This paper describes two simple, specific, accurate, and precise methods for estimation of olopatadine hydrochloride (OLO) in tablet dosage form. The first method is a stability-indicating isocratic RP-HPLC method. The analysis is performed on an RP-18 column using 0.1% orthophosphoric acid (adjusted to pH 4.5 with triethylamine)-acetonitrile (75 + 25, v/v) mobile phase at a flow rate of 1 mL/min. Paracetamol (PAR) was selected as the internal standard. Retention times of OLO and PAR were 11.30 +/- 0.02 and 4.70 +/- 0.03 min, respectively. For the HPTLC method, precoated silica gel 60 F254 aluminum sheets were used as the stationary phase; the mobile phase was methanol-chloroform-ammonia (8 + 2 + 0.1, v/v/v). The detection of the analyte band was carried out at 301 nm, and its Rf value was 0.46 +/- 0.03. The analytical methods were validated according to International Conference on Harmonization guidelines. Linear regression analysis data for the calibration plots showed a good linear relationship between response and concentration in the range of 0.1-1 microg/mL and 0.1-0.9 microg/band for HPLC and HPTLC, respectively.  相似文献

A simple, rapid, sensitive and specific liquid chromatography/electrospray ionization mass spectrometry method was developed and validated to quantify azithromycin in human plasma, using erythromycin as the internal standard (IS). A simple sample preparation method of protein precipitation with methanol was employed. Methanol, acetonitrile and water (12:30:58, v/v/v) were used as the isocratic mobile phase, with 0.1% formic acid and 0.1% ammonium acetate in water. Selected ion monitoring was specific for azithromycin and erythromycin. The assay was linear over the concentration range 4.69-600 ng/mL. The correlation coefficients for the calibration curves ranged from 0.9994 to 0.9998. The intra- and inter-day precisions, calculated from quality control samples, were less than 8.24%. The method was employed in a pharmacokinetic study after oral administration of 500 mg azithromycin dispersible tablet to 20 healthy volunteers.  相似文献

建立了蜂产品中10种头孢类药物(头孢喹肟、头孢噻肟、头孢洛宁、头孢哌酮、头孢匹林、头孢氨苄、头孢乙腈、头孢拉定、去乙酰基头孢匹林、头孢唑林)含量的液相色谱-串联质谱测定方法。蜂产品样品中头孢类药物用乙腈-水(80:20,v/v)溶液提取,离心,上清液经Oasis PRIME HLB固相萃取柱净化,氮吹后复溶,进行液相色谱-串联质谱分析。采用Acquity BEH C18色谱柱,以0.1%(v/v)甲酸水溶液-甲醇体系作为流动相进行梯度洗脱,ESI源正离子模式电离,多反应离子监测模式(MRM)检测,基质校准外标法定量。结果表明,10种头孢类药物在一定浓度范围内峰面积与质量浓度的相关系数(r2)大于0.999,线性关系良好;检出限为0.15~1.5 μg/kg,定量限为0.50~5.0 μg/kg;在阴性蜂产品样品中的加标回收率为75.0%~89.8%,相对标准偏差(RSD)为1.4%~4.6%(n=5)。该方法检测周期短,准确度和精密度高,能满足多种蜂产品样品中头孢类药物的检测需要。  相似文献

A new HPLC method for the determination of geniposide in rat serum with solid-phase extraction (SPE) for preconcentration is described. Geniposide and an internal standard (paeoniflorin) were extracted from serum by SPE using C18 cartridges. Analysis of the extract was then performed on a reversed-phase C18 column using acetonitrile-water (16:84, v/v) as the eluting solvent system, and UV detection at 238 nm was used to measure the analyte with a limit of quantitation about 0.1 microg/mL. The calibration curve for geniposide was linear (r = 0.9993) in the concentration range 0.1-16.0 microg/mL. The intra- and inter-day precision of the geniposide were determined and their RSD did not exceed 10%. The validated method has been successfully applied for pharmacokinetic studies of geniposide from rat serum after oral administration of Yin-Zhi-Ku decoction.  相似文献

A fast and selective HPLC method for assaying amphotericin B in biological samples was developed and validated. The chromatographic separation was achieved in less than 12 min on a reverse-phase C(18) column using an acetonitrile-acetic acid-water (52:4.3:43.7, v/v/v) mixture as mobile phase. The flow rate was 1 mL/min and the effluent was monitored at 406 nm. A linear response over the concentration range 0.1-10.0 microg/mL was obtained. Intra-day and inter-day RSDs were below 5% for all the sample types. This new HPLC method was applied to assay amphotericin B in plasma and several tissue samples such as kidney, liver, spleen and bone marrow. Application of this method to pharmacokinetic studies in mice and dog is provided.  相似文献

建立了高效液相色谱-三重四极杆质谱(HPLC-MS/MS)测定克氏原螯虾中二甲戊灵残留的分析方法。用含0.1%(v/v)乙酸的乙酸乙酯溶液提取克氏原螯虾中的二甲戊灵,于35℃旋蒸至干,经含0.1%(v/v)乙酸的甲醇-水(8∶2,v/v)溶解残渣后,用酸性氧化铝柱、石墨化炭黑(GCB)进行净化。采用Symmetry C18色谱柱(100 mm×2.1 mm,3.5 μm)进行分离,用加热大气压电喷雾电离(HESI)源、正离子模式进行扫描,在多反应监测模式(MRM)下检测。结果表明,二甲戊灵在1.0~20.0 μg/L范围内线性良好,相关系数为0.9960,检出限为0.2 μg/kg;二甲戊灵的加标回收率为63.3%~104.7%,精密度为6.9%~14.5%(n=7)。该方法简单、快速、灵敏度高,能够满足克氏原螯虾中二甲戊灵药物残留检测的需求。  相似文献

建立了改良的QuEChERS结合高效液相色谱-串联质谱(HPLC-MS/MS)同时测定水产品中7种阿维菌素类药物(阿维菌素、伊维菌素、多拉菌素、塞拉菌素、乙酰氨基阿维菌素、莫西菌素和埃玛菌素)的分析方法。样品经0.2%(v/v)氨化乙腈提取,3 g无水硫酸镁和2 g无水硫酸钠除水剂和沉淀蛋白质,100 mg十八烷基硅烷(C18)和500 mg无水硫酸镁净化。以0.1%(v/v)甲酸乙腈(含5 mmol/L乙酸铵)-0.1%(v/v)甲酸水溶液(含5 mmol/L乙酸铵)为流动相,采用Varian Pursuit ULTRA C8色谱柱(100 mm×2.0 mm,2.8μm)进行分离。在加热电喷雾离子(HESI)源、正离子模式下采用多反应监测模式检测,基质匹配标准曲线外标法定量。阿维菌素、伊维菌素、多拉菌素、塞拉菌素、乙酰氨基阿维菌素和莫西菌素在2~200μg/L范围内、埃玛菌素在0.2~20μg/L范围内呈线性相关,相关系数(r)均≥0.997 2,水产品中阿维菌素类药物的加标回收率为71.6%~112.8%,相对标准偏差(RSD)为4.7%~13.1%,不同水产品的基质效应均小于15%。阿维菌素、伊维菌素、多拉菌素、塞拉菌素、乙酰氨基阿维菌素和莫西菌素的定量限均为5μg/kg,埃玛菌素的定量限为0.25μg/kg。该法操作简便,重复性好,适用于水产品中7种阿维菌素类药物残留量的同时测定。  相似文献

A sensitive, rapid and specific quantitative liquid chromatography/tandem mass spectrometry (LC/MS/MS) method was developed and validated for the determination of apomorphine (APO) in canine plasma. The analytes were prepared using one-step liquid-liquid extraction, and analyzed on a Waters Symmetry C(18) column interfaced with triple quadrupole tandem mass spectrometer. A mixture of methanol/0.1% formic acid in water (70: 30, v/v) was employed as the isocratic mobile phase. Positive electrospray ionization was utilized as the ionization source. The analyte and clenbuterol (internal standard) were both detected using multiple reaction monitoring (MRM) mode. The limit of detection (LOD) obtained was 0.03 ng/mL. The assay was linear over the concentration range of 0.1-100 ng/mL, and provided good precision (RSD) and good accuracy (RE). The analyte was stable by using antioxidants throughout the whole study. The experimental results show that LC/MS/MS is a rapid and sensitive method to analyze APO in plasma. Finally, the proposed method was successfully applied to a pharmacokinetic study of APO after intranasal administration of 0.5 mg apomorphine to 10 healthy beagle dogs.  相似文献

A simple, sensitive and rapid liquid chromatography–tandem mass spectrometry (LC‐MS/MS) method was developed and validated for the determination of calceorioside B (CLB) in rat plasma. Detection was performed on a Thermo Scientific Hypersil Gold chromatography column using isocratic elution with a mobile phase of methanol–5 m m ammonium acetate–formic acid (70:30:0.1, v/v/v). Mass spectrometry was performed in selection reaction monitoring mode using a positive electrospray ionization interface. Good linearity was found for CLB in plasma in the linear range of 1.00–500 ng/mL (r > 0.9960). The validated method was successfully applied to the pharmacokinetic study of CLB in rats. A high-performance liquid chromatographic method was optimized and validated for the determination of desacetyl nitazoxanide (tizoxanide), the main active metabolite of nitazoxanide in human plasma, urine and breast milk. The proposed method used a CN column with mobile phase consisting of acetonitrile-12mM ammonium acetate-diethylamine in the ratio of 30:70:0.1 (v/v/v) and buffered at pH 4.0 with acetic acid, with a flow rate of 1.5 mL/min. Quantitation was achieved with UV detection at 260 nm using nifuroxazide as internal standard. A simplified direct injection of urine samples without extraction in addition to the urinary excretion pattern were calculated using the proposed method. Also, the effectiveness of protein precipitation and a clean-up procedure were investigated for biological plasma and human breast milk samples. The validation study of the proposed method was successfully carried out in an assay range between 0.2 and 20 μg/mL.  相似文献

建立了高效液相色谱-串联质谱(HPLC-MS/MS)检测蔬菜水果中双甲脒(AMZ)及其降解或代谢产物单甲脒(DMPF)、2,4-二甲基苯基甲酰胺(DMF)和2,4-二甲基苯胺(DMA)的分析方法。样品经0.1 mol/L NaOH溶液稀释、正己烷-异丙醇(2:1,v/v)混合提取液提取,然后采用HPLC-MS/MS对目标物进行定性确证和定量分析。采用Phenomenex Kinetex C18色谱柱(100 mm×4.6 mm,2.6 μm)分离,以0.1%(v/v)甲酸水溶液和甲醇为流动相进行梯度洗脱,采用电喷雾电离(ESI)源、正离子监测模式进行检测。在1.0~200.0 μg/L范围内,4种目标化合物呈良好的线性关系,相关系数(r)大于0.99;方法的定量限为0.01~0.4 μg/kg。4种目标化合物在0.5、5.0和20.0 μg/kg 3个水平下的加标回收率为62.5%~105.0%,RSD为7.5%~17.6%。该法简便快捷,准确可靠,灵敏度高,定量限满足目前国内外残留限量要求。  相似文献

The analyte and rutin, used as the internal standard, were analyzed on a Phenomenex Synergi Fusion RP column interfaced with a triple-quadrupole tandem mass spectrometer using positive electrospray ionization. Acetonitrile/water (20:80 v/v) was used as the isocratic mobile phase, with 0.1% formic acid in water. A simple sample preparation method of protein precipitation with perchloric acid was employed. The assay was linear over the concentration range 31.25-4000 pg/mL. Correlation coefficients generated by linear regression with a 1/x(2) weighting factor ranged from 0.9991 to 0.9996. The intra- and inter-day precision over the entire concentration range were less than 12.28%. The method was successfully applied to a pharmacokinetic study after oral administration of a 300 mg troxerutin drop pill to 18 healthy volunteers.  相似文献

A rapid, accurate and sensitive method for the determination of captopril in human plasma was developed by solid phase extraction and high performance liquid chromatography (HPLC), using precolumn derivatization of captopril with chromophore label o-phethaldialdehyde (OPA). The extraction of captopril from human plasma was carried out by an amino propyl cartridge. A 0.01 M solution of HCl in methanol showed the best recovery and was chosen for elution of captopril in cartridge. This methanolic solution was applied to react with aqueous solution of OPA and glycine as a coderivatization reagent. The process of derivatization was completed within 2 min at room temperature. The derivatized captopril was injected into a reverse phase HPLC system. Mobile phase was consisted of water:acetonitrile:trifluoroacetic acid (85:15:0.1 v/v/v) with a flow rate of 1 ml min?1 and detector was used at 345 nm. Linear dynamic range and limit of detection were found as 0.1–6 ppm and 0.1 ppm, respectively.  相似文献

建立了高效液相色谱同时测定化妆品中12种紫外吸收剂含量的方法。样品用甲醇提取,高速离心,过滤,以SB-C8柱(250 mm×4.6 mm, 5 μm)为分离色谱柱,甲醇和0.1%(v/v)甲酸水溶液为流动相,梯度洗脱,以311 nm为检测波长进行定性、定量分析。该方法前处理简单、易操作,12种紫外吸收剂分离效果良好;在1.0~500 mg/L范围内呈线性关系,相关系数大于0.9995;方法检出限为0.002~0.1 mg/L;实际样品中的加标回收率为97.4%~107.5%,相对标准偏差为1.54%~4.98%。该方法简便、准确,能够满足化妆品中紫外吸收剂的检测要求。  相似文献

Quantification of bradykinin peptides in limited amounts of rat muscle tissue dialysate has been performed using a packed capillary LC-ESI-TOF-MS method. The micro dialysate samples (450 microL) with added internal standard were loaded onto a 1 mm x 5 mm loading column packed with 5 microm Kromasil C18 particles by a carrier solution of 0.1% formic acid in ACN/water (5:95, v/v) at a flow rate of 250 microL/min for online preconcentration of the analytes. Back-flushed elution onto a 150 mm x 0.5 mm Zorbax C18 column packed with 5 microm particles was conducted using a linear solvent ACN/H2O gradient containing 0.1% formic acid. (Tyr8)-bradykinin was used as an internal standard and was added to the dialysis sample prior to injection. Baseline separation of bradykinin, arg-bradykinin and (tyr8)-bradykinin was achieved within 10 min. Positive ESI was performed in the m/z range of 200-1300. The method was validated in the range 0.2-1.0 ng/mL dialysate, yielding correlation coefficients of 0.995 and 0.990 for bradykinin and arg-bradykinin, respectively. The within-assay and between-assay precisions were between 4.3-9.6% and 6.2-10.6%, respectively. Both arg-bradykinin and bradykinin were detected in dialysate from rat muscle tissue, at concentrations of 0.1 and 0.4 ng/mL for bradykinin and arg-bradykinin, respectively, confirming the presence of arg-bradykinin in rat muscles.  相似文献

This paper describes validated high-performance column liquid chromatographic (HPLC) and high-performance thin-layer chromatographic (HPTLC) methods for simultaneous estimation of acetylsalicylic acid (ASA) and clopidogrel bisulfate (CLP) in pure powder and formulations. The HPLC separation was achieved on a Nucleosil C8 column (150 mm length x 4.6 mm id, 5 microm particle size) using acetonitrile-phosphate buffer, pH 3.0 (55 + 45, v/v) mobile phase at a flow rate of 1.0 mL/min at ambient temperature. The HPTLC separation was achieved on an aluminum-backed layer of silica gel 60F254 using ethyl acetate-methanol-toluene-glacial acetic acid (5.0 + 1.0 + 4.0 + 0.1, v/v/v/v) mobile phase. Quantitation was achieved with UV detection at 235 nm over the concentration range 4-24 microg/mL for both drugs, with mean recoveries of 99.98 +/- 0.28 and 100.16 +/- 0.66% for ASA and CLP, respectively, using the HPLC method. Quantitation was achieved with UV detection at 235 nm over the concentration range of 400-1400 ng/spot for both drugs, with mean recoveries of 99.93 +/- 0.55 and 100.21 +/- 0.83% for ASA and CLP, respectively, using the HPTLC method. These methods are simple, precise, and sensitive, and they are applicable for the simultaneous determination of ASA and CLP in pure powder and formulations.  相似文献

建立了超高效液相色谱-串联质谱(UPLC-MS/MS)同时测定茶叶、代用茶和饮料食品中63种非法添加化合物的分析方法。样品经甲醇超声提取后,采用Thermo Acclaim RSLC C18色谱柱(100 mm×2.1 mm,2.2 μm)分离,以5 mmol/L甲酸铵溶液(含体积分数为0.1%的甲酸)-0.1%(体积分数)甲酸乙腈为流动相进行梯度洗脱。在电喷雾离子源正离子模式下,采用动态多反应监测(dMRM)方式测定,外标法定量。63种待测化合物在相应的线性范围内呈良好的线性关系,相关系数(R2)均大于0.99;定量限为0.10~2.50mg/kg;在3个添加水平下,63种待测物的平均回收率为62.4%~129.4%,进样精密度和重复性的相对标准偏差为0.3%~9.6%(n=6)。该方法简便快捷、准确可靠,适用于茶叶、代用茶和饮料食品中非法添加具有解热镇痛效果的化合物检测。  相似文献

A simple, rapid, efficient, and environmentally friendly method for the determination of five triazine herbicides in water and soil samples was developed by using dispersive liquid-liquid microextraction (DLLME), coupled with high performance liquid chromatography-diode array detection (HPLC-DAD). The water samples were directly used for DLLME extraction. For soil samples, the target analytes were first extracted by water-methanol (99:1, v/v). In the DLLME extraction method, chloroform was used as an extraction solvent, and acetonitrile as a dispersive solvent. Under the optimum conditions, the enrichment factors of DLLME were in the range between 183-221. The linearity of the method was obtained in the range of 0.5-200 ng/mL for the water sample analysis, and 1-200 ng/g for the soil samples, respectively. The correlation coefficients ranged from 0.9968 to 0.9999. The limits of detection were 0.05-0.1 ng/mL for the water samples, and 0.1-0.2 ng/g for the soil samples. The proposed method has been successfully applied to the analysis of target triazine herbicides (simazin, atrazine, prometon, ametryn, and prometryn) in water and soil samples with satisfactory results.  相似文献

Abstract

High performance thin layer chromatographic (HPTLC) and high performance liquid chromatographic (HPLC) methods were developed for the simultaneous determination of Tinidazole and Furazolidone in suspension.

In the HPTLC method the separation of Tinidazole and Furazolidone was carried out on silica gel 60F254 HPTLC glass plate using chloroform:methanol:ammonia (9:1:0.1 v/v) as a mobile phase. Rf values obtained were 0.63 and 0.79 for Furazolidone and Tinidazole respectively. Densitometric evaluation was done at 335 nm. Linearity was obtained within the concentration range 10–50 μg/ml and 3.5–17.5 μg/ml for Tinidazole and Furazolidone respectively.

The second method is based on high performance liquid chromatography on a reversed phase column (μ Bondapak C18) using a mobile phase comprised of water: acetonitrile: triethylamine (80:20:0.1 v/v) adjusted to pH = 3.0 with dil. phosphoric acid. Retention times were 5.24 and 7.82 min for Tinidazole and Furazolidone respectively at a flow rate of 1.5 ml/min. Detection was done at 335 nm. Linearity was obtained within the concentration range 30–180 μg/ml and 10.5–63 μg/ml for Tinidazole and Furazolidone resp.  相似文献

A sensitive and specific high-performance liquid chromatographic method has been developed to measure the catechol-O-methyl-transferase (COMT) inhibitor 3,4-dihydroxy-4'-methyl-5-nitrobenzophenone (Ro 40-7592) in human plasma. The compound and the internal standard were extracted from plasma at pH 2 with n-butyl chloride-ethyl acetate (95:5, v/v). The extract was chromatographed on a reversed-phase column (Hypersil ODS, 5 microns) using a mixture of phosphate buffer (0.05 M, pH 2), methanol and tetrahydrofuran (45:55:5, v/v/v) as the mobile phase. Long-retained components were removed from the system by means of a simple column-switching system. Quantification of the catechol-O-methyltransferase inhibitor was performed by means of coulometric detection (0.1 V). The limit of quantification was about 1 ng/ml, using a 1-ml specimen of plasma. The recovery from human plasma was greater than 88%. The mean inter-assay precision was 5.3% in the range 2.5-1000 ng/ml. Linearity of the standard curve was obtained in the concentration range 2.5-500 ng/ml. The catechol-O-methyltransferase inhibitor was stable in human plasma when stored for six months at -20 degrees C and for 24 h at room temperature. The practicability of the new method was demonstrated by the analysis of more than 400 plasma samples from a tolerance study performed in human volunteers.  相似文献
