A simple calorimetric method for the determination of propranolol hydrochloride has been worked out. The method involves nitration of the drug with a mixture of potassium nitrate and sulphuric acid. The coloured nitro-derivative has an absorption maxima at 360 nm. The working concentration range is 10-50 mug/ml. The method is applicable to analysis of pharmaceutical preparations.  相似文献

The complexation equilibria of magnesium(II) with purpurin (PURP) are studied spectrophotometrically as a function of pH in 50% (v/v) ethanol-water medium at 20 degrees C. The uncharged complex formed at pH 9.5 allows precise and accurate determination of magnesium over the concentration range 0.8-4.3 mug ml(-1). The molar absorptivity of the Mg-PURP complex at 540 nm and detection limit for Mg are 9.2x10(3) l mol(-1) cm(-1) and 75 ng ml(-1), respectively. The proposed method is rapid and possesses reasonable selectivity. Under the optimum conditions, the use of first-derivative spectrophotometry has the advantage of high sensitivity than normal spectrophotometry and allows the determination of 0.2 mug ml(-1) of magnesium. The validity of the method is examined by analysing several SRM Portland cement samples and a variety of cement materials of variable magnesia content.  相似文献

Micellar electrokinetic capillary chromatography was developed to analyze plant hormones including gibberellic acid, abscisic acid, indole-3-acetic acid, alpha-naphthaleneacetic acid, 2,4-dichlorophenoxyacetic acid, kinetin-6-furfurylaminopurine and N6-benzyladenine. The influences of some crucial parameters including buffer concentration, pH value, micelle concentration and applied voltage on electrophoretic separation were investigated. Under optimum conditions (50 mM borate as the running buffer containing 50 mM sodium dodecylsulfate, pH 8.0; separation voltage: -15 kV; injection: hydrodynamic injection, 5 s at 50 mbar; temperature: 25 degrees C), a complete separation of seven plant hormones was accomplished within 30 min. Emphasis was placed on improving detection sensitivity in order to detect small amounts of hormones in plant tissue. Multiple wavelength detection and expanded bubble cell capillary were used with enrichment factors of 2 and 3, respectively. In addition, an on-line concentration method of large volume sample stacking was designed. Enrichment factors of up to approximately 10-600 were achieved for these hormones with detection limits down to 0.306 ng/ml. The method was successfully applied to analyzing abscisic acid in flowers of transgenic tobacco.  相似文献

A simple and sensitive spectrophotometric method is developed for the determination of titanium in aqueous medium. The metal ion forms a reddish brown coloured complex with 2,4-dihydroxybenzaldehyde isonicotinoyl hydrazone (2,4-DHBINH) in the pH range 1-7. The complex shows two absorption maxima, one at 430 nm and the other at 500 nm. The reagent shows appreciable absorbance of 430 nm and negligible absorbance at 500 nm at pH 1.5. Beer's law is obeyed in the range 0.09 to 2.15 mug ml(-1) of titanium(IV). The molar absorptivity and the Sandell's sensitivity of the method are 1.35 x 10(4) 1 mol(-1) cm(-1) and 0.0049 mug cm(-2), respectively. A method for the determination of titanium by first-order derivative spectrophotometry is also proposed. The methods have been employed successfully for the determination of titanium in several alloy and steel samples.  相似文献

Two spectrofluorimetric methods for the determination of dipyridamole in plasma are described. The thin-layer chromatographic-fluoridensitometric method utilizes 1 ml of plasma which is extracted at pH 10 with diethyl ether-dichloromethane (80:20). The organic phase is evaporated to dryness, reconstituted in 250 microliter dichloromethane and 5 microliter are spotted on a silica gel 60 plate. The plate is developed in ethyl acetate-methanol-ammonia (85:10:5), dried, dipped in a paraffin wax solution, dried, and scanned using 380 nm as excitation wavelength, a 430 nm cut-off filter, and collecting all emitted light on the photomultiplier. Quantitation was done by the external standard method, peak heights being measured and a calibration graph constructed. For the spectrofluorimetric method 1 ml of plasma is extracted at pH 10 with 8 ml of hexane-isoamyl alcohol (95:5) and the organic phase used directly for the measurement of the fluorescence intensity (excitation 405 nm, emission 495 nm). Quantitation was done by measuring the fluorescence of standards that were treated as above and constructing a calibration graph of concentration versus fluorescence intensity. Concentrations of unknowns were found by interpolation from this graph. The two methods were found to exhibit good correlation but the spectrofluorimetric method proved to be more amenable to the analysis of a large number of samples.  相似文献

A spectrofluorimetric method was described for the determination of drugs containing active methylene groups adjacent to carbonyl groups. The method was applied successfully to the determination of three life saving cardiovascular drugs, with narrow therapeutic indices: pentoxifylline (I), propafenone hydrochloride (II) and acebutolol hydrochloride (III), in laboratory-prepared mixtures, in commercial tablets and in plasma samples. The method involved the reaction of each of the tested drugs with N1-methyl nicotinamide chloride (NMNCl) in the presence of alkali, followed by addition of formic acid, where highly fluorescent reaction products were produced. The produced fluorescence were measured quantitatively at 472 nm (lambdaex 352 nm), 409 nm (lambdaex 310 nm) and 451 nm (lambdaex 266 nm) for (I), (II), and (III) respectively. The method was linear over concentration ranges of 10-1000 microg/ml , 0.2-12 microg/ml and 0.08-10 microg/ml in standard solutions for (I), (II), and (III) respectively. In spiked human plasma samples, calibration graphs were linear over concentration ranges of 20-1000 microg/ml, 0.2-15 microg/ml and 0.08-10 microg/ml for (I), (II), and (III) respectively. The method showed good accuracy, specificity and precision in both laboratory-prepared mixtures and spiked human plasma samples. The proposed method is simple, with low instrumentation requirements, suitable for quality control application, bioavailability and bioequivalency studies.  相似文献

Alpha-benzilmonoxime has been used for the extraction and determination of cobalt at microgram amount. The reagent reacts with cobalt(II) in the pH range of 8.8 - 9.3 to form a yellow-color chelate, which is extracted in chloroform, toluene, and some other non-polar solvents. The chelate is stable in chloroform for about one day. Under the optimum conditions of the a-benzilmonoxime concentration and pH of 9.0, Beer's law was obeyed in the concentration range of 0.08 - 2.2 microg/ml cobalt. The molar absorptivity of the extracted species was 2.55 x 10(4) dm3/mol cm at 380 nm, with a detection limit of 0.01 microg/ml cobalt. Relative standard deviations of 0.4, 0.8 and 2.3% were found for the determination of cobalt concentrations of 2.2, 1.1 and 0.08 microg/ml, respectively. The effect of diverse ions on the determination of 1.00 microg/ml of cobalt has been studied. The method was applied to the determination of cobalt in vitamin B12 and B-complex ampoules, a Co2O3-Co3O4 laboratory chemical mixture and some synthetic alloy samples. The method is sensitive, simple, rapid and accurate.  相似文献

A fluorescence method involving sample pre-treatment is investigated concerning the determination of cefoxitin. A fluorescent product is formed when samples containing cefoxitin are subjected to alkaline hydrolysis with 1.0M sodium hydroxide and heated for 60 min at 90 degrees . The fluorescence is measured in ethanol/water medium (50% v/v) at approximately pH 2.0 provided by adding of 0.1M hydrochloric acid. The fluorescence excitation and emission maxima were 317 and 400 nm, respectively. The quantitative range is between 0.020 and 1.40 mug/ml. A detection limit of 2 x 10(-3) mug/ml was found. The proposed method has been applied to the determination of cefoxitin in commercial injections, saline and glucosed physiological serum.  相似文献

A kinetic spectrophotometric method has been described for the determination of metoprolol tartrate in pharmaceutical formulations. The method is based on reaction of the drug with alkaline potassium permanganate at 25+/-1 degrees C. The reaction is followed spectrophotometrically by measuring the change in absorbance at 610 nm as a function of time. The initial rate and fixed time (at 15.0 min) methods are utilized for constructing the calibration graphs to determine the concentration of the drug. Both the calibration graphs are linear in the concentration range of 1.46 x 10(-6)-8.76 x 10(-6) M (10.0-60.0 microg per 10 ml). The calibration data resulted in the linear regression equations of log (rate)=3.634+0.999 log C and A=6.300 x 10(-4)+6.491 x 10(-2) C for initial-rate and fixed time methods, respectively. The limits of quantitation for initial rate and fixed time methods are 0.04 and 0.10 microg ml(-1), respectively. The activation parameters such as E(a), DeltaH(double dagger), DeltaS(double dagger) and DeltaG(double dagger) are also evaluated for the reaction and found to be 90.73 kJ mol(-1), 88.20 kJ mol(-1), 84.54 J K(-1) mol(-1) and 63.01 kJ mol(-1), respectively. The results are validated statistically and through recovery studies. The method has been successfully applied to the determination of metoprolol tartrate in pharmaceutical formulations. Statistical comparison of the results with the reference method shows excellent agreement and indicates no significant difference in accuracy and precision.  相似文献

A solid chelating compound phenanthrenequinonemonoxime (PQM) supported on naphthalene provides a rapid, sensitive and economical means of preconcentration and separation of palladium from standard solutions and from synthetic samples. Palladium forms a complex with PQM supported on naphthalene in a column at pH 2.2-5.4 with a flow-rate of 1 ml/min. The metal complex and naphthalene are dissolved out from the column with 5 ml of CHCl(3) and the absorbance is measured at 430 nm or 500 nm against a reagent blank. Beers law is obeyed in the concentration range 3.0-56.0 mug and 6.0-42.0 mug at 430 nm and 500 nm respectively. The molar absorptivities are 2.10 x 10(4) and 1.69 x 10(4) 1.mole(-1).cm(-1) at 430 and 500 nm respectively.  相似文献

N-m-Tolyl-n-phenylhydroxylamine is proposed for the spectrophotometric determination of titanium. The reagent forms a yellow chloroform-soluble complex with titanium in media with a hydrochloric acid concentration of at least 9M. The apparent molar absorptivity at 380 nm is 7.4 x 10(3) 1.mole(-1).cm(-1). The optimum final concentration range is 0-54 mug of titanium in 10 ml of chloroform. The complex contains the metal and reagent in 1:2 ratio. The proposed method has been successfully applied to the determination of titanium in coal, coal fly-ash, pond sediment and asphalt.  相似文献

Serum is rapidly digested with a mixture of nitric and perchloric acids at a temperature of 180 +/- 10 degrees C, and hydrochloric acid is used to reduce selenium(VI) to selenium(IV). Selenium is determined by hydride generation flame atomic absorption spectrometry. The results show that this method has the advantages of being sensitive, accurate, rapid and simple. After the serum is digested and diluted, 4.0 ml is taken for the determination. The characteristic concentration, detection limit, variation coefficient, recovery rate and linear range are 2.93 mug 1(-1), 1.55 mug l(-1), 1.6-5.0%, 97.3-99.2% and 0.0-320.0 mug l(-1) respectively. Serum at 4 degrees C and in frozen state can be preserved for at least 7 and 14 days, respectively.  相似文献

A new, simple, sensitive and selective catalytic method is developed for the determination of vanadium in natural and sea waters. The method is based on the catalytic effect of V(V) and/or V(IV) on the bromate oxidative-coupling reaction of metol with 2,3,4-trihydroxybenzoic acid (THBA). The reaction is followed spectrophotometrically by tracing the oxidation product at 380 and/or 570 nm after 5 min of mixing the reagents. The optimum reaction conditions are 6.4 x 10(-3) mol l-1 of metol, 2.0 x 10(-3) mol l-1 of THBA and 0.16 mol l-1 of bromate at 35 degrees C and in the presence of an activator-buffer solution of 1 x 10(-2) mol l-1 of tartrate (pH = 3.10). Following the recommended procedure, V(V) and/or V(IV) can be determined with linear calibration graphs up to 0.75 ng ml-1 and detection limits, based on the 3Sb criterion, of 0.008 and 0.018 ng ml-1 at 380 and 570 nm, respectively. The developed method was successfully applied, without any separation or preconcentration processes, to the determination of vanadium in natural and seawaters following the direct calibration and standard addition techniques, respectively.  相似文献

Two simple and accurate spectrophotometric methods are presented for the determination of beta-lactam drugs, flucloxacillin (Fluclox) and dicloxacillin (Diclox), in pure and in different pharmaceutical preparations. The charge transfer (CT) reactions between Fluclox and Diclox as electron donors and 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) pi-acceptor and potassium iodate via oxidation reduction reaction where the highly coloured complex species or the liberated iodine have been spectrophotometrically studied. The optimum experimental conditions have been studied carefully. Beer's law is obeyed over the concentration range of 2-450 microg ml(-1) for Fluclox and 10-450 microg ml(-1) for Diclox using DDQ reagent and at 50-550 microg ml(-1) for Fluclox and 50-560 microg ml(-1) for Diclox using iodate method, respectively. For more accurate results, Ringbom optimum concentration range is calculated and found to be 6-450 and 15-450 microg ml(-1) for Fluclox and Diclox using DDQ, respectively, and 65-550 and 63-560 microg ml(-1) for Fluclox and Diclox using iodine, respectively. The Sandell sensitivity is found to be 0.018 and 0.011 microg cm(-2) for DDQ method and 0.013 and 0.011 microg cm(-2) for iodate method for Fluclox and Diclox, respectively, which indicates the high sensitivity of both methods. Standard deviation (S.D.=0.01-0.80 and 0.07-0.98) and relative standard deviation (R.S.D.=0.13-0.44 and 0.11-0.82%) (n=5) for DDQ and iodate methods, respectively, refer to the high accuracy and precision of the proposed methods. These results are also confirmed by between-day precision of percent recovery of 99.87-100.2 and 99.90-100% for Fluclox and Diclox by DDQ method and 99.88-100.1 and 99.30-100.2% for Fluclox and Diclox by iodate method, respectively. These data are comparable to those obtained by British and American pharmacopoeias assay for the determination of Fluclox and Diclox in raw materials and in pharmaceutical preparations.  相似文献

A method is described for the determination of chloride in PtAl(2)O(3)/SiO(2) catalysts, based on its extraction with alkali followed by reaction with Hg(SCN)(2)Fe(3+) reagent and spectrophotometric measurement at 460 nm. Beer's law is obeyed in the concentration range 5-75 mug of chloride in 25 ml of aqueous solution. The molar absorptivity is 1.99 x 10(3) l.mole(-1).cm(-1). The rapid colour development, excellent reproducibility and absence of interference from the species most often present in the catalysts are the advantages of the method.  相似文献

A liquid chromatographic/turbo ionspray tandem mass spectrometric (LC/MS/MS) method was developed and validated for the determination of L-753,037, a potent endothelin receptor antagonist currently under development for the treatment of cardiovascular diseases, in human plasma. L-753,037 is extracted from 0.5 ml of human plasma using liquid-liquid extraction and analyzed by LC/MS/MS with a turbo ionspray interface. Method validation results showed that this method is very sensitive, reliable, selective and reproducible. L-753,048, an ethoxy analogue of L-753,037, was used as the internal standard. The method has a lower limit of quantitation (LOQ) of 50 pg ml(-1) with a linear calibration range of 0.05-50 ng ml(-1). The intra-day precision and accuracy (n = 5) were measured to be below 10% relative standard deviation (RSD) and between 97.4 and 102.8% of the nominal values, respectively, for all calibration standard concentrations within the calibration curve range. The inter-day precision and accuracy (n = 3 days, 5 replicates per day) were measured to be below 6.5% RSD and between 99.3 and 102.0% of the nominal values, respectively, for all quality control concentrations. The extraction recovery was determined to be approximately 99% on average. The analyte was found to be stable in plasma through three freeze-thaw cycles, for at least 4 h at ambient temperature and for up to 40 days under -20 degrees C freezer storage conditions. The analyte was also shown to be stable for at least 24 h in the reconstitution solution at room temperature and for up to 3 days as a dried extract at 4 degrees C. Additional variations in plasma concentration of the analyte due to the use of different sources of plasma were also evaluated.  相似文献

A sequential injection spectrophotometric method for stoichiometric studies, optimization and quantitative determination of ciprofloxacin and norfloxacin was developed. The work is based on the complexation reaction of ciprofloxacin and norfloxacin with iron(III) in sulfuric acid media and a spectrophotometric measurement of absorbances of the corresponding complexes at 447 and 430 nm respectively. The stoichiometries and formation constants were determined. A 1:2 iron(III) to drug mole ratio was found to give the most predominant complexes for both drugs with 5.00 x 10(-3) M H(2)SO(4) and at 0.20 M ionic strength utilizing Job's method and the molar ratio method. A numerical method was utilized for the calculation of the formation constants, the logarithms of which were found to be 7.756 +/- 0.121 and 7.839 +/- 0.056, for ciprofloxacin and norfloxacin respectively. A factorial design together with the all-model-search method was utilized for the optimization of the concentration and aspiration volume of iron(III) as these were the variables which most affected peak absorbance. Working dynamic ranges of 50-500 ppm and 50-400 ppm were obtained for ciprofloxacin and norfloxacin respectively. The method was found to be suitable for the determination of these compounds in pharmaceutical preparations.  相似文献

A rapid and sensitive method for the trace level determination of aluminium based on the formation of a 1:1 complex with chromotropic acid (1,8-dihydroxynaphthlene-3,6-disulfonic acid) in an methanol medium is reported. The fluorescence intensity of the system was 50 times greater than that of the system without aluminium. This method is very sensitive and selective for the direct determination of aluminium ion. The fluorescence is excited at 346 nm and measured at 370 nm. The optimum conditions are a chromotropic acid concentration of 5.0 ml (1.0x10(-4) M) and pH 4.0+/-0.5 (acetic acid-sodium acetate buffer). The fluorescence intensity is a linear function of the concentration of Al(III) in the range 2-100 ng ml(-1) and the detection limit is 1.0 ng ml(-1). The method has been applied successfully to the determination of trace amount of Al(III) in tap, river and sea-water samples.  相似文献

The kinetics of 1,3-dimethylbarbituric acid with some quinones, namely 1,4-benzoquinone, 1,4-naphthoquinone and p-chloranil in 50% methyl alcohol-water mixture have been investigated spectrophotometrically at 30-50 degrees C. The reaction follows overall second-order kinetics, first order each in reactant. From the dependence of the rate constants on temperature, activation parameters have been calculated. A plot of deltaH# versus deltaS# for the reaction gave a good straight line with an isokinetic temperature of 387.66 K. The rate of reaction increases with increasing dielectric constant of the medium. Based on this reaction, a spectrophotometric determination method of quinones is described. Beer's law was obeyed within the concentration range 2.7-61.5 microg ml(-1) quinone. The method was applied for determination of barbituric, thiobarbituric and 1,3-dimethylbarbituric acids with 1,4-naphthoquinone within a concentration range of 3.2-39.5 microg ml(-1) barbiturate. The reaction mechanism and reactivity have been discussed.  相似文献
