A fully functionalized pillar[5]arene was synthesized by ‘click chemistry’ and the possible synthetic strategies have been discussed. The host showed effective binding affinities toward neutral alkanediamines (Ka >103 M−1), which are much larger than those observed for simple methyl-substituted pillar[5]arene.  相似文献

CuI catalyzed 1,3-dipolar cycloaddition ‘click chemistry’ was used to prepare 18F-radiolabeled peptides. Three ω-[18F]fluoroalkynes were prepared in yields ranging from 36% to 81%. Conjugation of ω-[18F]fluoroalkynes to various peptides decorated with 3-azidopropionic acid via CuI mediated 1,3-dipolar cycloaddition yielded the desired 18F-labeled products in 10 min with yields of 54-99% and excellent radiochemical purity (81-99%). The total synthesis time was 30 min from the end of bombardment.  相似文献

We describe here a novel and convenient synthesis of head-to-tail cyclic peptide avoiding racemization. Linear depsipeptides including a serine residue as the key element for ester bond formation and acyl transfer were synthesized on 2-chlorotrityl chloride resin. After cleavage from the resin, intramolecular head-to-tail cyclization was performed in solution by C-terminal activation of urethane protected O-acyl serine residue. After removal of the Nα-serine protecting group, the final step consisted in O-N-acyl migration reaction on the ‘switch’ or ‘click’ element to restore native cyclic peptides.  相似文献

A novel ‘O-acyl isodipeptide unit’, Boc-Thr(Fmoc-Val)-OH 5 has been successfully used for the efficient synthesis of a difficult sequence-containing pentapeptide based on the ‘O-acyl isopeptide method’, in which racemization-inducible esterification could be omitted, suggesting that the use of O-acyl isodipeptide units allows the application of this method to fully automated protocols for the synthesis of long peptides or proteins.  相似文献

A very efficient modular reaction protocol was developed to attach various functionalities to a rigid cavitand scaffold. In this way, aryl, iodoaryl, benzyl, pyrrolidylmethyl groups, as well as a polyethylene-glycol chain were attached to the ‘triazol-level’ of the cavitand. The palladium-catalyzed ethynylation of the iodoarene moieties, followed by the copper(I)-catalyzed azide–alkyne cycloaddition produced novel cavitands with significantly elongated binding pockets. The dimensions of these molecules are calculated to be at least 9 Å×18 Å, which place them amongst the largest unimolecular hosts obtained by pure covalent synthesis. A cavitand-based click conjugate is used for selective complexation of Cu2+ and Fe3+, providing significant fluorescent quenching.  相似文献

We disclosed a novel ‘racemization-free segment condensation’ based on the ‘O-acyl isopeptide method’ in which an N-segment including C-terminal O-acyl isopeptide structure with urethane-protected Ser/Thr residue was employed for the segment condensation, suggesting that the use of this method contributes to the effective convergent synthesis of long peptides/proteins.  相似文献

The synthesis of a polypseudorotaxane, polyrotaxane, and polycatenane containing the electron-deficient cyclophane cyclobis(paraquat-p-phenylene) (CBPQT4+) subunit in the side chain is described. These interlocked supramolecular polymers have been prepared from an azide-functionalized polystyrene derivative and an acetylene-functionalized [2]rotaxane, [2]catenane and their parent tetracationic cyclophane via Cu(I)-catalyzed 1,3 dipolar cycloadditions (‘click chemistry’). The synthesis and characterization of the polymers and intermediates has been described using IR, 1H NMR, UV spectroscopies, and voltammetry. We have shown that the CBPQT4+ unit of the side chain polystyrene derivative has the ability to reversibly undergo complexation with a complementary dialkoxynaphthalene derivative.  相似文献

Non-destructive, rapid, instrumental tools in fruit production are required for predicting the optimum harvest window and monitoring fruit quality during shelf life. The degree of chlorophyll degradation is a sensitive indicator for fruit maturation and ageing. Adequate indices for chlorophyll prediction by means of non-destructive spectral analysis have been studied in the fields of photosynthesis research and remote sensing developments. However, an evaluation of these indices and multivariate linear regression models does not exist so far for predicting the fruit chlorophyll content.Spectral transmittance recordings in the visible wavelength range were carried out on apple fruit Malus domestica Borkh. ‘Elstar’ (n=99) and ‘Jonagold’ (n=117). The fruit chlorophyll a content of ‘Elstar’ apples was measured wet-chemically and predicted by means of the specific indices: NDVI, Tr698/Tr760, red-edge, TrII on Tr′(λ), TrII, and RVSI with the correlation coefficients of determination R2=0.84, 0.81, 0.75, 0.81, 0.15, and 0.67, respectively. Partial least-squares (PLS) calibration models were built using calibrated spectra (630-730 nm), first derivative of spectra and second derivative of spectra yielding multivariate correlation coefficients of determination R2=0.81, 0.86, and 0.92, respectively. Similar results were found for ‘Jonagold’ apple fruit. Linear regression of indices and PLS calibration models were empirically tested on the chlorophyll a content of apple fruit measuring the same cultivars grown in a different seasons and growing locations. In the case of ‘Elstar’ apple fruit, correlation coefficients of fruit chlorophyll a content analysed wet-chemically as well as fruit maturation measured as calendar weeks were similar to those in the calibration experiment. Application of linear regression equations of indices and PLS models on spectra of ‘Jonagold’ apple fruit led to less accurate results for those methods, which use wavelengths above 720 nm as indicative range.  相似文献

A method for the direct introduction of Aib-Pro into peptides on solid phase was developed. The Aib-Pro unit was introduced by means of the ‘azirine/oxazolone method’ using allyl N-(2,2-dimethyl-2H-azirin-3-yl)-l-prolinate as the synthon. After the reaction of the resin-bound amino or peptide acid with allyl N-(2,2-dimethyl-2H-azirin-3-yl)-l-prolinate, the allyl protecting group of the resulting extended peptide could be removed by a mild Pd0-promoted procedure. Cleavage of the peptide from the resin was performed with UV light at 352 nm and yielded C-terminal protected peptides. The method found a successful application in the syntheses of different Aib-Pro containing peptaibol segments. Furthermore, a protected derivative of the peptide antibiotic Trichovirin I 1B was prepared by segment condensation.  相似文献

The copper-catalyzed ‘click silylation’ has been exploited for the chemical modification of γ-azidopropyltriethoxysilane (AzPTES) with a wide range of terminal alkynes (1a1v) in a one-pot operation. The novel 1,2,3-triazole-triethoxysilane derivatives (2a2v) were synthesized by this procedure and comprehensively characterized by IR spectra, 1H and 13C NMR, and HRMS studies.  相似文献

A series of diversely 1-substituted 4-amino 1,2,3-triazoles were synthesized by [3+2] cycloaddition between azides and ynamides. This copper catalyzed process represents the first examples of a ‘click reaction’ employing ynamides and should expand the scope of the ynamide chemistry both synthetically and industrially. Various azides (even highly functionalized) were allowed to react with N-benzyl, N-tosyl ynamide to give the corresponding triazole adducts in high yield and with very high levels of regioselectivity.  相似文献

Derivatives of vitamin D3 carrying an 8-carbon linker at C-11 terminating in an active ester were synthesized from commercial vitamin D3 using a disassembly-reassembly strategy. Vitamin D3 was cleaved at the C6-C7 double bond and the ‘upper’ fragment was converted, via a series of reactions, to derivatives substituted at C-11 with an 8-carbon linker terminating in an ethyl ester. Reassembly with modified ‘lower’ fragments using Horner-Wittig olefination followed by linker ester hydrolysis and re-esterification with p-nitrophenol gave C-11 substituted p-nitrophenyl esters. These vitamin D derivatives were conjugated to 42-amino acid helix-loop-helix peptides by reaction of their p-nitrophenyl esters with lysyl side-chain amino groups on the peptides. The vitamin D—peptide conjugates, being potential specific binder candidates for vitamin D-binding protein, were characterized by mass spectroscopy and CD measurements.  相似文献

Biocatalytic racemization of aliphatic, (aryl)aliphatic and aromatic α-hydroxycarboxylic acids was achieved via a reversible oxidation-reduction sequence using a pair of stereo-complementary Prelog- and anti-Prelog d- and l-α-hydroxyisocaproate dehydrogenases from Lactobacillus confusus DSM 20196 and Lactobacillus paracasei DSM 20008, resp., overexpressed in Escherichia coli. The mild reaction conditions ensured essential ‘clean’ isomerization, undesired ‘over-oxidation’ of the substrate forming the α-ketoacid could be suppressed by exclusion of O2 and adjustment of the NAD+/NADH-ratio.  相似文献

A robust method allowing the routine determination of n(87Sr)/n(86Sr) with at least five significant decimal digits for large sets of mineral water samples is described. It is based on 2 consecutive chromatographic separations of Sr associated to multi-collector inductively coupled plasma mass spectrometry (MC-ICPMS) measurements. Separations are performed using commercial pre-packed columns filled with “Sr resin” to overcome isobaric interferences affecting the determination of strontium isotope ratios. The careful method validation scheme applied is described. It included investigations on all parameters influencing both chromatographic separations and MC-ICPMS measurements, and also the test on a synthetic sample made of an aliquot of the NIST SRM 987 certified reference material dispersed in a saline matrix to mimic complex samples. Correction for mass discrimination was done internally using the n(88Sr)/n(86Sr) ratio. For comparing mineral waters originating from different geological backgrounds or identifying counterfeits, calculations involved the well known consensus value (1/0.1194) ± 0 as reference. The typical uncertainty budget estimated for these results was 40 ‘ppm’ relative (k = 2). It increased to 150 ‘ppm’ (k = 2) for the establishment of stand alone results, taking into account a relative difference of about 126 ‘ppm’ systematically observed between measured and certified values of the NIST SRM 987. In case there was suspicion of a deviation of the n(88Sr)/n(86Sr) ratio (worst case scenario) our proposal was to use the NIST SRM 987 value 8.37861 ± 0.00325 (k = 2) as reference, and assign a typical relative uncertainty budget of 300 ‘ppm’ (k = 2). This method is thus fit for purpose and was applied to eleven French samples.  相似文献

The dipod 1,2-bis(8-hydroxyquinolinoxymethyl)benzene (3) and tetrapod 1,2,4,5-tetrakis(8-hydroxyquinolinoxymethyl)benzene (5) have been synthesized through nucleophilic substitution of respective 1,2-bis(bromomethyl)benzene (2) and 1,2,4,5-tetra(bromomethyl)benzene (4) with 8-hydroxyquinoline (1). For comparison, 1,3,5-tris(8-hydroxyquinolinoxymethyl)benzene derivatives (7a and 7b) have been obtained. The complexation behavior of these podands towards Ag+, Co2+, Ni2+, Cu2+, Zn2+, and Cd2+ metal ions has been investigated in acetonitrile by fluorescence spectroscopy. The sterically crowded 1,2,4,5-tetrapod 5 displays unique fluorescence ‘ON-OFF-ON’ switching through fluorescence quenching (λmax 395 nm, switch OFF) with <1.0 equiv of Ag+ and fluorescence enhancement (λmax 495 nm, switch ON) with >3 equiv Ag+ and can be used for estimation of two different concentrations of Ag+ at two different wavelengths. The addition of Cu2+, Ni2+, and Co2+ metal ions to tetrapod 5 causes fluorescence quenching, i.e., ‘ON-OFF’ phenomena at λmax 395 nm for <10 μM (1 equiv) of these ions but addition of Zn2+ and Cd2+ to tetrapod 5 results in fluorescence enhancement with a gradual shift of λem from 395 to 432 and 418 nm, respectively. Similarly, dipod 3 behaves as an ‘ON-OFF-ON’ switch with Ag+, an ‘ON-OFF’ switch with Cu2+, and an ‘OFF-ON’ switch with Zn2+. The placement of quinolinoxymethyl groups at the 1,3,5-positions of benzene ring in tripod 7a-b leads to simultaneous fluorescence quenching at λmax 380 nm and enhancement at λmax 490 nm with both Ag+ and Cu2+. This behavior is in parallel with 8-methoxyquinoline 8. The rationalization of these results in terms of metal ion coordination and protonation of podands shows that 1,2 placement of quinoline units in tetrapod 5 and dipod 3 causes three different fluorescent responses, i.e., ‘ON-OFF-ON’, ‘ON-OFF’, and ‘OFF-ON’ due to metal ion coordination of different transition metal ions and 1, 3, and 5 placement of three quinolines in tripod 7, the protonation of quinolines is preferred over metal ion coordination. In general, the greater number of quinoline units coordinated per metal ion in 5 compared with the other podands points to organization of the four quinoline moieties around metal ions in the case of 5.  相似文献

The design, synthesis, and photophysical properties of new BODIPY-based fluorogenic ‘click on’ dyes are reported. CuAAC reaction of non-fluorescent BODIPY azide with a series of non-fluorescent alkyne molecules resulted in fluorescent triazoles which displayed up to 532-fold enhancement of fluorescence in the red region. Imaging studies confirmed the general trend of cell permeability and a cholesterol linked derivative exhibited selective localization into intracellular membranes.  相似文献

The oxidative amidation of phenols effects the conversion of appropriately substituted phenols into 4-amidodienones (‘para-oxidative amidation’) or 2-amidodienones (‘ortho-oxidative amidation’) by the action of hypervalent iodine reagents. The reagent, (diacetoxyiodo)benzene (‘DIB’) is especially effective in these transformations. This paper focuses on techniques for the desymmetrization of the dienoes thus obtained, leading to the stereocontrolled creation of N-substituted spiro carbons. The methodology creates new opportunities in alkaloid synthesis, as apparent from a number of examples.  相似文献

A survey of conditions for the palladium catalyzed intramolecular Heck cyclization of protected amines has shown that the Herrmann-Beller palladacycle can be exploited under ‘cationic’ conditions to provide a robust and rapid route (<2 h) to the synthesis of single double bond isomer phenanthridines in excellent yield (76-99%). In addition, the same cyclization can be performed under ‘neutral’ conditions to provide phenanthridines with a double bond isomer profile suitable for exploitation in diversity-based applications. We have also shown that the highly reactive (tBu3P)2Pd catalyst can induce cyclization at low temperatures (≤50 °C), giving similar results to the ‘neutral’ conditions, and offering an alternative pathway for sensitive substrates.  相似文献

Inverse electron demand Diels–Alder additions (iEDDA) between 1,2,4,5-tetrazines and olefins have recently found widespread application as a novel ‘click chemistry’ scheme and as a mild technique for the modification of materials. Norbornenes are, due to their straightforward synthetic availability, especially interesting in the latter context. Therefore, the reactivity of different norbornene-based compounds was compared with unsubstituted norbornene and other alkenes using UV-vis measurements for the determination of reaction rates under pseudo first order conditions. Thereby, exo,exo-5-norbornene-2,3-dimethanol was found to be almost as reactive as unsubstituted norbornene whereas (±)-endo,exo-dimethyl-5-norbornene-2,3-dicarboxylate reacted only insignificanty faster than unstrained alkenes.  相似文献
