We report a switchable synthesis of acylindoles and quinoline derivatives via gold‐catalyzed annulations of anthranils and ynamides. α‐Imino gold carbenes, generated in situ from anthranils and an N,O‐coordinated gold(III) catalyst, undergo electrophilic attack to the aryl π‐bond, followed by unexpected and highly selective 1,4‐ or 1,3‐acyl migrations to form 6‐acylindoles or 5‐acylindoles. With the (2‐biphenyl)di‐tert‐butylphosphine (JohnPhos) ligand, gold(I) carbenes experienced carbene/carbonyl additions to deliver quinoline oxides. Some of these epoxides are valuable substrates for the preparation of 3‐hydroxylquinolines, quinolin‐3(4H)‐ones, and polycyclic compounds via facile in situ rearrangements. The reaction can be efficiently conducted on a gram scale and the obtained products are valuable substrates for preparing other potentially useful compounds. A computational study explained the unexpected selectivities and the dependency of the reaction pathway on the oxidation state and ligands of gold. With gold(III) the barrier for the formation of the strained oxirane ring is too high; whereas with gold(I) this transition state becomes accessible. Furthermore, energetic barriers to migration of the substituents on the intermediate sigma‐complexes support the observed substitution pattern in the final product.  相似文献

Gold‐catalyzed oxidations of alkynes by N‐oxides offer direct access to reactive α‐oxo gold carbene intermediates from benign and readily available alkynes instead of hazardous diazo carbonyl compounds. Despite various versatile synthetic methods developed based on this strategy, one of the hallmarks of α‐oxo carbene/carbenoid chemistry, that is, the Wolff rearrangement, has not been realized in this context. This study discloses the first examples that show the Wolff rearrangement can be readily realized by α‐oxo gold carbenes oxidatively generated from TBS‐terminated alkynes (TBS=tert‐butyldimethylsilyl). The thus‐generated silylketenes can be either isolated pure or subsequently trapped by various internal or external nucleophiles in one pot to afford α‐silylated carboxylic acids, their derivatives, or TBS‐substituted allenes.  相似文献

This report describes a gold(III)‐catalyzed efficient general route to densely substituted chiral 3‐formyl furans under extremely mild conditions from suitably protected 5‐(1‐alkynyl)‐2,3‐dihydropyran‐4‐one using H2O as a nucleophile. The reaction proceeds through the initial formation of an activated alkyne–gold(III) complex intermediate, followed by either a domino nucleophilic attack/anti‐endo‐dig cyclization, or the formation of a cyclic oxonium ion with subsequent attack by H2O. To confirm the proposed mechanistic pathway, we employed MeOH as a nucleophile instead of H2O to result in a substituted furo[3,2‐c]pyran derivative, as anticipated. The similar furo[3,2‐c]pyran skeleton with a hybrid carbohydrate–furan derivative has also been achieved through pyridinium dichromate (PDC) oxidation of a substituted chiral 3‐formyl furan. The corresponding protected 5‐(1‐alkynyl)‐2,3‐dihydropyran‐4‐one can be synthesized from the monosaccharides (both hexoses and pentose) following oxidation, iodination, and Sonogashira coupling sequences. Furthermore, to demonstrate the potentiality of chiral 3‐formyl furan derivatives, a TiBr4‐catalyzed reaction of these derivatives has been shown to offer efficient access to 1,5‐dicarbonyl compounds, which on treatment with NH4OAc in slightly acidic conditions afforded substituted furo[3,2‐c]pyridine.  相似文献

N-heterocyclic carbenes (NHCs) have received significant attention as gold nanoparticle stabilizers due to their strong binding affinity towards gold. However, their tunability is limited by the difficulty in obtaining nonsymmetric NHCs. In this regard, N-acyclic carbenes (NACs) are attractive alternatives due to their high synthetic versatility, allowing easy tuning of their steric and electronic properties towards specific applications. This work reports the first series of stable and monodisperse NAC-functionalized gold nanoparticles. These particles with sizes ranging 3.8 to 11.6 nm were characterized using NMR, UV/Vis and TEM. The nanoparticles display good stability at elevated temperatures and for extended periods both dried or dispersed in a medium, as well as in the presence of exogenous thiols. Importantly, these NAC-stabilized gold nanoparticles offer a promising and versatile alternative to NHC-stabilized gold nanoparticles.  相似文献

Cyclobutanones are synthetically versatile compounds that often require extensive effort to access. Herein, we report a facile synthesis of cyclobutanones based on the C(sp3)?H insertion chemistry of oxidatively generated gold carbenes. Various cyclobutanones were obtained in synthetically useful yields from substrates with minimal structural prefunctionalization. This discovery reveals new synthetic utilities of gold‐catalyzed oxidative transformations of alkynones.  相似文献

The formal (3+2) cycloaddition between terminal allenes and aryl or styryl gold(I) carbenes generated by a retro‐Buchner reaction of 7‐substituted 1,3,5‐cycloheptatrienes led to indenes and cyclopentadienes, respectively. These cycloaddition processes have been applied to the construction of the carbon skeleton of the cycloaurenones and the dysiherbols as well as to the total synthesis of (±)‐laurokamurene B.  相似文献

Substituent effects of fused six, and five‐membered aromatic rings are investigated on the stability, aromaticity, charge distribution, nucleophilic (N), and electrophilic (ω) characters of 20 singlet (s) and triplet (t) Hammick carbenes, at B3LYP/AUG‐cc‐pVTZ and M06‐2X/AUG‐cc‐pVTZ. Results display: (a) The higher thermodynamic and kinetic stability is revealed by carbenes situated between two nitrogen and/or two oxygen heteroatoms of two substituted rings, in a “W” arrangement toward the carbenic center; (b) Regardless of the arrangement, the order of thermodynamical and kinetic stabilization for fused rings is pyrrole > furan > thiophene > phosphole. (c) The substituted Hammick carbenes with two fused heterocyclics, in a given arrangement, show more stability than unsubstituted Hammick carbene; (d) While two five‐membered heterocyclic rings stabilize their corresponding substituted carbenes, two benzene rings destabilize Hammick carbene; (e) In all structures, s species emerges as ground state, exhibiting more stability than its t state; (f) The scrutinized s carbenes show higher N and lower ω than their corresponding t states.  相似文献

By reviewing the recent progress on the elucidation of the structure of gold carbenes and the definitions of metal carbenes and carbenoids, we recommend to use the term gold carbene to describe gold carbene‐like intermediates, regardless of whether the carbene or carbocation extreme resonance dominates. Gold carbenes, because of the weak metal‐to‐carbene π‐back‐donation and their strongly electrophilic reactivity, could be classified into the broader family of Fischer carbenes, although their behavior and properties are very specific.  相似文献

Patterned monolayers of N‐heterocyclic carbenes (NHCs) on gold surfaces were obtained by microcontact printing of NHC–CO2 adducts and NHC(H)[HCO3] salts. The NHC‐modified areas showed an increased conductivity compared to unmodified gold surface areas. Furthermore, the remaining surface areas could be modified with a second, azide‐functionalized carbene, facilitating further applications and post‐printing modifications. Thorough elucidation by a variety of analytical methods offers comprehensive evidence for the viability of the methodology reported here. The protocol enables facile access to versatile, microstructured NHC‐modified gold surfaces with highly stable patterns, enhanced conductivity, and the option for further modification.  相似文献

A gold(I)‐catalyzed asymmetric intermolecular tandem [3+3]‐cyclization reaction of 2‐(1‐alkynyl)‐2‐ alken‐1‐ones with nitrones has been developed by using Ming‐Phos as a chiral ligand. This method enables access to the stereodivergent synthesis of highly substituted furo[3,4‐d][1,2]oxazines in excellent efficiency and stereoselectivity (up to 99% yield, 99% ee, >20 : 1 dr).  相似文献

The intermolecular α‐allylation of enals and enones occurs by the condensation of variously substituted allenamides with allylic alcohols. Cooperative catalysis by [Au(ItBu)NTf2] and AgNTf2 enables the synthesis of a range of densely functionalized α‐allylated enals, enones, and acyl silanes in good yield under mild reaction conditions. DFT calculations support the role of an α‐gold(I) enal/enone as the active nucleophilic species.  相似文献

Gold(I) complexes are the most active catalysts for the biscyclopropanation of dienynes to form tetracyclic compounds. PtII and ZnII are also able to promote the biscyclopropanation, although less efficiently. The configurations obtained in all cases with the use of gold(I) catalysts can be explained by the pathway proceeding through anti cyclopropyl gold carbenes. Similar intermediates are most probably involved in reactions catalyzed by RuII and PtII. Two different cyclopropanation pathways have been found; they depend on the structures of the cyclopropyl gold carbenes (anti or syn) and the relative arrangements of the metal carbenes and the alkenes.  相似文献

An efficient strategy to construct salicyl ketones through gold‐catalyzed oxidation/C?H functionalization of ynones is reported. A variety of functionalized salicyl ketones are readily accessed by utilizing this non‐diazo approach, thus providing a viable alternative to synthetically useful salicyl ketones with a yield up to 98 %. The α‐oxo gold carbenes generated in situ through gold‐catalyzed oxidation of ynones can be trapped effectively by internal aryl and heteroaromatic groups. Electronic and steric effects were also investigated in this reaction. The anticancer activity of one salicyl ketone analogue was investigated and its cytotoxicity assays against the PC‐3 prostate cancer cell line and SKOV‐3 human ovarian carcinoma cell line yield IC50 were 0.81±0.05 and 0.87±0.15 μm , respectively, demonstrating that salicyl ketone analogues showed good anticancer activity.  相似文献

The gold(I)‐catalysed cycloisomerisation of appropriately substituted 1,6‐cyclopropene‐enes proceeds through regioselective electrophilic ring opening of the three‐membered ring to generate an alkenyl gold carbenoid that achieves the intramolecular cyclopropanation of the remote olefin. This strategy allows straightforward, highly efficient and diastereoselective access to a variety of substituted 3‐oxa‐ and 3‐azabicyclo[4.1.0]heptanes, as well as to bicyclo[4.1.0]heptan‐3‐ol derivatives. Since the isopropylidene group in the resulting cycloisomerisation products can be subjected to ozonolysis, 3,3‐dimethylcyclopropenes behave as interesting surrogates for α‐diazoketones.  相似文献

Carbophilic gold carbenes generated from the decomposition of α‐diazoesters show high reactivity towards enamides, leading to an unprecedented aza‐ene‐type reaction. The presence of 0.1 mol % of a chiral Brønsted acid co‐catalyst is sufficient to give synthetically relevant γ‐keto esters in excellent yields and selectivities (up to 99 % yield, 97 % ee ).  相似文献

Selective oxidation of amines using oxygen as terminal oxidant is an important area in green chemistry. In this work, we describe the use of graphite‐supported gold nanoparticles (AuNPs/C) to catalyze aerobic oxidation of cyclic and acyclic benzylic amines to the corresponding imines with moderate‐to‐excellent substrate conversions (43–100 %) and product yields (66–99 %) (19 examples). Oxidation of N‐substituted 1,2,3,4‐tetrahydroisoquinolines in the presence of aqueous NaHCO3 solution gave the corresponding amides in good yields (83–93 %) with high selectivity (up to amide/enamide=93:4) (6 examples). The same protocol can be applied to the synthesis of benzimidazoles from the reaction of o‐phenylenediamines with benzaldehydes under aerobic conditions (8 examples). By simple centrifugation, AuNPs/C can be recovered and reused for ten consecutive runs for the oxidation of dibenzylamine to N‐benzylidene(phenyl)methanamine without significant loss of catalytic activity and selectivity. This protocol “AuNPs/C+O2” can be scaled to the gram scale, and 8.9 g (84 % isolated yield) of 3,4‐dihydroisoquinoline can be obtained from the oxidation of 10 g 1,2,3,4‐tetrahydroisoquinoline in a one‐pot reaction. Based on the results of kinetic studies, radical traps experiment, and Hammett plot, a mechanism involving the hydrogen‐transfer reaction from amine to metal and oxidation of M‐H is proposed.  相似文献
