In this study, a sorbent was immobilized inside a needle resulting in the development of a needle trap (NT) device. This device was applied to extract organic components from gaseous samples and to introduce an enriched mixture into a conventional gas chromatography (GC) injector. Construction of this simple and integrated sampling/extraction/sample introduction device was optimized by considering different ways to immobilize a sorbent in the needle, packing single and multiple-layer sorbent beds, and applying different desorption strategies into the GC injector. A carrier gas system was modified to minimize the carryover for the needle trap with a sealed tip (NT-1), and a narrow-neckliner was used for the blunt-tip needle trap (NT-2). Breakthrough in the device was investigated by connecting two NT-2 devices in series. The needle trap performed very well as an exhaustive spot sampler, as well as in a time-weighted average (TWA) operation. The linear velocity of the mobile phase has no influence on the sampling rate of the needle trap. Validation results against the standard NIOSH 1501 method using charcoal tubes for indoor air surveys demonstrated good accuracy for the NT approach. The reproducibility of the NT-2 was about 1% for benzene. The detection limits for FID detection and for 25 ml gas sample were 0.23 ng/l, 2.10 ng/l and 1.12 ng/l for benzene, ethylbenzene and o-xylene, respectively.  相似文献

A sensitive and robust liquid chromatography/electrospray ion trap mass spectrometry (LC/MS/MS) method has been developed for the quantitative determination of noncovalently bound acridinium free acid in protein-acridinium conjugates. The lower level of quantitation (LOQ) for acridinium free acid was determined to be 0.6 ng. The assay was validated with a linear concentration range of 0.6-60 ng. The method requires minimum sample handling and is specific, reproducible, and provides a new aspect for protein-acridinium conjugate characterization.  相似文献

Pressurised hot water extraction (PHWE) was coupled on-line with liquid chromatography-gas chromatography (LC-GC) to determine brominated flame retardants in sediment samples. After extraction with pressurised hot water the analytes were adsorbed in a solid-phase trap. The trap was dried with nitrogen and the analytes were eluted to the LC column, where the extract was cleaned, concentrated and fractionated before transfer to the GC system. The fraction containing the brominated flame retardants was transferred to the GC system via an on-column interface. The PHWE-LC-GC method was linear from 0.0125 to 2.5 microg with limits of detection in the range 0.70-1.41 ng/g and limits of quantification 6.16-12.33 ng/g.  相似文献

Five different mass spectrometers interfaced to GC or LC were evaluated for their application to targeted and nontargeted screening of pesticides in two foods, spinach and ginseng. The five MS systems were capillary GC/MS/MS, GC-high resolution time-of-flight (GC/HR-TOF)-MS, TOF-MS interfaced with a comprehensive multidimensional GC (GCxGC/TOF-MS), an MS/MS ion trap hybrid mass (qTrap) system interfaced with an ultra-performance liquid chromatograph (UPLC-qTrap), and UPLC interfaced to an orbital trap high resolution mass spectrometer (UPLC/Orbitrap HR-MS). Each MS system was tested with spinach and ginseng extracts prepared through a modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) procedure. Each matrix was fortified at 10 and 50 ng/g for spinach or 25 and 100 ng/g for ginseng with subsets of 486 pesticides, isomers, and metabolites representing most pesticide classes. HR-TOF-MS was effective in a targeted search for characteristic accurate mass ions and identified 97% of 170 pesticides in ginseng at 25 ng/g. A targeted screen of either ginseng or spinach found 94-95% of pesticides fortified for analysis at 10 ng/g with GC/MS/MS or LC/MS/MS using multiple reaction monitoring (MRM) procedures. Orbitrap-MS successfully found 89% of 177 fortified pesticides in spinach at 25 ng/g using a targeted search of accurate mass pseudomolecular ions in the positive electrospray ionization mode. A comprehensive GCxGC/TOF-MS system provided separation and identification of 342 pesticides and metabolites in a single 32 min acquisition with standards. Only 67 or 81% of the pesticides were identified in ginseng and spinach matrixes at 25 ng/g or 10 ng/g, respectively. MS/MS or qTrap-MS operated in the MRM mode produced the lowest false-negative rates, at 10 ng/g. Improvements to instrumentation, methods, and software are needed for efficient use of nontargeted screens in parallel with triple quadrupole MS.  相似文献

Abstract

A chemical ionization and an electron impact GC/MS assaying approach is presented for determining dibucaine concentrations in biological fluids. Both use deuterium-labeled drug as the internal standard and rely on the same sample extraction and sample preparation procedure. Under chemical ionization conditions (CH4), the assaying limits are in the range of 1–80 ng/ml of serum. Under electron impact conditions, the analytical range is 20–800 ng/ml. The chemical ionization procedure has been found suitable for monitoring drug levels in man. One volunteer, who received a single 5-mg oral dose, showed peak serum drug concentrations of 23 ng/ml attained 2 hr after drug administration. The biologic half-life (ß phase) was 11 hr.  相似文献

Summary Methods are described for the analysis of environmental samples like water, sediment and suspended matter for the determination of all organotin compounds (OTs) that are currently used as biocides: tributyltin (TBT), triphenyltin (TPT), tricyclohexyltin (TCT) and fenbutatin oxide (FBTO). In water also five degradation products (di and mono substituted analogs) can be determined. Alkylation using a Grignard reagent was used to obtain OT derivatives amenable to gas chromatography (GC). Both methylation and pentylation have been employed for derivatization prior to GC analysis. The present results show that derivatization efficiencies for TPT, TCT and FBTO at trace levels are higher using methylation than pentylation. Detection limits for each type of sample matrix were determined using GC/Mass Selective Detection (GC/MSD) and GC/Atomic Emission Detection (AED). In sediment and suspended matter only tri-substituted OTs (i.e. the parent compounds) could be determined. Detection limits ranged from 0.2 to 10 ng/g dry weight. FBTO, not previously detected in environmental samples, was found at levels of 4 and 11 ng/g in a suspended matter sample and a sediment sample, respectively. In water the OTs and their degradation products were determined at levels of 1–10 ng/l (as tin) using 200 ml water samples.  相似文献

A direct ultra-high performance reverse-phase HPLC (UHPLC)--electrospray MS/MS method was developed for the simultaneous determination of 16 fluorinated benzoic acids (FBAs) in oil reservoir waters. The separation was achieved within 5 min in a non-linear gradient mode using a 1-ml sample aliquot. The method detection limits were in the lower ng/ml range (between 0.05 and 50 ng/ml, depending on the compound) owing to the use of the travelling-wave collision cell technology. The method developed was more sensitive, faster (by avoiding sample preconcentration and purification steps) and more robust than the GC/MS methods currently used in oil industries. The accuracy of the method was verified by comparison with GC/MS results. It was applied to the determination of FBAs in water samples coming from reservoir tracing campaigns.  相似文献

A simplified preconcentration method for a range of ultra-trace level pharmaceuticals in natural waters has been developed. Solid phase extraction was performed on-line using a micro-reversed-phase monolithic silica column, allowing for very rapid trace enrichment from large volume (500 ml) samples with minimal sample handling. Acceptable recoveries of >70% were obtained for the majority of compounds investigated and the monolithic columns could be washed and conditioned on-line with no sample carryover and used reproducibly for up to eight extractions each. The on-line SPE-LC-UV method was coupled to electrospray ionisation ion trap mass spectrometry (ESI-MS) to increase both selectivity and specificity. Detection limits were determined in spiked river and tap water samples and found to lie in the low ng/l region using sample volumes of 500 ml, loaded at a flow rate of 10 ml/min, and therefore, were suitable for ultra trace analysis.  相似文献

Pentazocine has been found to be measurable with much higher sensitivity by gas chromatography (GC)/surface ionization (SI) organic mass spectrometry (OMS) than by the conventional GC/electron ionization (EI) mass spectrometry. The compound was extracted from human whole blood and urine with Sep-Pak C(18) cartridges before analysis by GC/SIOMS; recoveries were > 96.6% for both samples. The calibration curves were linear in the range 6.25-100 ng ml(-1) and the detection limits were 500 pg ml(-1) of a sample by selected ion monitoring (SIM) with GC/SIOMS. The intra- and inter-day relative standard deviations for the determination of pentazocine in whole blood and urine were not greater than 9.6%. The sensitivity for pentazocine obtained by SI-SIM was about 60 times higher than that obtained by EI-SIM. To validate the present GC/SIOMS method for pentazocine, whole blood and urine samples collected from two volunteers 1-6 h after intramuscular injection of 15 mg of pentazocine were analyzed. The concentrations were 13.5-59.3 ng ml(-1) for whole blood and 0.39-4.00 microg ml(-1) for urine.  相似文献

A computer-controlled flow injection system was developed for the determination of cadmium in a hydrometallurgical zinc refining process stream. An anion-exchange method in acidic potassium iodide medium was used for the on-line separation of cadmium from the matrix zinc. 1-(4-Nitrophenyl)-3-(4-phenylazophenyl)triazene (Cadion) was used as the chromogenic reagent for the spectrophotometric detection of cadmium. In order to expand the dynamic range of the flow injection - spectrophotometry, a computer-aided time-based variable-volume injection method has been employed for the introduction of the sample into the flow injection system. Samples ranging from 0.56 to 350 microl can be delivered by controlling the time period of the sample introduction valve and the flow rate of the carrier solution. The system permits a throughput of 5 samples per hour. The reproducibility has been proven to be satisfactory with a relative standard deviation of less than 6.2% (sample injected: 0.56 microl of 850 microg Cd/ml; n=100) and 5.0% (350 microl of 0.14 microg Cd/ml; n=5). The determination limit was 20 microg Cd/ml with 0.56 microl sample injection and 0.05 microg Cd/ml with 350 microl sample injection (the absolute amount of cadmium injected into the system was 11 ng and 17.5 ng, respectively).  相似文献

A sensitive and specific capillary gas chromatographic (GC) assay was developed for the quantitation of the quaternary ammonium steroidal neuromuscular blocking drugs pancuronium (PANC), vecuronium (VEC) and pipecuronium (PIP), as well as the metabolites 3-desacetylpancuronium (3-desPANC) and 3-desacetylvecuronium (3-des VEC) in plasma, bile and urine; the putative metabolite 3-desacetylpipecuronium (3-des PIP) was extracted and quantitated only in urine. The procedure employed a single dichloromethane extraction of the iodide ion-pairs of the monoquaternary or bisquaternary ammonium compounds (including internal and external standards) from acidified, ether-washed biological fluid followed by the formation of stable O-tert.-butyldimethylsilyl derivatives at the 3-hydroxy steroidal position of the metabolites. An automated capillary GC system fitted with a nitrogen-sensitive detector and an integrator was then used to analyze and quantitate both parent compounds and their derivatized metabolites. Optimal extraction, derivatization and GC conditions, as well as short-term stability and recoveries of these drugs and metabolites in plasma, are reported. Electron ionization mass spectrometry combined with GC was used to confirm the identities of compounds eluted from the column. The assay demonstrated a 10(3)-fold linear range up to 5000 ng/ml for PANC, VEC, 3-des VEC and PIP, and lower limits of detection with adequate precision of 2 ng/ml for PANC, VEC and PIP, and 4 ng/ml for 3-des VEC; 3-des PANC was linear from 8 to 500 ng/ml while 3-des PIP was linear from 25 to 1000 ng/ml. The precision (coefficient of variation) of the calibration curves for underivatized drugs and their derivatized metabolites over the linear ranges was 2-20% and the reproducibility of the assay over a range of clinical concentrations of these drugs found in human plasma was 5-16% for PANC, 2-4% for VEC and 6-11% for PIP. No interferences were detected in the assay of plasma samples from 106 surgical patients.  相似文献

A method for detection, quantitation, and confirmation of more than 100 pesticides by gas chromatography (GC) with ion trap mass spectrometry (MS/MS) has been developed. The sensitivity of this method for many analytes is equal to or lower than those of selective GC detectors such as flame photometric detectors and electrolytic conductivity detectors. Using MS/MS, very low detection limits and good confirmation (1 precursor ion and 2 or more product ions) are achieved simultaneously. The entire list of pesticides is screened with 2 injections per sample. Samples are introduced onto the column by a temperature-programmed cold injection to maximize response. Each pesticide is run with its own unique set of parameters, which fragment the compound, retaining only the precursor ion. This ion is then refragmented to create a product spectrum. The selectivity of MS/MS gives a very clean spectrum, making compound identification and confirmation clear, even with a relatively dirty food matrix. If care is taken to maintain the injection port and guard column, this method can reliably identify and confirm more than 100 pesticides at the low parts-per-billion range.  相似文献

A novel Hadamard transform-gas chromatography/mass spectrometry (HT-GC/MS) system equipped with on-line sample collection systems is described. A Hadamard-injector was successfully designed and then coupled with an on-line adsorption/desorption system for detecting volatile organic compounds (VOCs) and a supercritical fluid extraction (SFE) system, respectively, by HT-GC/MS. Six VOCs and three pesticides were used as model compounds. In the former case, an activated-charcoal trap was used to trap VOCs from the indoor air. After 10 L of indoor air had passed through the trap, the condensed components were heated and simultaneously injected into the GC column through the Hadamard-injector, based on Hadamard codes. In a second experiment, a sample of rice was spiked with three types of pesticides and the sample then extracted using a commercially available supercritical fluid extractor. After extraction, the extracted components were transferred to a holding tank and simultaneously injected into the GC column also using the Hadamard-injector. The findings show that, in both cases, the combination of on-line sample collection methods and the use of the Hadamard transform resulted in improved sensitivity and detection. Compared to the single injection used in most GC/MS systems, the signal-to-noise (S/N) ratios were substantially improved after inverse Hadamard transformation of the encoded chromatogram.  相似文献

A method for the rapid and robust confirmation of 11‐nor‐?9‐tetrahydrocannabinol‐9‐carboxylic acid (THCA) in urine involving basic hydrolysis with NaOH and direct injection of the hydrolysate in a column‐switching LC‐MS‐MS system was developed and validated. THCA‐d3 was used as internal standard. Detection was performed in negative‐ion mode by monitoring the transitions from the [M‐CO2]‐ ion m/z 299.2→245.2 and and m/z 299.2→191.1 that were found to provide a better signal‐to‐noise ratio than the transition from the pseudomolecular ion at m/z 343. The high sensitivity of detection enabled the injection of a small volume (10 µl) of the NaOH hydrolysate which, together with the applied column switching system, proved to confer ruggedness to the method and to avoid the deterioration of the instrumental apparatus despite the large amount of inorganic ions in the hydrolysate. The LLOQ was established at 5 ng/ml, and the LLOD was calculated as 0.2 ng/ml (S/N =3). The method was submitted to thorough validation including evaluation of the calibration range (5–500 ng/ml), accuracy and precision, matrix effects, overall process efficiency, autosampler stability, carryover and cross‐talk, and 10‐times reduction of sample volume (0.1 ml). Proof of applicability was obtained by direct comparison with the reference GC‐MS method in use in the lab (the R2 between the two methods was 0.9951). Abstract

A simple, selective and sensitive HPLC method has been developed to measure R-831 levels in dogs and humans. It is an internal standard technique with a single step extraction and one wash. Samples are chromatographed on a reversed-phase system with ultraviolet detection. The lowest detectable concentration for plasma is 25 ng R-831/ml with a 1 ml sample and the linear range is 25–8000 ng R-831/ml. The lowest detectable concentration for urine is 250 ng R-831/ml with a 0.1 ml sample and the linear range is 250–8000 ng R-831/ml. This method has been used to quantitate levels of R-831 in bioavailability and toxicity studies in dogs, and in pharmacokinetic and efficacy studies in humans.  相似文献

A new analytical approach, based on derivatization with 2,2,2-trichloroethyl chloroformate and gas chromatography/mass spectrometry (GC/MS), was investigated for qualitative and quantitative analyses of a large range of amphetamine-related drugs and ephedrines in plasma, urine and hair samples. Sample preparation involved alkaline extraction of analytes from biological samples using Extrelut columns, after addition of the internal standard 3,4-methylenedioxypropylamphetamine (MDPA), and subsequent derivatization to produce 2,2,2-trichloroethylcarbamates. GC/MS analyses, in splitless mode using a slightly polar 30-m capillary column, were performed with quadrupole or ion trap instruments. MS acquisition modes were electron ionization (EI) in full-scan or selected ion monitoring (SIM) modes (quadrupole), and full-scan MS or MS/MS modes with chemical ionization (CI) conditions (ion trap). EI spectra of 2,2,2-trichloroethylcarbamates showed variably abundant molecular ions as well as abundant diagnostic fragment ions, both characterized by ion clusters reflecting the isotope distribution of three chlorine atoms in the derivatized molecules. CI spectra showed abundant protonated molecules. Quantitative studies using EI SIM conditions gave recoveries in the range 74-89%, linear response over ranges of 10-2000 ng/mL (plasma and urine) and 0.20-20 ng/mg (hair), with corresponding limits of detection in the ranges 2-5 ng/mL and 0.1-0.2 ng/mg. Potential applications (following full method validation) include clinical and forensic toxicology, as well as doping control.  相似文献

Abstract

Sample preparation procedures and gas chromatography methodology are presented for the determination of tabun, sarin, soman, and VX in aqueous solutions. Extraction recoveries from chloroform were quantitative. Peak area ratios of organophosphorus compounds (OPs) to internal standard versus concentrations of OP were linear over the range of 10-1000 (μg/ml when determined by the flame ionization detector and 10-800 μg/ml when determined by the flame photometric detector. Imprecision occurring at low ng concentrations of VX was caused by its adsorption on the analytical column. Acceptable precision was regained by the addition of a weak base, such as atropine, to the sample extract prior to its injection onto the gas chromatograph (GC).  相似文献

An unattended, automated, on-line, cryogen-free, remotely controlled gas chromatography (GC) system was developed and has been deployed for more than 1 year for the continuous determination of C(2) to C(6) hydrocarbons at an observatory located at 2225 m elevation, on the summit caldera of an inactive volcano on the island of Pico, Azores. The GC instrument is tailored to the measurement challenges at this remote and high altitude site. All consumable gases are prepared in situ. Total power use remains below 700 W at all times. Sample collection and analysis is performed without use of cryogen. Hydrocarbons are concentrated on a one-stage trapping/injection system consisting of a Peltier-cooled multi-bed solid adsorbent trap. Analytes are detected after thermal desorption and separation on an alumina-PLOT (porous-layer open tubular) column by flame ionization detection (FID). Sample focusing, desorption, separation and detection parameters were thoroughly investigated to ensure quantitative collection and subsequent injection onto the GC system. GC operation is controlled remotely and data are downloaded daily. Sample volumes (600 and 3000 ml) are alternated for analysis of C(2) to C(3) and C(3) to C(6) hydrocarbons, respectively. Detection limits are in the low parts per trillion by volume (pptv) range, sufficient for quantification of the compounds of interest at their central North Atlantic lower free troposphere background concentrations.  相似文献

A reliable solid-phase extraction (SPE) method for the simultaneous determination of 2,4,6-trichloroanisole (TCA) and 2,4,6-tribromoanisole (TBA) in wines has been developed. In the proposed procedure 50 mL of wine are extracted in a 1 mL cartridge filled with 50 mg of LiChrolut EN resins. Most wine volatiles are washed up with 12.5 mL of a water:methanol solution (70%, v/v) containing 1% of NaHCO3. Analytes are further eluted with 0.6 mL of dichloromethane. A 40 microL aliquot of this extract is directly injected into a PTV injector operated in the solvent split mode, and analysed by gas chromatography (GC)-ion trap mass spectrometry using the selected ion storage mode. The solid-phase extraction, including sample volume and rinsing and elution solvents, and the large volume GC injection have been carefully evaluated and optimized. The resulting method is precise (RSD (%) < 6% at 100 ng L(-1)), sensitive (LOD were 0.2 and 0.4 ng/L for TCA and TBA, respectively), robust (the absolute recoveries of both analytes are higher than 80% and consistent wine to wine) and friendly to the GC-MS system (the extract is clean, simple and free from non-volatiles).  相似文献
