The determination of proteins with arsenazo-DBN and Al3+ by Rayleigh light-scattering (RLS) is described. The weak RLS of arsenazo-DBN and BSA can be enhanced greatly by addition of Al3+ in the pH range 5.3-7.0; this resulted in two enhanced RLS signals at 420-440 nm and 460-480 nm. The reaction between arsenazo-DBN, Al3+, and proteins was studied and a new method was developed for quantitative determination of proteins. This method is very sensitive (0.34-41.71 microg mL(-1) for bovine serum albumin, BSA, and 0.29-53.41 microg mL(-1) for human serum albumin, HSA), rapid (< 2 min), simple (one step), and tolerant of most interfering substances. The effects of different surfactants were also examined. When these proteins were determined in four human serum samples the maximum relative error was not more than 2% and the recovery was between 97 and 103%.  相似文献

The determination of proteins with tetrasodium 2-(3,6-disulfo-8-hydroxynaphthylazo)-1,8-dihydroxynaphthalene-3,6-disulfonate (Beryllon II) by Rayleigh light scattering (RLS) was studied. The weak RLS of the Beryllon II-bovine serum albumin (BSA) complex can be greatly enhanced by the addition of Al3+ in the pH range 5.6-7.2; there was a maximum RLS platform at 400-420 nm. Based on the reaction between Beryllon II, Al3+ and proteins, a new method for the determination of proteins was developed. This method is very sensitive [0.20-41.42 micrograms ml-1 for BSA and 0.18-48.15 micrograms ml-1 for human serum albumen (HSA)], rapid (< 2 min), simple (one step) and tolerant towards most interfering substances. The effects of different surfactants were also examined. Four samples of protein in human serum were determined; the maximum relative error was no more than 5% and the recovery was 96-105%.  相似文献

A rapid sampling/mixing system has been designed which conforms to certain criteria necessary for its use as a clinical analyser. These include low solution volume (<100mul) for each determination on a sample, rapid cycle time (<2 sec to take an aliquot, mix and transfer reactants to an observation cell), good precision (reproducibility better than 0.2%), and ready automation (requiring only two electronic signals to perform a complete cycle). This device has been incorporated into an automated spectrophotometer to be used for a variety of clinical methods. Equilibrium methods for the determination of calcium and albumin are presented that require measurement times of only 6 and 7 sec, respectively. A reaction-rate procedure for total protein is presented that requires only 3 sec per sample. Precisions obtained with these procedures are typically better than 1% in the normal serum range. Results on samples from hospital patients are compared with values obtained on an SMA 12/60 instrument.  相似文献

This paper researched the determination of proteins with 2-(2-arsonophehenylazo)-7-[(2,6-dibromo-4-sulfophenylazo)-1,8-dihydroxynaphthalene-3,6-disulfonic acid (Arsenazo-DBS) by Rayleigh light-scattering (RLS). The reaction parameters, such as acidity, volume of buffer solution and concentration of Arsenazo-DBS, were examined by orthogonal array design (OAD). Under optimal conditions, the weak RLS of Arsenazo-DBS and BSA can be enhanced greatly and two enhanced RLS signals were produced at 340-350 and 400-420 nm. Based on this reaction, a new quantitative determination method for proteins has been developed. This method is proved to be very sensitive (the determination limits are 0.077 μg ml−1 for bovine serum albumen (BSA) and 0.074 μg ml−1 for human serum albumen (HSA)), rapid (<2 min), simple (one step) and tolerance of most interfering substances. The effects of different surfactants were also examined. The amount of proteins in human serum samples was determined and the maximum relative error was no more than 2% and the recovery was between 95 and 105%.  相似文献

An automated non-chiral high-performance liquid chromatographic method is described for the determination of the new anti-proliferative agent B859-35 in serum. This method employs sample clean-up of 1 ml of biofluid by liquid-solid extraction with the AASP (Advanced Automatic Sample Preparation) system. First separation is achieved on a LiChrospher-60-RP-Select-B column. A fraction of this elute is then collected by solid-phase trapping. Thereafter, the final chromatogram is developed on a narrow-bore Hypers1-CPS column and quantified with ultraviolet detection at 230 nm. The limit of quantitation of the assay is 250 ng/ml. Linearity was proven in the range 0.25-100 ng/ml. Typical figures for precision at these concentrations are 7.4 and 3.3%, and for accuracy 8.0 and 1.3%, respectively. An application of this method to the study of pharmacokinetics of B859-35 in serum samples of cancer patients is given.  相似文献

A simple, rapid and automatic fluorimetric method for the determination of total ascorbic acid is described. The method makes use of the stopped-flow mixing technique in order to achieve the rapid oxidation of ascorbic acid by dissolved oxygen to dehydroascorbic acid, which then reacts with o-phenylenediamine to form a fluorescent quinoxaline. The initial rate and fluorescence signal of this system are directly proportional to the ascorbic acid concentration. The calibration graph was linear over the range 0.1-30 microg ml(-1) (kinetic method) and 0.25-34 microg ml(-1) (equilibrium method). The precision (% RSD) was close to 0.5%. The method has been used for the determination of ascorbic acid in pharmaceutical formulations, fruit juices, soft drinks and blood serum.  相似文献

最佳实验条件下,基于脱氧尿苷与人血清白蛋白(HSA)相互作用,导致血清白蛋白的同步荧光光谱发生特异性变化,且体系的同步荧光强度和溶液中白蛋白的浓度呈良好的线性关系,建立了以脱氧尿苷为探针,运用固定波长同步荧光光谱快速测定生物样品中蛋白质总含量的新方法.深入考察了△λ值、反应介质、试剂用量、离子强度、加入顺序、反应时间等因素对测定的影响.在最佳实验条件下,体系的同步荧光强度与血清白蛋白在2.76~524.4μg/mL范围内的线性相关系数为0.9991,方法的检出限可达0.11μg/mL.运用本方法对人血清、唾液和尿液等生物样品进行测定并进行加标回收实验,回收率在98.0%~102.5%之间.对11份空白溶液进行平行测定,相对标准差为1.2%.用经典的考马斯亮蓝G-250(CBB)法做了对照实验,两种方法测定结果基本一致.还考察了一些常见离子和有机物存在时对蛋白质测定的影响.本方法已用于血清、唾液和尿样等生物样品中蛋白质总量的快速测定.  相似文献

A rapid and automated method was developed for the determination of bacterial contamination and using Escherichia coli as a model microorganism. The method involves the use of a sensor connected to a flow injection (FI) system. The sample is introduced through a flow injection system into a piezoelectric quartz crystal (PQC) flow-cell. The resulting change of the resonance frequency is related to the bacterial contamination in the sample. The parameters associated with the flow system and the conditions for introducing the sample culture were optimized. Calibration curves are linear in the range from 3.2?×?107 to 3.2?×?109 cfu per mL-1, with a correlation coefficient of 0.997. The reproducibility was between 3.1 and 7.6%, and the detection limit is 1.1?×?107 cfu per mL-1. The method allowed the determination of bacterial contamination in residual water and in samples of milk and chicken stock within 5 h, while the conventional plate count method requires 24 to 48 h. The results obtained by these two methods are in good agreement.

A new Rayleigh light scattering (RLS) assay of protein is presented in this paper. At the optimum pH 4.10, the weak RLS of Arsenazo-DBN can be greatly enhanced by the addition of proteins due to the interaction between protein and Arsenazo-DBN. Based on this, the reactions of Arsenazo-DBN and proteins, including bovine serum albumin, human serum album, gamma-globulin, egg albumin, lysozyme and trypsin, were studied. A new quantitative determination method for proteins has been developed. The linear range for human serum albumin, for example, is 0.085-34.62 micrograms mL-1 with a detection limit of 44.8 ng mL-1. Besides high sensitivity, the method is characterized by good reproducibility, rapidity of reaction, good stability, and few interfering substances. The determination of the proteins in human serum and urine samples by this method give results very close to those obtained using Coomassie Brilliant Blue G-250 colorimetry, with relative standard deviations of 0.7-2.5%.  相似文献

The development of automated stopped-flow spectrophotometric systems, their potential in automated routine determinations using kinetic and fast equilibrium techniques and several examples of applications are briefly reviewed. The use of a compact, inexpensive laboratory-made stopped-flow system for the measurement of reaction rates and a fast equilibrium method for the determination of acetaminophen in formulations and serum are described. The reaction-rate method is based on monitoring the oxidation of acetaminophen by iron(III) in the presence of the chelating agent 2,4,6-tris(2-pyridyl)-s-triazine to form a highly absorbing complex of iron(II). The calibration graph is linear in the range 20–200 μg ml?1, with a precision of 0.8–2.6%, a detection limit of 5.5 μg ml?1 and a measurement throughput of 120 per hour. Common excipients do not interfere and the analysis of commercial formulations gave results similar to those of a reference method. The optimization of the experimental conditions was done by a kinetic study of the reaction and some kinetic parameters are given.The method for the determination of acetaminophen in serum is based on a rapid measurement of the absorbance of the reaction mixture after a delay time of 15 s in the presence of chlorpromazine, which catalyses the reaction. Acetaminophen is isolated by extraction with ethyl acetate and the calibration graph is linear in the range 0.5–6 μg ml?1 with a detection limit of 0.04 μg ml?1 and a precision of 1.5%. The proposed method showed a decreased interference from drugs that also react with iron(III).  相似文献

Ketamine was used for anaesthesia originally but has emerged as an abused drug in recent years. The prevalence of ketamine abuse demands a direct and rapid determination method. It is known that in-tube solid phase microextraction (in-tube SPME) can perform extraction with a capillary linked directly to a HPLC system, providing an automated and accurate extraction procedure. In this paper, an in-tube SPME coupled to HPLC method was developed for the determination of ketamine in urine samples with a poly(methacrylic acid-ethylene glycol dimethacrylate) monolithic capillary column as the extraction phase, which is expected to provide higher extraction efficiency than open tubular capillaries. After optimizing the extraction conditions, ketamine was extracted directly from urine samples in a wide dynamic linear range of 50-10,000 ng mL(-1), with the detection limit obtained as 6.4 ng mL(-1). The intra-day and inter-day precision for the method was 1.6% and 1.7%, respectively. The urine samples from suspect addicts have been successfully analyzed within 20 min. The re-usability of the monolithic column was also confirmed as no decrease of the extraction efficiency was shown after urine extraction.  相似文献

Fully automated structure determination of proteins in solution (FLYA) yields, without human intervention, three-dimensional protein structures starting from a set of multidimensional NMR spectra. Integrating existing and new software, automated peak picking over all spectra is followed by peak list filtering, the generation of an ensemble of initial chemical shift assignments, the determination of consensus chemical shift assignments for all (1)H, (13)C, and (15)N nuclei, the assignment of NOESY cross-peaks, the generation of distance restraints, and the calculation of the three-dimensional structure by torsion angle dynamics. The resulting, preliminary structure serves as additional input to the second stage of the procedure, in which a new ensemble of chemical shift assignments and a refined structure are calculated. The three-dimensional structures of three 12-16 kDa proteins computed with the FLYA algorithm coincided closely with the conventionally determined structures. Deviations were below 0.95 A for the backbone atom positions, excluding the flexible chain termini. 96-97% of all backbone and side-chain chemical shifts in the structured regions were assigned to the correct residues. The purely computational FLYA method is suitable for substituting all manual spectra analysis and thus overcomes a main efficiency limitation of the NMR method for protein structure determination.  相似文献

An integrated microchip electrophoresis (MCE) system with online immunoreaction and laser induced fluorescence (LIF) detection has been developed for simultaneous determination of multi-analytes. In this system, the multiplexed immunoreactions between multiple antibody-immobilized glass beads with analytes and respective fluorescently labeled antigens were performed in a sample reservoir. After online incubation, the immunoreaction solution was injected into a one-way separation channel, and free fluorescently labeled antigens were separated and detected in the separation channel. With the help of glass beads, the immunocomplex can not move into the separation channel, which simplifies the separation of fluorescently labeled antigens. With the use of phenobarbital (PB), phenytoin (PHT), carbamazepine (CBZ) and theophylline (Th) as proof-of-principle analytes, the one-way multiplexed immunoassay could be completed within 20 min, resulting in a response curve over the range of 4.0-400 nM for each analyte. Detection limits (S/N = 3) for the drugs tested were in the range of 1.8 × 10(-9) to 2.5 × 10(-9) M. Compared with the conventional immunoassays, this assay is simple, rapid, sensitive and low cost, and provides an accurate procedure for a multiplex immunoassay. The present method has been applied for the simultaneous determination of PB, PHT, CBZ and Th in human serum, which showed a promise of automated clinical application.  相似文献

A simple, sensitive and selective method was proposed for the determination of proteins by using a resonance light scattering technique. The weak resonance light scattering (RLS) of Bordeaux red (BR) can be enhanced greatly in the pH range 3.87-3.96 by the addition of micro amounts of proteins, resulting in four characteristic peaks in the wavelength range 250-600 nm. At the maximal wavelength of 363 nm, the enhanced RLS is proportional to the concentration of proteins in the range 0.12-10.8 microg ml-1 for bovine serum albumin (BSA) and 0.24-18.0 microg ml-1 for human serum albumin (HSA). The detection limits were 40.0 and 52.9 ng ml-1 for BSA and HSA, respectively. The present method has been applied to the determination of total proteins in human serum, urine and saliva samples. The obtained results are in good agreement with those obtained by the Bradford method with relative standard deviations (R.S.D.) of 0.9-2.3%.  相似文献

A column-switching high-performance liquid chromatographic method was developed for the determination of vincristine in serum. Sample preparation was carried out by means of on-line column-extraction, using a C18 reversed-phase preconcentration column. This technique is simple (minimizing manual sampling errors), rapid (reduction of time and costs) and can be easily automated. Both ultraviolet and electrochemical detection are possible, but the latter shows a cleaner chromatogram and is, by the use of a new electrochemical detector, far more sensitive (detection limit 0.3 microgram/l at a signal-to-noise ratio of 3). A matrix study was carried out (using human serum and urine and two kinds of calf's serum). Although it appeared that the system was matrix-dependent, no difference in matrix effects could be found in the serum or plasma of different patients. Controls for human serum analysis should be prepared in human serum. With the method described, pharmacokinetic studies of vincristine in children can be performed.  相似文献

A rapid method for the determination of flunitrazepam and desmethylfflunitrazepam in human serum in the range 10-300 ng/ml is described. Both drugs are isolated from biological material by means of a single extraction, part of the organic phase is evaporated to dryness and the residue is dissolved in a small volume of benzene. Without further purification, the substance is determined gas chromatographically with an electron-capture detector configuration of 63Ni-type. The method permits the quantitative determination of at least 25-300 ng/ml with an overall recovery of flunitrazepam of 99.7 +/- 4.9% and of desmethylflunitrazepam of 98.6 +/- 7.8% from serum. All calculations were carried out by a data system that was programmed for this purpose. The limit of detection for flunitrazepam is of the order of 1 ng/ml in serum. The method is sufficiently sensitive and specific for therapy control purposes. The time needed for an analysis is less than 1 h.  相似文献

An automated, continuous-flow system is utilized to determine specific conductance and pH simultaneously in natural waters. A direct electrometric procedure is used to determine values in the range pH 4–9. The specific conductance measurements are made with an electronically modified, commercially available conductivity meter interfaced to a separate module containing the readout control devices and printer. The system is designed to switch ranges automatically to accommodate optimum analysis of widely varying conductances ranging from a few μmhos cm-1 to 15,000 μmho cm-1. Thirty samples per hour can be analyzed. Comparison of manual and automated procedures for 40 samples showed that the average differences were 1.3% for specific conductance and 0.07 units for pH. The relative standard deviation for 25 replicate values for each of five samples was significantly less than 1% for the specific conductance determination; the standard deviation for the pH determination was ? 0.06 pH units.  相似文献

An isocratic high-performance liquid chromatographic method specifically developed to allow simple and rapid determination of beta-carotene concentrations in serum and plasma is reported. Using a method modified from a previously published technique, serum and plasma proteins are denatured by exposure to perchloric acid, and beta-carotene is subsequently extracted into an organic matrix consisting of ethyl acetate-tetrahydrofuran (1:1); no evaporation step is required. Separation is achieved using isocratic elution from a reversed-phase C18 column with UV detection at 436 nm. Recovery of beta-carotene from water and plasma was greater than 98.1%; beta-carotene was stable in the extraction matrix for at least 4 h. Three anticoagulants (oxalate, citrate, and EDTA) caused losses of beta-carotene; perchloric acid and tetrahydrofuran could also destroy beta-carotene under certain conditions. Each run required less than 15 min; within-day coefficient of variation for identical samples averaged 2.3%, between-day coefficient of variation was 4.4% and sensitivity was better than 10 ng/ml. Stability of beta-carotene in plasma was also examined. This method permits a simple, rapid, sensitive, precise, and accurate determination of beta-carotene using 0.5 ml of serum or heparinized plasma.  相似文献
