应用主成分分析(Principal component analysis,PCA)和聚类分析法(Cluster analysis,CA)对9种(27个)常见食用植物油及100个餐饮废油的低场核磁共振(Low-field nuclear magnetic resonance,LF-NMR)(T2)弛豫特性数据进行分析。结果表明:在正常食用油种类区分方面,主成分分析的效果较优,9种食用油在主成分分布图上按种类正确分组,边界清晰。而在正常食用油与餐饮废油的区分方面,聚类分析效果较优,引入30个待测样本后,聚类分析(127个样品,欧式距离=5)的正确率为94.49%,分析误判率为5.51%,分组效果良好。LF-NMR结合化学模式识别可实现对油脂种类及餐饮废弃油脂的鉴别。  相似文献

Microalgae have a great potential for the production of healthy food and feed supplements. Their ability to convert carbon into high-value compounds and to be cultured in large scale without interfering with crop cultivation makes these photosynthetic microorganisms promising for the sustainable production of lipids. In particular, microalgae represent an alternative source of polyunsaturated fatty acids (PUFAs), whose consumption is related to various health benefits for humans and animals. In recent years, several strategies to improve PUFAs’ production in microalgae have been investigated. Such strategies include selecting the best performing species and strains and the optimization of culturing conditions, with special emphasis on the different cultivation systems and the effect of different abiotic factors on PUFAs’ accumulation in microalgae. Moreover, developments and results obtained through the most modern genetic and metabolic engineering techniques are described, focusing on the strategies that lead to an increased lipid production or an altered PUFAs’ profile. Additionally, we provide an overview of biotechnological applications of PUFAs derived from microalgae as safe and sustainable organisms, such as aquafeed and food ingredients, and of the main techniques (and their related issues) for PUFAs’ extraction and purification from microalgal biomass.  相似文献

建立了利用短的微径色谱柱直接测定食用油中未衍生化长链脂肪酸的快速气相色谱方法。C12~C22脂肪酸在2 min之内实现基线分离,定量标准曲线的相关系数大于0.988,最低检测限(S/N=3)为2.80~9.60 mg/L,回收率为74.5%~86.5%。该方法快速、简便、准确,分析通量大,已成功地用于香油、色拉油及调和油等3种食用油中长链脂肪酸含量的测定。  相似文献

建立了动态固相微萃取-气相色谱(FID)测定环境空气中挥发性脂肪酸(VFAs)的分析方法,选择乙酸(AA)、丙酸(PA)、丁酸(BA)、异丁酸(iso-BA)、戊酸(VA)和异戊酸(iso-VA)6种代表性VFAs为研究对象,研制了动态萃取装置并建立了动态萃取机制。研究了固相微萃取材料、萃取时间、萃取温度、萃取流速、解析温度及时间对VFAs萃取效率的影响,在此基础上优化了气相色谱法检测条件。在优化条件下,6种脂肪酸的线性范围均达到4个数量级,相关系数均大于0.990。方法的检出限(S/N=3)为1.0~4.7μg/L,相对标准偏差(RSD,n=3)为1.1%~6.8%。与常规方法相比,该方法大大提高了检测范围和检测灵敏度,并成功用于环境空气中挥发性脂肪酸的分析检测。  相似文献

Capillary ElectroChromatography (CEC) on a fused silica capillary column (40 cm L × 100 μm i.d.) packed with 3 μm octadecylsilica (ODS) was evaluated for the analysis of the triglycerides and their fatty acids in fish oils, and more especially in the oil of Moroccan Sardinia pilchardus. The very high complexity of the lipids in fish oil is well illustrated by CEC with a nonaqueous mobile phase consisting of acetonitrile/isopropanol/n-hexane in the ratio 57/38/5 to which 50 mM ammonium acetate has been added. In order to unravel the complexity, the oil was hydrolyzed and the free fatty acids (FFAs), the methyl esters (FAMEs), and the phenacyl esters (FAPEs) were analyzed by CEC on the same column used for the analysis of the triglycerides. Isocratic elution was achieved with the mobile phase acetonitrile/50 mM MES pH 6 in ratio 9/1. The migration characteristics of FFAs, FAMEs, and FAPEs are compared. The analysis of FAPE derivatives has the advantage that quantification applying UV detection is possible and moreover that the number of double bonds in the fatty acid chains can be elucidated by measuring the UV abundance ratio 240/210 nm.  相似文献

通过模式识别方法区分花生油、大豆油、米糠油、棕榈油和菜籽油。采用气相色谱法分析5种植物油脂的脂肪酸,用面积归一化法计算每个植物油脂样品的各脂肪酸相对含量。以每个植物油脂中9个脂肪酸的相对含量为变量,采用SPSS13.0软件的模式识别技术对119个植物油脂样品进行区分。由主成分分析图可知,花生油、大豆油、米糠油、棕榈油和菜籽油被清晰地分为5组。判别分析建立的判别方程能较好地实现样品的判别,自身验证和交互验证的准确率均为100%。另取每种植物油脂各5个样品(共25个)进行验证,识别准确率为100%。对调和有棕榈油的花生油进行主成分分析,在主成分分析图上,调和油的分布点在花生油分布区域与棕榈油分布区域之间。  相似文献

用碱催化法将青海湖裸鲤鱼油甲酯化,以气相色谱/质谱法分析鱼油中的脂肪酸。青海湖裸鲤可食用部分中鱼油含量为25.13%。从鱼油中共鉴定出47种脂肪酸,包括直链、单支链、多支链饱和脂肪酸,单不饱和、多不饱和脂肪酸,环丙烷基、呋喃基脂肪酸等。不饱和脂肪酸含量为73.6%,其中多不饱和脂肪酸含量为25.4%,以C18∶2(4.9%),C18∶3(3.1%),C20∶4(1.3%),C20∶5(二十碳五烯酸(EPA), 9.4%)和C22∶6(二十二碳六烯酸(DHA), 6.7%)为主。单不饱和脂肪酸含量为48.2%,主要由C16∶1(20.3%),C18∶1(25.9%)构成。饱和脂肪酸含量为25.7%,主要有C14∶0(3.4%),C16∶0 (19.4%)和C18∶0(1.1%)。青海湖裸鲤鱼油中还存在不常见的环丙烷基和呋喃基脂肪酸及多种奇数碳链和支链脂肪酸。因此,青海湖裸鲤是功能性脂肪酸的重要膳食来源。  相似文献

In the present work, the applicability of attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy, coupled with chemometric tools in recognizing essential oils (EOs) for routine control, was evaluated. EOs belonging to Mentha, Cymbopogon, and Lavandula families and to S. rosmarinus and T. vulgaris species were analyzed, and the performance of several untargeted approaches, based on the synergistic combination of ATR-FTIR and Partial Least Squares Discriminant Analysis (PLS-DA), was tested to classify the species and chemotypes. Different spectra pre-processing methods were employed, and the robustness of the built models was tested by means of a Receiver Operating Characteristic (ROC) curve and random permutations test. The application of these approaches revealed fruitful results in terms of sensitivity and specificity, highlighting the potentiality of ATR-FTIR and chemometrics techniques to be used as a sensitive, cost-effective, and rapid tool to differentiate EO samples according to their species and chemotype.  相似文献

研究了105℃和180℃氧化条件下葵花籽油理化指标(过氧化值、共轭二、三烯、茴香胺值、酸价、碘值和极性化合物)的变化规律,就理化指标对油脂总体品质变化的贡献度进行了主成分分析,建立了可有效表征葵花籽油在相应条件下品质变化的综合评价指标。结果表明:随着氧化时间的延长、温度的升高,共轭二、三烯、茴香胺值、酸价和极性化合物的升高趋于显著(P≤0.05),而碘值则均呈下降趋势;通过对各理化指标的总体变化率以及与油脂不饱和度和常用理化指标的相关性分析,发现过氧化值、共轭二烯、茴香胺值(105℃)或共轭二烯、茴香胺值、极性化合物(180℃)可有效表征葵花籽油的品质变化;成功建立了评判葵花籽油品质变化的综合指标,并确定其临界值分别为-0.189(105℃)和0.727(180℃);此外,可通过对共轭二烯和茴香胺值的主成分分析有效区分在105℃或180℃下氧化葵花籽油的品质变化。  相似文献

用氢氧化四甲胺/甲醇酯化法使深海鱼油内的三酸甘油酯中的脂肪酸和游离脂肪酸转化成脂肪酸甲酯,用气相色谱和气相色谱/质谱进行分析,鉴定出41个组分,并对其中的主要组分顺式-5,8,11,14,17-十二碳五稀酸(EPA)和顺式-4,7,10,13,16,19-二十二碳六烯酸(DHA)建立了定量分析方法。方法的相对标准偏差分别为4.3%(EPA)和4.7%(DHA)。EPA和DHA的回收率分别为100.  相似文献

Abstract

Principal component (PCA) and factor analysis (FA) are evaluated for the interpretation of the information contained in large datasets resulting from the study of environmental samples by gas chromatography (GC) and GC coupled to mass spectrometry (GC-MS). A case involving the identification and quantitation of 64 variables (hydrocarbons and fatty acids) in 87 water samples (dissolved and particulate fractions) of a coastal system (Ebre Delta) has been selected for examination.

PCA has evidenced important differences between the dissolved and particulate materials, as well as between the particulates collected in the bays and those obtained in the river and channels. PCA has also allowed the identification of outlier samples in the dissolved fraction. Independent application of FA to each of these groups has provided a useful method for the characterization of diverse algal, terrestrial, microbial and anthropogenic inputs. Direct correspondences between these source inputs and factor loadings have provided a selection of representative components of each contribution in the coastal system.  相似文献

Hyperspectral images contain both spectral and spatial image information and were investigated to characterize the freshness of fish. However, most studies of this application have focused on spectral signals rather than image features. The goal of this work was to investigate the ability of spectral and image textural variables for predicting the chemical and physical qualities of fish, respectively, and to optimize the variables for the specific quality determination. The chemical (total volatile basic nitrogen, TVB-N) and physical (texture profile analysis, TPA) properties were investigated. Partial least square (PLS) was applied to develop fish quality prediction models with the spectral and textural variables from the hyperspectral images. The results showed that the TVB-N content of fish fillets was accurately predicted using the spectra. Meanwhile, the TPA parameters were determined through the image textural features with high accuracy, which indicated image textural features were highly related with the TPA parameters. Moreover, spectral and textural features were also extracted from fish eyes and gills and were further used to predict the intact fish quality, taking advantage of the freshness sensitivity of the eyes and gills. The results illustrate that spectra from fish eyes and gills are a potential tool to predict the TVB-N content and TPA parameters for intact fish.  相似文献

基于浓度参量同步荧光光谱技术,对不同溢油类型不同油源原油样品集、引入外扰相似油源样品集进行光谱数据采集,获取其浓度同步荧光光谱矩阵Concentration-Synchronous-Matrix-Fluorescence(CSMF),利用主成分分析方法对两套不同层次的原油相关样品集进行了多类分类识别。结果表明:主成分载荷图可以很好地反映各个原油相关样品在油源上的相似程度,结合支持向量机可以实现不同溢油类型及不同油源原油的准确分类,对于引入风化和海水外扰相似油源溢油样品集,两类分类区分的结果远远高于多类分类识别的结果。通过详细的主成分分析讨论,为溢油油种鉴别提供了一种利用多类分类识别,逐步缩减嫌疑样本数量,最后通过两两分类实现溢油样品准确识别的新思路。  相似文献

The present study investigated and compared the quality and chemical composition of Moroccan walnut (Juglans regia L.) oil. This study used three extraction techniques: cold pressing (CP), soxhlet extraction (SE), and ultrasonic extraction (UE). The findings showed that soxhlet extraction gave a significantly higher oil yield compared to the other techniques used in this work (65.10% with p < 0.05), while cold pressing and ultrasonic extraction gave similar yields: 54.51% and 56.66%, respectively (p > 0.05). Chemical composition analysis was carried out by GC–MS and allowed 11 compounds to be identified, of which the major compound was linoleic acid (C18:2), with a similar percentage (between 57.08% and 57.84%) for the three extractions (p > 0.05). Regarding the carotenoid pigment, the extraction technique significantly affected its content (p < 0.05) with values between 10.11 mg/kg and 14.83 mg/kg. The chlorophyll pigment presented a similar content in both oils extracted by SE and UE (p > 0.05), 0.20 mg/kg and 0.16 mg/kg, respectively, while the lowest content was recorded in the cold-pressed oil with 0.13 mg/kg. Moreover, the analysis of phytosterols in walnut oil revealed significantly different contents (p < 0.05) for the three extraction techniques (between 1168.55 mg/kg and 1306.03 mg/kg). In addition, the analyses of tocopherol composition revealed that γ-tocopherol represented the main tocopherol isomer in all studied oils and the CP technique provided the highest content of total tocopherol with 857.65 mg/kg, followed by SE and UE with contents of 454.97 mg/kg and 146.31 mg/kg, respectively, which were significantly different (p < 0.05). This study presents essential information for producers of nutritional oils and, in particular, walnut oil; this information helps to select the appropriate method to produce walnut oil with the targeted quality properties and chemical compositions for the desired purpose. It also helps to form a scientific basis for further research on this plant in order to provide a vision for the possibility of exploiting these oils in the pharmaceutical, cosmetic, and food fields.  相似文献

High content of citric acid in lemon juice leads to poor sensory experience. The study aimed to investigate the dynamics changes in organic acids, phenolic compounds, and antioxidant activities of lemon juice fermented with Issatchenkia terricola WJL-G4. The sensory evaluation of fermented lemon juice was conducted as well. Issatchenkia terricola WJL-G4 exhibited a potent capability of reducing the contents of citric acid (from 51.46 ± 0.11 g/L to 8.09 ± 0.05 g/L within 60 h fermentation) and increasing total phenolic level, flavonoid contents, and antioxidant activities compared to those of unfermented lemon juice. A total of 20 bioactive substances, including 10 phenolic acids and 10 flavonoid compounds, were detected both in fermented and unfermented lemon juice. The lemon juice fermented for 48 h had better sensory characteristics. Our findings demonstrated that lemon juice fermented with Issatchenkia terricola exhibited reduced citric acid contents, increased levels of health-promoting phenolic compounds, and enhanced antioxidant activities.  相似文献

Osteonecrosis of femoral head (ONFH) is a disease characterized by an impaired blood flow in the bone. The pathogenesis is still unknown, which makes an exact diagnosis troublesome and heavily dependent on experience. Exploring the information of molecular level by modern spectroscopy may help to discover the underlying pathogenesis and find its diagnostic application in clinical medicine. The study focuses on the combination of near-infrared (NIR) spectroscopy and classification models for discriminating ONFH and normal tissues. A total of 128 surgical specimens was prepared and NIR spectra were recorded by an integrating sphere. The experiment data set was divided into three subsets, i.e., the training set, validation set, and test set. Successive projection algorithm-linear discriminant analysis (SPA-LDA) was used to compress variables and build the diagnostic model. Partial least square-discriminant analysis (PLS-DA) was used as the reference. Principal component analysis (PCA) was used for exploratory analysis. The results showed that compared to PLS-DA, SPA-LDA provided a more parsimonious model using only seven variables and achieved better performance, i.e., sensitivity of 90.5 and 85%, and specificity of 100 and 95.5% for the validation and test sets, respectively. It indicated that NIR spectroscopy combined with SPA-LDA algorithm was a feasible aid tool for discriminating ONFH from normal tissue.  相似文献

福尔马林固定石蜡包埋(Formalin-fixed and paraffin-embedding,FFPE)是最常见的临床组织保存技术,FFPE组织具有标准化的制备流程、易于存储、标本量大、包含较完整的临床回顾性信息等特点,而成为疾病生物标志物发掘的良好载体。近年来,基于临床FFPE组织的特点,发展了系列蛋白质组学研究方法,包括样本制备、消解、分离到蛋白质质谱鉴定等多个领域,总体呈现出微量、高灵敏度、高通量的技术特点,并已成功用于肿瘤精准医学等临床蛋白质组研究。该综述将对FFPE组织切片样本的蛋白质组学方法,以及其在肿瘤研究中的应用进行概述,从而为临床精准医学蛋白质组学的研究提供借鉴思路。  相似文献

采用气相色谱-串联四极杆飞行时间质谱(GC/Q-TOF MS)建立了稻米的代谢组学分析方法,并将其应用于产地溯源。利用D-最优设计对提取溶剂进行优化,通过偏最小二乘分析法考察了不同溶剂对代谢物提取效率的影响。考察了衍生试剂种类、衍生温度及时间对代谢物检测的影响,最终确定分析条件为:甲醇-水(1∶1)为提取溶剂,N-甲基-N-三甲基硅基三氟乙酰胺-三甲基氯硅烷(MSTFA-TMCS,99∶1)于60℃条件下衍生1 h。选取10种代谢物考察仪器精密度、方法重现性和衍生产物稳定性,相对标准偏差(RSD)均小于9%,证明了方法的可靠性。结合Mass Profiler Professional(MPP)软件,以主成分分析(PCA)法区分黑龙江省4个地区的29个绥粳4号稻米样本,分类效果显著。对显著变化(P0.05,倍率变化≥2)的差异性代谢物进行鉴定,确定了11种特征标记物。结果表明,利用GC/Q-TOF MS建立代谢组学分析方法,结合主成分分析鉴别稻米产地具有可行性,为大米市场的规范和资源开发利用提供了依据。  相似文献

In this study, supercritical fluid extraction (SFE), ultrasonic-assisted extraction (UAE), and microwave-assisted extraction (MAE) were applied to explore the most suitable extraction method for fatty acids of Potentilla anseris L. from 12 different producing areas of the Qinghai-Tibetan Plateau. Meanwhile, the important experimental parameters that influence the extraction process were investigated and optimized via a Box-Behnken design (BBD) for response surface methodology (RSM). Under optimal extraction conditions, 16 fatty acids of Potentilla anserina L. were analyzed via high-performance liquid chromatography (HPLC) with fluorescence detection, using 2-(4-amino)-phenyl-1-hydrogen-phenanthrene [9,10-d] imidazole as the fluorescence reagent. The results showed that the amounts of total fatty acids in sample 6 by applying SFE, UAE, and MAE were, respectively, 16.58 ± 0.14 mg/g, 18.11 ± 0.13 mg/g, and 15.09 ± 0.11 mg/g. As an environmental protection technology, SFE removed higher amounts of fatty acids than did MAE, but lower amounts of fatty acids than did UAE. In addition, the contents of the 16 fatty acids of Potentilla anserina L. from the 12 different producing areas Qinghai-Tibetan Plateau were significantly different. The differences were closely related to local altitudes and to climatic factors that corresponded to different altitudes (e.g., annual mean temperature, annual mean precipitation, annual evaporation, annual sunshine duration, annual solar radiation.). The temperature indices, photosynthetic radiation, ultraviolet radiation, soil factors, and other factors were different due to the different altitudes in the growing areas of Potentilla anserina L., which resulted in different nutrient contents.  相似文献
