We describe here the fast LC‐MS/MS separation of a mixture of neurotransmitters consisting of dopamine, epinephrine, norepinephrine, 3,4‐dihydroxybenzylamine (DHBA), salsolinol, serotonin, and γ‐aminobutyric acid (GABA). The new UltiMate® 3000 Rapid Separation system (RSLC) was successfully coupled to the 4000 QTRAP mass spectrometer operating in multiple‐reaction monitoring (MRM) mode. The separation was attained using a 100 mm length, 2.2 μm particle size Acclaim column at a flow rate of 0.5 mL/min. The column back pressure was 350 bar, while the total run time including column re‐equilibration was 5.2 min. The peak resolution was minimally affected by the fast separation. The RSLC‐MRM separation was found to have a precision range based on peak area for 50 replicate runs of 2–5% CV for all analytes, and the reproducibility of the retention time for all analytes was found to range from 0–2% CV. The described method represents an almost seven times shorter analysis time of neurotransmitters using LC/MRM which is very useful in screening large quantities of biological samples for various neurotransmitters.  相似文献

1,3-Butadiene, a common air pollutant formed in the combustion of organic matter, has been assessed by the U.S. EPA to be a strongly carcinogenic compound. This risk assessment is very uncertain because of the lack of information on the dose of the powerful carcinogenic metabolite diepoxybutane (DEB). This report presents an analytical method for in vivo dose monitoring of a unique marker for DEB. For a large number of alkylating agents in vivo doses are monitored by measurement by gas chromatography/mass spectrometry (GC/MS) of adducts to N-terminal valine in hemoglobin (Hb), using a modified Edman degradation method. This method is applicable to monofunctional epoxides from butadiene. However, in reaction with N-terminal valine, DEB forms an adduct which is ring-closed to a pyrrolidine, N,N-(2,3-dihydroxy-1,4-butadiyl)valine, with a tertiary amino group that prevents detachment of the alkylated valine by the Edman reagent. Therefore a method has been developed based on the analysis by liquid chromatography/electrospray ionization mass spectrometry (LC/ESI-MS) of the N-modified N-terminal peptides enriched after trypsin digestion of globin. In this study Hb samples from mice injected intraperitoneally with (+/-)-DEB were examined qualitatively and quantitatively with regard to the ring-closed adduct. The N-terminal pyrrolidine-heptapeptide was identified in treated mice. The highest adduct levels were obtained in samples from animals given the highest dose of DEB and the adduct levels were below the detection level in control mice.  相似文献

建立了依赖色谱保留时间的智能化选择反应监测质谱方法,并与非依赖色谱保留时间的智能化选择反应监测质谱分析方法对不同体系(牛血清白蛋白酶切物、6种标准蛋白质混合物酶切物、腾冲嗜热菌蛋白提取液酶切物)的分析结果进行了系统比较。结果表明,引入色谱保留时间后的智能化选择反应监测质谱方法能够显著提高肽段及蛋白质的鉴定量,并且在复杂体系(如腾冲嗜热菌蛋白提取液酶切物)中效果尤为明显,鉴定到的肽段及蛋白质的覆盖率可分别达到目标肽段和蛋白质数量的89.62%和92.41%,并且灵敏度高、重复性好,能够实现对质荷比相同但保留时间有差异的肽段的准确鉴定。该方法将在复杂生物样本目标蛋白质组高通量、高灵敏度的鉴定、验证和确认中发挥独特作用。  相似文献

Performance of gravitational field-flow fractionation (GFFF) is improved here with respect to the ability to fractionate and distinguish different varieties of wine-making yeast from Saccharomyces cerevisiae. A new GFFF channel with non-polar walls has been employed to enhance fractionation selectivity and reproducibility. Since GFFF retention depends from first principles on particle size, Coulter counter measurements were performed in order to compare size distribution profiles with GFFF profiles. From such a comparison, GFFF was shown to be able to reveal differences in yeast cells other than size. This could make use of GFFF for screening different varieties of wine-making yeast towards future quality assessment procedures based on a possible correlation between yeast cell morphology indexes and quality indexes.  相似文献

Pressure dependence of several reversed phase liquid chromatographic solvents, including methanol, acetonitrile and mixtures of methanol: water and acetonitrile: water has been investigated. Typical ions, [MH]+, [2M+H]+ and [3M+H]+, were recorded as function of source pressure and as a function of the amount of water in the mixture. Ion formation processes in methanol: water and acetonitrile: water are discussed. The influence of liquid chromatographic mass spectrometric solvent pressure on the mass spectra of samples has been demonstrated on an amino acid sample.  相似文献

Cross-contamination among wells of a high-throughput, high-density assay is a risk that cannot be detected or controlled by the performance of calibration standards and quality control samples. In the current practice, carryover and cross-contamination is detected only when analytes are detected in blank, zero, placebo, pre-dose samples, in a low standard or low quality control sample. There is no mechanism that allows bioanalytical scientists to determine if cross-contamination has occurred among other samples. As a result, erroneous results can be released to clients even though a batch meets the acceptance criteria. We tested a new approach that quantifies the cross-contamination of each sample and allows the scientist to make quality decisions with documentation. The approach will also detect carryover in over 90% of the wells. Briefly, two additional analytes were added as contamination markers. The markers were added to a multi-well plate alternatively creating a pattern of a checkerboard. The spiked multi-well plate was then used to perform the assay. If both markers were detected in a well, the sample was considered contaminated. The amount of the unexpected marker detected in a well measures the degree of contamination and may be used to make deactivation decisions. Depending on the relative impact of the contamination, a scientist can choose to tolerate the bias, reject the sample, reject the batch or raise the lower limit of quantitation for the batch. A guideline for rejection decisions is presented for discussion.  相似文献

A fully automatic membrane inlet mass spectrometric (MIMS) on-line instrumentation for the analysis of aroma compounds in continuous beer fermentation processes was constructed and tested. The instrumentation includes automatic filtration of the sample stream, flushing of all tubing between samples and pH control. The calibration standards can be measured periodically. The instrumentation has also an extra sample line that can be used for off-line sample collection or it can be connected to another on-line method. Detection limits for ethanol, acetic acid and eight organic beer aroma compounds were from μg l−1 to low mg l−1 levels and the standard deviations were less than 3.4%. The method has a good repeatability and linearity in the measurement range. Response times are shorter than or equal to 3 min for all compounds except for ethyl caproate, which has a response time of 8 min. In beer aroma compound analysis a good agreement between MIMS and static headspace gas chromatographic (HSGC) measurements was found. The effects of different matrix compounds commonly present in the fermentation media on the MIMS response to acetaldehyde, ethyl acetate and ethanol were studied. Addition of yeast did not have any effect on the MIMS response of ethanol or ethyl acetate. Sugars, glucose and xylose, increased the MIMS response of all studied analytes only slightly, whereas salts, ammonium chloride, ammonium nitrate and sodium chloride, increased the MIMS response of all three studied compounds prominently. The system was used for on-line monitoring of continuous beer fermentation with immobilised yeast. The results show that with MIMS it is possible to monitor the changes in the continuous process as well as delays in the two-phase process.  相似文献

Protein glycosylation has a significant medical importance as changes in glycosylation patterns have been associated with a number of diseases. Therefore, monitoring potential changes in glycan profiles, and the microheterogeneities associated with glycosylation sites, are becoming increasingly important in the search for disease biomarkers. Highly efficient separations and sensitive methods must be developed to effectively monitor changes in the glycoproteome. These methods must not discriminate against hydrophobic or hydrophilic analytes. The use of activated graphitized carbon as a desalting media and a stationary phase for the purification and the separation of glycans, and as a stationary phase for the separation of small glycopeptides, has previously been reported. Here, we describe the use of activated graphitized carbon as a stationary phase for the separation of hydrophilic tryptic glycopeptides, employing a chip‐based liquid chromatographic (LC) system. The capabilities of both activated graphitized carbon and C18 LC chips for the characterization of the glycopeptides appeared to be comparable. Adequate retention time reproducibility was achieved for both packing types in the chip format. However, hydrophilic glycopeptides were preferentially retained on the activated graphitized carbon chip, thus allowing the identification of hydrophilic glycopeptides which were not effectively retained on C18 chips. On the other hand, hydrophobic glycopeptides were better retained on C18 chips. Characterization of the glycosylation sites of glycoproteins possessing both hydrophilic and hydrophobic glycopeptides is comprehensively achieved using both media. This is feasible considering the limited amount of sample required per analysis (<1 pmol). The performance of both media also appeared comparable when analyzing a four‐protein mixture. An integrated system has been developed which combines liquid (LC) and gas (GC) chromatographic separation with a single mass spectrometer (MS). On-line solid-phase extraction (SPE) of 10–200 ml aqueous samples on a short (10 × 2.0 mm I.D.) precolumn packed with a styrene-divinylbenzene copolymer is used for analyte enrichment. The trace-enrichment procedure was automated by means of a PROSPEKT cartridge-exchange/solvent-selection/valve-switching unit. After sample loading, the precolumn is eluted on-line in two subsequent runs, first onto the GC-MS system and, next, onto the LC-MS system using a particle beam (PB) interface. Prior to entering the PB-MS, the LC eluent passes through the flow cell of a UV diode-array detector (DAD). Both GC-MS and LC-PB-MS generate classical electron ionisation (EI) and chemical ionisation (CI) spectra which are useful for the identification of low- and sub-μg/l concentrations of environmental pollutants covering a wide polarity and volatility range. The LC-DAD data provide additional means for quantitation and yield complementary spectral information. All three detection systems (GC-MS, LC-DAD, LC-PB-MS) and the trace-enrichment procedure are fully automated and controlled from the keyboard of the central computer. With such a ‘MULTIANALYSIS’ system GC-MS, LC-DAD and LC-MS data of the same sample can be obtained within 3 h. The system was optimised with nine chlorinated pesticides in drinking water as test mixture. With 100-ml samples detection limits in GC-MS were 0.0005−0.03 μg/l, and in LC-PB-MS 0.5–7 μg/l, both in the full-scan (EI) mode. Negative chemical ionisation (NCI) with methane as reagent gas improved the sensitivity of six halogenated compounds 3- to 30-fold and provided relevant information for structural elucidation of unknown compounds in real-world samples. LC-DAD detection limits varied from 0.01 to 0.05 μg/l. Relative standard deviations (R.S.D.) of retention times were less than 0.2% in all systems, R.S.D.s of peak areas were 5–15% for GC-MS and LC-PB-MS and less than 5% for LC-DAD. The ‘MULTIANALYSIS’ system was used to analyse surface water samples and river sediment extracts; several pollutants were detected and identified.  相似文献

Protein glycosylation has a major influence on functions of proteins. Studies have shown that aberrations in glycosylation are indicative of disease conditions. This has prompted major research activities for comparative studies of glycoproteins in biological samples. Multiple reaction monitoring (MRM) is a highly sensitive technique which has been recently explored for quantitative proteomics. In this work, MRM was adopted for quantification of glycopeptides derived from both model glycoproteins and depleted human blood serum using glycan oxonium ions as transitions. The utilization of oxonium ions aids in identifying the different types of glycans bound to peptide backbones. MRM experiments were optimized by evaluating different parameters that have a major influence on quantification of glycopeptides, which include MRM time segments, number of transitions, and normalized collision energies. The results indicate that oxonium ions could be adopted for the characterization and quantification of glycopeptides in general, eliminating the need to select specific transitions for individual precursor ions. Also, the specificity increased with the number of transitions and a more sensitive analysis can be obtained by providing specific time segments. This approach can be applied to comparative and quantitative studies of glycopeptides in biological samples as illustrated for the case of depleted blood serum sample. Reversed-phase high-performance liquid chromatography (HPLC) is directly coupled to helium microwave-induced plasma mass spectrometry (He MIP-MS) for the element-selective detection of halogenated organic compounds. Absolute detection limits are approximately 50 pg Br for brominated compounds, 1 pg I for iodinated compounds, and 10 ng Cl for chlorinated compounds. The linear dynamic range for Br- and I-containing compounds is 3-4 orders of magnitude. However, the linear range for chlorinated species is severely limited by high background at m/z = 35. The relative standard deviation for repetitive injections is less than 10%. The helium microwave-induced plasma is operated at moderate powers (300-350 W) and with a total helium consumption of 6-8 L/min. The effect of organic solvents on the background mass spectrum is investigated.  相似文献

A robust and fitted routine method resulting from an analytical optimisation has been applied for the determination of acrylamide in several foods including mainly potato and cereal products. For the sample treatment, different materials were evaluated for filtration and purification of the extract. To increase the performances in terms of sensitivity, a preconcentration to small volume was introduced before liquid chromatography coupled to tandem mass spectrometry analysis on a μ-Bondapak C18 column using d3-acrylamide as internal standard. For identification, relative retention time and two diagnostic ions were monitored. A limit of detection of 10 μg kg−1, a limit of quantitation of 20 μg kg−1, mean recoveries ranging from 100 to 115%, coefficients of variation from 1.36 to 8.06% for repeatability and from 3.3 to 18.2% for reproducibility within the laboratory and a measurement uncertainty of 42% were obtained during an in-house validation procedure. Results of tests, validation data and Z-score obtained during participation to proficiency studies are presented.  相似文献

A liquid chromatography/mass spectrometry (LC/MS) method for separation and characterization of ergosterol biosynthetic precursors was developed to study the effect of Posaconazole on sterol biosynthesis in fungi. Ergosterol biosynthetic precursors were characterized from their electron ionization mass spectra acquired by a normal-phase chromatography, particle beam LC/MS method. Fragment ions resulting from cleavage across the D-ring and an abundant M - 15 fragment ion were diagnostic for methyl substitution at C-4 and C-14. Comparison of the sterol profile in control and treated Candida albicans incubations showed depletion of ergosterol and accumulation of C-4 and C-14 methyl-substituted sterols following treatment with Posaconazole. These C-4 and C-14 methyl sterols are known to be incapable of sustaining cell growth. The results demonstrate that Posaconazole exerts its antifungal activity by inhibition of ergosterol biosynthesis. Furthermore, Posaconazole appears to disrupt ergosterol biosynthesis by inhibition of lanosterol 14alpha-demethylase.  相似文献

A high resolution mass spectrometric read-out system for a vacuum fusion apparatus is described. Rapid analyses are possible and the rate at which various gases are evolved can be monitored. Gas mixture assays are accomplished by scanning the nominal mass 28 peak. Serial measurements of CO+, N2+ and C2H4+ are made. Thus detection and measurement of evolved hydrocarbons are possible. The results for silver and tin from a conventional vacuum fusion apparatus and the mass spectrometer apparatus are compared.  相似文献

A solid-phase extraction (SPE) procedure with the use of superparamagnetic Fe(3)O(4) nanoparticles as extracting agent was developed for HPLC-ESI-MS/MS analysis. Four most heavily used triazine pesticides (herbicides) were taken as the test compounds. The NPs showed an excellent capability to retain the compounds tested, and a quantitative extraction was achieved within 10min under the testing conditions, i.e. 100 microL NP solution was added to 400 mL sample in a beaker with stirring. After extraction, the superparamagnetic NPs were easily collected by using an external magnet. Very importantly, analytes retained on the Fe(3)O(4) NPs could be quantitatively recovered by dissolving the NPs with an HCl solution, allowing subsequent HPLC-ESI-MS/MS quantification. A capillary HPLC-ESI-MS/MS method with the present NP-based SPE procedure was developed for the determination of triazines including atrazine, prometryn, terbutryn, and propazine. Atrazine-d(5) was used as internal standard. The method had an LOD of 10 pg/mL atrazine, and a linear calibration curve over a range from 30 pg to 50.0 ng/mL. Simultaneous determination of the four triazine pesticides in water samples taken from local lakes was demonstrated.  相似文献

A liquid chromatographic-tandem mass spectrometric multiresidue method for the simultaneous quantitative determination of the tetrahydroimidazole, levamisole and the benzimidazoles thiabendazole, oxfendazole, oxibendazole, albendazole, fenbendazole, febantel and triclabendazole in milk has been developed and validated. The anthelmintic residues were extracted with ethyl acetate. The liquid chromatographic separation was performed on a reversed-phase C18 column with gradient elution. The analytes were detected by tandem quadrupole mass spectrometry after positive electrospray ionisation by multiple reaction monitoring. The confirmatory method is very sensitive and each component can be detected at a residue level lower than 1 microgram/l. The method is validated according to the revised European Union requirements and all parameters were found conform the criteria. The evaluated parameters were linearity, specificity, stability, recovery, precision (repeatability and within-laboratory reproducibility) and analytical limits (detection limit, decision limit and detection capability). This analytical method is applied in the Belgian monitoring programme for classical anthelmintic veterinary drugs in raw farm cow's milk.  相似文献

Several recent experiments demonstrated the enhancement of turbulent heat transfer with evaporation as compared with that of a dry body. The present study was an attempt to obtain more accurate data on the heat and mass transfer coefficients with water evaporation. In addition, a method of error evaluation was developed and applied to correct the experimental errors in the recently reported results. Most of the corrected values and the present experimental data revealed that the heat transfer coefficient with evaporation agreed with that of the dry body without evaporation within experimental error.  相似文献
