Abstract— The Stern-Volmer constants for fluorescence quenching by tetramethylethylene decrease in the order DMC ≫ DHP > F-2 > 8-MOP. The same order was observed for the quantum yields of [2+2] cycloaddition reaction with tetramethylethylene on direct irradiation. In [2+2] photocycloaddition of F-2 with tetramethylethylene in ethanol, the ratio of quantum yields deduced from singlet and triplet states of F-2; φ3010, is about 5. The excited triplet state is the reactive state for the [2+2] photocycloaddition of F-2 with tetramethylethylene in solution but the excited singlet state of F-2 becomes very important in biological conditions.  相似文献

Abstract— Absorption and emission spectra of several N-arylbenzamides have been measured. The quantum yields for their fluorescence were found to be dependent on matrix viscosity and temperature. Singlet-triplet splittings for these compounds were determined from their emission spectra and found to be abnormally small for π. π* states (˜ 1500 cm-1). Indeed, the phosphorescence maxima of N-arylbenzamides occur slightly to the blue relative to their fluorescence maxima. Intersystem crossing efficiencies were determined for several of these compounds and are consistent with S1→ S 0 radiationless decay.  相似文献

Abstract— Quantum yields for the lumiflavin-sensitized oxidation of guanosine monophosphate and adenosine monophosphate in solution have been measured as functions of oxygen and nucleotide concentration. The quantum yield increases with oxygen concentration at low oxygen concentrations, but quenching of the excited flavin molecule by oxygen results in a fall in quantum yield at higher concentrations. It has also been established that the reciprocal of the quantum yield is linearly related to the reciprocal of the nucleotide concentration. A mechanism in which molecular oxygen reacts with an excited complex formed between triplet lumiflavin and the nucleotide is consistent with these observations.
A value for the second-order rate constant for the quenching of triplet lumifiavin by oxygen of 2·65 × 109 M -1 sec-1 has been obtained.  相似文献

Encapsulating a xanthene dye in phospholipid vesicles produces vesicle solutions that contain dye at very high microscopic concentrations, but have a low overall optical density, thereby eliminating reabsorption. Using this system, we have studied the effects of concentration on the fluorescence lifetime of one such dye, sulforhodamine 101. We have observed that the lifetime decreases as a function of encapsulated dye concentration, which is indicative of collisional quenching. The lifetime decreases from 4.5 nsec for sulforhodamine in dilute aqueous solution to 0.69 ns at an encapsulated concentration of 33 m M . The bimolecular rate constant for this event is 2.6 1010 M -1 s-1, consistent with a diffusion controlled event. However, the quenching constant calculated from changes in intensity is 2.2 1011 M -1 s-1. Thus, collisional quenching is not the predominant mechanism of quenching. The absorption spectra of dye in vesicles indicate an important contribution from static complex formation. Förster distance calculations indicate that energy transfer can also occur to a significant extent, with a predicted efficiency of transfer of 34% at a dye concentration of only 1 m M  相似文献

Abstract— The fluoroquinolone antibiotics can induce skin photosensitivity in some patients and this has been ascribed to the generation of reactive oxygen species, such as singlet oxygen (O2[1Δg]). We have studied the photochemical properties of the different ionized forms of the fluoroquinolone norfloxacin upon complexation with Mg2+ and Ca2+ ions, as it is proposed that the antibiotic exists mainly as a complex in the blood plasma. We found that the norfloxacin cation (pH < 6) shows no photodegradation after UVA irradiation and has a low quantum yield of O2(1Δg) generation. The norfloxacin cation does not complex. Ca2+ or Mg2+ ions; when these ions are added to the solution, we observed no changes in the fluorescence quantum yields (φflu) and singlet oxygen yields (φΔ). In contrast, the neutral (6 ± pH > 8.5) and anionic (pH > 9) forms of norfloxacin are able to complex calcium and magnesium, and their generation of O2 (1Δg) is decreased by complexation. The neutral zwitterionic form and the anionic form also quench singlet oxygen by both chemical and physical pathways regardless of complex formation, while physical quenching is observed for the cation. At pH > 7.4, norfloxacin photobleaches and complexation to Ca2+ and Mg2+ increases the rate at which photobleaching occurs. Thus, both the pH of the medium and complexation with metal cations may affect the phototoxic potential of this antibiotic.  相似文献

Abstract— Absorption changes attributed to the triplet state of carotenoids and to primary electron donors (P-700. P-680): and fluorescence quenching at several wavelengths have been measured with a single apparatus. following flash excitation with a dye or a ruby laser. Spinach chloroplasts as well as subchloroplast particles enriched in Photosystem-1 (F1), Photosystem-2 (F1) or the light-harvesting Chl a/h (FIII) have been examined at temperatures varying between 5 and 294 K.
The triplet state of carotenoids has been identified on the basis of its difference spectrum (having a peak at 515 nm) and decay kinetics (⋍ 7 µs at low temperature; accelerated by O2 at 294 K). It is formed in all of the materials studied. The quantum yield of carotenoid triplet formation in chloroplasts increases at low temperature, but less than the fluorescence yield.
In most cases the fluorescence quenching recovers approximately with the same kinetics as the decay of the carotenoid triplets. The fluorescence recovery is, however, significantly faster for chloroplasts at 730 nm. Fluorescence quenching occurs in all types of materials. The ratio of fluorescence quenching to the concentration of carotenoid triplets varies with the material, being maximum in chloroplasts and minimum in Fm particles.
We conclude that the formation of the carotenoid triplet state is not limited to a few sites in the chloroplast and that a carotenoid triplet is a quencher of chlorophyll fluorescence. A detailed comparison of carotenoid triplets and fluorescence quenching gives some information concerning the organization of the pigments in the photosynthetic apparatus.  相似文献

Abstract— The quenching of 8-methoxypsoralen (8-MOP) fluorescence by protons was observed to occur at the diffusion controlled rates in aqueous solutions at room temperature. Enhanced basicity of 8-MOP in the excited state compared to the ground state is expected on theoretical grounds. The fluorescence yield. which we determined as 6.3 × 10--4 at pH 1 is surprisingly low and indicative of extremely fast radiationless decay pathways. The fluorescence lifetime of 8-MOP in neutral aqueous solution is on the order of 1–2 ns.  相似文献

Abstract— Mono- and multilayers of chlorophyll a (Chl a )– lecithin have been prepared on quartz slides, by means of the Blodgett-Langmuir technique, for fluorescence studies. Self-quenching of the Chl a fluorescence has been observed in Chl a -lecithin single layer excited with a laser light at 632.8 nm. The fluorescence yield is reduced by 50% at a concentration of 7 ± 1012 Chl a molecules cm−2. Chl a fluorescence quenching, by adding N,N -distearoyl-1,4-diaminoanthraquinone (SAQ), has been studied. in a single layer, in pure Chl a and also at various dilutions of Chl a in lecithin. The results are explained in terms of a dynamic quenching rather than in terms of a permanent complex formation, at the ground state, between Chl a and SAQ. The fluorescence quenching has been interpreted as the result of an electron transfer from excited Chl a to SAQ, and rate constants of 8.3 ± 10−5 cm2 molecule−1 S−1 and 2.4 ± 10−4 cm2 molecule−1 s−1 have been found for pure diluted Chl a , respectively. Ten per cent of the diluted Chl a fluorescence always remains unquenchable and independent of the quinone concentration. In multilayers, where SAQ and Chl a are in different layers, there is no fluorescence quenching for pure or diluted Chl a even when the chromophores are in two adjacent layers. This happens only if SAQ is not able to diffuse from one layer to another. A minimum value of 22.4 nm has been found for the singlet exciton diffusion length in pure Chl a multilayers.  相似文献

CURCUMIN-DERIVED TRANSIENTS: A PULSED LASER AND PULSE RADIOLYSIS STUDY   总被引:1,自引:0,他引:1  
Abstract In this paper we report a time-resolved investigation of transients derived from curcumin, which may be intimately involved in the processes leading to its biological activity. Fluorescence and triplet quantum yields are respectively 0.06 and 0.11. The high percentage of internal conversion is proposed to proceed via H-transfer within the thermodynamically favored enol structure of what is formally a 1,3-diketone. The triplet energy (191 ± 2 kJ mol−1), natural lifetime (1.5 μs) and self-quenching rate constant (5.0 × 108 L mol−1 s−1) have been determined. Oxygen quenching of the triplet leads to the production of singlet oxygen with unit efficiency. Curcumin quenches the latter species very inefficiently (2.5 × 105 L mob−1 s−1). The curcumin radical has been produced via three mechanistically distinct methods. This species is unreactive toward oxygen but is repaired by vitamins C and E and anthralin.  相似文献

Abstract— The electronic absorption, fluorescence emission and photoreaction quantum yields for thymine-psoralen (1), psoralen-psoralen (2) and thymine-thymine (3) linked by triethyleneoxide chains are determined in ethanol and in water. Compounds (1), (2) and (3) are able to complex Li+ cations, new and striking effects of which have been revealed in their fluorescence properties and photoreactivities. An important hypochromism effect is observed in the UV spectrum of compound (1) in contrast to compounds (2) and (3).  相似文献

Abstract— The fluorescence decay profiles, relative quantum yield, and transmission of the phycoerythrin a subunit, isolated from the photosynthetic antenna system of Nostoc sp., were measured using single picosecond laser excitation. The fluorescence decay profiles were found to be intensity independent for the intensity range investigated (4 × 1013 and 4 × 1015 photons-cm-2 per pulse). The decay profiles were fitted to a model assuming both chromophores absorb and fluoresce. The inferred total deactivation rates for the two chromophores, in the absence of energy transfer and when the effects of the response time of the streak camera and the finite pulse width are properly included, are 1.0 × 1010s' and 1.0 × 109 s 1 for the s and f chromophores. respectively, whereas the transfer rate between the two fluorophorcs is estimated to be 1.0 × 1010 s−1 giving a s→ f transfer rate on the order of (100 ps)−1. Steady-atate polarization measurements were found to be equal to those calculated using the rate parameters inferred from the kinetic model fit to the fluorescence decays. The apparent decrease in the relative fluorescence quantum yield and increase of the relative transmission with increasing excitation intensity is suggestive of ground state depletion and upper excited state absorption. Evidence suggests that exciton annihilation is absent within isolated α subunits for the intensity range investigated (4 × 1013 to 4 × 1015 photons-cm 2 per pulse).  相似文献

Abstract— The absorption spectra, lifetimes, extinction coefficients and intersystem crossing quantum yields of the lowest triplet T 1 of 4-thiouridine have been determined both in acetonitrile and in water. Abstract— The quantum yield of the photodynamic inactivation of lysozyme increases in the sequence acridine orange, methylene blue, proflavine and acriflavine (1:5:6:12). At least up to protein concentrations of 0.1 m M , singlet oxygen is exclusively responsible for the inactivation of the enzyme. For methylene blue, acriflavine and proflavine the quantum yields decrease considerably with increasing dye concentrations. From measurements in H2O and D2O buffer solutions it was concluded that in the case of methylene blue the effect is mainly caused by the quenching of singlet oxygen [rate constant (3–4) × 108 M −1 s−1]. For the acridine sensitizers both singlet oxygen and dye triplet quenching processes have to be taken into consideration. It has been found that all sensitizers act as competitive inhibitors of the enzymatic reaction of lysozyme. However, the dye-protein interaction near the active center cannot be responsible for the observed dye self-quenching effect.  相似文献

Abstract— Ascorbic acid and ascorbate in chlorophyll ethanol solution were found to be fairly efficient quenchers of the chlorophyll triplet state; comparable to the efficiency of ascorbic acid as a quencher in aqueous pyridine solution.
It has been well established that ascorbic acid quenches the triplet state of chlorophyll in aqueous pyridine solution.(1,2) The bimolecular quenching constant, kQ , is very much less than that for O2 or quinine.(3,4)
Information regarding the quenching of the triplet state of chlorophyll by ascorbic acid in ethanolic solution is lacking. There has been some question as to whether ascorbic acid reduces photoexcited chloro-phyll-ethanolic solution because of its high oxidation potential, or because like the ascorbate ion, it acts only as a quencher; both ascorbic acid and ascorbate in high concentrations gave low quantum yields.(5) The quenching of the triplet state by ascorbic acid and ascorbate was determined by the flash-photolytic method.  相似文献

Abstract— The long-lived (> 1 μsec) transients formed in the flash excitation of the representative photosensitizers methylene blue, eosin Y and pyrene have been investigated and various criteria have been used to distinguish between triplet state intermediates and chemical intermediates. Previous assignments of the triplet transients of methylene blue appeared less secure in view of the photochemical reactivity of this dye and its lack of phosphorescence. Earlier assignments of monomeric and dimeric triplet transients of methylene blue are substantiated, however, by the observations that the rate constant for quenching by oxygen is approximately 1/9th diffusion controlled and the formation rates are commensurate with singlet decay rates and by the observation of triplet-triplet annihilation. Additional evidence in support of monomer triplet assignments for methylene blue and eosin Y is provided by the effect of heavy atom quenchers Cs+, Hg2+ and T1+ on decay rates. Due to chemical reactivity, quenching by Iappears less suitable as a diagnostic test for triplet state intermediates. The effect of N3, which is known to quench singlet oxygen molecules and to alter the course of photosensitized oxidations, on the triplet decay of methylene blue, eosin Y and pyrene is also investigated.  相似文献

Abstract— The mechanism of the photoreduction of 9,10-anthraquinone (AQ) in alcohol and hexane has been studied by flash photolysis. The fluorescence spectrum of the photoproduct, 9,10-dihydroxy anthracene shows a large shift between hexane and ethanol. The quantum yields of photoreduction for AQ are solvent-dependent, the reaction between the solvent radical and AQ determining the quantum yield.
The absorption spectrum of the 9,10-anthrasemiquinone (AQH.) has a long-wavelength absorption band with peaks at 631 and 678 nm. The second-order decay constants for AQH. were estimated to be 1.3 × 109, 6.7 × 108 and 2.0 × 108 M -1 sec-1 in ethanol, 2-propanol and ethylene glycol, respectively.
A long-wavelength absorption band was observed for 9,10-anthrasemiquinone radical anion, having peaks at 776 and 860 nm; epsi;max= 1900 at 776 nm. This spectrum is compared with the spectra of 9,10-dihydroxy anthracene mono- and di-anions. The 9,10-anthrasemiquinone radical anion was found to photoreduce quantitatively to 9,10-dihydroxy anthracene mono-anion with a quantum yield of 0.1.  相似文献

Abstract— The influence of chloride ion on the rate of decay of triplet methylene blue in 0.01 M acid in the absence and presence of ferrous ions was investigated by means of laser flash-photolysis monitored by kinetic spectrophotometry. Chloride weakly accelerates decay of 3MBH in aqueous solution in the absence of Fe(II). Quenching of 3MBH2+ by Fe(II) is more strongly catalyzed by Cl- in both water and 50 v/v% aq. CH3CN. The uncatalyzed quenching constant, k 5, is of the order of 1 × 106 M -1 s-1 while in 4.8 M aqueous chloride ( μ – 7.2 M ) k 5= (37.2 ± 1.8) × 106 M -1 s-1. A possible role of chloride is as a bridging species in quenching via electron transfer between 3MBH2+ and Fe(II).  相似文献

Abstract— The dynamics of the flavin bound to the flavocytochrome b2 from Hansenula anomala were studied by fluorescence intensity quenching and quenching emission anisotropy with iodide. The fluorescence intensity of bound flavin is decreased 13-fold as compared to the free molecule. The remaining fluorescence decays with two lifetimes equal to 0.963 ± 0.040 and 4.635 ± 0.008 ns and fractional intensities of 0.036 ± 0.002 and 0.964 ± 0.002, respectively. The bimolecular diffusion constant was found to be 3.33 × 109 M -1 s-1 when the flavin is bound to the enzyme and 8.3 × 109 Mv s-1 when the flavin is free in solution. Thus, the flavin in flavocytochrome b2 is accessible to the solvent, but the amino acid residues of the binding site inhibit the diffusion of iodide. The rotational correlation time of bound flavin was found to be 2.015 ± 0.365 ns, a value higher than that (155 ps) of free flavin in solution. Our results are discussed on the basis of local dynamics of the flavin.  相似文献

Abstract— The fluorescence yields and lifetimes of indole, five of its alkyl detivatives, tryptophan, and tryptamine have been determined in degassed, heavy and light water at room temperature. All of the compounds have radiative lifetimes nearly identical to the parent compound indole, and a comparison of these results with recently reported data on tryptophyl derivatives disclosed a striking uniformity in radiative lifetimes between indole and many amino acids and peptides which contain the indole group as the fluorescence unit. The fluorescence rate k f in H2O, was found to be 4.5 × 107 sec-1. The nonradiative decay rates were found to vary between 5.1 and 46 × 107 sec-1 and from a study of the deuterium-solvent isotope effect and the deuterium-substituent effect a mechanism for nonradiative deactivation is proposed which includes an isotopically dependent proton transfer and a pathway involving energy loss via the ring carbon hydrogen vibrations. Tryptophan at pH 7 was found to have a unique nonradiative decay scheme not evidenced at a pH 1 or pH 10.  相似文献

Abstract— The quantum yields of phosphorescence and fluorescence and the phosphorescence lifetime have been determined for biacetyl in benzene, p-xylene, n-heptane, and water, and for benzil in n-heptane at 22C. The following values for the fluorescence quantum yield and natural phosphorescence lifetime were obtained: For biacetyl in benzene, 2.7 times 10-3 and 7.8 msec, in p-xylene, 2.5 times 10-3 and 10.8 msec, in n-heptane, 2.9 times 10-3 and 10.8 msec, in water, 1.1 times 10-3 and 10.0 msec, and for benzil in n-heptane, 1.1 times 10-3 and 10.4 msec.  相似文献
