Polylactide (PLA)‐grafted dextran was synthesized with a trimethylsilyl protection method to produce novel biodegradable, biomedical materials. PLA‐grafted dextrans with various lengths and numbers of graft chains were synthesized. The properties of solution‐cast films prepared from PLA‐grafted dextrans were investigated with thermal and dynamic mechanical analyses. The graft‐copolymer films exhibited lower glass‐transition temperatures, melting temperatures (Tm's), and crystallinities as well as higher viscosity properties as compared with poly‐L ‐lactide film. The Tm and crystallinity and mechanical properties at 37 °C could be adjusted by controlling the molecular structure such as the lengths and numbers of graft chains. Furthermore, the biodegradability of PLA‐grafted dextran films was investigated through the weight change of film and the molecular weight change of polymer during the in vitro degradation test. PLA‐grafted dextrans exhibited different degradation behavior from poly‐L ‐lactide with the introduction of a polysaccharide segment and branched structure as well as the change of end‐functional group. The degradation rate of PLA‐grafted dextran and the cast film prepared from PLA‐grafted dextran could be adjusted by controlling the sugar content or the length of graft chains. © 2003 Wiley Periodicals, Inc. The syntheses of polypropylene-graft-poly(l -lactide) copolymers (PP-g-PLAs) via copper (I)-catalyzed azide-alkyne cycloaddition “click” reaction (CuAAC) using azide side-chain functionalized polypropylene (PP-N3) and alkyne end-functionalized poly(l -lactide) (PLA-Alkyne) were reported. The CuAAC was then applied to azide and different feeding ratios of alkyne functional polymers to give PP-g-PLAs that were characterized by FTIR, 1H-NMR, GPC, DSC, and WCA measurements. The CuAAC click reaction was achieved by two different feeding ratio (PP-N3:PLA-Alkyne = 1:5 and 1:10) and thermal, biodegradable, and surface properties of obtained graft copolymers were investigated. The molar ratio of PLA were calculated as 72.7 (PP-g-PLA-1) and 78.4% (PP-g-PLA-2) by 1H-NMR spectroscopy. The water contact angle (WCA) values of PP-g-PLA-1 (81o ± 1.3) and PP-g-PLA-2 (75o ± 1.6) copolymers were compared with commercial chlorinated polypropylene (PP-Cl) (90o ± 1.0), suggesting a more hydrophilic nature of desired graft copolymers produced. Conversely, the enzymatic biodegradation studies revealed that the weight losses of graft copolymers were determined as 13.6 and 22.1%, which is about 4% for commercial PP-Cl sample. Thus, it was clear that this simple and facile method was effective in promoting biodegradation of commercial polypropylene and attractive particularly for worldwide environmental remediation goals. © 2018 Wiley Periodicals, Inc. Thermogelling poly(ε-caprolactone-co-D,L -lactide)–poly(ethylene glycol)–poly(ε-caprolactone-co-D,L -lactide) and poly(ε-caprolactone-co-L -lactide)–poly(ethylene glycol)–poly(ε-caprolactone-co-L -lactide) triblock copolymers were synthesized through the ring-opening polymerization of ε-caprolactone and D,L -lactide or L -lactide in the presence of poly(ethylene glycol). The polymerization reaction was carried out in 1,3,5-trimethylbenzene with Sn(Oct)2 as the catalyst at various temperatures, and the yields were about 96%. The molecular weights and polydispersities (Mw/Mn) by gel permeation chromatography were in the ranges of 5140–6750 and 1.35–1.45, respectively. The differential scanning calorimetry results showed that the melting temperatures of the poly(ε-caprolactone) components were between 30 and 40 °C. By the subtle tuning of the chemical compositions and microstructures of these triblock copolymers, the aqueous solutions underwent sol–gel transitions as the temperature increased, with the suitable lower critical solution temperature in the range of 17–28 °C at different concentrations. Transesterification in the polymerization process generated the redistribution of sequences, which remarkably affected the sol–gel transition temperature. The amphiphilic copolymers formed micelles in aqueous solutions with a diameter of 62 nm and a critical micelle concentration of about 0.032 wt % at 20 °C. Micelles aggregated as the temperature increased, leading to gel formation. The sol–gel transition was studied, with a focus on the structure–property relationship. It is expected to have potential applications in drug delivery and tissue engineering. © 2007 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem 45: 4091–4099, 2007  相似文献

Novel microspheres from copolymers of 1,5-dioxepan-2-one (DXO) and L -lactide (L -LA) were prepared by oil-in-water solvent evaporation and oil-in-oil solvent removal. The two preparation techniques were evaluated for sphere formulation and incorporation of two different drugs. Sustained release of these therapeutic substances was obtained. The consequences of altering the DXO : LA ratio, preparation method, and drug hydrophilicity were explored and identified as factors governing sphere quality, in vitro degradation, and drug release characteristics. We show that these relationships provide a powerful means of controlling the microsphere performance. © 1999 John Wiley & Sons, Inc. J Polym Sci A: Polym Chem 37: 1877–1884, 1999  相似文献

Core‐shell structured nanoparticles of poly(ethylene glycol) (PEG)/polypeptide/poly(D ,L ‐lactide) (PLA) copolymers were prepared and their properties were investigated. The copolymers had a poly(L ‐serine) or poly(L ‐phenylalanine) block as a linker between a hydrophilic PEG and a hydrophobic PLA unit. They formed core‐shell structured nanoparticles, where the polypeptide block resided at the interface between a hydrophilic PEG shell and a hydrophobic PLA core. In the synthesis, poly(ethylene glycol)‐b‐poly(L ‐serine) (PEG‐PSER) was prepared by ring opening polymerization of N‐carboxyanhydride of O‐(tert‐butyl)‐L ‐serine and subsequent removal of tert‐butyl groups. Poly(ethylene glycol)‐b‐poly(L ‐phenylalanine) (PEG‐PPA) was obtained by ring opening polymerization of N‐carboxyanhydride of L ‐phenylalanine. Methoxy‐poly(ethylene glycol)‐amine with a MW of 5000 was used as an initiator for both polymerizations. The polymerization of D ,L ‐lactide by initiation with PEG‐PSER and PEG‐PPA produced a comb‐like copolymer, poly(ethylene glycol)‐b‐[poly(L ‐serine)‐g‐poly(D ,L ‐lactide)] (PEG‐PSER‐PLA) and a linear copolymer, poly(ethylene glycol)‐b‐poly(L ‐phenylalanine)‐b‐poly(D ,L ‐lactide) (PEG‐PPA‐PLA), respectively. The nanoparticles obtained from PEG‐PPA‐PLA showed a negative zeta potential value of ?16.6 mV, while those of PEG‐PSER‐PLA exhibited a positive value of about 19.3 mV. In pH 7.0 phosphate buffer solution at 36 °C, the nanoparticles of PEG/polypeptide/PLA copolymers showed much better stability than those of a linear PEG‐PLA copolymer having a comparable molecular weight. © 2011 Wiley Periodicals, Inc. Poly(D ,L -lactide)–poly(ϵ-caprolactone)–poly(ethylene glycol)–poly(ϵ-caprolactone)–poly(D ,L -lactide) block copolymer (PLA–PCL–PEG–PCL–PLA) was prepared by copolymerization of ϵ-caprolactone (ϵ-CL) and D ,L -lactide (D ,L -LA) initiated by potassium poly(ethylene glycol)ate in THF at 25°C. The copolymers with different composition were synthesized by adjusting the mole ratio of reaction mixture. The resulted copolymers were characterized by 1H-NMR, 13C-NMR, IR, DSC, and GPC. Efforts to prepare copolymers with the corresponding structure of PCL–PLA–PEG–PLA–PCL and D ,L -lactide/ϵ-caprolactone random copolymers were not successful. © 1997 John Wiley & Sons, Inc.  相似文献

The thermal properties, crystallization, and morphology of amphiphilic poly(D ‐lactide)‐b‐poly(N,N‐dimethylamino‐2‐ethyl methacrylate) (PDLA‐b‐PDMAEMA) and poly (L ‐lactide)‐b‐poly(N,N‐dimethylamino‐2‐ethyl methacrylate) (PLLA‐b‐PDMAEMA) copolymers were studied and compared to those of the corresponding poly(lactide) homopolymers. Additionally, stereocomplexation of these copolymers was studied. The crystallization kinetics of the PLA blocks was retarded by the presence of the PDMAEMA block. The studied copolymers were found to be miscible in the melt and the glassy state. The Avrami theory was able to predict the entire crystallization range of the PLA isothermal overall crystallization. The melting points of PLDA/PLLA and PLA/PLA‐b‐PDMAEMA stereocomplexes were higher than those formed by copolymer mixtures. This indicates that the PDMAEMA block is influencing the stability of the stereocomplex structures. For the low molecular weight samples, the stereocomplexes particles exhibited a conventional disk‐shape structure and, for high molecular weight samples, the particles displayed unusual star‐like shape morphology. © 2011 Wiley Periodicals, Inc. The hydrolytic degradation of a series of poly‐L ‐lactide (PLLA)‐polyisobutylene (PIB) multiblock copolymers was studied in phosphate buffer solution (pH = 7.4) at 37 °C. The multiblock copolymers were synthesized by chain extension of PLLA‐b‐PIB‐b‐PLLA triblock copolymers, which were obtained by ring‐opening polymerization of L ‐lactide initiated by hydroxyallyl telechelic PIB. The degradation strongly depended on the PLLA segment length. At constant PIB segment length, the multiblock copolymer with the shortest PLLA segment length (DPn = 10), showed significant weight loss after 8 weeks, whereas weight loss for DPn = 36 was only observed after 24 weeks. The gel‐permeation chromatographic analysis showed a similar decrease in the number‐average molecular weight (Mn) with time further supporting the weight loss data. Dynamic mechanical analysis showed a decrease in ultimate stress and modulus with time. The crystallinity of multiblock copolymers changed significantly with degradation time as indicated from differential scanning calorimetric analysis. © 2010 Wiley Periodicals, Inc. Using native cellulose as the starting material, cellulose acetate-graft-ploy (l-lactide) (CA-g-PLA) copolymers were successfully synthesized by “one-pot” process in an ionic liquid 1-allyl-3-methylimidazolium chloride (AmimCl). In this process, cellulose was first reacted with acetic anhydride, yielding cellulose acetate (CA), and then ring opening graft copolymerization of l-lactide was carried out from the residual hydroxyl groups of CA in the same solution using 4-dimethylaminopridine (DMAP) as the catalyst. Both acetyl and ploy (l-lactide) contents in CA-g-PLA copolymers could be well controlled by changing reaction conditions. The structures and thermal properties of CA-g-PLA copolymers were characterized. The glass transition temperature Tg of copolymers decreased with increasing PLA content. Compared to the pure PLA and cellulose-graft-PLA copolymers, the CA-g-PLA copolymers possessed better thermo mechanical properties in a temperature range of 60–130 °C. When the molar substitution of PLA (MSPLA) was above 1.71, the CA-g-PLA copolymers exhibited thermoplastic behavior and could be processed by conventional thermal processing methods, such as injection molding and melt spinning.  相似文献

Diblock copolymerizations of L - or D,L -lactide (LA) with trimethylene carbonate (TMC) or 2,2-dimethyltrimethylene carbonate (2,2-DTMC) with SmMe(C5Me5)2-(tetrahydrofuran) as an initiator and triblock copolymerizations of L - or D,L -LA/cyclic carbonates/L - or D,L -LA with [Sm(C5Me5)2]2(PhCCCCPh) as an initiator generated the desired block copolymers. This article describes the comparison of biodegradabilities by proteinase K and a compost and mechanical properties between the resulting di- or triblock copolymers and random copolymers composed of L - or D,L -LA and cyclic carbonates. The scanning electron microscopic profiles of resulting polymers were measured to understand the morphological change during biodegradation. © 2003 Wiley Periodicals, Inc. Poly[(caprolactone‐co‐lactide)‐b‐perfluoropolyether‐b‐(caprolactone‐co‐lactide)] copolymers (TXCLLA) were prepared by ring‐opening polymerization of D ,L ‐dilactide (LA2) and caprolactone (CL) in the presence of α,ω‐hydroxy terminated perfluoropolyether (Fomblin Z‐DOL TX) as macroinitiator and tin(II) 2‐ethylexanoate as catalyst. 1H NMR analysis showed that LA2 is initially incorporated into the copolymer preferentially with respect to CL. A blocky structure of the polyester segment was also indicated by the sequence distribution analysis of the monomeric units. Differential scanning calorimetry analysis showed the compatibility between poly(lactide) (PLA) and poly(caprolactone) (PCL) blocks inside the amorphous phase with glass‐transition temperature values increasing from ?60 to ?15 °C by increasing the PLA content. Copolymers with high average length of CL blocks were semicrystalline with a melting temperature ranging from +35 to +47 °C. Surface analysis showed a high surface activity of TXCLLA copolymers with values of surface tension independent from the PLA/PCL content and very close to those of pure TX. © 2005 Wiley Periodicals, Inc. A series of multihydroxyl (2, 4, and 8) terminated poly(ethylene glycol)s and their biodegradable, biocompatible, and branched barbell‐like (PLGA)nb‐PEG‐b‐(PLGA)n (n = 1, 2, 4) copolymers have been synthesized. The lengths of the PLGA arms were varied by controlling the molar ratio of monomers to hydroxyl groups of PEG ([LA+GA]0/[? OH]0 = 23, 45, 90). Chemical structures of synthesized barbell‐like copolymers were confirmed by both 1H and 13C‐NMR spectroscopies. Molecular weights were determined by 1H‐NMR end‐group analysis and gel permeation chromatography. The result of hydrolytic degradation indicated that the rate of degradation increased with the increase of arm numbers or with the decrease of arm lengths. The thermal properties were evaluated by using differential scanning calorimetry and a thermogravimetric analysis. The results indicated that the thermal properties of barbell‐like copolymers depended on the structural variations. The morphology of (PLGA)n‐PEG‐(PLGA)n copolymers self‐assembly films were investigated by atomic force microscope, the results indicated that the microphase separation existed in (PLGA)n‐PEG‐(PLGA)n copolymers. Because of the favorable biodegradability and biocompatibility of the PLGA and PEG, these results may therefore create new possibilities for these novel structural amphiphilic barbell‐like copolymers as potential biomaterials. © 2008 Wiley Periodicals, Inc. Poly(ε‐caprolactone)/polylactide blend (PCL/PLA) is an interesting biomaterial because the two component polymers show good complementarity in their physical properties. However, PCL and PLA are incompatible thermodynamically and hence the interfacial properties act as the important roles controlling the final properties of their blends. Thus, in this work, the PCL/PLA blends were prepared by melt mixing using the block copolymers as compatibilizer for the studies of interfacial properties. Several rheological methods and viscoelastic models were used to establish the relations between improved phase morphologies and interfacial properties. The results show that the interfacial behaviors of the PCL/PLA blends highly depend on the interface‐located copolymers. The presence of copolymers reduces the interfacial tension and emulsified the phase interface, leading to stabilization of the interface and retarding both the shape relaxation and the elastic interface relaxation. As a result, besides the relaxation of matrices (τm) and the shape relaxation of the dispersed PLA phase (τF), a new relaxation behavior (τβ), which is attribute to the relaxation of Marangoni stresses tangential to the interface between dispersed PLA phase and matrix PCL, is observed on the compatibilized blends. In contrast to that of the diblock copolymers, the triblock copolymers show higher emulsifying level. However, both can improve the overall interfacial properties and enhance the mechanical strength of the PCL/PLA blends as a result. © 2010 Wiley Periodicals, Inc. Poly(lactide) (PLA), poly(ε‐caprolactone) (PCL) and poly(trimethylene carbonate) (PTMC) homopolymers of high molecular weight were prepared using potassium‐based catalyst. Polymerizations were carried out in toluene at room temperature. The chemical structure of the polymers was investigated by 1H and 13C NMR. The physical properties investigated by GPC and DSC for the polymers obtained are similar to those prepared using tin octanoate based catalyst. Using a sequential polymerization procedure, PLA‐b‐PCL, PLA‐b‐PTMC, and PCL‐b‐PTMC diblock copolymers were synthesized and characterized in terms of their composition and physical properties. The formation of diblock copolymers was confirmed by NMR and DSC measurements. In vitro cytotoxicity tests have been carried out using MTS assay and the results show the biocompatibility of these polymers in the presence of the fibroblast cells. © 2008 Wiley Periodicals, Inc. The syntheses of {‐poly(L ‐lactide) (PLLA)‐b‐polyisobutylene (PIB)‐}n multiblock copolymers were accomplished for the first time by chain extension of PLLA‐b‐PIB‐b‐PLLA triblock copolymers. Well‐defined PLLA‐b‐PIB‐b‐PLLA triblock copolymers with predictable Mns, low PDIs (1.10–1.18) and excellent blocking efficiencies were prepared by anionic ring‐opening polymerizations of L ‐lactide initiated with hydroxyallyl telechelic PIB (HO‐Allyl‐PIB‐Allyl‐OH) in toluene at 110 °C. The triblock copolymers were successfully chain extended with 4,4′‐methylenebis(phenylisocyanate) (MDI) to obtain the multiblock copolymers with good gravimetric yields of ~86 to 96%. The chain‐extended polymers were soluble in a range of common organic solvents. The block copolymers showed two glass transition temperatures in differential scanning calorimetric analysis for the PIB and PLLA blocks indicating microphase separation, which was supported by atomic force microscopy images. The as‐synthesized compression molded multiblock copolymers exhibited tensile strengths in the range of 8–24 MPa with elongations at break in the range of 2.5–400%. The static and dynamic mechanical properties showed a strong dependence on the relative PLLA content in the copolymer. The dynamic mechanical analysis also indicated microphase separation at higher PLLA compositions. © 2009 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem 47: 3490–3505, 2009  相似文献

Biocompatible and biodegradable ABC and ABCBA triblock and pentablock copolymers composed of poly(ε‐caprolactone) (PCL), poly(L ‐lactide) (PLA), and poly(ethylene glycol) (PEO) with controlled molecular weights and low polydispersities were synthesized by a click conjugation between alkyne‐terminated PCL‐b‐PLA and azide‐terminated PEO. Their molecular structures, physicochemical and self‐assembly properties were thoroughly characterized by means of FT‐IR, 1H‐NMR, gel permeation chromatography, differential scanning calorimetry, wide‐angle X‐ray diffraction, dynamic light scattering, and transmission electron microscopy. These copolymers formed microphase‐separated crystalline materials in solid state, where the crystallization of PCL block was greatly restricted by both PEO and PLA blocks. These copolymers self‐assembled into starlike and flowerlike micelles with a spherical morphology, and the micelles were stable over 27 days in aqueous solution at 37 °C. The doxorubicin (DOX) drug‐loaded nanoparticles showed a bigger size with a similar spherical morphology compared to blank nanoparticles, demonstrating a biphasic drug‐release profile in buffer solution and at 37 °C. Moreover, the DOX‐loaded nanoparticles fabricated from the pentablock copolymer sustained a longer drug‐release period (25 days) at pH 7.4 than those of the triblock copolymer. The blank nanoparticles showed good cell viability, whereas the DOX‐loaded nanoparticles killed fewer cells than free DOX, suggesting a controlled drug‐release effect. © 2010 Wiley Periodicals, Inc. A series of di‐ and triblock copolymers [poly(L ‐lactide‐b‐ε‐caprolactone), poly(D,L ‐lactide‐b‐ε‐caprolactone), poly(ε‐caprolactone‐b‐L ‐lactide), and poly(ε‐caprolactone‐b‐L ‐lactide‐b‐ε‐caprolactone)] have been synthesized successfully by sequential ring‐opening polymerization of ε‐caprolactone (ε‐CL) and lactide (LA) either by initiating PCL block growth with living PLA chain end or vice versa using titanium complexes supported by aminodiol ligands as initiators. Poly(trimethylene carbonate‐b‐ε‐caprolactone) was also prepared. A series of random copolymers with different comonomer composition were also synthesized in solution and bulk of ε‐CL and D,L ‐lactide. The chemical composition and microstructure of the copolymers suggest a random distribution with short average sequence length of both the LA and ε‐CL. Transesterification reactions played a key role in the redistribution of monomer sequence and the chain microstructures. Differential scanning calorimetry analysis of the copolymer also evidenced the random structure of the copolymer with a unique Tg. © 2012 Wiley Periodicals, Inc. A technique is described for the preparation of arborescent graft copolymers containing poly(tert‐butyl methacrylate) (PtBMA) segments. For this purpose, tert‐butyl methacrylate is first polymerized with 1,1‐diphenyl‐2‐methylpentyllithium in tetrahydrofuran. The graft copolymers are obtained by addition of a solution of a bromomethylated polystyrene substrate to the living PtBMA macroanion solution. Copolymers incorporating either short (Mw ≈ 5000) or long (Mw ≈ 30,000) PtBMA side chains were prepared by grafting onto linear, comb‐branched (G0), G1, and G2 bromomethylated arborescent polystyrenes. Branching functionalities ranging from 9 to 4500 and molecular weights ranging from 8.8 × 104 to 6.3 × 107 were obtained for the copolymers, while maintaining a low apparent polydispersity index (Mw/Mn ≈ 1.14–1.25). Arborescent polystyrene‐graft‐poly(methacrylic acid) (PMAA) copolymers were obtained by hydrolysis of the tert‐butyl methacrylate units. Dynamic light scattering measurements showed that the arborescent PMAA copolymers are more expanded than their linear PMAA analogues when neutralized with NaOH. This effect is attributed to the higher charge density in the branched arborescent copolymer structures. © 2008 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem 46: 2335–2346, 2008  相似文献

A set of well‐defined poly(ethylene‐alt‐propylene)‐b‐polylactide (PEP‐PLA) diblock copolymers containing volume fractions of PLA (fPLA) ranging between 0.08 and 0.91 were synthesized by a combination of living anionic polymerization, catalytic hydrogenation, and controlled coordination‐insertion ring‐opening polymerization. The morphological behavior of these relatively low‐molecular‐weight PEP‐PLA diblock copolymers was investigated with a combination of rheology, small‐angle X‐ray scattering, and differential scanning calorimetry. The ordered microstructures observed were lamellae (L), hexagonally packed cylinders (C), spheres (S), and gyroid (G), a bicontinous cubic morphology having Ia3 d space group symmetry. The G morphology existed in only a small region between the L‐C morphologies in close proximity to the order–disorder transition (ODT). Transformations from L to G were observed upon heating in several samples. The efficacy of the reverse G to L transition in one sample was cooling rate dependent. The PEP‐PLA Flory–Huggins interaction parameter as a function of temperature χPEP‐PLA(T) was estimated from TODT's by mean‐field theory and subsequently used in the construction of the experimental PEP‐PLA morphology diagram (χN versus fPLA). The resultant morphology diagram was symmetric there were the well‐defined L‐C morphology boundaries. The low molecular weight of the materials imparted no significant deviation from previously documented diblock systems. © 2002 Wiley Periodicals, Inc.
