Cytochrome c6A is a unique dithio-cytochrome of green algae and plants. It has a very similar core structure to that of bacterial and algal cytochromes c6 but is unable to fulfill the same function of transferring electrons from cytochrome f to photosystem I. A key feature is that its heme midpoint potential is more than 200 mV below that of cytochrome c6 despite having His and Met as axial heme-iron ligands. To identify the molecular origins of the difference in potential, the structure of cytochrome c6 from the cyanobacterium Phormidium laminosum has been determined by X-ray crystallography and compared with the known structure of cytochrome c6A. One salient difference of the heme pockets is that a highly conserved Gln (Q51) in cytochrome c6 is replaced by Val (V52) in c6A. Using protein film voltammetry, we found that swapping these residues raised the c6A potential by +109 mV and decreased that of c6 by almost the same extent, -100 mV. X-ray crystallography of the V52Q protein showed that the Gln residue adopts the same configuration relative to the heme as in cytochrome c6 and we propose that this stereochemistry destabilizes the oxidized form of the heme. Consequently, replacement of Gln by Val was probably a key step in the evolution of cytochrome c6A from cytochrome c6, inhibiting reduction by the cytochrome b6f complex and facilitating establishment of a new function.  相似文献

In biosensors based on direct electron transfer in redox proteins, efficient electron-transfer pathways between the immobilized redox protein and the electrode surface have to be established so to allow a fast electron transfer and concomitantly avoiding free-diffusing redox species. In this review, prerequisites for the direct electron transfer of redox proteins and immobilization of redox proteins on the electrode surfaces are addressed. Based on the specific nature of different proteins and non-manual immobilization procedures, possible biosensor designs are discussed, namely biosensors based on (1) ferritin; (2) cytochrome c; (3) myoglobin; (4) hemoglobin; (5) horseradish peroxidase; (6) catalase; (7) glucose oxidase; and (8) xanthine oxidase.  相似文献

This paper reports the results of the research on the interaction between the highly active cytochrome b(6)f complex and plastocyanin, both isolated from the same source - spinachia oleracea plants. An equilibrium constant K between the cytochrome f of the cytochrome b(6)f complex and plastocyanin has been estimated by two independent spectroscopic techniques: steady-state absorption spectroscopy and stopped-flow. The second-order rate constants k2 for forward and backward electron transfer between cytochrome f and plastocyanin have been found between 1.4-2 x 10(7) and 8-10 x 10(6) M(-1)s(-1), respectively, giving the value of an equilibrium constant of about 2+/-0.4 or a difference in redox potential between plastocyanin and cytochrome f of cytochrome b(6)f complex of ca. 17 mV. The value of K=1.7+/-0.3 has been estimated from steady-state experiments in which the initial and final concentrations of participating components after mixing have been estimated via differential spectra analysis or spectra deconvolution. We propose a method of evaluation of the final plastocyanin concentration after the electron transfer reaction between cytochrome bf complex and plastocyanin that overcomes the interference by the strong chlorophyll absorption in the spectral region where oxidised plastocyanin has its low extinction absorption band. The data from both experiments, in the system devoid of quinol being the electron donor to cytochrome b(6), suggest that in case of electron transfer from cytochrome f to plastocyanin electron transfer can either bypass cytochrome f or the Rieske iron-sulfur protein can be reduced prior to its movement to the quinol binding site of cytochrome b(6). The role of the Rieske protein in forward and backward electron transfer reactions is discussed.  相似文献

A general equation is derived for the shift in half wave potential, ΔE , for ligand reduction on complexation with a metal. The formation of several complexes with differing stoichiometry and protonation states is considered explicitly. The dependence of ΔE on metal ion concentration, holding the ligand concentration constant, can be used to determine stability constants or their ratios. The meaning of ΔE depends on the stoichiometric changes following ligand oxidation state change, in a manner analogous to the case where the metal is the redox site.  相似文献

Axial iron ligation and protein encapsulation of the heme cofactor have been investigated as effectors of the reduction potential (E degrees ') of cytochrome c through direct electrochemistry experiments. Our approach was that of partitioning the E degrees ' changes resulting from binding of imidazole, 2-methyl-imidazole, ammonia, and azide to both cytochrome c and microperoxidase-11 (MP11), into the enthalpic and entropic contributions. N-Acetylmethionine binding to MP11 was also investigated. These ligands replace Met80 and a water molecule axially coordinated to the heme iron in cytochrome c and MP11, respectively. This factorization was achieved through variable temperature E degrees ' measurements. In this way, we have found that (i) the decrease in E degrees ' of cytochrome c due to Met80 substitution by a nitrogen-donor ligand is almost totally enthalpic in origin, as a result of the stronger electron donor properties of the exogenous ligand which selectively stabilize the ferric state; (ii) on the contrary, the binding of the same ligands and N-acetylmethionine to MP11 results in an enthalpic stabilization of the reduced state, whereas the entropic effect invariably decreases E degrees ' (the former effect prevails for the methionine ligand and the latter for the nitrogenous ligands). A comparison of the reduction thermodynamics of cytochrome c and the MP11 adducts offers insight on the effect of changing axial heme ligation and heme insertion into the folded polypeptide chain. Principally, we have found that the overall E degrees ' increase of approximately 400 mV, comparing MP11 and native cytochrome c, consists of two opposite enthalpic and entropic terms of approximately +680 and -280 mV, respectively. The enthalpic term includes contributions from both axial methionine binding (+300 mV) and protein encapsulation of the heme (+380 mV), whereas the entropic term is almost entirely manifest at the stage of axial ligand binding. Both terms are dominated by the effects of water exclusion from the heme environment.  相似文献

Structural and energetic reorganizations in redox reaction of type 1 copper proteins are studied by density functional and ab initio molecular orbital calculations. Model complexes of the active site with varying number of ligands, from Cu(SCH(3))(0/+) to Cu(SCH(3))(Im)(2)(S(CH(3))(2))(0/+), where Im denotes imidazole, are investigated. Following the findings of structural instability in Cu(I)(SCH(3))(Im)(2) and its stabilization by the addition of the axial methionine (Met) ligand model, the structure and energetics are examined as functions of the Cu-S(Met) distance in the range of 2.1-3.3 A?. The reorganization energies in both redox states exhibit a minimum at the Cu-S(Met) distance of ~2.4?A?, whereas the ionization potential increases monotonically. The changes of reorganization energies correlate well with one of the Cu-N(His) distances rather than the Cu-S(Cys) distance. The estimated Arrhenius factor for oxidation of plastocyanin by P700(+) (in photosystem I) changes by an order of magnitude when the Cu-S(Met) distance fluctuates between 2.4 and 3.0 A?, whereas the factor for reduction of plastocyanin by cytochrome f is nearly constant. Together with the data from our previous classical molecular dynamics simulation of solvated protein, we argue that the electron transfer rate is affected, and thus may be controlled, by the fluctuation of a weakly bound axial Met ligand. We also present the assessment of various exchange-correlation functionals, including those with the long-range correction, against the CCSD(T) reference and on the basis of a perturbative adiabatic connection model. For Cu(SCH(3)) and Cu(SCH(3))(Im), simple correlations have been found between the reorganization energies and the amount of Hartree-Fock exchange.  相似文献

The photoinduced covalent redox label 8-thiouredopyrene-1,3,6-trisulfonate (TUPS) has been attached to two lysine residues (K8 and K39) at opposite sides of horse heart cytochrome c, as well as to cysteines, at the same positions, introduced by site-directed mutagenesis. Electron transfer between TUPS and the heme of cytochrome c deviates from the expected monoexponential kinetic behavior. Neither the overall rate nor the individual exponential components of electron transfer, as followed by kinetic absorption spectroscopy, correlate with the length of the covalent link connecting the dye with the protein. Molecular dynamics calculations show that TUPS can approach the protein surface and occupy several such positions. This heterogeneity may explain the multiexponential electron-transfer kinetics. The calculated optimal electron-transfer pathways do not follow the covalent link but involve through space jumps from the dye to the protein moiety, effectively decoupling the length of the covalent link and the electron-transfer rates.  相似文献

In ionic liquids, the diffusion coefficients of a redox couple vary considerably between the neutral and radical ion forms of the molecule. For a reduction, the inequality of the diffusion coefficients is characterized by the ratio gamma = D(red)/D(ox), where D(red) and D(ox) are the diffusion coefficients of the electrogenerated radical anion and of the corresponding neutral molecule, respectively. In this work, measurements of gamma have been performed by scanning electrochemical microscopy (SECM) in transient feedback mode, in three different room temperature ionic liquids (RTILs) sharing the same anion and with a series of nitro-derivative compounds taken as a test family. The smallest gamma ratios were determined in an imidazolium-based RTIL and with the charge of the radical anion localized on the nitro group. Conversely, gamma tends to unity when the radical anion is fully delocalized or when the nitro group is sterically protected by bulky substituents. The gamma ratios, standard potentials of the redox couple measured in RTILs, and those observed in a classical organic solvent were compared for the investigated family of compounds. The stabilization energies approximately follow the gamma ratios in a given RTIL but change considerably between ionic liquids with the nature of the cation.  相似文献

The electron tunneling of the protein-polypeptide interactions was observed in the study of direct electron transfer of the myoglobin (Mb) on the electrode surface. The Mb was selected as a redox active protein and gelatine was selected to couple with Mb to form an electron tunneling. The electrochemical results indicated the presence of the electron tunneling and the direct electron transfer. The circular dichroism spectra suggested that the beta-sheet chain of gelatine could interact with alpha-helical chain to form an electron tunneling to promote the protein direct electrochemistry. The SDS-PAGE results proved that the electron tunneling between Mb and gelatine was noncovalent hydrogen bonds. The immobilized Mb showed a couple of quasi-reversible redox peaks with a formal potential of -0.37V (vs SCE) in 0.1 M pH 7.0 PBS. The modified electrodes displayed a rapid amperometric response to the reduction of oxygen, H2O2, and nitrite.  相似文献

The complex formation of flavin mononucleotide (FMN) with tryptamine, indoleacetate, and trytophan was investigated in the presence of polyelectrolytes; that is, sodium polyethylene sulfonate (NaPES), sodium polystyrene sulfonate (NaPSS), and a copolymer of diethyldiallylammonium chloride and sulfur dioxide (DECS). The complexation of FMN and tryptamine was strongly retarded by the macrocations and macroanions, whereas that of FMN and indoleacetate was enhanced by the macrocations. Furthermore, the equilibrium constants of FMN-tryptophan complex were insensitive to the addition of polyelectrolytes. These results suggest that the complexation of FMN was strongly influenced by the electrostatic interactions between the reactant ions and macroions, in addition to those between the reactant ions, in conformity with the secondary salt effect.  相似文献

Experimental evidence has been obtained for the involvement of encounter complexes between both enantiomers of a pi,pi* triplet excited ketone and a chiral phenol or indole. Current competing models for the two-electron oxidation of quinol (QH2) at the cytochrome bc1 complex and related complexes impose distinct requirements for the reaction intermediate. At present, the intermediate species of the enzymatic oxidation process have not been observed or characterized, probably due to their transient nature. Here, we use a biomimetic oxidant, excited-state Ru(bpy)2(pbim)+ (bpy=2,2'-dipyridyl, pbim=2-(2-pyridyl)benzimidazolate) in an aprotic medium to probe the oxidation of the ubiquinol analogue, 2,3-dimethoxy-5-methyl-1,4-benzoquinol (UQH2-0), and the plastoquinol analogue, trimethyl-1,4-benzoquinol (TMQH2-0), using time-resolved and steady-state spectroscopic techniques. Despite its simplicity, this system qualitatively reproduces key features observed during ubiquinol oxidation by the mitochondrial cytochrome bc1 complex. Comparison of isotope-dependent activation properties in the native and synthetic systems as well as analysis of the time-resolved direct-detection electron paramagnetic resonance signals in the synthetic system allows us to conclude that (1) the initial and rate-limiting step in quinol oxidation, both in the biological and biomimetic systems, involves electron and proton transfer, probably via a proton-coupled electron-transfer mechanism, (2) a neutral semiquinone intermediate is formed in the biomimetic system, and (3) oxidation of the QH*/QH2 couple for UQH2-0, but not TMQH2-0, exhibits an unusual and unexpected primary deuterium kinetic isotope effect on its Arrhenius activation energy (DeltaGTS), where DeltaGTS for the protiated form is larger than that for the deuterated form. The same behavior is observed during steady-state turnover of the cyt bc1 complex using ubiquinol, but not plastoquinol, as a substrate, leading to the conclusion that similar chemical pathways are involved in both systems. The synthetic system is an unambiguous n=1 electron acceptor, and it is thus inferred that sequential oxidation of ubiquinol (by two sequential n=1 processes) is more rapid than a truly concerted (n=2) oxidation in the cyt bc1 complex.  相似文献

Intermolecular electron transfer (ET) between the free phenothiazine donor (PH) and its cation radical (PH*+) proceeds via the [1:1] precursor complex (PH)(2)*+ which is transiently observed for the first time by its diagnostic (charge-resonance) absorption band in the near-IR region. Similar intervalence (optical) transitions are also observed in mixed-valence cation radicals with the generic representation: P(br)P*+, in which two phenothiazine redox centers are interlinked by p-phenylene, o-xylylene, and o-phenylene (br) bridges. Mulliken-Hush analysis of the intervalence (charge-resonance) bands afford reliable values of the electronic coupling element H(IV) based on the separation parameters for (P/P*+) centers estimated from some X-ray structures of the intermolecular (PH)(2)*+ and the intramolecular P(br)P*+ systems. The values of H(IV), together with the reorganization energies lambda derived from the intervalence transitions, yield activation barriers DeltaG(ET)() and first-order rate constants k(ET) for electron-transfer based on the Marcus-Hush (two-state) formalism. Such theoretically based values of the intrinsic barrier and ET rate constants agree with the experimental activation barrier (E(a)) and the self-exchange rate constant (k(SE)) independently determined by ESR line broadening measurements. This convergence validates the use of the two-state model to adequately evaluate the critical electronic coupling elements between (P/P*+) redox centers in both (a) intermolecular ET via the precursor complex and (b) intramolecular ET within bridged mixed-valence cation radicals. Important to intermolecular ET mechanism is the intervention of the strongly coupled precursor complex since it leads to electron-transfer rates of self-exchange that are 2 orders of magnitude faster (and activation barrier that is substantially lower) than otherwise predicted solely on the basis of Marcus reorganization energy.  相似文献

An ultrathin, ordered, and packed protein film, consisting of the 2-mercaptoacetic acid (MAA), polydimethyldiallylammonium chloride (PDDA), and wild-type (WT) photosynthetic reaction center (RC; termed as WT-RC) or its pheophytin (Phe)-replaced counterpart (termed as Phe-RC), was fabricated by self-assembling technique onto gold electrode for facilitating the electron transfer (ET) between RC and the electrode surface. Near-infrared (NIR)-visible (Vis) absorption and fluorescence (FL) emission spectra revealed the influence of pigment substitution on the cofactors arrangement and excitation relaxation of the proteins, respectively. Square wave voltammetry (SWV) and photoelectric tests were employed to systematically address the differences between the WT-RC films and mutant ones on the direct and photo-induced ET. The electrochemical results demonstrated that ET initiated by the oxidation of the primary donor (P) was obviously slowed down, and the formed P+ had more population as well as more positive redox potential in the Phe-RC films compared with those in the WT ones. The photoelectrochemical results displayed the dramatically enhanced photoelectric performances of the mutant ones, further suggesting the slow-down formation of final charge-separated state in Phe-RC. The functionalized protein films introduced in this paper provided an efficient approach to sensitively probe the redox cofactors and ET differences resulting from only minor changes in pigment arrangement in the pigment–protein complex. The favored ET process observed for the membrane proteins RC was potentially valuable for a deep understanding of the multi-step biological ET process and development of versatile bioelectronic devices.  相似文献

Gold(III) porphyrins of the type (P-R)AuPF(6), where P = 5,10,15,20-tetrakis(3,5-di-tert-butylphenyl)porphyrin and R is equal to H (1), NO(2) (2), or NH(2) (3) which is substituted at one of the eight beta-pyrrolic positions of the macrocycle, were investigated as to their electrochemistry and spectroelectrochemistry in nonaqueous media. Each compound undergoes three reductions, the first of which involves the central metal ion to give a Au(II) porphyrin or a Au(III) porphyrin pi-anion radical depending upon the nature of the porphyrin ring substituent. A similar metal-centered reduction also occurs for compounds 1, 3, and Au(III) quinoxalinoporphyrin, (PQ)AuPF(6) (4), where PQ = 5,10,15,20-tetrakis(3,5-di-tert-butylphenyl)quinoxalino[2,3-b]porphyrin, and these results on the three Au(III) porphyrins overturn the long held assumption that reductions of such complexes only occur at the macrocycle. In contrast, when a NO(2) group is introduced on the porphyrin ring to give (P-NO(2))AuPF(6) (2), the site of electron transfer is changed from the gold metal to the macrocycle to give a porphyrin pi-anion radical in the first reduction step. This change in the site of electron transfer was examined by electrochemistry combined with thin-layer UV-vis spectroelectrochemistry and ESR spectroscopy of the singly reduced compound produced by chemical reduction. The reorganization energy (lambda) of the metal-centered electron transfer reduction for (P-H)AuPF(6) (1) in benzonitrile was determined as lambda = 1.23 eV by analyzing the rates of photoinduced electron transfer from the triplet excited states of an organic electron donor to 1 in light of the Marcus theory of electron transfer. The lambda value of the metal-centered electron transfer of gold porphyrin (1) is significantly larger than lambda values of ligand-centered electron transfer reactions of metalloporphyrins.  相似文献
