A gradient polyethylene (PE) surface was created through corona treatment, in which the corona power increased along the 5 cm length of the PE. The gradient surface was treated with polyethyleneimine and then biotin. Fluorescein-conjugated streptavidin (SAV) caged within 5-carboxymethoxy-2-nitrobenzyl (CMNB) adsorbed onto the biotinylated gradient PE surface following molecular recognition principles. Photoirradiation decomposed the CMNB cage and allowed the fluorescein-conjugated SAV to fluoresce, the intensity of which increased gradually along the PE surface.  相似文献

We covalently immobilized poly(ethylene oxide) (PEO) chains onto a fluorinated ethylene propylene copolymer (FEP) surface. On the FEP surface, aldehyde groups were first deposited by plasma polymerization of acetaldehyde or acrolein. Then, amino‐PEO chains were immobilized through Schiff base formation, which was followed by reduction stabilization with sodium cyanoborohydride. The PEO‐grafted polymer surfaces thus prepared were characterized by X‐ray photoelectron spectroscopy (XPS), atomic force microscopy, contact‐angle measurements, and protein adsorption. The dramatic increase in the C O intensity of the high‐resolution XPS C 1s spectrum, together with an overall increase in oxygen content, indicated the successful attachment of PEO chains onto the acetaldehyde plasma surfaces. The amount of grafted PEO chains depended on the superfacial density of the plasma‐generated aldehyde groups. The grafted monoamino‐PEO chains formed a brushlike structure on the polymer surface, whereas the bisamino‐PEO chains predominately adopted a looplike conformation. The PEO surface had a regular morphology with greater roughness than the aldehyde surface underneath. Surface hydrophilicity increased with the grafting of PEO. Also, the bisamino‐PEO‐grafted surface had slightly higher surface hydrophilicity than its monoamino‐PEO counterpart. A collagen gradient was constructed to interrogate cell adhesion on a poly(L-lactide) (PLLA) membrane surface. Utilizing a microinfusion pump, gradients of amino groups were generated on the PLLA surface by an aminolysis method. Immobilization of collagen onto the gradient surfaces was performed by glutaraldehyde (GA) coupling to form the collagen gradients. The -NH(2) and immobilized collagen density profiles on the PLLA membranes were quantitatively determined by ninhydrin and hydroproline (Hyp) analysis, respectively. By using fluorescein isothiocyanate (FITC) labeled collagen (FITC-Col), the profile of the as-prepared collagen gradient was directly monitored by a fluorescence microscope. The scanning force microscopy and water contact angle studies revealed that the morphology and wettability of the modified membranes changed progressively as a function of position along the gradient surface. Rabbit auricular chondrocytes were cultured on the collagen gradient membranes to test their cellular response. The attachment and spreading behaviors of the chondrocytes were dependent on the surface collagen density. These results indicate a surface on which the variation of collagen gradient strongly modifies the biological response of chondrocytes.  相似文献

In this paper we present a study of using oxygen plasma for chemically modifying inert hydrocarbon self-assembled monolayers of octadecyltrichlorosilane (OTS-SAMs) and rendering active surfaces for protein immobilization. Detailed surface modification and protein immobilization were characterized by using ellipsometry, X-ray photoelectron spectroscopy (XPS), Fourier transform infrared-attenuated total reflectance spectroscopy, and fluorescence microscopy. Our XPS results showed that the surface reaction between OTS-SAMs and oxygen plasma can generate new surface functional groups such as alcohol (C-O), aldehyde (C=O), and carboxylic acid (O-C=O), and their compositions can be controlled by using different treatment times and powers. A short treatment time ( approximately 1 s) and high power (10 W) can lead to a higher density of aldehyde groups, which can serve as linker groups for protein immobilization through the formation of Schiff bases with the amine groups of proteins. By using the fluorescence immunostaining method, we confirmed that human immunoglobulin (IgG) can be immobilized on a glass slide, only if the surface was decorated with OTS-SAMs and if the OTS-SAMs were pretreated with oxygen plasma. The protein immobilized on the oxygen-plasma-treated surface can only be recognized by using a highly specific antibody, FITC-anti-IgG, but not FITC-anti-biotin.  相似文献

A method for the development of continuous density gradients of immobilized oligonucleotide probes (20mer) along the length of microfluidic channels is demonstrated. The development of continuous density gradients was achieved using variable electrokinetic transport of probes in hybrid glass-polydimethylsiloxane microfluidic chips. The probes were terminated with an amine functional group, and were delivered by electrokinetic pumping to the flat glass channel wall after it had been densely coated with covalently immobilized aldehyde groups. This method provided probe immobilization densities ranging from 4.5(±0.8)×10(13) to 2.5(±0.8)×10(11) molecules cm(-2), with longitudinal dilution and differential mass transport of the injected plug of probes being the primary factors responsible for the gradient of density. The utility of the resulting density gradient of immobilized probes to control the selectivity of hybridization was demonstrated at room temperature by discrimination between a fully complementary oligonucleotide target, and a target strand containing 3 base pair mismatches (3 BPM) based on the spatial pattern of hybridization for sub-picomole quantities of targets. Single nucleotide polymorphism (SNP) discrimination was possible when temperature control was implemented to improve resolution of the mismatch discrimination, allowing SNP discrimination at 35 °C with a contrast ratio of almost 5 to 1.  相似文献

The process of surface functionalization involving silanization, biotinylation and streptavidin bonding as platform for biospecific ligand immobilization was optimized for thin film polyimide spin-coated silicon wafers, of which the polyimide film serves as a wave guiding layer in evanescent wave photonic biosensors. This type of optical sensors make great demands on the materials involved as well as on the layer properties, such as the optical quality, the layer thickness and the surface roughness. In this work we realized the binding of a 3-mercaptopropyl trimethoxysilane on an oxygen plasma activated polyimide surface followed by subsequent derivatization of the reactive thiol groups with maleimide-PEG2-biotin and immobilization of streptavidin. The progress of the functionalization was monitored by using different fluorescence labels for optimization of the chemical derivatization steps. Further, X-ray photoelectron spectroscopy and atomic force microscopy were utilized for the characterization of the modified surface. These established analytical methods allowed to derive information like chemical composition of the surface, surface coverage with immobilized streptavidin, as well as parameters of the surface roughness. The proposed functionalization protocol furnished a surface density of 144 fmol mm−2 streptavidin with good reproducibility (13.9% RSD, n = 10) and without inflicted damage to the surface. This surface modification was applied to polyimide based Mach-Zehnder interferometer sensors to realize a real-time measurement of streptavidin binding validating the functionality of the MZI biosensor. Subsequently, this streptavidin surface was employed to immobilize biotinylated single-stranded DNA and utilized for monitoring of selective DNA hybridization. These proved the usability of polyimide based evanescent photonic devices for biosensing application.  相似文献

Surface amine gradients that exhibit a wide variety of profiles, including those that incorporate spatially distinct regions having steep and gradual variations in chemical functionality, have been prepared by the sol-gel process using a controlled-rate infusion method. In this work, a substrate that incorporates dimethyl and Si-OH groups is temporally modified with an aminoalkoxysilane (NH(2)(CH(2))(3)Si(OC(2)H(5))(3)) to build a gradient film for which the amine content changes over a 10-20 mm distance. Both X-ray photoelectron spectroscopy (XPS) and contact angle measurements confirm the presence of a chemical gradient across the surface of the film. As expected, a greater density of amine functionalities and lower contact angle were found at the bottom of the gradient relative to the top. The local steepness of the gradient was systematically controlled by changing the rate of infusion. Fast rates of infusion created gradient surfaces where the amine content changed slowly along the surface and never reached saturation, whereas slow rates of infusion formed a surface exhibiting a steep rise in amine content followed by saturation. The steepness of the gradient was also changed at predefined positions along its length by programming the rate of infusion. Gradients prepared using six-step, three-step, and two-step programmed infusion rates are shown. The data fit nicely to a kinetic model that assumes first-order kinetics. The ability to manipulate the gradient profile is particularly vital for applications that rely on mass transport and/or those that require spatial control of gradient properties.  相似文献

The electrostatic adsorption onto charged surfaces of comb copolymers comprising a polyelectrolyte backbone and pendent PEG side chains, such as poly(l-lysine)-g-poly(ethylene glycol) (PLL-g-PEG), has in previous studies provided protein-repellent thin coatings, particularly on metal oxide surfaces. A drawback of this approach is, however, the instability of such adsorbed layers under extreme pH values or high ionic strength. We have overcome this limitation in the present study by covalently immobilizing PLL-g-PEG copolymers onto aldehyde plasma-modified substrates. Silicon wafers, optical waveguide chips, and perfluorinated ethylene-co-propylene (FEP) polymer substrates were first coated with a thin plasma polymer layer using a propionaldehyde plasma, followed by covalent immobilization of PLL-g-PEG via reductive amination between amine groups of the PLL backbone with aldehyde groups on the plasma-deposited interlayer. The stability in high salt media and the protein resistance of different molecular architectures of immobilized PLL-g-PEG layers were quantitatively investigated by XPS, an optical waveguide technique (OWLS), and ToF-SIMS. The adsorption of bovine serum albumin was found to be below the detection limit (<2 ng/cm(2)), as for electrostatically adsorbed PLL-g-PEG layers. However, after 24 h of exposure of covalently immobilized layers of PLL-g-PEG to high ionic strength buffer (2400 mM NaCl), no significant change in the protein resistance was observed, whereas under the same conditions electrostatically adsorbed PLL-g-PEG coatings lost their protein resistance. Moreover, covalent immobilization via an aldehyde plasma interlayer enabled the application of PLL-g-PEG layers onto substrates such as FEP onto which electrostatic binding is not possible. These findings create a generic platform for the covalent immobilization of PLL-g-PEG onto a wide variety of substrates.  相似文献

The homobifunctional crosslinker 1,4‐phenylene diisothiocyanate (PDC) was coupled to amine‐terminated magnetic particles, and human IgG, streptavidin, protein G and protein A were immobilized on the activated magnetic particles. The coupling of PDC to the amine‐terminated magnetic particles was completed in 10 min, and 1 mg of activated magnetic particles was able to immobilize 95 (g of protein G, 120 µg of protein A, 160 µg of streptavidin and 280 µg of IgG. Ultraviolet‐visible spectroscopy, FTIR spectroscopy and electron micrography were used to characterize the functional particles. The results indicated that PDC was successfully coupled to the surface of the amine‐terminated magnetic particles, and the proteins were effectively immobilized on their surface. The activity of protein G immobilized on the activated magnetic particles was confirmed by its ability to purify IgG from plasma.  相似文献

<正>A generic method was described to change surface biocompatibihty by introducing reactive functional groups onto surfaces of polymeric substrates and covalently binding them with biomolecules.A block copolymer with protected carboxylic acid functionality,poly(styrene-b-tert-butyl acrylate)(PS-PtBA),was spin coated from solutions in toluene on a bioinert polystyrene(PS) substrate to form a bilayer structure:a surface layer of the poly(tert-butyl acrylate)(PtBA) blocks that order at the air-polymer interface and a bottom layer of the PS blocks that entangle with the PS substrate.The thickness of the PtBA layer and the area density of tert-butyl ester groups of PtBA increased linearly with the concentration of the spin coating solution until a 2 nm saturated monolayer coverage of PtBA was achieved at the concentration of 0.4%W/W.The protected carboxylic acid groups were generated by exposing the tert-butyl ester groups of PtBA to trifluoroacetic acid (TFA) for bioconjugation with FMRF peptides via amide bonds.The yield of the bioconjugation reaction for the saturated surface was calculated to be 37.1%based on X-ray photoelectron spectroscopy(XPS) measurements.The success of each functionalization step was demonstrated and characterized by XPS and contact angle measurements.This polymer functionalization/modification concept can be virtually applied to any polymeric substrate by choosing appropriate functional block copolymers and biomolecules to attain novel biocompatibility.  相似文献

Commercially available polystyrene (PS) slides were plasma nanotextured (nano-roughened) through treatment in oxygen plasma discharges to create substrates with increased surface area for microarray applications. Conditions of plasma treatment were determined for maximum and uniform oligonucleotide immobilization on these nanotextured PS slides. Oligonucleotides were immobilized onto the surface in the form of biotinylated oligonucleotide/streptavidin conjugates to take advantage of increased protein binding capacity of the substrate. It was found that the amount of oligonucleotides that could be immobilized was increased up to ten times on plasma treated as compared with untreated slides. The sensitivity of detection of labelled hybridized probes was improved by a factor of 20. Optimized nanotextured PS slides were subsequently used to develop a microarray for the detection of three deleterious BRCA1 gene mutations by immobilizing oligonucleotides corresponding to wild and mutant-type sequences. The microarray developed on the nanotextured PS slides provided higher specific hybridization signal and discrimination ratios as compared with flat untreated PS slides.  相似文献

In this study heparin was covalently immobilized onto LDPE-VEMAC sheet fabricated by the introduction of carboxyl groups to the surface of low-density polyethylene (LDPE) using a plasma technique. The plasma irradiation time influenced the density of carboxyl groups on the LDPE-VEMAC sheet. Heparin was immobilized on the LDPE-VEMAC sheet using a condensation reagent, N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide hydrochloride (EDC). We confirmed the immobilization of heparin from the ATR-FT-IR spectrum of the sheet obtained. Where heparin was directly immobilized on the LDPE-VEMAC sheet, the density of the immobilized heparin depended on that of the carboxyl groups. Heparin was also immobilized with a spacer, hexamethylene diamine, and the density of such heparin was about 1.6 times that of the directly immobilized heparin. This result suggests that the introduction of a spacer may be an effective way to increase the density of immobilized heparin.  相似文献

A microfluidic network (μFN) etched into a silicon wafer was used to deliver protein solutions containing different concentrations of the axonal guidance molecule ephrinA5 onto a silicone stamp. In a subsequent microcontact printing (μCP) step, the protein was transferred onto a polystyrene culture dish. In this way, stepwise substrate-bound concentration gradients of ephrinA5 were fabricated spanning a total distance of 320 μm. We tested the response of chick retinal ganglion cell (RGC) axons, which are guided in vivo by ephrin gradients, to these in vitro gradients. Temporal, but not nasal axons stop at a distinct zone in the gradient, which is covered with a certain surface density of substrate-bound ephrinA5. Within the temporal RGC population, all axons respond uniformly to the gradients tested. The position of the stop zone depends on the slope of the gradient with axons growing further into the gradient in shallow gradients than in steep gradients. However, axons stop at lower ephrinA5 concentrations in shallow gradients than in steep gradients, indicating that the growth cone can adjust its sensitivity during the detection of a concentration gradient of ephrinA5. Ca2+‐regulated photoprotein obelin was genetically fused with a minimum‐sized core streptavidin. Hybrid protein (SAV–OL) was produced by bacterial expression and applied as a specific bioluminescent probe in diverse solid‐phase assays. The obtained results clearly demonstrate specific activity of each domain indicating its proper folding with favorable space orientation. SAV–OL has been shown to be a much more sensitive label than the chemical conjugate of a full‐length streptavidin with obelin.  相似文献

Evaluation of the streptavidin‐biotin binding at the surface of chitin film was carried out with voltammetry. Immobilization of streptavidin was attempted to the protonated chitin film, based on an electrostatic interaction that hardly causes any change in the protein structure. The streptavidin‐biotin binding was estimated from changes in the electrode response of biotin labeled with an electroactive compound. Although the response of daunomycin as an electroactive compound did not change at an electrode covered with streptavidin/chitin film, the response of the labeled biotin decreased. This observation shows that streptavidin is immobilized on the chitin film and the biotin binds with immobilized streptavidin. Consequently, it was clear that the chitin film is useful as a reaction field for protein‐ligand binding. Generally, a binding event between protein and its ligand in the living body occurs on the cell surface. The electrochemical evaluation of protein‐ligand binding on a natural polysaccharide like chitin membrane surface is important.  相似文献

Micron and submicron-scale features of aldehyde functionality were fabricated in polymer films by photolithography to develop a platform for protein immobilization and assembly at a biologically relevant scale. Films containing the pH-reactive polymer poly(3,3'-diethoxypropyl methacrylate) and a photoacid generator (PAG) were patterned from 500 nm to 40 mum by exposure to 365 nm (i-line) light. Upon PAG activation and hydrolysis of acetals, aldehyde groups formed. After the films were incubated with a biotinylated aldehyde reactive probe, the X-ray photoelectron spectroscopy results were consistent with biotin being attached to the surface. The background was subsequently passivated by flood exposure and incubation with an aminooxy-terminated poly(ethylene glycol), resulting in a 98% reduction in nonspecific protein adsorption. Protein patterning and assembly was demonstrated using streptavidin, biotinylated anthrax toxin receptor-1, and the protective antigen moiety of anthrax toxin and confirmed by fluorescence microscopy and atomic force microscopy (AFM). AFM demonstrated that 500 nm protein features were achieved. Because of the abundance of biotinylated proteins, this methodology provides a platform for protein immobilization and assembly for various applications in biotechnology.  相似文献

Two approaches to producing gradients of vertically aligned single-walled carbon nanotubes (SWCNTs) on silicon surfaces by chemical grafting are presented here. The first approach involves the use of a porous silicon (pSi) substrate featuring a pore size gradient, which is functionalized with 3-aminopropyltriethoxysilane (APTES). Carboxylated SWCNTs are then immobilized on the topography gradient via carbodiimide coupling. Our results show that as the pSi pore size and porosity increase across the substrate the SWCNT coverage decreases concurrently. In contrast, the second gradient is an amine-functionality gradient produced by means of vapor-phase diffusion of APTES from a reservoir onto a silicon wafer where APTES attachment changes as a function of distance from the APTES reservoir. Carboxylated SWCNTs are then immobilized via carbodiimide coupling to the amine-terminated silicon gradient. Our observations confirm that with decreasing APTES density on the surface the coverage of the attached SWCNTs also decreases. These gradient platforms pave the way for the time-efficient optimization of SWCNT coverage for applications ranging from field emission to water filtration to drug delivery.  相似文献

Protein adsorption and dissociation on cell membrane surfaces is a topic of important study to reveal biological processes including signal transduction and protein trafficking. We demonstrated here the establishment of a mimic model system for the spatial control of protein adsorption/elimination on a lipid bimembrane using a photochemical technique. The novel photoeliminative linker that we synthesized here consists of three distinct components: a substrate (biotin), a photoeliminative group (4-(4-(1-hydroxyethyl)-2-methoxy-5-nitrophenoxy)butanoic acid), and a lipid bimembrane-adsorbent group (farnesyl). The photoeliminative linker was inserted on the entire surface of the lipid bimembrane and two-dimensionally eliminated by spatial UV irradiation onto the membrane to create a biotin pattern. A target protein, streptavidin was selectively immobilized on the patterned biotin, although it was almost not attached on the nonirradiated region. The streptavidin array was selectively dissociated by UV irradiation onto the entire membrane.  相似文献

Herein we report the topochemical modification of polymer surfaces with perfluorinated aromatic azides. The aryl azides, which have quaternary amine or aldehyde functional groups, were linked to the surface of the polymer by UV irradiation. The polymer substrates used in this study were cyclic olefin copolymer and poly(methyl methacrylate). These substrates were characterized before and after modification using reflection-absorption infrared spectroscopy, sessile water contact angle measurements, and X-ray photoelectron spectroscopy. Analysis of the surface confirmed the presence of aromatic groups with aldehyde or quaternary amine functionality. Enzyme immobilization and patterning onto polymer surfaces were studied using confocal microscopy. Enzymatic digests of protein were carried out on modified probes manufactured from thermoplastic substrates, and the resulting peptide analysis was completed using matrix-assisted laser desorption/ionization mass spectrometry. The use of functionalized perfluorinated aromatic azides allows the surface chemistry of thermoplastics to be tailored for specific lab-on-a-chip applications.  相似文献

Surface modification is an important technique in fields, such as, self‐cleaning, surface patterning, sensing, and detection. The diselenide bond was shown to be a dynamic covalent bond that can undergo a diselenide metathesis reaction simply under visible light irradiation. Herein we develop this diselenide dynamic chemistry into a versatile surface modification method with a fast response and reversibility. The diselenide bond could be modified onto various substrates, such as, PDMS, quartz, and ITO conductive film glass. Different functional diselenide molecules could then be immobilized onto the surface via diselenide metathesis reaction. We demonstrated that by using this modification method we could achieve liquid motion in a capillary tube under light illumination. We also show that this approach has the potential to serve as an efficient modification method for surface bioconjugation, which has practical applications in clinical usage.  相似文献
