The parameters affecting the fractionation performance in size-exclusion chromatography (SEC) of broad polymer samples were investigated. Some equations were derived which enable the prediction of polydispersity (PD) in an SEC fraction. Good agreements were obtained between the calculated data and the experimental values. Based on these equations, SEC fractionation conditions were optimized. In the off-line SEC-matrix-assisted laser desorption ionization (MALDI) time-of-flight (TOF) mass spectrometry (MS), two different modes can be employed, i.e., using MALDI-MS to provide an absolute calibration curve for SEC, or using SEC as a sample preparation step for MALDI-MS measurements. It was demonstrated that it is more reliable to use the latter combination, because most problems inherent in SEC can be circumvented. Some guidelines for the optimization of off-line SEC fractionation-MALDI-TOF-MS were given. It was found that under optimized conditions normally only a few SEC fractions are already sufficient to separate a highly polydisperse sample into portions of low PD that can accurately be measured by MALDI-TOF-MS.  相似文献

Matrix assisted laser desorption ionisation-time of flight-mass spectrometry (MALDI-TOF-MS), laser desorption ionisation-time of flight-mass spectrometry (LDI-TOF-MS) and temperature resolved analytical pyrolysis field ionisation mass spectrometry (Py-FIMS) have been applied for the first time on two pyrolytic lignins (PL's), precipitated from different aged bio oil, and four PL-fractions for molar mass characterization. The results were compared with data from size exclusion chromatography (SEC). SEC was the only method that allowed a mathematical calculation of molar mass characteristics such as average molecular weight (Mw), dispersity (D), and the molar mass at the peak maximum of the elugram (Mp). The SEC Mp values of PL-fractions differ from visually interpolated MALDI-TOF-MS measurements by 20%. MALDI-TOF-MS spectra showed detailed structures of the molar mass distribution (MMD) of PL and PL-fractions. Especially, the spectrum of one PL showed various local maxima with intervals of 170–200 Da. The size of these intervals could represent the average absolute molar mass of PL-monomers. MALDI-TOF-MS was limited by the influence of superposing matrix signals in the spectrum at low molar masses. LDI-TOF-MS showed clearer spectra than MALDI-TOF-MS in mass ranges below 400 Da. No signals were obtained for fractions with higher masses or whole PL. Intervals between main signals were mostly 14–16 Da. In spectra of different PL-fractions, corresponding main signals can vary between 2 and 4 Da. These mass differences indicate variations in the aliphatic region of the PL molecules. Py-FIMS spectra contained masses of thermally ejected, but unfragmented monomers and dimers. It was the only method, which allowed partial identification of monomeric and dimeric structures of all samples. The detected monomers correspond to known lignin derived compounds in bio oil, the detected dimers have some similarities to phenylcoumaran structures. PL from aged bio oil showed an increased content of higher oligomers and a higher average molecular weight.  相似文献

In this study, various organic compounds, with different activation modes, have been tested as catalysts for the ring‐opening polymerization (ROP) of cyclic butylene terephthalate oligomers (CBT) in bulk at 210 °C, using tert‐butylbenzyl alcohol (tBnOH) as initiator. Among them, 1,3,5‐triazabicyclo[4.4.0]dec‐5‐ene (TBD) appeared to be the most efficient, achieving high monomer conversions in short reaction times (within minutes). Analysis by size‐exclusion chromatography (SEC) of the poly(butylene terephthalate) (PBT) synthesized using this catalyst also showed that the polymerization follows the expected theoretical M n trend for molecular weights up to 50 kg·mol?1. Chain‐end fidelity relatively to the alcohol initiator has been confirmed by MALDI‐TOF mass spectroscopy, which showed that all polymer chains possess the tert‐butylbenzyl moiety as chain‐end. Finally, to demonstrate the potential of this system for the synthesis of PBT‐based block copolymers, a monomethyl ether poly(ethylene glycol) (PEG) of 5000 g·mol?1 has been employed as initiator for the ROP of CBT. A PEO‐b‐PBT block copolymer of 15,000 g·mol?1 could thus been obtained, as confirmed by the shift of the SEC traces towards higher molecular weights and the same diffusion coefficient determined for 1H NMR signals of the PEO block and the PBT block. © 2017 Wiley Periodicals, Inc. Four generations of poly(aryl ether) dendrimers containing aryl sulfide or aryl sulfone groups, and aryl fluoride terminal functionality, have been synthesized using the divergent initiator core method and bis(4-fluorophenyl) sulfone as the core precursor. The strategy is based on the divergent approach and an activation/condensation sequence that involves oxidation of the aryl sulfide group and the displacement of the activated halide moiety by a phenolate ion. The phenolate is easily generated in situ from an aryl carbonate. No reaction intermediates were detected when the condensation reaction was carried out utilizing a very active metal carbonate, such as cesium carbonate, in conjunction with magnesium hydroxide or calcium carbonate which removes some of the fluoride ions formed. The samples were characterized by HPLC, MALDI-TOF-MS, NMR, and SEC. Imperfections in some of the molecules of the dendrimers, formed by reaction of the core precusors with an impurity present in the phenol, were also identified by MALDI-TOF-MS. © 1997 John Wiley & Sons, Inc. Size exclusion chromatography (SEC), gradient polymer elution chromatography (GPEC) and liquid chromatography at critical conditions (LC-CC) have been developed and applied to observe chemical changes in poly(bisphenol A)carbonate (PC) due to hydrolytic degradation. Especially LC-CC appeared to be very successful to observe differences in functionality of PC as result of hydrolytic degradation. Observed differences due to degradation could be identified by (semi) on-line coupling to matrix assisted laser desorption-ionization time-of-flight mass spectrometry (MALDI-TOF-MS). The differences in functionality could be attributed to the formation of different end-groups, i.e. OH end-groups. In addition, comprehensive two-dimensional liquid chromatography (2D-LC) has been applied successfully to study the hydrolytic degradation of PC. LC-CC x SEC showed that the formation of PC with different end-groups occurred over the whole molecular mass range. This information could not be obtained with the separate liquid chromatographic techniques, thereby illustrating the added value of 2D-LC.  相似文献

Dendrimers with molecular weights ranging from ca. 2700 to 11 000 and from 16 to 64 homoallyl ether end groups were cross-linked using the Grubbs ring-closing metathesis reaction. A combination of SEC, MALDI-TOF-MS, and AFM were used to characterize the cross-linked nanoparticles. The data suggest a significant decrease in volume with cross-linking and a concomitant increase in rigidity, both of which can be controlled independently with a fair degree of precision.  相似文献

The applicability of size exclusion chromatography (SEC) to analyze (upgraded) pyrolysis oil samples has been studied using model compounds, pyrolysis oils and hydrodeoxygenated pyrolysis oils. The assumptions needed for the conversion of the chromatogram to the Mw-distribution were validated. It was shown that the conversion of elution volume to molecular weight (based on polystyrene calibration curves) can introduce substantial errors in the prediction of the molecular weight. The conversion of RID response to W(log M) (as plotted on the y-axis of the Mw-distribution) is based on the assumption of a compound independent RID response factor and linear response to concentration. While the latter was shown to be true within the concentration range studied, the former was not true: the RID response factor depends on the type of (upgraded) pyrolysis oil. It was shown that within a single pyrolysis oil sample, the RID response for the low molecular weight fraction was a factor 3 lower than the high molecular weight fraction. Furthermore long term column fouling can influence SEC results that cannot be corrected with regular polystyrene recalibrations.Based on the results we recommend SEC not to be used as a quantitative method for characterization (upgraded) pyrolysis oil samples, but as a tool to compare (upgraded) pyrolysis oil samples, preferably prepared using incremental operating conditions and expected to have similar molecular composition. This work has further shown that (i) the ∫UVDdv/∫RIDdv ratio can be used as an indication of the sum of the relative aromaticity and conjugated double bond content for (upgraded) pyrolysis oil, and (ii) the negative peak area appearing in the low molecular weight part of the chromatogram can be used to estimate the water content of (upgraded) oil samples.  相似文献

Polymers of ε-caprolactone were synthesized by microwave-assisted polymerization initiated with polyethylene glycols (PEG 200 and PEG 300) and monodisperse diols (mono-, di-, tri-, tetra- and hexaethylene glycol) and tin octoate as catalyst. These polymers were characterized by different chromatographic techniques (SEC, LAC and LCCC) and MALDI-TOF-MS. A comparison with commercially available polycaprolactone diols with molecular weight 530 and 830 showed that the new polymers had a much higher content of triblock structures, while the commercial samples contained considerable amounts of diblocks.  相似文献

The synthesis of well-defined block copolymers from styrene and methyl acrylate via ATRP is discussed in this contribution. Kinetic studies on these block copolymerizations as well as characterization studies were performed to investigate the monomer composition in the respective PS and PMA blocks. MALDI-TOF-MS was performed to clarify the exact number of repeating units of each block and the total number of units in the block copolymer. Block copolymers up to 22 kDa could be analyzed by MALDI-TOF-MS, whereby polymers with PMA as first block showed a large second distribution corresponding to PMA homopolymers. However, SEC demonstrated that only a small amount of homopolymer was present indicating that care needs to be taken with interpreting MALDI-TOF-MS data, which is a qualitative rather than a quantitative technique.  相似文献

The hydrolytic degradation of poly(bisphenol A)carbonate (PC) has been characterized by various liquid chromatography techniques. Size exclusion chromatography (SEC) showed a significant decrease in molecular mass as a result of hydrolytic degradation, while ‘liquid chromatography at critical conditions’ (LC-CC) was very successful for observing differences in functionality due to degradation, i.e. the formation of OH end-groups. To characterize and identify the observed differences semi on-line coupling of liquid chromatography to matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and Fourier transform infrared spectroscopy (FT-IR) has proved to be very useful.Comprehensive two-dimensional liquid chromatography (2D-LC) was also applied to study the hydrolytic degradation of poly(bisphenol A)carbonate. LC-CC × SEC showed that the formation of poly(bisphenol A)carbonate with OH end-groups occurred over the whole molecular mass range. This information could not be obtained with the separate liquid chromatographic techniques, thereby illustrating the usefulness of 2D-LC.The main degradation processes of poly(bisphenol A)carbonate under the applied hydrolysis conditions turned out to be disappearance of cyclic PC oligomers, chain scission of PC and (subsequent) formation of OH end-groups. FT-IR, SEC and LC-CC have been used to follow the hydrolytic degradation with time. LC-CC proved to be the most promising technique to detect the degradation of poly(bisphenol A)carbonate at an early stage.  相似文献

Block copolymers of ethylene oxide (EO) and propylene oxide (PO) are characterized by combination of two-dimensional chromatography and MALDI-TOF-MS. Liquid chromatography under critical conditions (LCCC) is used as first dimension and fractions are collected, mobile phase evaporated and diluted in the mobile phase used in second dimension (SEC or LAC). This two-dimensional chromatography in combination of MALDI-TOF-MS gives information about purity of reaction products, presence of the byproducts, chemical composition and molar mass distribution of all the products.  相似文献

Vacuum distillation residues from two petroleum crudes, a coal liquefaction extract and a coal tar pitch have been fractionated by planar chromatography (PC) using two solvent sequences: pyridine–acetonitrile and tetrahydrofuran (THF)–toluene. Fractions recovered from PC were examined by UV-fluorescence spectroscopy (UV-F), size-exclusion chromatography (SEC). UV-F and SEC of the whole samples showed differences in aromatic cluster size and molecular mass (MM) ranges which could be related to the different origins of the samples. The MM ranges indicated by SEC were greater for the vacuum residues than for coal-derived materials. However, the UV-F spectra of the fractions indicated that the petroleum residue fractions contained similar aromatic types, whereas the fractions from coal liquids contained significantly different aromatic types. SEC profiles of the fractions indicated a separation of coal-derived samples by increasing molecular size with increasing immobility in PC, whereas for petroleum fractions, the same trend was not apparent. MALDI-mass spectra of the set of original samples showed broadly similar ranges of MM distributions but additional work is necessary to identify appropriate matrices and procedures in order to improve the MALDI spectra.  相似文献

Size exclusion chromatography (SEC) is widely used in the characterization and quality control of therapeutic proteins to detect aggregates. Aggregation is a carefully monitored quality attribute from the earliest stages of clinical development owing to the possibility of eliciting an immunogenic response in the patient. During early stage molecule assessment for cell culture production, small-scale screening experiments are performed to permit rapid turn-around of results so as to not delay timelines. We report the development of a capillary SEC methodology for preliminary molecule assessment to support the evaluation of therapeutic candidates at an early stage of development. By making several key modifications to a commercially available liquid chromatography system, we demonstrate a platform process to perform capillary SEC with excellent precision, picogram sensitivity and good ruggedness. The limit of quantitation was determined to be approximately 15 pg; picogram sensitivity for SEC analysis of monoclonal antibodies had not been achieved prior to this work. In addition, we have developed a method to capture low levels of antibody (1 μg/mL) from harvested cell culture fluid (HCCF) for capillary SEC analysis. Up to 40% recovery efficiency was achieved using this micro-recovery method on 3 mL HCCF samples. Using early stage cell culture transient transfection samples, which typically have much lower titers than stable cell line samples, we demonstrate a consistent method for analyzing aggregates in low protein concentration HCCF samples for molecule assessment and early stage candidate screening.  相似文献

A comparison between various methods to determine diffusion coefficients of polymers in dilute solutions has been made. It is shown that Taylor dispersion analysis (TDA), dynamic light scattering (DLS), hydrodynamic chromatography (HDC), and size exclusion chromatography (SEC) can all be used to accurately determine diffusion coefficients when the polymer samples have low polydispersities. By the analysis of a series of practically representative styrene acrylonitrile copolymer (SAN) samples, it is shown that polydispersity of the samples and the presence of low‐molecular‐mass material cause considerable differences between the methods. It was found that TDA is mostly disturbed by the presence of low‐molecular‐mass material, whereas DLS is more sensitive to the polydispersity of the polymer. With broad samples, DLS gives the Z‐average diffusion coefficient. SEC can be used to obtain a diffusion coefficient distribution as well as an average diffusion coefficient of a polydisperse sample. Although, the same was expected for HDC, it was found that this method could only be successfully used for polymer samples having low polydispersities. Deviations between SEC, HDC, and TDA found for narrow samples were not related to the chemical composition of the samples. © 1999 John Wiley & Sons, Inc. Phosphorescent iridium(III) complexes have been attracting increasing attention in applications as luminescent chemosensors. However, no instance of an iridium(III) complex being used as a molecular logic gate has hitherto been reported. In the present study, two iridium(III) complexes, [Ir(ppy)(2)(PBT)] and [Ir(ppy)(2)(PBO)], have been synthesized (PBT, 2-(2-Hydroxyphenyl)-benzothiazole; PBO, 2-(2-hydroxyphenyl)-benzoxazole), and their chemical structures have been characterized by single-crystal X-ray analysis. Theoretical calculations and detailed studies of the photophysical and electrochemical properties of these two complexes have shown that the N^O ligands dominate their luminescence emission properties. Moreover, [Ir(ppy)(2)(PBT)], containing a sulfur atom in the N^O ligand, can serve as a highly selective chemodosimeter for Hg(2+) with ratiometric and naked-eye detection, which is associated with the dissociation of the N^O ligand PBT from the complex. Furthermore, complex [Ir(ppy)(2)(PBT)] has been further developed as an AND and INHIBIT logic gate with Hg(2+) and histidine as inputs.  相似文献

Amphiphilic block copolymers were synthesised by ring opening polymerization of ε-caprolactone with polyethylene glycol monomethyl ether (PEG-MME) using different catalysts (boron trifluoride, sodium hydride, and tin octoate) in a one pot procedure. The products obtained were characterized with respect to their molar mass distribution and content of homopolymers using size exclusion chromatography (SEC), Liquid chromatography under critical conditions (LCCC) and MALDI-TOF-MS. The homopolymers of caprolactone could be separated from the block copolymer by LCCC on a reversed phase column in tetrahydrofurane-water mobile phases with evaporative light scattering detection (ELSD). Residual monomer could be determined under the same conditions using density detection, a separation of the copolymer from residual initiator could be achieved on a normal phase column in acetone-water mobile phases.  相似文献

Two methods of analysis of organic components of colour layers of art works have been tested: IR microspectroscopy of indigo, Cu-phthalocyanine, and Prussian blue, and MALDI-TOF-MS of proteinaceous binders and a protein-containing red dye. The IR spectra distortion common for smooth outer surfaces and polished cross sections of colour layer of art works is suppressed by reflectance measurement of microtome slices. The detection limit of the three blue pigments examined is ~0.3 wt% in reference colour layers in linseed oil binder with calcite as extender and lead white as a drying agent. The sensitivity has been sufficient to identify Prussian blue in repaints on a Gothic painting. MALDI-TOF-MS has been used to identify proteinaceous binders in two historical paintings, namely isinglass (fish glue) and rabbit glue. MALDI-TOF-MS has also been proposed for identification of an insect red dye, cochineal carmine, according to its specific protein component. The enzymatic cleavage with trypsin before MALDI-TOF-MS seems to be a very gentle and specific way of dissolution of the colour layers highly polymerised due to very long aging of old, e.g. medieval, samples.  相似文献

This paper describes how size exclusion chromatography (SEC) can be used to rapidly characterize Au/Pd core/shell nanoparticles (NPs). We monitored the sizes of Au/Pd core/shell NPs by effecting SEC separation using a mobile phase of 10 mM sodium dodecyl sulfate (SDS); the plot of retention time with respect to the standard size of the Au NPs was linear (R 2 = 0.991) for diameters falling in the range from 12.1 to 59.9 nm; for five consecutive runs, the relative standard deviations of these retention times were less than 0.4%. Under the optimized separation conditions, we found that the addition of the surfactant SDS stabilized the Au/Pd core/shell NP samples. In addition, SEC analysis revealed that the sizes of the Au/Pd core/shell NPs could be controlled via modification of the rate of addition of the reducing agent and the use of adequate volumes of the seed and shell precursor metal ion solutions. When using these conditions to analyze the Au/Pd core/shell NPs produced through seed-assisted synthesis, a good correlation existed between the sizes determined through SEC and transmission electron microscopy. Our results suggest that SEC is a useful technique for monitoring the sizes of NPs and nanomaterials in general.  相似文献
