The mononuclear palladium(II) (1) and platinum(II) (2) complexes containing phenylglycine have been synthesized and characterized by elemental analysis, IR spectra, and 1H NMR spectra. The structure of 1 was determined by X-ray diffractometry. The interaction between the complexes and fish sperm DNA (FS-DNA), adenosine-5′-triphosphate (ATP), and adenine (Ade) were investigated by UV absorption spectra, the interaction mode of the complex binding to DNA was studied by fluorescence spectra and viscometry. The results indicate that the two complexes have different binding affinities to DNA, complex 2 > complex 1. Gel electrophoresis assay demonstrates that the two complexes have the ability to cleave pBR322 plasmid DNA. Cytotoxicity experiments were carried out toward four different cancer cell lines, and 1 shows lower inhibitory efficiency than 2, consistent with the binding affinities towards DNA.  相似文献

We report the synthesis and full characterization for a series of cyclometallated complexes of Pt(II) and Pd(II) incorporating the fluxional trithiacrown ligand 1,4,7-trithiacyclononane ([9]aneS3). Reaction of [M(C insertion mark N)(micro-Cl)]2 (M = Pt(II), Pd(II); C insertion mark N = 2-phenylpyridinate (ppy) or 7,8-benzoquinolinate (bzq)) with [9]aneS3 followed by metathesis with NH4PF6 yields [M(C insertion mark N)([9]aneS3)](PF6). The complexes [M(C insertion mark P)([9]aneS3)](PF6) (M = Pt(II), Pd(II); Cinsertion markP = [CH2C6H4P(o-tolyl)2-C,P]-) were synthesized from their respective [Pt(C insertion mark P)(micro-Cl)]2 or [Pd(C insertion mark P)(micro-O2CCH3)]2 (C insertion mark P) starting materials. All five new complexes have been fully characterized by multinuclear NMR, IR and UV-Vis spectroscopies in addition to elemental analysis, cyclic voltammetry, and single-crystal structural determinations. As expected, the coordinated [9]aneS3 ligand shows fluxional behavior in its NMR spectra, resulting in a single 13C NMR resonance despite the asymmetric coordination environment of the cyclometallating ligand. Electrochemical studies reveal irreversible one-electron metal-centered oxidations for all Pt(II) complexes, but unusual two-electron reversible oxidations for the Pd(II) complexes of ppy and bzq. The X-ray crystal structures of each complex indicate an axial M-S interaction formed by the endodentate conformation of the [9]aneS3 ligand. The structure of [Pd(bzq)([9]aneS3)](PF6) exhibits disorder in the [9]aneS3 conformation indicating a rare exodentate conformation as the major contributor in the solid-state structure. DFT calculations on [Pt([9]aneS3)(ppy)](PF6) and [Pd([9]aneS3)(ppy)](PF6) indicate the HOMO for both complexes is primarily dz2 in character with a significant contribution from the phenyl ring of the ppy ligand and p orbital of the axial sulfur donor. In contrast, the calculated LUMO is primarily ppy pi* in character for [Pt([9]aneS3)(ppy)](PF6), but dx2-y2 in character for [Pd([9]aneS3)(ppy)](PF6).  相似文献

Summary A series of new complexes [RuL2(pdphen)]2+, where pdphen is the planar ligand pyridino[3,2-f] [1,7]phenanthroline and L = 2,2-bipyridine, phenanthroline, 2,9-dimethylphenanthroline or 5-nitrophenanthroline, were prepared and characterized. The binding of [RuL2-(pdphen)] 2+ to calf thymus DNA was investigated using absorption, fluorescence and circular dichroism (c.d.) spectroscopies. All of the complexes show absorption hypochromicity associated with binding to calf thymus DNA. [Ru(bipy)2pdphen]2+ and [Ru(phen)2pdphen]2+ also show fluorescence intensities and excited state life-time increases. The c.d. spectra of dialyzates from solutions of racemic complexes versus calf thymus DNA indicate enantioselectivity associated with binding to DNA.  相似文献

The reaction of Ni(dtc)(PR3)Cl (dtc=diethyldithiocarbamate, R=Ph or Bu) with HSC6H4Cl-4 or HSCH2C6H4Cl-4 and Et3N gave two types of complex. For PPh3, the products were [Ni(dtc)(μ-SC6H4Cl-4)]2 (1) and [Ni(dtc)(μ-SCH2C6H4Cl-4)]2 (2); whilst PBu3 gave Ni(dtc)(PBu3)(SC6H4Cl-4) (3). The structure of freshly prepared 3 was determined to be monomeric, as indicated by X-ray diffraction studies. However, at room temperature in solution, 3 was observed to slowly convert to 1. Structural identification of 1 and 2 and similar dimers, and structural identification of 3 and analogous monomers, were investigated by mass spectrometry. Electron impact mass spectrometry (EIMS) failed to confirm the proposed structures due to extensive decomposition in the mass spectrometer. In the electron impact (EI) mode, all complexes invariably decomposed to Ni(dtc)2; on the other hand, fast atom bombardment (FAB) ionisation gave the expected molecular ions for all compounds.  相似文献

Platinum(II) complexes with various selenones (L) having the general formula [PtL2Cl2] were prepared and characterized by elemental analysis and, IR and NMR (1H, 13C, and 77Se) spectroscopies. A decrease in the IR frequency of the >C=Se mode and an upfield shift in 13C NMR for the >C=Se resonance of selenones are consistent with their selenium coordination to platinum(II). The NMR data show that the complexes are stable in solution and do not undergo equilibration at 297 K. The geometrical structures of the complexes were predicted theoretically (with DFT method) using Gaussian09 program. DFT calculations predicted that the trans configurations were up to 1.7 kcal/mol more stable than the cis forms in gas phase, while in solution form the cis isomers were predicted to be more stable. The UV–vis spectra of the two complexes, 6 and 7 were also recorded at room temperature for 24 h and it was observed that the complexes were stable and did not undergo decomposition. The in vitro antitumor properties of the complexes as well as of cisplatin were evaluated on two human cancer cell lines, HeLa (cervical cancer cells) and MCF7 (breast cancer cells) using MTT assay. The results indicated that the prepared complexes exerted significant inhibition on the selected cancer cells.  相似文献

The synthesis of a new phthalonitrile derivative (4-(benzo[d]thiazol-2-ylthio)phthalonitrile), its metal-free phthalocyanine, and metallo-phthalocyanines are described. Phthalocyanines are formed in high yield with template synthesis. These compounds showed considerable solubility in CHCl3, DMF and DMSO, a primary condition for potential applications. The newly synthesized compounds have been characterized by elemental analyses, UV-Visible spectra, IR and 1H-NMR.  相似文献

Mixed ligand complexes having the formulae Cu(RPO)2Py2, Cu(RPO)2Im2 and Cu(DBO)2Py2 [RPO = resacetophenone oxime, DBO = 2,4-dihydroxybenzophenone oxime, Py = pyridine and Im = imidazole] have been synthesized and characterized by UV–Vis, IR, ESR, cyclic voltammetry and magnetic susceptibility methods. Absorption studies revealed that each of these octahedral complexes is an avid binder of calf thymus DNA. The apparent binding constants for mixed ligand complexes are in order of 104–105 M−1. Based on the data obtained in the DNA binding studies a partial intercalative mode of binding is suggested for these complexes. The nucleolytic cleavage activity of the adducts was carried out on double stranded pBR322 circular plasmid DNA by using a gel electrophoresis experiment in the presence and absence of oxidant (H2O2). All the metal complexes cleaved supercoiled DNA by hydrolytic and oxidative paths. The oxidative path dominates the hydrolytic cleavage. The hydrolytic cleavage of DNA is evidenced from the control experiments showing discernable cleavage inhibition in the presence of the hydroxyl radical inhibitor DMSO or the singlet oxygen quencher azide ion.  相似文献

Platinum(II) and palladium(II) complexes of the trithiacrown [9]aneS(3) containing a range of Group 15 donors are reviewed. These complexes have the general formula [M([9]aneS(3))(L(2))](n+) where L represents at least one Group 15 donor. Complexes involving pnictogens, with the exception of bismuth, are observed. The complexes generally have an elongated square pyramidal geometry with a long distance interaction to the third sulphur of the [9]aneS(3) which forms the apex of the square pyramid. This axial metal-sulphur distance is quite sensitive to the donor properties of L. Poorer donors such as Sb and As ligands show short axial distances whereas the better N donor ligands show longer distances. Pt(II) complexes of the formula [Pt([9]aneS(3))(EPh(3))(2)](2+) (E = P, As, Sb) show a considerable distortion towards a trigonal bipyramidal geometry due to intramolecular π-π interactions. Over seventy of these types of complexes have been crystallographically characterized and are discussed in this article. Other unique features of the complexes, including NMR spectroscopy, redox chemistry, and electronic spectroscopy, are also discussed.  相似文献

A series of di- and trithiosemicarbazone ligands as well as their Pd(II) and Pt(II) 1,3,5-triaza-7-phosphaadamantane (PTA) complexes have been synthesised using templated reactions between various substituted salicylaldimine thiosemicarbazone ligands and metal precursors of the general formula cis-[M(PTA)2Cl2], where M = Pd or Pt. Characterization of these complexes was achieved using various analytical and spectroscopic techniques: elemental analysis, ESI-MS, FT-IR, and NMR (1H, 13C{1H} and 31P{1H}) spectroscopy. The data revealed tridentate (O-N-S) coordination of the thiosemicarbazone moieties via the imine nitrogen, thiolato sulfur and phenolic oxygen to each metal center. In vitro biological evaluation of selected compounds was conducted against WHCO1 oesophageal cancer cells. Some of the multimeric compounds display some promising biological activity.  相似文献

Abstract

The lysozyme- and DNA-binding affinities of the biologically active cisplatin analogues, Pd(II) (1) and Pt(II) (2) complexes of functionalized N,N-pyridylbenzimidazole ligand, are reported. The electronic transitions were studied by time-dependent density functional theory. Complex 1 exhibits interesting antifungal activity against Candida albicans (MIC?=?32?μg?mL?1; 64?nM) and Cryptococcus neoformans (MIC?=?16?μg?mL?1, 32?nM). Complex 1 is covalently bound to DNA and lysozyme via the elimination of the chloride ligand(s). When complex 2 interacts with lysozyme, two adduct peaks are observed in the mass spectrum corresponding to binding of Pt(II) ion to lysozyme.  相似文献

The reaction of two equivalents of the functional phosphine ligand N-(diphenylphosphino)-1,3,4-thiadiazol-2-amine Ph2PNHC=NNCHS (2) with [PdCl2(NCPh)2] in the presence of NEt3 gives the neutral, P,N-chelated complex cis-[Pd(Ph2PN=CNN=CHS)2] ([Pd(2-H)2], 3b), which is analogous to the Pt(II) analogue cis-[Pt (Ph2PN=CNN=CHS)2] ([Pt(2-H)2], 3a) reported previously. These complexes function as chelating metalloligands when further coordinated to a metal through each of the CH-N atoms. In the resulting complexes, each endo-cyclic N donor of the thiadiazole rings is bonded to a different metal centre. Thus, the heterodinuclear palladium/platinum complexes cis-[Pt(Ph2PN=CNN=CHS)2PdCl2]([Pt(2-H)2·PdCl2], 4a) and cis-[Pd(Ph2PN=CNN=CHS)2PtCl2]([Pd(2-H)2·PtCl2], 4b) were obtained by reaction with [PdCl2(NCPh)2] and [PtCl2(NCPh)2], respectively. In contrast, reaction of 3a with [AuCl(tht)] occurred instead at the P-bound N atom, and afforded the platinum/digold complex cis-[Pt{Ph2PN(AuCl)=CNN=CHS}2] ([Pt(2-H)2(AuCl)2], 5). For comparison, reaction of 4a with HBF4 yielded cis-[Pt(Ph2PNH=CNN=CHS)2PdCl2](BF4)2([H24a](BF4)2, 6), in which the chelated PdCl2 moiety is retained. Complexes 3b, 4a·CH2Cl2, 4b·0.5C7H8, 5·4CHCl3 and 6 have been structurally characterized by X-ray diffraction.  相似文献

Novel Cu(II) (1) and Zn(II) (2) complexes with 4-(1-(4-morpholinophenyl)ethylideneamino)pyrimidine-5-carbonitrile) (L) have been synthesized and characterized by various spectroscopic and analytical techniques. DFT (density functional theory) studies result confirms that, LMCT mechanism have been done between L and M(II) ions. The antimicrobial studies indicate that the ligand L and complexes 1 & 2 exhibit higher activity against the E. coli bacteria and C. albicans fungi. The groove binding mode of ligand L and complexes 1 & 2 with CT-DNA have been confirmed by electronic absorption, competitive binding, viscometric and cyclic voltammetric studies. The electronic absorption titration of ligand L and complexes 1 & 2 with DNA have been carried out in different buffer solutions (pH 4.0, 7.0 & 10.0). The Kb values of ligand L and complexes 1 & 2 are higher in acidic buffer at pH 4.0 (Kb = 2.42 × 105, L; 2.8 × 105, 1; 2.65 × 105, 2) and the results revealed that, the interaction of synthesized compounds with DNA were higher in the acidic medium than basic and neutral medium. Furthermore, CT-DNA cleavage studies of ligand L and complexes 1 & 2 have been studied. The in vitro anticancer activities results show that complexes 1 & 2 have moderate cytotoxicity against cancer cell lines and low toxicity on normal cell line than ligand L.  相似文献

The absolute necessity to fight some class of tumor is perceived as serious health concerns, so the discovery and development of effective anticancer agents are urgently needed. (E)‐4‐((2‐hydroxyphenyl)diazenyl)‐3‐phenyl‐1H‐pyrazol‐5(4H)‐one, HL, and its Ni(II), Pd(II) and Pt(II) complexes were synthesized and the biological activity was evaluated for antitumor, antioxidant and antimicrobial activity as well as DNA cleavage. Their structures were assigned depending on the elemental analysis, conductivity, magnetic moment, spectral measurements (IR, 1HNMR, mass and UV–Vis) and thermal analysis. 3D molecular modeling using DFT method confirmed that the geometrical structures agree well with the suggested experimental ones. The antitumor activity was evaluated against four different cell lines using MTT assay. The ligand HL showed a potent cytotoxic activity compared to 5‐fluorouracil as a reference drug. For metal complexes, the order of activity was: Pd(II) > Ni(II) > Pt(II). A remarkable antioxidant activity for the ligand HL was recorded. It was higher than that of the metal complexes. Results of antimicrobial experiments revealed that all compounds were moderate to highly active against selected bacterial strains but inactive as antifungal except Pd(II) which showed a moderate antifungal activity. Gel electrophoresis showed insignificant nucleases activity for the ligand or its metal complexes even in the presence of H2O2 providing protection of DNA from damage. The antitumor activity of our compounds may be not due to DNA cleavage but may be referred to a mechanism similar to that of 5‐fluorouracil which interfere with DNA replication. The present work suggests the use of this ligand in the design and development of new anticancer drugs.  相似文献

Two novel Platinum(II) and Palladium(II) complexes of 2,2′-bipyridine (bpy) with ethyldithiocarbamate (Et-dtc) were synthesized. These complexes were characterized by spectroscopic methods such as ultraviolet-visible, infrared and 1H NMR as well as conductivity measurements and chemical analysis. In these complexes, the dithiocarbamate ligand coordinates with Pt(II) or Pd(II) center as bidentate with two sulfur atoms. These water soluble complexes were tested for their in vitro anti-tumor activity against chronic myelogenous leukemia cell line, K562. They show Cc50 values lower than those of cisplatin. The mode of binding of the complexes to calf thymus DNA, were investigated by circular dichroism, ultraviolet difference and fluorescence spectroscopy. These complexes can denature DNA at very low concentrations (~100 μM). Both complexes exhibit cooperative binding and presumably intercalate into DNA. Remarkably, most of the experimental results indicate that the tendency of the Pd(II) complex to interact with DNA and its anti-tumor activity against K562 is more than that of its Pt(II) analog.  相似文献

Four dinuclear cadmium(II) complexes, [Cd2(L1)(μ2-Cl)Cl2] (1), [Cd2(L2)(μ2-Cl)Cl2] (2), [Cd2(L3)(μ2-Cl)Cl2] (3), and [Cd2(L4)3ClO4] (4), where HL1 = 4-methyl-2,6-bis(1-(2-piperidinoethyl)iminomethyl)-phenol, HL2 = 4-methyl-2,6-bis(1-(2-pyrrolidinoethyl)iminomethyl)-phenol, HL3 = 4-methyl-2,6-bis(1-(2-morpholinoethyl)iminomethyl)-phenol and HL4 = 4-methyl-2,6-bis(cyclohexylmethyl)iminomethyl)-phenol, were synthesized. They were characterized by elemental analysis, FT-IR, UV–Vis, fluorescence and electronspray ionization mass spectroscopy. Complexes 1 and 4 were also characterized by single crystal X-ray analysis. The cadmiums atoms in 1 are linked by μ2-chloride in a distorted square pyramidal geometry, whereas cadmium atom in 4 is in a distorted octahedral environment. The complexes show emission bands around 500 nm with excitation at 395 nm.  相似文献

Synthetic methods toward ruthenium(II) complexes incorporating the benzo[i]dipyrido[3,2-a:2',3'-c]phenazine-11,16-quinone ligand, qdppn, are reported. In several cases, it was found that complexes containing coordinated benzo[i]dipyrido[3,2-a:2',3'-c]phenazine, dppn, could be chemically or photochemically oxidized to their qdppn analogues. Since this method was not possible in all the cases, a new, higher yielding, convenient synthesis of qdppn was developed. The crystal structure of the complex [Ru(phen)(2)(qppn)](PF(6))(2) (phen = 1,10-phenanthroline) which was synthesized from free qdppn reveals that a combination of π-π stacking between coordinated phen and qdppn units, as well as anion-ligand hydrogen bonding, define large hexagonal channels which are occupied by anions and solvent molecules. Electrochemical and photophysical studies reveal that the new qdppn-based complexes are not luminescent and, in contrast to their dppn analogues, they are also poor singlet oxygen sensitizers. Time-resolved studies and density functional theory (DFT) calculations indicate that optical properties of the new complexes are due to a short-lived charge separated state involving the quinone moiety of qdppn. The DNA binding properties of the new complexes have also been investigated. It was found that they are intercalators, displaying binding affinities which are comparable to their dppn analogues.  相似文献

The thioethers 4-tert-butyl-2,6-bis((2-(dimethylamino)ethylimino)methyl)phenyl(tert-butyl)sulfane (tBu-L3) and 4-tert-butyl-2,6-bis((2-(dimethylamino)ethylimino)methyl)phenyl(tert-butyl)sulfane (tBu-L4) react with PdCl2(NCMe)2 to give the dinuclear palladium thiophenolate complexes [(L3)Pd2Cl2]+ (2) and [(L4Pd2(mu-Cl)]2+ (3) (HL3= 2,6-bis((2-(dimethylamino)ethylimino)methyl)-4-tert-butylbenzenethiol, HL4 = 2,6-bis((2-(dimethylamino)ethylamino)methyl)-4-tert-butylbenzenethiol). The chloride ligands in could be replaced by neutral (NCMe) and anionic ligands (NCS-, N3-, CN-, OAc-) to give the diamagnetic Pd(II) complexes [(L3)Pd2(NCMe)2]3+ (4), [(L3)Pd2(NCS)2]+ (5), [(L3)Pd2(N3)2]+ (6), [{(L3)Pd2(mu-CN)}2]4+ (7) and [(L3)Pd2(OAc)]2+ (9). The nitrile ligands in and in [(L3)Pd2(NCCH2Cl)2]3+ are readily hydrated to give the corresponding amidato complexes [(L3)Pd2(CH3CONH)]2+ (8) and [(L3)Pd2(CH2ClCONH)]2+ (10). The reaction of [(L3)Pd2(NCMe)2]3+ with NaBPh4 gave the diphenyl complex [(L3)Pd2(Ph)2]+ (11). All complexes were either isolated as perchlorate or tetraphenylborate salts and studied by IR, 1H and 13C NMR spectroscopy. In addition, complexes 2[ClO4], 3[ClO4]2, 5[BPh4], 6[BPh4], 7[ClO4]4, 9[ClO4]2, 10[ClO4]2 and 11[BPh4] have been characterized by X-ray crystallography.  相似文献
