A feasibility research was performed to study the possibilities of using a molecularly imprinted polymer as sorbent material in solid-phase extraction for the separation of active inhibitors of epidermal growth factor receptor (EGRF) from Caragana Jubata, a Chinese traditional Tibetan medicine. A molecularly imprinted polymer using quercetin, an active anti-EGFR inhibitor (IC50 = 15 microM), as the template and acrylamide as the functional monomer was prepared. The polymer was evaluated as a selective sorbent in molecularly imprinted solid-phase extraction. The EtOAc extract of Caragana Jubata was loaded on the polymer, and two novel active anti-EGFR inhibitors were found to be selectively retained after washing the polymer with appropriate solvent to disrupt the non-specific interactions occurring between the sample and the polymer matrix, which were identified as (E)-piceatannol (IC50 =4.9 microM) and butein (IC50 = 10 microM). The present work affords us a new potential method for selective separation of bioactive components from herb by using molecularly imprinted polymer as a solid-phase extraction adsorbent.  相似文献

Surface molecularly imprinted polymers for mezlocillin were used for the selective solid-phase extraction of mezlocillin from eggs. The molecularly imprinted polymers were prepared using mezlocillin as the template, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker, silica particle as the support and were characterized by infrared spectroscopy, scanning electron microscopy, and thermogravimetric analysis. The adsorption properties of polymers were investigated in detail. The obtained polymers provided high imprinting efficiency to mezlocillin with an imprinting factor of 3.72 and were used as selective sorbents for mezlocillin. Molecularly imprinted solid-phase extraction followed by high-performance liquid chromatography was used for the determination of mezlocillin in eggs and the conditions were evaluated. The average recovery of mezlocillin in fortified eggs was between 69.8 and 82.3% with a relative standard deviation less than 6.9%. The results demonstrate the application of molecularly imprinted polymers for the extraction of mezlocillin from biological samples.  相似文献

A molecularly imprinted polymer solid-phase extraction method is used to extract esculetin from the ash bark of Chinese traditional medicine. Ratio of ethanol and water as washing solution were investigated. Data of accumulative adsorption on molecularly imprinted polymers from the continuous loading experiment suggests that there are two different kinds of recognition sites in molecularly imprinted polymers. By selecting the washing and eluting solution a scheme was designed to separate esculetin and its analogues including esculin, coumarin, 7-methoxylcoumarin and daphnetin. Finally, by applying the revised scheme esculetin was extracted from the ash bark of Chinese traditional medicine that was purchased from two big drugstores, respectively, with both molecularly imprinted polymers and non-molecularly imprinted polymers.  相似文献

Cocaine is a well-known drug of abuse which, when ingested nasally or by smoking, undergoes a number of biotransformation and degradation reactions. In the present work, a synthetic analogue of the cocaine metabolite benzoylecgonine was prepared and used as a template molecule in the preparation of a series of molecularly imprinted polymers (MIPs). Molecularly imprinted solid-phase extraction (MISPE) conditions were established under which benzoylecgonine in aqueous samples could be selectively extracted and quantified at clinically relevant concentrations (μg/ml). Under optimised MISPE conditions, recoveries of analyte were high (>70%) and excellent discrimination between imprinted and non-imprinted materials observed.  相似文献

A novel molecularly imprinted polymers based on multiwalled carbon nanotubes synthesized by precipitate polymerization was applied as a selective sorbent for separation and determination of rhein (4,5-dihydroxyanthraquinone-2-carboxylic acid) from the root of kiwi fruit samples coupled with high performance liquid chromatography (HPLC). The molecularly imprinted polymers were prepared with methacrylic acid and 4-vinylpyridine as bifunctional monomers. The chemical structure of the molecularly imprinted polymers was characterized by Fourier transform infrared spectrometer. The equilibrium rebinding experiment and competitive adsorption experiment showed that these imprinted polymers exhibited good adsorption ability toward rhein. The Langmuir adsorption equilibrium constant, K(m) , and theoretical maximum adsorption capacity, Q(m) , were estimated to be 0.43 and 6.77 mg g(-1) , respectively. Compared with molecularly imprinted polymers prepared with methacrylic acid or 4-vinylpyridine solely, the molecularly imprinted polymers synthesized with bifunctional monomers showed enhanced molecular imprinting effect and higher adsorption capacity for the template rhein. The performances of the molecularly imprinted polymers utilized as solid phase extraction sorbent were investigated in detail. The molecularly imprinted polymers prepared by the method proposed in this work could successfully apply to extraction and determination of rhein from the root of kiwi fruit samples coupled with HPLC.  相似文献

A facile method to fabricate molecularly imprinted polymers (MIPs) on glass microspheres in a column was developed. The column was prepacked with glass microspheres, and then the prepolymerization mixture was injected into the interstitial volume of the column. The polymerization took place in situ and the column could be directly used for high-performance liquid chromatography after the template had been removed. The template consumption was reduced greatly because the prepolymerization mixture just filled the interstitial volume between the glass microspheres in the column. The MIPs obtained exhibited better kinetic properties, higher efficiency, and low back pressure of the column. Emodin imprinted polymers were prepared by this method and were used for solid-phase extraction.  相似文献

以罗丹明B为模板分子,以钛酸丁酯为交联剂,采用溶胶-凝胶法在酸性条件下制备了罗丹明B二氧化钛溶胶-凝胶分子印迹聚合物;利用傅立叶变换红外光谱和透射电镜分析了分子印迹聚合物的结构,使用热重分析测定了其热稳定性,采用静态吸附和动态吸附方法考察了其吸附性能,并与非印迹聚合物进行了对比.结果表明,与非印迹聚合物相比,印迹聚合物具有网络状多孔隙凝胶微结构及对模板分子的特异性识别结合位点,因而对模板分子具有更好的吸附性能和特异选择性.其原因可能在于,模板分子上的羰基与钛酸丁酯水解产生的羟基产生氢键作用,使钛酸围绕于模板分子周围,并通过缩聚形成凝胶;当模板分子洗脱后,孔隙得到保留并形成网络状凝胶,从而能够吸附更多的模板分子.  相似文献

The quercetin molecularly imprinted polymer microspheres were prepared by reversible addition-fragmentation chain transfer free radical polymerization combined with precipitation method (RAFTPP) with the dibenzyl trithiocarbonate (DBTTC) as RAFT reagent and quercetin as template molecule. The effects of solvents and cross-linking agent on the morphology and size of polymers were investigated, and the polymers were preliminarily screened by scanning electron microscope. The adsorption experiment and adsorption model analysis were carried out on the molecularly imprinted polymers (R-MIP) prepared under optimal condition and on the molecularly imprinted polymers prepared with a traditional precipitation polymerization (T-MIP). Consequently it was determined that, QR-MIP max was 37.71 mg g?1 and QT-MIP max was 29.61 mg g?1. The results showed that the R-MIP had a good adsorptive property and was obviously superior to T-MIP. R-MIP was used as solid-phase extraction filler in combination with the HPLC method to conduct enrichment, separation and measurement of the quercetin in honeysuckle and clove leaves, which effectively removed the matrix interference and the recovery rate of this method was 93.8–102.9%.

A multitemplate molecularly imprinted polymer was synthesized using fragments similar to gonyautoxins 1 and 4 including 2,4,6-triaminopyrimidine, 4-hydroxy-2-butanone, and imidazole as dummy templates by bulk polymerization. Scanning electron microscopy and infrared spectroscopy showed a porous structure and the formation of organic groups in the imprinting process of the polymer. One millilitre of 0.1?mol?L?1 acetic acid and 1?mL of 95% methanol were the optimized washing and eluting solutions during the molecularly imprinted solid-phase extraction. The adsorption capacity of the multitemplate molecularly imprinted polymer was lower than for the monotemplate molecularly imprinted polymer prepared in a previous study in seawater in which Alexandrium tamarense and Alexandrium minutum were cultivated. This result may due to the inappropriate combination of the dummy template fragments in the synthesis resulting in the unfitness of the imprinting sites for gonyautoxins 1 and 4. In general, it is inappropriate to use fragments with the similar parts to the analyte as the dummy templates. However, this work indicated the optimal molecularly imprinted polymer for the selective extraction of gonyautoxins 1 and 4.  相似文献

In this work, a novel method is described for the determination of bromhexine in biological fluids using molecularly imprinted solid-phase extraction as the sample cleanup technique combined with high performance liquid chromatography (HPLC). The water-compatible molecularly imprinted polymers (MIPs) were prepared using methacrylic acid as functional monomer, ethylene glycol dimethacrylate as cross-linker, chloroform as porogen and bromhexine as the template molecule. The novel imprinted polymer was used as a solid-phase extraction sorbent for the extraction of bromhexine from human serum and urine. Various parameters affecting the extraction efficiency of the polymer have been evaluated. The optimal conditions for molecularly imprinted solid-phase extraction (MISPE) consisted of conditioning 1 mL methanol and 1 mL of deionized water at neutral pH, loading of 5 mL of the water sample (25 μg L−1) at pH 6.0, washing using 2 mL acetonitrile/acetone (1/4, v/v) and elution with 3× 1 mL methanol/acetic acid (10/1, v/v). The MIP selectivity was evaluated by checking several substances with similar molecular structures to that of bromhexine. Results from the HPLC analyses showed that the calibration curve of bromhexine using MIP from human serum and urine is linear in the ranges of 0.5-100 and 1.5-100 μg L−1 with good precisions (3.3% and 2.8% for 5.0 μg L−1), respectively. The recoveries for serum and urine samples were higher than 92%.  相似文献

以乙腈为分散剂,采用沉淀聚合法合成了反乌头酸分子印迹聚合物微球。研究了合成反应条件对聚合物形貌的影响,发现聚合前主客体氢键络合物和功能单体氢键低聚体是控制微球形成及其粒径大小的关键因素。通过振荡吸附法对聚合物的结合特性进行了评价,发现印迹聚合物微球对模板分子的识别选择性优于块状印迹聚合物和非印迹聚合物。  相似文献

A variety of bulk polymers for the selective separation of chloramphenicol were synthesised from 2-vinylpyridine, diethylaminoethyl methacrylate or methacrylic acid monomers. Chromatographic evaluation indicated that chloramphenicol was retained under nonpolar elution conditions (k = 58.65) through selective hydrogen bonding and ionic interactions. The retention of chloramphenicol under aqueous elution conditions (k > 100) results from nonselective hydrophobic interactions. Under nonpolar elution conditions, the functional monomer employed imparted a significant influence on the recognition properties of the corresponding polymer. After solid-phase extraction using a molecularly imprinted polymer as sorbent and either an organic or aqueous washing solvent, nearly 100% recovery from the chloramphenicol standard solution was achieved, and nearly 90% recovery could be attained from spiked honey samples. The molecularly imprinted polymer was well suited to suppress matrix effects, and provided optimal preconcentration of the target molecule (chloramphenicol) prior to chromatographic analysis.  相似文献

A semi-covalent imprinted polymer was prepared by precipitation polymerisation using propazine methacrylate as template molecule, ethylene glycol dimethacrylate as cross-linker and toluene as porogen. After removal of propazine by basic hydrolysis of the covalent bond, the optimum loading, washing and elution conditions for the solid-phase extraction of the selected triazines were established. The binding sites present in the polymeric matrix were characterised by fitting the experimental results of several rebinding studies to the Langmuir-Freundlich isotherm. Subsequently, an analytical methodology based on molecularly imprinted solid-phase extraction (MISPE) was developed for the determination of several triazinic herbicides in soil and vegetable samples. Following this procedure, a good degree of clean-up of the sample extracts was easily achieved, allowing the HPLC-UV determination of selected triazines in complex samples at low concentration levels.  相似文献

A novel molecularly imprinted polymer monolith was prepared by the room temperature ionic liquid-mediated in situ molecular imprinting technique, using norfloxacin (NOR) as the template, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker. The optimal synthesis conditions and recognition properties of NOR-imprinted monolithic column were investigated. The results indicated that the imprinted monoliths exhibited good ability of selective recognition against the template and its structural analog. Using the fabricated material as solid-phase extraction sorbent, a sample pre-treatment procedure of molecularly imprinted solid-phase extraction coupling with HPLC was developed for determination of trace quinolone residues in animal tissues samples. The recoveries ranging from 78.16 to 93.50% for eight quinolones antibiotics such as marbofloxacin, NOR, ciprofloxacin, danofloxacin, difloxacin, oxolinic acid, flumequine and enrofloxacin were obtained.  相似文献

以多壁碳纳米管表面接枝的L-苯丙氨酸为结合位点, 甲基丙烯酸为功能单体, 乙二醇二甲基丙烯酸酯为交联剂, 采用沉淀聚合技术, 在碳纳米管表面制备了对硝基苯酚印迹复合材料. 采用红外光谱和扫描电镜研究了该印迹复合材料的结构和形貌, 结果表明, 在碳纳米管表面接枝了一层稳定的印迹材料. 采用高效液相色谱研究了该印迹材料的等温吸附性能, 结果表明, 该印迹材料对模板分子具有较大的吸附容量(Qmax=80.5 μmol/g)和良好的选择吸附性能(选择因子达2.5). 以该印迹材料作为固相萃取吸附剂, 研究了它对对硝基苯酚和其它结构类似物混合溶液的动态吸附性能, 结果表明, 印迹复合材料对对硝基苯酚的吸附容量不受结构类似物浓度的影响, 能较好地应用于对硝基苯酚的分离富集检测.  相似文献

Dibenzofuran (DBF) recognition was studied by a molecularly imprinted polymer (MIP) made of commercially available polymers: polyvinyl chloride (PVC), polysulfone, polystyrene, and polyacrylonitrile. Preparation of MIPs was carried out by phase inversion of the polymer. Respective polymer solutions were coagulated with DBF template in water (non-solvent). In addition, reference polymers without the template were prepared in same manner as unimprinted polymers. After the DBF template was extracted from the solidified polymers by washing with methanol (MeOH), recognition experiments of DBF by the imprinted polymers were carried out for DBF in MeOH solutions. Equilibrium constants for DBF were calculated from saturation binding kinetics. Equilibrium experiment results reveal that recognition was effective for DBF binding to the DBF-imprinted polymer, especially for the imprinted PVC.  相似文献

A molecularly imprinted polymer (MIP) using phenytoin as template and methacrylamide as the functional monomer was prepared. The selectivity was measured by comparing capacity factors of phenytoin and other structurally related compounds. The polymer was evaluated as a selective sorbent in molecularly imprinted solid-phase extraction (MISPE). Several washing solvents were tested to study their ability to disrupt the non-specific interactions occurring between the sample and the polymer matrix and the role of water in the recognition process was also investigated. It was shown that the key step of successful sample extraction is the right choice of the washing solvent. Plasma samples spiked with phenytoin were analyzed by the MISPE methodology developed in this work. Method validation (intra- and inter-day precision, recovery, specificity) was carried out. The calibration curve showed good linearity in the 2.5–40 μg/ml range corresponding to therapeutically relevant plasma levels. The intra- and inter-day precision values were below the 15% limit established for bioanalytical methods. The results showed that the method could be successfully applied for the determination of phenytoin in plasma samples.  相似文献

Molecularly imprinted polymers (MIPs) were prepared using bisphenol A (BPA) as a template by precipitation polymerization. The polymer that had the highest binding selectivity and ability was used as solid-phase extraction (SPE) sorbents for direct extraction of BPA from different biological and environmental samples (human serum, pig urine, tap water and shrimp). The extraction protocol was optimized and the optimum conditions were as follows: conditioning with 5 mL methanol–acetic acid (3:1), 5 mL methanol, 5 mL acetonitrile and 5 mL water, respectively, loading with 5 mL aqueous samples, washing with 1 mL acetonitrile, and eluting with 3 mL methanol. MIPs can selectively recognize, effectively trap and preconcentrate BPA over a concentration range of 2–20 μM. Recoveries ranged from 94.03 to 105.3 %, with a relative standard deviation lower than 7.9 %. Under the optimal condition, molecularly imprinted SPE recoveries of spiked human serum, pig urine, tap water and shrimp were 65.80, 82.32, 76.00 and 75.97 %, respectively, when aqueous samples were applied directly. Compared with C18 SPE, a better baseline, better high-performance liquid chromatography separation efficiency and higher recoveries were achieved after molecularly imprinted SPE.   相似文献
