Novel histone deacetylase(HDAC) inhibitors 9a–l were designed and synthesized by coupling the carboxyl group of salicylic acid(SA) with N-hydroxycinnamamides through various alkylol amines, and their in vitro biological activities were evaluated. The N-hydroxycinnamamide/SA hybrids 9b–f and 9h showed good to moderate anti-tumor activities. Notably, compound 9e had a greater potency, comparable to vorinostat(SAHA), in human colon carcinoma cells, which was probably, or at least partially, attributable to the positive effects of the chain length noted in alkylol amines. Furthermore, the HDAC inhibitory activities of 9e against Hela cell nuclear were also similar to that of vorinostat(SAHA), while the tested compounds 9c–f did not exhibit any isoform selectivity in the inhibition of HDACs. In addition, compound 9e could selectively inhibit tumor cells, but not inhibit non-tumor cell proliferation in vitro. Our findings suggest that the N-hydroxycinnamamide/SA hybrids may hold significant promise as therapeutic agents for the intervention of human cancers.  相似文献

A series of novel L-amino acid esters prodrugs of acyclic nucleoside phosphonates was synthesized and their anti-HBVactivity was evaluated in HepG2 2.2.15 cells. Compound 1d exhibited more potent anti-HBV activity and lower cytotoxicity than those of adefovir dipivoxil with EC50 and CC50 values of 0.207 mmol/L and 2530 mmol/L, respectively.  相似文献

A novel dioxime derivative (2E,2′E,3E,3′E)-3,3′-(pyrimidine-4,5-diylbis(azanylylidene))bis(butan-2-one)dioxime was synthesized with a yield of 65%. IR, elemental analysis, mass spectroscopy and 1H-NMR were used to characterize the structure of the synthesized compound. 99mTc-dioxime was radio-synthesized with a high radiochemical yield of 97.8 ± 0.5% and in vitro stability of 6 h under the optimum conditions. The preclinical evaluation of 99mTc-dioxime in solid tumor-bearing mice showed high accumulation in solid tumor cells with a high Target/Non-Target ratio of 5.14 at 30 min post-injection. This study suggests that 99mTc-dioxime derivative is a promising candidate as a new 99mTc-based tumor-imaging agent after further preclinical studies.  相似文献

A biphenyl-based axially chiral amino acid (S)-2 has been designed and synthesized. The new amino acid (S)-2 has been found to be a more efficient catalyst than (S)-1 in the direct asymmetric aldol reaction of acetone with aldehydes. For instance, the use of only 0.1 mol % of (S)-2 was sufficient to complete the reaction between acetone and 4-nitrobenzaldehyde, giving the corresponding aldol adduct in good yield with an excellent enantioselectivity.  相似文献

A series of amino organophosphorus imidazoles were designed and synthesized as a novel structural type of antimicrobial agents.Bioactive evaluation in vitro showed that compound 3f exhibited equipotent or superior anti-methicillin-resistant Staphylococcus aureus(anti-MRSA) and anti-S. cerevisiae efficiencies(minimal inhibitory concentration(MIC)=2 μg/mL) to clinical drugs,and the combinations with antibacterial or antifungal drugs enhanced the antimicrobial efficiency. Highly active molecule 3f showed low propensity for bacteria to develop resistance, and the preliminary action mechanism studies demonstrated that 3f was membrane-active, but had no significant intercalation towards MRSA DNA. The computational study on 3f reasonably explained its high antimicrobial activity. Experimental data revealed that ground-state 3f-HSA complexes were formed mainly through hydrophobic interactions and hydrogen bonds with a spontaneous process, and the non-radioactive energy transfer from HSA to 3f occurred beyond F鰎ster resonance energy transfer theory. The participation of metal ions in 3f-HSA supramolucular system could increase the concentration of free compound 3f, and shorten its storage time and half-life in the blood to improve the maximum antimicrobial efficacy.  相似文献

Summary The aminoacid glutamine in aqueous solution and in conditions of high temperature and long term storage is partly transformed into pyroglutamic acid which exhibits potential neurotoxic effects.Commercially available aminoacid mixtures supplemented with glutamine are heat-sterilized and some losses of glutamine and formation of pyroglutamic acid may occur.The aim of the work was to set up an easy and reliable HPLC method which allows the determination of pyroglutamic acid as a degradation product of glutamine. The column was a 5 m Hypersil ODS (100×4.6 mm) and the mobile phase 100% 0.007 M phosphate buffer pH 3.5.Stability studies in different conditions of temperature and time of storage were performed on aminoacid mixture available in the commerce.  相似文献

A calix[6]arene carboxylic acid derivative was utilized as an extractant for various amino compounds including amino acids. The host compound exhibited a high extractability compared to that of commercially available extractants such as di(2-ethylhexyl)phosphoric acid (D2EHPA). The important factors for amino acid extraction were found to be its cyclic structure and its cavity size, the functional carboxylic groups, and the length of the alkyl chains. The hydrophobicity and the ionic property of guest molecules also affect the extraction efficiency. Calix[6]arene forms a stable complex with a guest molecule by entrapping it into the cavity. The molecular geometries of calixarene before and after extraction were also discussed using computational calculations with the PM3 type of semi-empirical method.  相似文献

99mTc-Sestamibi has been playing an important role in the cardiac imaging for the last decades. Previously, we reported that [99mTc(CO)3(MIBI)3]+ demonstrated a significant location in myocardium with a lower liver uptake as compared with 99mTc-Sestamibi. In this work, we found that new [99mTc(CO)2(MIBI)4]+ could be prepared with high radiochemical purity. The inter-transformations between [99mTc(CO)3(H2O)(MIBI)2]+, [99mTc(CO)3(MIBI)3]+, and [99mTc(CO)2(MIBI)4]+ were investigated and biodistribution was performed to evaluate the [99mTc(CO)2(MIBI)4]+ as a myocardial perfusion imaging agent. The results showed that one more CO was replaced by MIBI slowing down the pharmacokinetics. The structure characterization was performed on their corresponding rhenium complexes, and the results indicated that there were differences between 99mTc-CO-MIBI and Re-CO-MIBI in preparation and hydrophobic characteristics.  相似文献

Summary An optimum gas chromatographic separation of all protein amino acids in one run on capillaries coated with Chirasil-Val is difficult to achieve. Overlap of enantiomers of different amino acids may occur because the relative retention times depend upon the overall polarity of the stationary phase, the film thickness and the actual temperature programm. Employment of different derivatives formed by esterification with isopropanol, n-propanol, isobutanol and n-butanol and by acylation with trifluoroacetic, pentafluoropropionic and heptafluorobutyric anhydrides yields patterns of relative elution of all amino acid enantiomers. Thus, even critical pairs of amino acid enantiomers can be separated or shifted in their relative retention times. All amino acid enantiomers can be separated and quantitatively estimated.  相似文献

The enhanced choline uptake and phosphorylation in tumor cells has motivated the development of radiolabeled choline derivatives as diagnostic markers for imaging cell membrane proliferation and noninvasive detection of prostate, brain and breast tumors. In the present work, we report a facile strategy for the synthesis of choline functionalized macrocyclic chelating agent (DO3A-EA-choline) and its radiocomplexation with (67)Ga for potential tumor imaging applications. The synthesis of the desired compound featured quaternization of N,N-dimethylaminoethanol with 1,2-dibromoethane followed by subsequent alkylation with trisubstituted cyclen (DO3A). All intermediates and final compounds have been fully characterized by spectroscopic techniques, namely, (1)H, (13)C NMR and mass spectroscopy. The compound has been successively labeled with (67)Ga-citrate in ammonium acetate buffer (pH 6.5) at 80 °C. MTT assays have been performed on the HEK cell line to determine the cytotoxicity of the compound. Cell uptake studies carried out on the U-87 MG cell line exhibited saturable binding of the radioconjugate in picomolar range with a K(d) value of 0.528 pM. The in vivo biodistribution and blood kinetics studies exhibited rapid clearance of the radiolabeled complex and excretion through the renal and hepatobiliary route. The present studies demonstrate the potential applications of (67)Ga-DO3A-EA-choline as a radiopharmaceutical for molecular imaging using ((67/68)Ga) SPECT and PET modalities.  相似文献
