysis. The standard curve obtained from the gas chromatograph was linear between 0.5 and 20 mug/ml of 6-MP. Serum samples were stored in the freezer for two weeks without significant loss of drug. No interference was encountered from normal serum constituents or xanthines, such as caffeine or theophylline added to serum.  相似文献

In this paper, multi-wall carbon nanotubes fuctionalized with carboxylic groups modified electrode (MWNT-COOH CME) was fabricated. This chemically modified electrode (CME) can be used as the working electrode in the liquid chromatography for the determination of 6-mercaptopurine (6-MP). The results indicate that the CME exhibits efficiently electrocatalytic oxidation for 6-MP with relatively high sensitivity, stability and long-life. The peak currents of 6-MP are linear to its concentrations ranging from 4.0×10−7 to 1.0×10−4 mol l−1 with the calculated detection limit (S/N=3) of 2.0×10−7 mol l−1. Coupled with microdialysis, the method has been successfully applied to the pharmacokinetic study of 6-MP in rabbit blood. This method provides a fast, sensible and simple technique for the pharmacokinetic study of 6-MP in vivo.  相似文献

Using 1-ml plasma samples, levels of 6-mercaptopurine (6MP) as low as 5 ng/ml and azathioprine (AZA) as low as 40 ng/ml can be detected using a high-performance liquid chromatography reversed-phase column procedure following extraction. Both compounds were stable in frozen plasma for seven weeks. AZA stability in blood was temperature dependent; the half-lives of AZA breakdown to 6MP at 37 degrees were 28 and 46 min in blood drawn from two rhesus monkeys. Plasma levels of 6MP were measured in a rhesus monkey following 6MP (1.47 mg/kg) and AZA (3 mg/kg) intravenous administration. 6MP levels were also measured in three renal transplant patients on daily 50- and 100-mg AZA doses. Peak levels (45-75 ng/ml) were reached within an hour and 6MP levels were detected for up to 7 h.  相似文献

The conformational stability and the C–O and O–H internal rotations in oxiranemethanol were investigated at the DFT-B3LYP/6-311G**, MP2/6-311G** and MP4(SDQ)/6-311G** levels of theory. Three minima were predicted in the CCOH potential energy scans of the molecule to have relative energies of about 2 kcal/mol or less and all were calculated to have real frequencies upon full optimization of structural parameters at the DFT and the MP2 levels of calculations. The Cg1 (H bond inner) conformation was predicted to be the lowest energy conformation for oxiranemethanol in excellent agreement with an earlier microwave study. The equilibrium mixture was calculated from Gibb's free-energy changes to be about 79% Cg1, 17% G1g and 3% Gg1 at the B3LYP/6-311G** level and about 87% Cg1, 11% G1g and 2% Gg1 at the MP2/6-311G** level for oxiranemethanol at 298.15 K. No conclusive evidence was obtained for the presence of high-energy form in the liquid phase of oxiranemethanol. The vibrational frequencies of oxiranemethanol in its three stable forms were computed at the B3LYP level and complete vibrational assignments were made for the lowest energy Cg1 form on basis of calculated and experimental data of the molecule.  相似文献

An investigation employing the ab initio molecular orbital (MO) and density functional theory (DFT) methods to calculate structural optimization and conformational interconversion pathways for the two diastereoisomeric forms, (±) and meso configurations of 1,3,7,9-tetraphospha-cyclododeca-1,2,7,8-tetraene (1) was undertaken. Two axial symmetrical conformations are found for (±)-1 configuration. (±)-1-TB axial symmetrical form is found to be about 0.35 and 0.99 kcal mol?1 more stable than (±)-1-Crown axial symmetrical conformation, as calculated by HF/6-31G*//HF/6-31G* and B3LYP/6-31G*//HF/6-31G* levels of theory, respectively. The unsymmetrical meso-1-TBCC form is found to be the most stable geometry, among the various conformations of meso-1 configuration. HF/6-31G*//HF/6-31G* and B3LYP/6-31G*//HF/6-31G* results showed that between the two most stable conformations of (±) and meso configurations, (±)-1-TB is more stable than meso-1-TBCC by about 3.35 and 2.43 kcal mol?1, respectively. In addition, MP2/6-31G* and B3LYP/6-311+G** results showed that the (±)-1-TB form is about 1.10 and 2.36 kcal mol?1 more stable than the meso-1-TBCC form. Further, NBO results revealed that in the most stable form of meso configuration (meso-1-TBCC), the sum of the π* allenic antibonding orbital occupancies (Σ π *occupancy) is greater than dl configuration ((±)-1-TB). Also, NBO results indicated that in the (±)-1-TB conformer, the sum of σ and π allenic moieties bonding orbital deviations (Σ σ dev+Σ π dev) from their normal values, is lower than in the meso-1-TBCC form.  相似文献

Despite being used as an anti-leukemic drug, the poor solubility of 6-mercaptopurine (6-MP) limits its use in topical and parenteral applications. Dendrimers are commonly used as drug carriers to improve their solubility in aqueous solution. In this work, the interactions between 6-MP and the amine-terminated poly(amidoamine) dendrimers (PAMAM−NH2) were investigated by various NMR technology. The chemical shift titrations disclosed that the 6-MP interacted with the surface of PAMAM−NH2 mainly through electrostatics. The determination of diffusion coefficient and relaxation measurements further confirmed the presence of interactions in 6-MP/PAMAM−NH2 complexes. In addition, the encapsulation of 6-MP within the cavity of PAMAM−NH2 was revealed through nuclear Overhauser effect spectroscopy and Saturation Transfer Double Difference analysis. Finally, the binding strength (H-8 is 100% and H-2 is 70%) of 6-MP to PAMAM−NH2 was quantitatively expressed using epitope maps. This study provides a systematic methodology for qualitative and quantitative studies of the interactions between dendrimers and drug molecules in general.  相似文献

Trans influence of triphenylphosphines and pseudohalogens on Ni-S bonds of NiS2PN and NiS2PC chromophores has been investigated by synthesizing and characterizing them. The complexes show the characteristic thioureide IR band at ∼ 1530 cm−1. Electronic spectrum of the cyanide analogue shows a strong blue shift relative to others. X-ray structures of [Ni(pipdtc)(4-MP)(NCS)] (1), [Ni(pipdtc)(PPh3)(NCS)] (2) and [Ni(pipdtc)(PPh3)(CN)] (3) (pipdtc = piperidinecarbodithioate anion, 4-MP = tri(4-methylphenyl)phopshine) are reported. Ni-S bond distance trans to 4-MP(1) is longer than the distances in (2) and (3) and Ni-S bond distances trans to Ni-NCS/CN decrease as follows: (3) > (2) > (1). Particularly, 4-MP shows a highly significant trans influence than triphenylphosphine on Ni-S bond. Similarly, CN exerts a marginally significant trans influence compared to NCS-. Thioureide C-N distances are relatively very short due to the drift of electron density towards the metal. The Ni-N-C angle (163.5(2)°) observed in (2) indicates deviation from linearity to a larger extent compared to that in (1) (176.3(3)°) due to the steric effect of the 4-methyl group. The reduction potentials (CV) for the mixed ligand complexes are much less compared to that of the parent NiS4 chromophore due to the π-acidic phosphines.   相似文献

2-Ethylhexyl 4-methoxycinnamate (EHMC) is one of the most commonly used sunscreen ingredient. In this study we investigated photodegradation of EHMC in the presence of such common oxidizing and chlorinating systems as H2O2, H2O2/HCl, H2O2/UV, and H2O2/HCl/UV. Reaction products were detected by gas chromatography with a mass spectrometric detector (GC-MS). As a result of experimental studies chloro-substituted 4-methoxycinnamic acid (4-MCA), 4-methoxybenzaldehyde (4-MBA) and 4-methoxyphenol (4-MP) were identified. Experimental studies were enriched with DFT and MP2 calculations. We found that reactions of 4-MCA, 4-MBA and 4-MP with Cl2 and HOCl were in all cases thermodynamically favorable. However, reactivity indices provide a better explanation of the formation of particular chloroorganic compounds. Generally, those isomeric forms of mono- and dichlorinated compounds which exhibits the highest hardness were identified. Nucleophilicity of the chloroorganic compounds precursors were examined by means of the Fukui function.   相似文献

Ab initio Hartree–Fock calculations at the HF/6-31G* level of theory for geometry optimization and the MP2/6-31G*//HF/6-31G* and B3LYP/6-311G(2df,p)//HF/6-31G* levels for a single point total energy calculation are reported for the important energy-minimum conformations of 1,1-dioxo-thiane (2), 1,1-dioxo-1,2-dithiane (3), 1,1-dioxo-1,3-dithiane (4), 1,1-dioxo-1,4-dithiane (5), 1,1,2-trioxo-1,2-dithiane (6), 1,1,3-trioxo-1,3-dithiane (7), 1,1,4-trioxo-1,4-dithiane (8), 1,1,2,2-tetroxo-1,2-dithiane (9), 1,1,3,3-tetroxo-1,3-dithiane (10), and 1,1,4,4-tetroxo-1,4-dithiane (11). According to the MP2/6-31G*//HF/6-31G* calculations, compound 5 is more stable than 3 and 4 by 7.8 and 8.9 kJ mol?1, respectively. The axial geometries of 6 and 8 are more stable than the equatorial forms by 21.4 and 19.1 kJ mol?1, respectively, but the equatorial form of 7 is 4.1 kJ mol?1 more stable than the axial geometry. Compound 11 is more stable than 9 and 10 by 49.3 and 31.0 kJ mol?1, respectively.  相似文献

Dopamine (DA) can be oxidized to dopamine quinone (DAquinone) through a one-step, two-electron redox reaction. The electron transfer property of DA and its supramolecular complex with aspartic acid (Asp) has been investigated by the theoretical calculations. We calculated the standard redox potentials (E o) of DA/DAquinone at the MP2/6-31G(d,p)//B3LYP/6-31G(d,p), MP2/6-31+G(d,p)//B3LYP/6-31+G(d,p), MP2/6-31G(d,p)//B3LYP/6-311G(d,p), and MP2/6-311+G(d,p)//B3LYP/6-311+G(d,p) levels. Comparing the experimental value, the redox potentials of DA/DAquinone obtained at MP2//B3LYP/6-311G(d,p) and MP2//B3LYP/6-311+G(d,p) levels can be considered as the upper and lower estimates. DA can form supramolecular complex (DA-Asp) with Asp through hydrogen bond (H-bond). Therefore, the values of 0.631 and 0.628 V obtained at MP2//B3LYP/6-311G(d,p) and MP2//B3LYP/6-311+G(d,p) levels for DA-Asp/DAquinone-Asp can be proposed as the upper and lower estimates of a probable (about 0.630 V) value of the corresponding redox potential. The calculated E o values of DA-Asp/DAquinone-Asp at the four theoretical levels are upper than those of DA/DAquinone, which indicates that the formation of H-bonds weaken the electron-donating ability of DA.  相似文献

Abstract

A simple, rapid and specific HPLC method has been developed to determine acetazolamide concentrations in human plasma. The assay procedure requires only 250 μl of sample with direct injection of the organic supernatant after protein precipitation with acetonitrile. Chlorothiazide was used as an internal standard. A reversed-phase C18 μBondapak column was employed for the chromatographic separation. The eluent was monitored at 265 nm using a UV variable wavelength detector. The retention times for acetazolamide (ACZ) and chlorothiazide (CTZ) were 6 and 8 min respectively. A linear relationship (r).995) was obtained over the 1-20 μg/ml concentration range. The limit of sensitivity for ACZ was 0.5 μg/ml, with greater than 85% recovery of ACZ and internal standard. The method was applied to human plasma samples obtained after administration of a 250 mg acetazolamide tablet.  相似文献

The complexes trans-[Ni(4-MP)2(NCS)2]·MeCN (1) and trans-[Ni(3-MP)2(NCS)2] (2) (4-MP = tri(4-methylphenyl)phosphine, 3-MP = tri(3-methylphenyl)phosphine) were prepared and characterized by IR, UV–visible, NMR spectra, CV, TGA and single crystal X-ray crystallography. Both the complexes have planar geometry and are diamagnetic. The Ni–P distances in both complexes are relatively short as a result of strong back donation from nickel to phosphorus. The phenyl rings in the 3-MP analogue (2) show increased pitching with reference to the plane formed by the ipso carbons due to increased steric effects. For complex (2), the N–Ni–N and P–Ni–P angles are significantly lower than the almost linear N–Ni–N and N–Ni–P angles observed for both complex (1) and trans-[Ni(PPh3)2(NCS)2]. This observation indicates that the 3-methylphosphine ligand forces complex (2) to distort towards a tetrahedral geometry. IR spectra of both complexes show strong bands around 2,090 cm−1 due to N-coordinated thiocyanate, while the electronic spectra contain d–d transitions around 452 nm. Cyclic voltammograms show that the irreversible one-electron reduction potentials increase in the following order: trans- [Ni(PPh3)2(NCS)2] < trans- [Ni(3-MP)2(NCS)2] < trans-[Ni(4-MP)2(NCS)2], revealing the electron releasing effect of the methyl groups. The planar complexes exhibit interallogony in coordinating solvents.  相似文献

Novel organophilic nanohybrid materials (K‐TDD) were obtained by the grafting of 1,2‐tetradecanediol (TDD) onto the surface of kaolinite (K). XRD, IR, TGA‐DTG, and SEM characterization showed that TDD grafting results in a partial exfoliation of kaolinite layers. This material was used to modify a glassy carbon electrode (GCE/K‐TDD) and applied for the trace analysis of methyl parathion (MP). The signal of MP recorded on GCE/K‐TDD was more intense compared to the unmodified GCE or to one modified with a film of natural kaolinite. Several parameters that can affect the stripping response were systematically investigated to optimize the sensitivity of the organokaolinite‐modified electrode. A linear calibration curve for MP was obtained in the concentration range from 2×10−6 to 14×10−6 mol .L−1 in acetate buffer (pH 6), giving a detection limit of 9×10−8 mol .L−1. The sensitivity of the method was found to be 2.42 μA/μM for the range of concentrations that gives a linear calibration curve. The electrode was shown to be very stable, with the electrochemical response of MP decreasing by only 1.5 % after a series of nine measurements. The interference of various inorganic ions and organic compounds likely to influence the stripping determination of the MP were also examined. The results showed that the GCE/K‐TDD electrode was effective in solutions containing interfering species and could be applied for the quantification of MP pesticide in natural water.  相似文献

Using the avidin-biotin binding system, an enzyme immunoassay procedure was developed to measure the membrane-associated placental tissue protein 1 (MP1) in serum. The standard curve covered the range from 10 to 1000 ng/ml of MP1. The intra- and inter-assay coefficient of variations (C.Vs) were less than 5 and 10%, respectively. Recoveries of MP1 added to serum ranged between about 96 and 101%. The MP1 serum level was over 10 and under 112 ng/ml in non-pathological men, and under 240 ng/ml in non-pathological women. The MP1 level in the ovulatory phase was higher than in other phases of the menstrual cycle. In pregnancies during 6-39 weeks, the MP1 level ranged from 10 to 540 ng/ml, and it increased during the third trimester of gestational age. In benign gynecologic diseases, the MP1 concentration in serum ranged from 10 to 215 ng/ml. The MP1 levels in benign diseases were compared with those in ovarian malignancies, in endometrial carcinoma, and in uterine cervical cancer. The immunohistochemical location of MP1 was detected in the cell membrane of ovarian cystadenocarcinoma.  相似文献

Abstract

A method is described for the quantitative analysis in plasma of Ro 15–0778, an arotinoid, (E)-1, 2, 3, 4-tetrahydro-1, 1, 4, 4-tetramethyl-6-(1-methyl-2-phenylethenyl) naphthalene and its metabolite Ro 14–6113, (E)-4- 2-(5,6,7,8-tetrahydro-5,5,8,8-tetramethyl-2-naphthalenyl)-2 methylethenyl) phenol. Following a simple extraction, the compounds are determined by reversed-phase high-performance liquid chromatography (HPLC) and detection at 300 nm. The experimental error is below 12% in the concentration range 6–4350 ng/ml. The detection limit is about 2 ng/ml. This method was applied to plasma specimens collected from patients receiving single or multiple dose administration of this compound.  相似文献
