A high performance liquid chromatographic (HPLC) method for analysis of 4 free and 8 conjugated bile acids in submicromolar quantities in serum is described using precolumn derivatization with 4-bromomethyl-7-methoxycoumarin (BMC) and fluorescence detection. Bile acids were extracted from serum with 0.4 M sodium bicarbonate, adsorbed onto a Sep-Pak C18 cartridge and eluted with methanol. The extract was derivatized with BMC in acetonitrile using 18-crown-6 crown ether as catalyst and the BMC labelled glycine conjugates and free bile acids were analysed using acetonitrile + methanol + water gradient elution and detection at 320/385 nm. Using a novel and simple approach, taurine conjugates were isolated by extracting the dried, derivatized material with water, in contrast to previous methods which required column chromatography cleanup to isolate the taurine conjugates prior to derivatization. The isolated taurine conjugates were then hydrolysed enzymatically, extracted, derivatized and analysed as free-bile acids. Recoveries of individual bile acids varied from 83-96% for free and glycine conjugates and 72-83% for taurine conjugates. Coefficients of variation were in the range of 5.1-12.5%. In addition to the simpler and shorter procedure for taurine conjugates, this method has increased sensitivity over most other procedures and improved HPLC separation for the various bile acids and conjugates with equivalent recovery and reproducibility compared with other published methods.  相似文献

A chromatographic procedure for the preparative isolation of six different 6-alkylsalicylic acids (syn. ginkgolic acids) with as alkyl substituents C13:0, C15:0, C15:1, C17:1, C17:2 and, tentatively C17:3 from Ginkgo biloba leaves was developed. The procedure consisted of a combination of normal-phase, reversed-phase and argentation chromatography. The compounds were characterised by means of UV, 1H-NMR and 13C-NMR spectroscopy, and mass spectrometry after silylation. A 15 cm C18 RP-HPLC column connected in series with a 20 cm silver(I) loaded cation exchanger HPLC column in combination with the solvent methanol–water (93:7) acidified with 0.1% formic acid was capable of separating the ginkgolic acids C13:0, C15:1, C17:2, C15:0 and C17:1 within 21 min on an analytical scale. The separation is based on a combination of reversed-phase mechanisms and double bond complexation. Detection took place by UV at 311 nm. The separation is a good starting point for the development of a quantitative procedure for the five major ginkgolic acids in Ginkgo leaves and standardised extracts.  相似文献

Summary This report describes a modified method for the separation and analysis of polyunsaturated fatty acids such as 20:4, 20:5, and 22:6, using HPLC. The results show that these fatty acids are well separated from the saturated acids. Since the unsaturated fatty acids elute earlier than saturated acids, and this method does not require the fractionation of free fatty acids using thin layer chromatography, a necessary step for the gas chromatographic analysis, the recoveries of polyunsaturated fatty acids were significantly higher as compared to those from gas chromatography. Furthermore, HPLC and gas chromatographic methods gave identical results for the acyl chain composition of phosphatidylserine. The advantages of using HPLC over gas chromatography in determining the acyl chain composition of free fatty acids and phospholipids are also discussed.  相似文献

The main constituents of plant oils are complex mixtures of TGs differing in acyl chain lengths, number and positions of double bonds, and regioisomerism. A non-aqueous reversed-phase HPLC method with acetonitrile-2-propanol gradient and 30 + 15 cm NovaPak C18 columns makes possible an unambiguous identification of the highest number of TGs ever reported for these oils, based on positive-ion APCI mass spectra. A new approach to TG quantitation is based on the use of response factors with three typical detection techniques for that purpose (APCI-MS, evaporative light-scattering detection, and UV at 205 nm). Response factors of 23 single-acid TGs (saturated TGs from C7 to C22, 7 unsaturated TGs), 4 mixed-acid TGs, diolein and monoolein are calculated from their calibration curves and related to OOO. Due to differences between saturated and unsaturated acyl chains, the use of response factors significantly improves the quantitation of TGs. 133 TGs containing 22 fatty acids with 8-25 carbon atoms and 0-3 double bonds are identified and quantified in 9 plant oils (walnut, hazelnut, cashew nut, almond, poppy seed, yellow melon, mango, fig, date) using HPLC/APCI-MS with a response factor approach. Average parameters and relative fatty acid concentrations are calculated with both HPLC/APCI-MS and GC/ FID.  相似文献

A parallel library of 108 4-aryl-1,4-dihydropyrimidine (DHPM) enantiomers, which are potential selectors for chiral HPLC separations, was synthesized using the single-step Biginelli multicomponent condensation. The individual compounds were screened by observing the enantioselectivity for resolution on a "brush-type" L-(3,5-dinitrobenzoyl)leucine-based chiral stationary phase, and separation factors alpha up to 12 were achieved. The best candidates from the library contained an ortho-substituted aromatic group at C4 carbon atom of the pyrimidine ring and an alkyl substituent at N1 nitrogen atom. Resolution of the enantiomers of the lead compound, 4-(9-phenanthryl)-DHPM 8, using semipreparative chiral HPLC followed by attachment to monodisperse macroporous aminomethacrylate beads, provided the novel polymer based chiral stationary phase with good enantioselectivities in the resolution of several pi-acidic aryl-dihydropyrimidines and derivatized profens. In addition, 3,5-dinitrobenzamido derivatives of alpha-amino acids could be resolved under normal phase HPLC conditions with separation factors up to 8.  相似文献

A method for the determination of individual free and conjugated bile acids in serum using microcolumn liquid chromatography coupled with a laser-induced fluorescence detector is described. Bile acids are separated into free/glycine-conjugate and taurine-conjugate fractions using a Sep-Pak SIL cartridge. The taurine-conjugated bile acid fraction is subjected to enzymatic hydrolysis. Subsequently, free and conjugated bile acids are labeled using 4-(bromomethyl)-7-methoxycoumarin as a fluorogenic reagent, producing stable derivatives that can be excited by the 325 nm line of a He/Cd laser. Prior to their fluorimetric detection, the individual components of a bile acid serum profile are separated by reversed-phase microcolumn liquid chromatography.  相似文献

A HPLC-fluorescence method for identification of drying oils from binding media or protective film used in pictorial works of art prior to conservation or restoration is proposed. Fluorescence derivatization of fatty acids released by hydrolysis of structural drying oils is studied. The derivatization reagent was 4-(bromomethyl)-7-methoxycoumarin with 18-crown-6 as catalyst. Mobile phase was programmed from methanol-water (90:10 v/v) to methanol-water (100:0 v/v) in 25 min. The excitation and emission wavelengths were 325 and 395 nm, respectively. Under these chromatographic conditions, coumarin derivatives of myristic, palmitic, oleic and stearic acids were satisfactorily resolved. The method shows good sensitivity, with a detection limit of 6.0 x 10(-8) mmol, and good linearity between 1.0 x 10(-7) and 1.8 x 10(-4) mmol of each analyte. Peak area ratios among fatty acids derivatives, especially the stearic acid/palmitic acid peak area ratio, are useful to identify the drying oils. The proposed method has been successfully applied to artistic samples from items of the cultural heritage of Valencia (Spain).  相似文献

A combined system of flow injection on-line dialysis sample pretreatment and high performance liquid chromatographic separation/detection (FID-HPLC) was developed for simultaneous determination of six organic acids (tartaric, malic, lactic, acetic, citric and succinic acids). A sample or mixed standard solution (400 μL) was injected into a donor stream (water) of FID system and was pushed further through a dialysis cell, while an acceptor solution (water) was held in the opposite side of the dialysis membrane. The dialysate containing organic acids in the acceptor solution was then flowed to an injection loop of the HPLC valve, where it was further injected into the HPLC system and analysed under normal HPLC conditions, using a reversed-phase (C18) analytical column and UV detection (210 nm). The order of elution was tartaric, malic, lactic, acetic, citric and succinic acids with the analysis time of 8 min. The FID system could be operated in parallel with HPLC separation, providing sample throughput of 7.5 h−1. Dialysis efficiencies of six organic acids were in range of 4.6-9.5%. Calibration graphs for all the mentioned organic acids were linear over the range of 250-7500 mg L−1. Precisions for all the organic acids were within 5.4%. The proposed system was successfully applied for analysis of some Thai wines. By spiking wine samples with mixed acid standard solutions, the percentage recoveries in range of 84-104 were found. This system has advantages of fast and high degrees of automation for dialysis sample pretreatment, on-line sample separation and dilution, good clean-up for prolongation of life-time of the HPLC column and low consumption of chemicals and materials.  相似文献

The separation of fatty acids derivatised with Nile Blue (NB) by CE with detection using a red light-emitting diode (LED) was examined. NB was selected as the derivatisation agent due to its high molar absorption coefficient of 76,000 M(-1) cm(-1) at 633 nm, making it well suited for sensitive absorbance detection using a red 635 nm LED. NB-labelled fatty acids were separated by both MEKC using SDS micelles, i-PrOH and n-BuOH and by NACE in a number of solvents including MeOH, EtOH and ACN. The sensitivity of NACE was superior to MEKC, with detection limits of 5x10(-7)-7x10(-7) M obtained for each acid, approximately 20 times lower than the MEKC method. The NACE detection limits are approximately 100 times lower than previous reports on the separation of fatty acids by CE using indirect absorbance detection, ten times lower than using indirect fluorescence detection and are inferior only to those obtained using precapillary derivatisation and direct fluorescence detection. The efficiency of the NACE method was also superior to MEKC and allowed the separation of unsaturated fatty acids to be examined, although it was not possible to baseline-resolve linoleic (C18:2) and linolenic (C18:3) acids in a reasonable time. The method was used to analyse the fatty acid profile of two edible oils, namely sunflower and sesame oils, after alkali hydrolysis, where it was possible to identify both the saturated and unsaturated fatty acids in each sample.  相似文献

Naphthenic acids are a complex mixture of carboxylic acids with the general formula CnH(2n+Z)O2 and they are natural, toxic components of crude oils. GC-MS analyses of tert-butyldimethylsilyl esters of naphthenic acids are used to estimate component distribution within naphthenic acids mixtures. Our evaluations of the GC-MS method showed that ions from column bleed erroneously appear as C14 Z = -4 acids and that correcting for heavy isotopes of C and Si do not significantly affect ion distribution plots. Overall, the GC-MS method appears to overestimate the relative proportion of low-molecular-mass acids.  相似文献

Eicosapentaenoic and docosahexaenoic acids are important bio‐active fatty acids in fish oils. Monolithic HPLC columns both in the polymeric cation exchange (silver‐ion) and RP formats were compared with corresponding packed columns for the isolation of these acids from tuna oil ethyl esters. Monolithic columns in both formats enabled rapid (typically 5–10 min) separations compared with packed columns (30 min). Polymeric monolithic silver‐ion disc column rapidly furnished mixtures of eicosapentaenoic and docosahexaenoic esters (90% purity) within 5–10 min, but was unable to resolve individual esters. A preparative version of the same column (80 mL bed volume) enabled isolation (>88% purity) of 100 mg quantities of eicosapentaenoic and docosahexaenoic esters from esterified tuna oil within 6 min. Baseline separation of eicosapentaenoic and docosahexaenoic esters was achieved on all RP columns. The results show that there is potential to use polymeric monolithic cation exchange columns for scaled‐up preparation of eicosapentaenoic and docosahexaenoic ester concentrates from fish oils.  相似文献

Ethanolamides of polyunsaturated fatty acids (9Z,12Z,15Z-octadecatrienoic; 5Z,8Z,11Z,14Z,17Z-eicosapentaenoic; and 4Z,7Z,10Z,13Z,16Z,19Z-docosahexaenoic) and mixtures of ethanolamides of fatty acids from natural fats (oils) were synthesized. It was found that the polyunsaturated fatty acids did not isomerize under the proposed conditions for aminolysis of the ester bond. Conditions for analyzing the complicated mixtures of ethanolamides of fatty acids by HPLC-MS were determined.  相似文献

This study presents a new HPLC method for the simultaneous determination of seven major components, namely chlorogenic acid, caffeic acid, loganin, sweroside, secoxyloganin, rutin and luteolin 7-O-glucoside in Caulis Lonicerae Japonicae, a commonly used traditional Chinese medicinal herb derived from the caulis of Lonicera japonica Thunb. These seven compounds, belonging to the chemical types of phenolic acids, iridoids and flavonoids, were separated on a C18 column (250 x 4.6 mm, 5.0 microm) with the column temperature at 30 degrees C. The mobile phase was composed of (A) aqueous acetic acid (0.4%, v/v) and (B) acetonitrile using a gradient elution of 10% B at 0-12 min, 10-17% B at 12-25 min and 17% B at 25-35 min. The flow rate was 1.0 mL/min and detection wavelength was set at 245 nm. The limit of detection (S/N = 3) ranged from 0.10 to 0.23 microg/mL and the limit of quantification (S/N = 10) ranged from 0.69 to 3.56 microg/mL. All calibration curves showed good linear regression (r2 > 0.9990) within the test ranges. The intra- and inter-day precisions as determined from sample solutions were below 1.24 and 2.28%, respectively. The recoveries for seven compounds were found to range from 94.2 to 103.6%. This verified method has been successfully applied to evaluation of commercial samples of Caulis Lonicerae Japonicae from different markets in China.  相似文献

A method has been developed for separation of linolenic acid and its seven isomers by HPLC on a silver-ion-loaded column. The standard 18:3 isomers, isolated from a heated linseed oil or prepared by isomerization of linolenic acid, were converted into phenacyl esters and detected by UV at 238 nm. The use of low temperature (10 °C) combined with a gradient of dichloromethane and methanol enabled separation of all the cis/trans isomers. The peaks were identified by comparison of ECL values with those of a standard mixture, by chromatographing collected HPLC fractions on a polar GC column. HPLC quantification was compared with GC analysis. There was satisfactory agreement between the tow methods. This method could be used for seperation, collection and quantification of 18:3 fatty acids with trans double bonds in different oils and foods.  相似文献

A gas chromatographic method was developed for the determination of low-molecular-weight carboxylic acids in aqueous samples based on a derivatization procedure compatible with aqueous solutions. The technique uses nitrogen-selective detection with a thermionic-specific detector after derivatization of carboxylic acids as 2- nitrophenylhydrazides. The hydrazides were extracted with ethyl acetate prior to injection into the gas chromatograph. The derivatives appear to be stable in ethyl acetate at 0–5° C for long periods and, therefore, can be stored for analysis at a later date. The detection limits of different short-chain acids are in the range 0.8–1.4 pmol per injected sample. The relative standard deviation is less than 10% at the 1 μM level. Examples of the use of the method are given for the determination of carboxylic acids in anoxic marine sediment pore waters, coastal sea water and Black Sea water samples.  相似文献

Oleanolic acid (OA) and ursolic acid (UA) are the ubiquitous triterpenic acids in plant kingdom and have multiple biological effects. In the present study, a highly sensitive and selective method using a dual-sensitive probe 2-(5-benzoacridine)ethyl-p-toluenesulfonate (BAETS) as pre-column labeling reagent has been developed for rapid determination of the triterpenes OA and UA by HPLC with fluorescence detection (FLD) and online mass spectrometry identification. Response surface methodology as an efficient tool was employed to optimize the ultrasonic-assisted extraction of triterpenic acids from Swertia plants and the pre-column derivatization reaction, respectively, which ensured the highest triterpenic acids recoveries within the shortest extraction time and the sufficient labeling of the analyzed components. Fast separation of the isomers OA and UA could be achieved on a Hypersil BDS C8 column within 7 min. Both of OA and UA gave the good correlation coefficients of 0.9999. This developed method offered the satisfactory detection limits of 1.10 and 1.30 ng mL−1 for UA and OA, respectively. When applied to Swertia species, it showed good reproducibility.  相似文献

Naphthenic acids are found in wastewaters from petroleum refineries and oil sands extraction plants. Currently, the concentrations of these toxic carboxylic acids are determined by extracting them into methylene chloride and measuring the absorption of the carboxyl group by Fourier-transform infrared (FTIR) spectroscopy. An improved HPLC method, that is simpler and faster than the FTIR method, was used to detect the 2-nitrophenylhydrazides of the naphthenic acids at concentrations as low as 5 mg l(-1). Analyses of 58 oil sands water samples showed that the naphthenic acids concentrations determined by FTIR were on average 11% higher than those determined by HPLC.  相似文献

Crude oil contains such an extensive range of compounds that a complete analysis is impossible. Fractionation by chemical properties is often used to simplify analytical handling. This work presents a high performance liquid chromatography (HPLC) method using normal phase chromatography on a cyano-bonded phase column to separate acid extracts from crude oils into four fractions; non-polar compounds, saturated carboxylic acids, phenols and polyfunctional acids. The method has been developed both in analytical scale for characterisation of acid extracts, and in preparative scale to provide sufficient sample amounts for further analysis by complementary methods.  相似文献

A new triflate-type fluorescence chiral derivatizing reagent, (S)-(+)-1-methyl-2-(6,7-dimethoxy-2,3-naphthalimido)ethyl trifluoromethanesulfonate, [S-(+)-MDNE-OTf], has been developed for the determination of the enantiomers of carboxylic acids. By introducing the two methoxy groups on the naphthalimido ring moiety, the red shift in the fluorescence spectrum and a high resolution in reversed-mode separation of the diastereomers of chiral carboxylic acids have been achieved. The detection limits (S/N=3) with ultraviolet and fluorescence detection are 8 fmol (λmax=283 nm) and 4 fmol (λex=283 nm, λem=467 nm), respectively.  相似文献
