Simple Stereospecific Syntheses of (R)-2-Fluorohexanoic Acid Ethyl Ester

Simple stereospecific syntheses of 2-fluorohexanoic acid 5 were accomplished by conversion of optically pure L-(+)-norleucine to the optically pure hydroxy acid 3 via the classic diazotization reaction followed by substitution of the hydroxy functionality by the fluoro group. This was accomplished stereospecifically using DAST reagent or more practically by nucleophilic displacement of the corresponding mesyloxy group with fluoride ion.     

6-取代苯氧基-7-氯-4-羟基-3-喹啉羧酸乙酯的合成及抗球虫活性研究

设计并合成了10个新的6-取代苯氧基-7-氯-4-羟基-3-喹啉羧酸乙酯类化合物; 合成路线短, 产率高, 原料易得, 易实现工业化; 所有化合物的结构均经¹H NMR, MS, IR和元素分析所证实; 并选择了其中7个化合物进行抗球虫活性实验, 结果表明: 其中4个6-取代苯氧基-7-氯-4-羟基-3-喹啉羧酸乙酯化合物具有抗球虫效果, 并有可能作为抗球虫药物使用.

Synthesis of 7-hydroxy(phenyl)ethylguanines by alkylation of 2-amino-6-chloropurine with allyl-protected bromohydrins

Protecting the hydroxyl group in both 2-bromo-2-phenylethanol and 2-bromo-1-phenylethanol enhanced the alkylation of 2-amino-6-chloropurine to give corresponding 7- and 9-alkylated products. Subsequent hydrolysis and deprotection led to 7- and 9-hydroxy(phenyl)ethylguanines. 7-Hydroxy(phenyl)ethylguanines are major guanine adducts formed by interaction of styrene 7,8-oxide with DNA.

Metal complexes of 6-chloropurine

Summary Five Cu(II), Pd(II), Cd(II), Pt(IV), and Au(III) complexes of 6-chloropurine have been obtained. The complexes were characterized by elemental analysis, IR,¹H-NMR and¹³C-NMR spectroscopy. On the basis of these data the structure of the complexes and the coordination of the ligand have been proposed. Thus, the physical and chemical methods supported evidence that in acidic medium, with exception of the Cu(II) complex, 6-chloropurine acts in the monoprotonated form neutralizing the charge of [PdCl₄]^2–, [CdCl₄]^2–, [AuCl₄]^– and [PtCl₆]^2– anions. The thermal behaviour of the complexes has also been studied.

Metallkomplexe von 6-Chlorpurin

Zusammenfassung Es wurden fünf Komplexe von 6-Chlorpurin mit Cu(II), Pd(II), Cd(II), Pt(IV) und Au(III) erhalten. Die Komplexe wurden mittels Elementaranalysen, IR,¹H-NMR und¹³C-NMR charakterisiert. Auf der Basis dieser Daten wurde eine Komplexstruktur und eine bestimmte Koordination der Liganden vorgeschlagen. Physikalische und chemische Methoden beweisen, daß im sauren Bereich [mit der Ausnahme von Cu(II)] das 6-Chlorpurin in der monoprotonierten Form koordiniert, wobei die Ladung von [PdCl₄]^2–, [CdCl₄]^2–, [AuCl₄]^– und [PtCl₆]^2– jeweils neutralisiert wird. Das thermische Verhalten der Komplexe wurde ebenfalls untersucht.

     

2-甲硫基-4-羟基-5-嘧啶甲酸乙酯的工艺优化

以S-甲基异硫脲半硫酸盐和乙氧基亚甲基丙二酸二乙酯为主要原料,乙醇为溶剂,合成出2-甲硫基-4-羟基-5-嘧啶甲酸乙酯,研究了2-甲硫基-4-羟基-5-嘧啶甲酸乙酯的工艺优化,考察了投料比、反应温度、反应时间等因素对产率的影响。 获得较好的反应条件:n(C₂H₆N₂S·1/2H₂SO₄):n(C₁₀H₁₆O₅):n(NaOH)=1.2:1:1.8,在室温下缓慢滴加NaOH溶液,搅拌反应6 h,酸化得白色固体产物,产率为81.1%。产物结构经熔点测定仪、¹H NMR、ESI-MS、FT-IR等技术手段得到验证。     

Synthesis and Crystal Structure of 4-（4,6-Dimethoxylpyrimidin-2-yl）- 3-thiourea Carboxylic Acid Ethyl Ester

4-（4,6-Dimethoxyl-pyrimidin-2-yl）-3-thiourea carboxylic acid ethyl ester was synthesized by the reaction of 2-amino-4,6-dimethoxyl pyrimidine, potassium thiocyanate and methyl chloroformate in ethyl acetate. Single crystals suitable for X-ray measurement were obtained by recrystallization with the solvent of dimethyl formamide at room temperature. The crystal structure was determined by X-ray diffraction analysis. Crystallographic data： C10H14N4O4S, M, = 286.31, monoclinic, space group C2/c with a = 2.5309（3）, b = 0.67682（6）, c = 1.74237（19） nm, β = 114.744（3）°, V= 2.7106（5） nm3, Dc = 1.403 g/cm3, p = 0.225 mm-1, F（000） = 1200, Z= 8, R= 0.0514 and wR= 0.1529.
