A simple method to obtain predetermined constant cooling rates for insect supercooling point (SCP) determination is described. A transient heat transfer equation was used to design polystyrene cubes of different sizes to yield constant rates of cooling at their centers when held at a constant surrounding temperature. Cubes of 0.29 x 0.29 x 0.29 m and 0.19 x 0.19 x 0.19 m were found to produce cooling rates of about -0.5 and -1 degree C per min, respectively, from 0 to -40 degree C. The observed temperature variations at the geometrical center of the cubes were similar to those predicted by the equation. Temperature plots showed a nearly constant rate of cooling. Supercooling points of Tribolium castaneum (Herbst) at different stages of development were recorded using polystyrene cubes. These SCPs compared favorably with published values. This method of obtaining cooling rates is economical, flexible, and allows for multiple simultaneous SCP measurements.  相似文献

In this paper, we propose a simple method for processing a 2D wrapped phase map that contains a spatial carrier signal in order to completely eliminate, or greatly reduce, the number of phase wraps in the image. The 2D Fourier transform of the wrapped phase map is calculated. Then the spectrum is shifted to the origin in frequency space. After that, the inverse 2D Fourier transform is computed. Finally, a four-quadrant arctangent function is used to calculate the angle of the complex array that was produced by the inverse 2D Fourier transform. This produces a phase map with a smaller number of 2π phase jumps than the original phase map. In some cases, all of the phase wraps are eliminated and there is therefore no need to unwrap the resultant phase map. The reduction of the number of 2π phase jumps can reduce the execution time and improve the noise performance of some phase unwrapping algorithms such as the Flynn method. The validation of the proposed algorithm is demonstrated experimentally and also via computer-simulation.  相似文献

A new method for the determination of roxithromycin based on the fluorescence quenching of 3-mercaptopropionic acid-capped CdTe quantum dots (MPA-CdTe QDs) was developed. In ethanol medium, the fluorescence of CdTe quantum dots at 552 nm was quenched in the presence of roxithromycin. Based on this a simple, sensitive, and selective method for rapid determination of roxithromycin was described. Reaction time, interfering substances on the fluorescence quenching, and mechanism of the interaction of CdTe QDs with roxithromycin were investigated. After optimization, the proposed method allows the determination of roxithromycin over the range 25.0-350.0 μg ml−1. The detection limit is 4.6 μg ml−1. The proposed method was successfully applied to commercial capsules and tablets with satisfactory results. The recovery of the method was in the range of 96.8-102.5%.  相似文献

A method of determination of chemical shift anisotropy (CSA) tensor principal components under MAS condition is presented. It is a simple, one-dimensional, and robust alternative to the commonly exploited, but more complicated 2D-PASS. The required CSA components are delivered by simultaneous numerical analysis of a few regular MAS spectra acquired under different spinning rates.  相似文献

The total reflectance of a slab turbid material is measured with an integrating sphere. Each slab was measured with a glossy and diffusing (etched) rear surface, both with and without a fully reflecting pill placed behind it. Formulae derived show that the results, together with the refractive index, permit calculation of the reflectance of the parity diffuse radiation at the internal rear surface. A rapid and simple assay (6 min, two steps) is described for determination of cell viability of hepatocytes subjected to cold preservation protocols. In this method, cells are incubated with the fluorescent marker propidium iodide (PI) and the fluorescence intensity is measured before (direct fluorescence--Fd) and after (total fluorescence--Ft) addition of digitonin, which allows the dye to enter the hepatocytes. The Fd originated from non-viable cells that have membrane damage and taken up PI. The Ft originated from all cells in the sample. The ratio between the two fluorescence values is used as an indicator of cell viability. The assay was challenged versus two classical viability tests: LDH retention and Trypan Blue exclusion. Our assay shows good correlation only with Trypan Blue test. In addition, a fluorescence confocal microscopy protocol was used to evaluate the possible toxicity of PI in hepatocyte suspensions.  相似文献

We have used hepatocyte suspensions to study how hypothermic storage in a modified University of Wisconsin (mUW) solution affects liver cell metabolism and cell membrane properties. At present the UW solution is the gold standard of organ preservation. However it contains several ingredients which either are expensive or cannot easily be obtained worldwide. The aim of the present study was the development of a novel preservation solution for rat hepatocytes effective and comparable with the mUW, with enhanced buffer capacity and less expensive. In particular, we investigated the effects of the buffering agent BES, a derivate of aminosulfonic acid, on liver cells metabolism during cold storage and rewarming. In the development of this novel preservation solution we have included three key components: gluconate as impermeant anion, sucrose to give additional osmotic support and the aminosulfonic acid BES for its buffer capacity. Our results shown that BGS solution was equally effective to mUW to protect rat hepatocytes against cold preservation injury due to ischemia and reoxigenation. Also, BGS solution is a good alternative with its high buffer capacity, best indices of respiration activity and it is considerably less expensive than the mUW solution. The use of this solution for the storage of isolated hepatocytes may facilitate hepatocyte research in situations in which the more complex and expensive mUW solution is not available since the cost of one liter of BGS is about 1/3 that an equal volume of mUW solution.  相似文献

Size-adjustable silver nanoparticles were selectively in situ produced and immobilized on glass surface by simply UV exposing poly(vinylpyrrolidone) (PVP) film containing silver nitrate and 4,4′-diazostilbene-2,2′-disulfonic acid disodium salt (DAS) with the presence of a photomask, during which the crosslink between PVP and DAS completed and the silver nitrate was reduced to silver nanoparticles under the assistance of UV. Because the reduction speed of silver nitrate is much slower than the crosslink reaction of PVP, and a mount of unreduced silver nitrate still existed in the formed PVP micro-scale patterns after removing unexposed PVP by water, we can easily adjust the size of silver nanoparticles through further controlling the reduction of unreduced silver nitrate in the formed PVP micro-scale patterns. Optical microscope, UV–vis-spectroscopy, AFM and SEM-EDX were used to characterize the morphology and distribution of those silver nanoparticles immobilized on glass surface. The results indicate that it is an effective way to immobilize silver nanoparticles with high selectivity.  相似文献

A simple statistical method is proposed for afterpulse probability measurement in high-speed single-photon detectors. The method is based on in-laser-period counting without the support of time-correlated information or delay adjustment, and is readily implemented with commercially available logic devices. We present comparisons among the proposed method and commonly used methods which use the time-correlated single-photon counter or the gated counter, based on a 1.25-GHz gated infrared single-photon detector. Results show that this in-laser-period counting method has similar accuracy to the commonly used methods with extra simplicity, robustness, and faster measuring speed.  相似文献

A simple and accurate approach for analysis of dielectric periodic structures is proposed in this paper. The method combines the coupled-mode theory with Floquet theory and joins the eigenmode expansion with the space harmonic expansion properly. As a result, the whole analysis procedure is significantly simplifid.
