Indirect iodination of 2-chloro-nicotinonitrile gave 2-chloro-5-iodonicotinonitrile, which was cyclized with methylhydrazine to lead to 3-amino-5-iodopyrazolo[3,4-b]pyridine. Position 3 was then protected by pivaloyl group and the resulting 5-iodo-3-pivaloylaminopyrazolo[3,4-b]pyridine was engaged in palladium-promoted coupling reactions with various reagents to give 3-pivaloylamino-5-substituted compounds. Deprotection and iododediazoniation followed by cross-coupling reactions in position 3 afforded novel unsymmetrical 3,5-disubstituted pyrazolo[3,4-b]pyridine species.  相似文献

Various novel polycyclic thiopyranoindol annulated [3,4-c]quinolone derivatives were synthesized via domino Knoevenagel-hetero-Diels–Alder reactions of indoline-2-thions and novel N-acrylated anthranilaldehydes in refluxing ethanol as a solvent in the presence of 20 mol% ZnBr2 as a Lewis acid catalyst. All reactions proceed with high yields with excellent regio- and stereoselectivity.  相似文献

A new synthetic route has been developed for the preparation of indolo[3,2,1-d,e]phenanthridines and isochromeno[3,4-a]carbazoles via palladium catalyzed intramolecular biaryl coupling reactions. The coupling reactions proceeded smoothly and in high yields under ligand-free conditions with the catalytic system Pd(OAc)2/Cs2CO3/TBAB. Under optimized reaction conditions no halogen-reduced products were observed.  相似文献

A convenient route to novel 3-iodo-1H-pyrazolo[3,4-b]pyridines via iododediazonation of 3-amino-1H-pyrazolo[3,4-b]pyridines, which are obtained by copper-catalysed cyclisation of 2-chloro-3-cyanopyridine with hydrazines. We describe also efficient coupling reactions from 3-iodo derivatives with various reagents according to Suzuki, Heck, Stille, and Sonogashira conditions.  相似文献

A microwave-assisted chemoselective synthesis of novel pyrazolo[3,4-b]thieno[3,4-e]pyridines has been achieved via tandem Michael addition–cyclization–tautomerization–oxidative aromatization sequence of reactions. Compounds with ortho-substituted phenyl rings exhibited axial chirality due to restricted rotation around C–C single bond.  相似文献

An efficient synthesis of 12 pyrazolo[3,4-b]pyridin-6-one derivatives was achieved using either microwave or ultrasound irradiation, resulting in yields of 40-60% and 60-95%, respectively. Under our conditions, these reactions occurred with notably reduced reaction times as compared to literature reports involving traditional heating. Furthermore, these reactions were highly regioselective and produced only one pyrazolo[3,4-b]pyridin-6-one isomer, whose identity was confirmed by NOESY spectroscopy.  相似文献

3-Alkynylquinoxaline-2-carbonitriles have been synthesized from 3-chloroquinoxaline-2-carbonitrile via the Sonogashira reaction. Treatment of 3-alkynylquinoxaline-2-carbonitriles with an alkylamine or ammonia has been shown to produce stable enamines, e.g., (Z)-3-(2-aryl-2-aminovinyl)quinoxaline-2-carbonitriles. Their base-induced cyclization gave the previously unknown pyrido[3,4-b]quinoxalin-1(2H)-imines or pyrido[3,4-b]quinoxalin-1-amines. 3-Alkynylquinoxaline-2-carbonitriles were directly transformed into pyrido[3,4-b]quinoxalin-1(2H)-imines by heating with an alkylamine and K2CO3 in DMF. The ionization constants, and absorption and fluorescent properties of the resulted pyrido[3,4-b]quinoxalin-1(2H)-imines were measured.  相似文献

Phytochemical investigation on the stems of Trigonostemon flavidus resulted in the isolation of five new 3,4-seco-diterpenoids, trigoflavidones A-E (1-5), structurally related to the main co-occurring known 3,4-seco-sonderianic acid (6) and 3,4-seco-sonderianol (7). Compound 4 possesses new 3,4-seco rearranged ent-pimarane skeletal type, characteristic of a vinyl group at C-8, while 5 features a unique five-membered ring (C1) fused with a cyclopropane ring (C2). The structures of the new compounds were established by a combination of spectroscopic data and computational methods. Compounds 1-7 were tested for their cytotoxicities on HL-60, SMMC-7721, A-549, MCF-7, and SW480 human tumor cell lines.  相似文献

The synthesis of dipyrrolo[3,4-a:3,4-c]carbazole-1,3,4,6-tetraones, structurally related to granulatimide is reported. These compounds can be considered as granulatimide analogues in which a maleimide heterocycle replaces the imidazole moiety. The synthesis of pyridino[2,3-b]dipyrrolo[3,4-e:3,4-g]indole-1,3,4,6-tetraones is also reported. In these compounds, a 7-azaindole unit replaces the indole moiety present in the granulatimide and isogranulatimide structures.  相似文献

The 1,3-dipolar cycloaddition of unstabilized azomethine ylides with 2- and 3-nitroindoles furnishes the expected hexahydropyrrolo[3,4-b]indole cycloadducts in good to excellent yields. The cycloadducts can be denitrated with Bu3SnH/AIBN, and cycloadduct 5 was oxidized with MnO2 to yield the known pyrrolo[3,4-b]indole 13.  相似文献

The first convenient synthesis of 2-S-cysteinyl-3,4-dihydroxyphenylalanine (2-S-cysteinyldopa) in 30% overall yield is reported, which capitalizes on the anomalous regiochemistry of the oxidative coupling of 3,4-dihydroxycinnamic acid derivatives with thiol compounds, leading to 2-S rather than the usual 5-S conjugates.  相似文献

The syntheses of dipyrrolo[3,4-a:3,4-c]carbazole-1,4,6-triones and dipyrrolo[3,4-a:3,4-c]carbazole-3,4,6-triones are reported. These compounds can be considered as granulatimide analogues in which a maleimide replaces the imidazole moiety and a five-membered lactam ring replaces the upper maleimide. The Chk1 inhibitory properties of the more soluble compounds have been evaluated and their in vitro antiproliferative activities toward three tumor cell lines: murine leukemia L1210, and human colon carcinoma HT29 and HCT116. Due to their insolubility, the biological activities of the other compounds in this series could not be evaluated. All the tested compounds proved to be potent Chk1 inhibitors.  相似文献

The reaction of 1-methyl-2-nitromethylenepyrrolidine with arylisothiocyanates in the presence of a base was found to proceed at the 3-СН2 group of the nitroenamine followed by further cyclization to afford 4-R-imino-1-methyl-1,2,3,4-tetrahydro-6H-thieno[3,4-b]pyrrol-6-one oximes. Under analogous conditions, alkylisothiocyanates add to 1-methyl-2-nitromethylenepyrrolidine giving 1-methyl-2-nitromethylenepyrrolidine-3-carbothioic acid amides, and no cyclization was observed. A tentative mechanism for the formation of thieno[3,4-b]pyrrolе derivatives is proposed.  相似文献

Unexpected reactions between 1-α-aminobenzyl-2-naphthol, 1-aminomethyl-2-naphthol, N-benzyl-1-α-aminobenzyl-2-naphthol and 6,7-dimethoxy-3,4-dihydroisoquinoline to furnish naphth[1,2-e][1,3]oxazino[2,3-a]isoquinolines are reported. The reaction conditions involved classical heating at 80 °C in MeCN for 22 h (57-62%), or the use of microwave conditions (100 °C), which allowed a reduction of the reaction time to 90 min and resulted in somewhat higher yields (73-82%).  相似文献

Intramolecular base catalyzed C-C bond formation led to exclusive stereoselective syntheses of trans-6,7-dihydro-5H-[1,2,4]triazolo[3,4-b][1,3,4]thiadiazines, trans-7,8-dihydro-6H-[1,2,4]triazino[3,4-b][1,3,4]thiadiazin-4-ones, and trans-3,4-dihydro-2H-[1,3,4]thiadiazino[2,3-b]quinazolin-10-one. trans-6,7-Dihydro-5H-[1,2,4]triazolo[3,4-b][1,3,4]thiadiazines isomerize slowly in CDCl3 and more rapidly in DMSO-d6 into the corresponding cis-stereoisomers. The other trans-6,7-dihydro-[1,3,4]thiadiazines isomerize also in DMSO-d6 into the corresponding cis-stereoisomers. Pyrolytic conversion of these heterocyclic condensed dihydrothiadiazines into their corresponding pyrazolo[5,1-b][1,2,4]triazoles, pyrazolo[5,1-c][1,2,4]triazin-4-ones and pyrazolo[4,3-b]quinazolin-9-ones via desulfurization ring contraction is described.  相似文献

Dimethyl-1,2-diphenyl-3-methyl-cyclobutene-(1)-cis-3,4-dicarboxylate 2 leads in a thermal reaction to an equilibrium with (E, Z)-dimethyl-3,4-diphenyl-5-methyl-muconate (4). The equilibrium is shifted to the cyclic compound by pressure. Dimethyl-3,4-diphenyl-cyclobutene-(1,2-diphenyl-cyclobutene-(1)-cis-3,4-dicarboxylate (3) isomerizes thermally to (E, Z)-dimethyl-2,5-dimethyl-3,4-diphenylmuconate (6). Both reactions are accelerated by pressure. The activation volumes ΔV0+ are given for each ringopening reaction.  相似文献

Pyrazolo[3,4-d]pyrimidines represent an important class of heterocyclic compounds well-known for their anticancer activity exerted by the inhibition of eukaryotic protein kinases. Recently, pyrazolo[3,4-d]pyrimidines have become increasingly attractive for their potential antimicrobial properties. Here, we explored the activity of a library of in-house pyrazolo[3,4-d]pyrimidines, targeting human protein kinases, against Staphylococcus aureus and Escherichia coli and their interaction with ampicillin and kanamycin, representing important classes of clinically used antibiotics. Our results represent a first step towards the potential application of dual active pyrazolo[3,4-d]pyrimidine kinase inhibitors in the prevention and treatment of bacterial infections in cancer patients.  相似文献

The reactions of 3-formylchromones with alkyl isocyanides in dry dichloromethane at room temperature lead to new types of organic fluorophores (1Z)-3-(alkylimino)-1-[(chromone-3-yl)methylene]-1,3-dihydro-9H-furo[3,4-b]chromen-9-one, which exhibited strong blue emission in solution. The reactions involve a [4+1] cycloaddition followed by an activated electrophilic aromatic substitution at the furan ring and dehydration sequence.  相似文献
