The time-resolved fluorescence properties of the bacteriophage T4 capsid protein gp23 are investigated. The structural characteristics of this protein are largely unknown and can be probed by recording time-resolved and decay-associated fluorescence spectra and intensity decay curves using a 200 ps-gated intensified CCD-camera. Spectral and decay data are recorded simultaneously, which makes data acquisition fast compared to time-correlated single-photon counting. A red-shift of the emission maximum within the first nanosecond of decay is observed, which can be explained by the different decay-associated spectra of fluorescence lifetimes of the protein in combination with dipolar relaxation. In addition, iodide quenching experiments are performed, to study the degree of exposure of the various tryptophan residues. A model for the origin of the observed lifetimes of 0.032 +/- 0.003, 0.39 +/- 0.06, 2.1 +/- 0.1 and 6.8 +/- 0.8 ns is presented: the 32 ps lifetime can be assigned to the emission of a buried tryptophan residue, the 0.4 and 2.1 ns lifetimes to two partly buried residues, and the 6.8 ns lifetime to a single tryptophan outside the bulk of the folded gp23.  相似文献

The time-resolved fluorescence spectra of the main arterial fluorescent compounds were retrieved using a new algorithm based on the Laguerre expansion of kernels technique. Samples of elastin, collagen and cholesterol were excited with a pulsed nitrogen laser and the emission was measured at 29 discrete wavelengths between 370 and 510 nm. The expansion of the fluorescence impulse response function on the Laguerre basis of functions was optimized to reproduce the observed fluorescence emission. Collagen lifetime (5.3 ns at 390 nm) was substantially larger than that of elastin (2.3 ns) and cholesterol (1.3 ns). Two decay components were identified in the emission decay of the compounds. For collagen, the decay components were markedly wavelength dependent and hydration dependent such that the emission decay became shorter at higher emission wavelengths and with hydration. The decay characteristics of elastin and cholesterol were relatively unchanged with wavelength and with hydration. The observed variations in the time-resolved spectra of elastin, collagen and cholesterol were consistent with the existence of several fluorophores with different emission characteristics. Because the compounds are present in different proportions in healthy and atherosclerotic arterial walls, characteristic differences in their time-resolved emission spectra could be exploited to assess optically the severity of atherosclerotic lesions.  相似文献

A fiber-optic-based system for remote measurement of time-resolved fluorescence emission spectra is described and characterized. A pulsed nitrogen laser is used to induce fluorescence and a time-gated, one-dimensional photodiode array is used to measure the decay of the fluorescence emission spectra. The results compare favorably with reported values for well characterized compounds having fluorescence decay times in the range 4–50 ns. The potential of using time-resolved fluorimetry (TRF) over fiber-optic cables as a means of improving the specificity of remote fluorescence determinations of spectrally similar polycyclic aromatic hydrocarbons in sea water is demonstrated.  相似文献

ANS is widely used as a probe for locating binding sites of proteins and studying structural changes under various external conditions. However, the nature of ANS-binding sites in proteins and the accompanying changes in fluorescence properties are controversial. We examined the steady-state and time-resolved fluorescence of the ANS-protein complexes for tear lipocalin (TL) and its mutants in order to discern the origin of lifetime components via analysis that included the multiexponential decay and the model-free maximum entropy methods. Fluorescence lifetimes of ANS-TL complexes can be grouped into two species, 14.01-17.42 ns and 2.72-4.37 ns. The log-normal analyses of fluorescence spectral shapes reveal the heterogeneous nature of both long- and short-lifetime species. The constructed time-resolved emission, amplitude (TRES) and area normalized (TRANES), and decay-associated spectra are consistent with a model that includes heterogeneous modes of ANS binding with two separate lifetime components. The two lifetime components are not derived from solvent relaxation, but rather may represent different binding modes.  相似文献

Summary Fiber optical recorded time-resolved fluorescence emission spectra distorted by wavelength dispersion have been investigated. Calculated and measured transit times for long fibers (50 m) are in good agreement. A simple correction algorithm was proposed resulting in time-resolved spectra of second-order, bilinear structure. The analysis of various varnishes from different origins, which are commonly found on historical musical instruments was carried out for the first time with both fluorescence excitation emission spectroscopy and laser-induced time-resolved fluorescence spectroscopy. Samples studied include varnishes prepared using shellac, and selected diterpenoid and triterpenoid resins from plants, and mixtures of these materials. Fluorescence excitation emission spectra have been collected from films of naturally aged varnishes. In parallel, time-resolved fluorescence spectroscopy of varnishes provides means for discriminating between short- (less than 2.0 ns) and long-lived (greater than 7.5 ns) fluorescence emissions in each of these complex materials. Results suggest that complementary use of the two non destructive techniques allows a better understanding of the main fluorophores responsible for the emission in shellac, and further provides means for distinguishing the main classes of other varnishes based on differences in fluorescence lifetime behaviour. Spectrofluorimetric data and time resolved spectra presented here may form the basis for the interpretation of results from future in situ fluorescence examination and time resolved fluorescence imaging of varnished musical instruments.  相似文献

The excited-state processes of protochlorophyllide a, the precursor of chlorophyll a in chlorophyll biosynthesis, are studied using picosecond time-resolved fluorescence spectroscopy. Following excitation into the Soret band, two distinct fluorescence components, with emission maxima at 640 and 647 nm, are observed. The 640 nm emitting component appears within the time resolution of the experiment and then decays with a time constant of 27 ps. In contrast, the 647 nm emitting component is built up with a 3.5 ps rise time and undergoes a subsequent decay with a time constant of 3.5 ns. The 3.5 ps rise kinetics are attributed to relaxations in the electronically excited state preceding the nanosecond fluorescence, which is ascribed to emission out of the thermally equilibrated S(1) state. The 27 ps fluorescence, which appears within the experimental response of the streak camera, is suggested to originate from a second minimum on the excited-state potential-energy surface. The population of the secondary excited state is suggested to reflect a very fast motion out of the Franck-Condon region along a reaction coordinate different from the one connecting the Franck-Condon region with the S(1) potential-energy minimum. The 27 ps-component is an emissive intermediate on the reactive excited-state pathway, as its decay yields the intermediate photoproduct, which has been identified previously (J. Phys. Chem. B 2006, 110, 4399-4406). No emission of the photoproduct is observed. The results of the time-resolved fluorescence study allow a detailed spectral characterization of the emission of the excited states in protochlorophyllide a, and the refinement of the kinetic model deduced from ultrafast absorption measurements.  相似文献

Urinary cytology is a noninvasive and unconstraining technique for urothelial cancer diagnosis but lacks sensitivity for detecting low-grade lesions. In this study, the fluorescence properties of classical Papanicolaou-stained urothelial cytological slides from patients or from cell lines were monitored to investigate metabolic changes in normal and tumoral cells. Time- and spectrally-resolved fluorescence imaging was performed at the single cell level to assess the spectral and temporal properties as well as the spatial distribution of the fluorescence emitted by urothelial cells. The results reveal quite different fluorescence distributions between tumoral urothelial cells, characterized by a perimembrane fluorescence localization, and the normal cells which exhibit an intracellular fluorescence. This is not caused by differences in the fluorescence emission of the endogenous fluorophores NAD(P)H, flavoproteins or porphyrins but by various localization of the EA 50 Papanicolaou stain as revealed by both the spectral and time-resolved parameters. The present results demonstrate that the use of single-cell endofluorescence emission of Papanicolaou-stained urothelial cytological slides can allow an early ex vivo diagnosis of low-grade bladder cancers.  相似文献

Time-resolved area normalized emission spectroscopy (TRANES) is a new method for the analysis of fluorescence of dyes in complex chemical and biological systems (A S R Koti, M M G Krishna and N Periasamy, 2001,J. Phys. Chem. 105, 1767). The model-free method extends the power of time-resolved emission spectroscopy (TRES) analysis and removes the ambiguity in the interpretation when the emission spectrum is time-dependent. Observation of an isoemissive point in TRANES analysis of fluorescence is an unambiguous indication for the presence of two emissive species in the sample. The isoemissive point occurs at a wavelength where the ratio of the radiative rates of the two species is equal to the ratio of their total radiative rates. The polarity-sensitive nile red dye shows time-dependent emission spectra in the organized bilayer assemblies of TX micelle and bilayer egg-phosphotidylcholine (egg-PC) membrane. Time-dependent spectra in complex systems support many important models (solvation model and heterogeneity in the ground and/or excited state). TRANES analysis shows that the fluorescence emission of nile red in TX micelle and egg-PC membrane is due to two emissive species solubilized in different sites.  相似文献

The time-resolved emission properties of selected anthracene derivatives, namely anthracene (1a), 2-methylanthracene (1b), 2-chloroanthracene (1c), 2-methoxyanthracene (1d), 2-methoxy-6-methylanthracene (1e), 2-(N,N'-dicyclohexylureidocarbonyl)-6-methoxyanthrace ne (1f), 2-(6-methoxyanthr-2-yl)-4,4-dimethyl-2-oxazoline (1g), 2-(6-methoxyanthr-2-yl)-pyridine (1h), and N-cyclohexylanthracene-2-carboxamide (1i) were investigated. In contrast to anthracene (1a), 1b, and 1c, the 2-methoxy-substituted anthracene derivatives 1d-1h exhibit two emission lifetimes. The determination of the lifetimes at different emission wavelengths and additional time-resolved emission spectroscopy (TRES) reveal that the dual emission originates from two different, interconvertible emissive species, with the s-cis and s-trans conformation relative to the exocyclic C2-O bond. The energy difference between the two emissive species is very small (<0.1 eV) both in the ground and in the excited state. The larger energy difference between the conformers in the excited-state is responsible for the interconversion within the singlet excited-state lifetimes of the s-cis into the s-trans conformations leading to coupled decay kinetics. The proposed mechanism for the dual emission was qualitatively supported by theoretical studies on CASSCF and DFT level. In addition, the emission lifetimes of the fluoride- and pH-sensitive fluorescent probes 1f and 1g change upon addition of fluoride or acid, respectively, so that in these cases the detection of the fluorescence lifetime may be used complementary to the steady-state fluorimetric detection of these analytes.  相似文献

The development of organic materials with white-light emission and thermally activated delayed fluorescence (TADF) properties in the solid state remain a challenge. Herein, a series of white-light-emitting organic luminogens have been developed and are found to show aggregation-induced delayed fluorescence (AIDF) characteristics. The AIDF emitters present dual-emission consisted of prompt fluorescence and TADF in the crystalline state. Their white-light emissions can be easily tuned by altering the chemical structure and connecting position of the heterocyclic aromatic substituent. Under the stimuli of mechanical force and solvent vapor, the compounds exhibit remarkable and reversible mechanochromism, in which their emission colors are switchable between white and yellow. Upon grinding, they also display linearly tunable luminescence colors, as well as force-induced TADF enhancement, which may be associated with the more compact molecular packing and the restriction of intramolecular motions. The results from time-resolved emission scanning and theoretical calculation suggest that the dual-emission of the AIDF luminogens likely results from the twisted intramolecular charge transfer transitions of the molecules, and the reversible mechanochromism properties probably stem from the interconversion of the quasi-axial and the quasi-equatorial conformations.  相似文献

Photophysical properties of a natural plant alkaloid, ellipticine (5,11-dimethyl-6H-pyrido[4,3-b]carbazole), which comprises both proton donating and accepting sites, have been studied in different solvents using steady state and time-resolved fluorescence techniques primarily to understand the origin of dual fluorescence that this molecule exhibits in some specific alcoholic solvents. Ground and excited state calculations based on density functional theory have also been carried out to help interpretation of the experimental data. It is shown that the long-wavelength emission of the molecule is dependent on the hydrogen bond donating ability of the solvent, and in methanol, this emission band arises solely from an excited state reaction. However, in ethylene glycol, both ground and excited state reactions contribute to the long wavelength emission. The time-resolved fluorescence data of the system in methanol and ethylene glycol indicates the presence of two different hydrogen bonded species of ellipticine of which only one participates in the excited state reaction. The rate constant of the excited state reaction in these solvents is estimated to be around 4.2-8.0 × 10(8) s(-1). It appears that the present results are better understood in terms of solvent-mediated excited state intramolecular proton transfer reaction from the pyrrole nitrogen to the pyridine nitrogen leading to the formation of the tautomeric form of the molecule rather than excited state proton transfer from the solvents leading to the formation of the protonated form of ellipticine.  相似文献

In order to test whether lignin fluorescence originates from discrete fluorophores, fluorescence emission spectra of the lignin model dehydrogenative polymer (DHP) were analyzed by the band deconvolution method and time-resolved analysis of both the excitation and emission spectra. Two series of 22 fluorescence emission spectra of DHP in chloroform/methanol (3:1, v/v) solution, and as a solid suspension in water, were deconvoluted into three fluorescence and one Raman Gaussian components. Emission spectra were obtained by stepwise variation of the excitation wavelength from 360 to 465 nm. Deconvolution was performed by nonlinear fitting of all three Gaussian parameters: area, width and position. Position of all components in a series was treated as a random variable and its approximate probability distribution (APD) calculated from a series of histograms with increasing number of abscissa intervals. A five peak multimodal APD profile was obtained for both series of DHP emission spectra. The mean fluorescence lifetime varied with wavelength both in the emission and the excitation decay-associated spectra (DAS), where four kinetic components were resolved. The shapes of the excitation spectra of the four components were quite different and gradually shifted bathochromically. The multicomponent nature of the DHP emission spectra along with the changes in the mean fluorescence lifetime and the form of the excitation DAS of the four components give evidence of the heterogeneous origin of fluorescent species emitting in the visible.  相似文献

The differentiation of proteins commonly found as binding media in paintings is presented based on spectrally resolved and time-resolved laser-induced fluorescence (LIF) and total emission spectroscopy. Proteins from eggs and animal glue were analysed with pulsed laser excitation at 248 nm (KrF excimer) and 355 nm (third harmonic of Nd:YAG) for spectrally resolved measurements, and at 337 nm (N2) and 405 nm (N2 pumped dye laser) for spectrally resolved lifetime measurements and fluorescence lifetime imaging (FLIM). Total emission spectra of binding media are used for the interpretation of LIF spectra. Time-resolved techniques become decisive with excitation at longer wavelengths as fluorescence lifetime permits the discrimination amongst binding media, despite minimal spectral differences; spectrally resolved measurements of fluorescence lifetime have maximum differences between the binding media examined using excitation at 337 nm, with maximum observed fluorescence at 410 nm. FLIM, which measures the average lifetime of the emissions detected, can also differentiate between media, is non-invasive and is potentially advantageous for the analysis of paintings. We present the results of the deterministic identifiability analysis based on similarity transformation for models of one-state excited-state events of cylindrically symmetric rotors in isotropic environments undergoing rotational diffusion described by Brownian reorientation. Such an analysis on error-free time-resolved fluorescence (anisotropy) data can reveal whether the parameters of the considered model can be determined. The fluorescence delta-response functions I(parallel)(t) and I(perpendicular)(t), for fluorescence polarized respectively parallel and perpendicular to the electric vector of linearly polarized excitation, are used to construct, in convenient matrix form, expressions of the sum S(t) = I(parallel)(t) + 2I(perpendicular)(t), the difference D(t) = I(parallel)(t) - I(perpendicular)(t), and the time-resolved fluorescence anisotropy r(t) = D(t)/S(t). The identifiability analysis of r(t) demonstrates that the rotational diffusion coefficients D(parallel) and D(perpendicular) for rotation respectively about and perpendicular to the symmetry axis can be uniquely resolved. However, the polar and azimuthal angles defining the absorption and emission transition moments in the molecular reference frame are not individually identifiable. Lanthanide-based luminescent sensors are noteworthy because their long-lifetime luminescence enables time-resolved fluorescence measurements. After exploring suitable antenna groups, we designed and synthesized lanthanide-based luminogenic sensors detecting protease activities. This sensor yielded strong luminescence on addition of proteases such as calpain I and leucine aminopeptidase (LAP). Since the luminescence lifetimes of the probes were very long, the sensors could be applied to time-resolved measurements that exclude background fluorescence signals derived from proteins or other impurities. This sensing principle could be applicable to general time-resolved fluorescence assays for other proteases.  相似文献

Absorption, steady state fluorescence and time-resolved fluorescence spectra of omeprazole (OMP) have been studied in solvents of different polarity and pH. With an increase in the polarity of the solvents, blue shift is observed in the longer wavelength whereas red shift is noticed in the shorter wavelength band. The dual emission observed in non-polar solvents suggests that the energy of the twisted intramolecular charge transfer (TICT) state is lower than that of the locally excited (LE) state. The normal Stokes-shifted band originates from the LE state, and the large Stokes-shifted band is due to the emission from a TICT state. The Stokes shift of OMP is correlated with various solvent polarity scales like ET(30) and f?(D,n).  相似文献

Hypericin is the active ingredient of the off-the-shelf antidepressant St. John's Wort. It is an effective phototoxic agent and its systemic administration at therapeutic doses could induce particular damage in the eye due to continuous light exposure. Hypercin is strongly fluorescent and its fluorescence properties can be monitored to investigate noninvasively its localization and interactions. To this aim, time-resolved microspectrofluorimetry and fluorescence lifetime imaging were used to assess the spectral and temporal properties as well as the spatial distribution of the fluorescence emitted by retinal pigment epithelium (RPE) cells treated with Hyp at concentrations in the micromolar range (0.5-10 microM). In the presence of hypericin, the emission peaks at 600-605 nm and the fluorescence decay is best fitted with three lifetimes (5.5-7 ns, 1.9-2.5 ns and <0.8 ns). Spectral and temporal differences were observed between high (> or =5 microM) and low hypericin concentrations. In particular, upon increasing concentration, the emission spectrum of the slow component broadens and its lifetime shortens. The latter change is observed also when high concentrations are reached locally, due to more efficient localization within the cell.  相似文献

Femtosecond/picosecond time-resolved fluorescence study of hydrophilic polymer fine particles (polyacrylamide, PAAm) was reported. Ultrafast fluorescence dynamics of polymer/water solution was monitored using a fluorescent probe molecule (C153). In the femtosecond time-resolved fluorescence measurement at 480 nm, slowly decay components having lifetimes of tau(1) approximately 53 ps and tau(2) approximately 5 ns were observed in addition to rapid fluorescence decay. Picosecond time-resolved fluorescence spectra of C153/PAAm/H2O solution were also measured. In the time-resolved fluorescence spectra of C153/PAAm/H2O, a peak shift from 490 to 515 nm was measured, which can be assigned to the solvation dynamics of polymer fine particles. The fluorescence peak shift was related to the solvation response function and two time constants were determined (tau(3) approximately 50 ps and tau(4) approximately 467 ps). Therefore, the tau(1) component observed in the femtosecond time-resolved fluorescence measurement was assigned to the solvation dynamics that was observed only in the presence of polymer fine particles. Rotational diffusion measurements were also carried out on the basis of the picosecond time-resolved fluorescence spectra. In the C153/PAAm/H2O solution, anisotropy decay having two different time constants was also derived (tau(6) approximately 76 ps and tau(7) approximately 676 ps), indicating the presence of two different microscopic molecular environments around the polymer surface. Using the Stokes-Einstein-Debye (SED) equation, microscopic viscosity around the polymer surface was evaluated. For the area that gave a rotational diffusion time of tau(6) approximately 76 ps, the calculated viscosity is approximately 1.1 cP and for tau(7) approximately 676 ps, it is approximately 10 cP. The calculated viscosity values clearly revealed that there are two different molecular environments around the polyacrylamide fine particles.  相似文献
