Two new indole‐diterpenoids 4b‐deoxy‐1′‐O‐acetylpaxilline (1) and 4b‐deoxypenijanthine A (2) were isolated from the fermentation broth and the mycelia of the soil fungus Penicillium sp. CM‐7, along with three known structurally related compounds, 1′‐O‐acetylpaxilline (3), paspaline (4) and 3‐deoxo‐4b‐deoxypaxilline (5). The structures of compounds 1 and 2 were elucidated by extensive spectroscopic methods, especially 2D NMR, and their absolute configurations were suggested on the basis of the circular dichroism spectral analysis and the NOESY data. Penicillium citrium produces several fructose oligomers, namely kestose, neokestose, nystose, 1‐furanosylnystose and a novel pentasaccharide. The pentasaccharide consists of four fructoses and one glucose but its structure is different from that of 1‐furanosylnystose. One new meroterpene, arisugacin K (1), and four known ones, arisugacin J (2), arisugacin G (3), arisugacin C (4), and territrem C (5), were isolated from the culture of Penicillium echinulatum pt‐4, an endophytic fungus isolated from the marine red alga Chondrus ocellatus. The structure and the absolute configuration of 1 were unambiguously elucidated by spectroscopic analyses and quantum chemical calculations, which further resulted in the assignments of absolute configurations for the other arisugacins and territrems. Compound 1 showed inhibitory activity against Escherichia coli with an inhibition diameter 8 mm at 30 µg/disk and 4 exhibited lethality against Artemia salina with an LC50 of 48.6 µg/ml. Two new trichothecenes, named 8α‐hydroxyroridin H and myrothecin A, along with six known compounds, 8α‐acetoxy roridin H, isororidin K, verrucarin A, verrucarin J, verrucarin L and 8α‐acetoxy verrucarin L, were isolated from the fermentation broth of a halotolerant fungus Myrothecium sp. GS‐17, which was separated from the soil sample of a salina. Structure elucidation and NMR signal assignments were achieved on the basis of spectroscopy. The complete 1H and 13C NMR assignments are reported for the new natural products, 7-hydroxy-3-(hydroxymethyl)-1-methoxy-9H-xanthen-9-one (1) and 2,5-dihydroxy-8-methoxy-6-methyl-9-oxo-9H-xanthene -1-carboxylic acid (2). Both of these secondary metabolites were isolated from the fermentation culture of the endophytic fungus Xylaria sp. FRR 5657. 1D and 2D NMR experiments that included 1H, gCOSY, gHSQC and gHMBC were used for the determination of the structure and assignment of these xanthones.  相似文献

Three new fusarielins, 3‐epi‐fusarielin H ( 1 ), 3‐O‐methyl‐fusarielin H ( 2 ), and 3‐O‐methyl‐epi‐fusarielin H ( 3 ), were isolated from the fungus Fusarium sp. together with the known analogues, fusarielins F ( 4 ) and G ( 5 ). The structures of these compounds were elucidated by analysis of their ESI‐HRTOFMS, 1D and 2D NMR spectroscopic data. The new compounds exhibited weak antibacterial effect against Staphylococcus aureus.  相似文献

Two new natural products, named paecilin A (1) and B (2), together with two known compounds secalonic acid D (3) and (11)-cytochalasa-6(12),13-diene-1,21-dione-16,18-dimethyl-7-hydroxy-10-phenyl-(7S*,13E,16S*,18S*) (4), were isolated from the mangrove endophytic fungus, Paecilomyces sp. (tree 1-7) from the South China Sea. 1D and 2D NMR experiments including COSY, HMQC, and HMBC were used for the determination of their structures. In our cytotoxicity assays, secalonic D (3) showed cytotoxicity toward KB cells with IC(50) < 1 microg ml(-1) and inhibiting human topoisomerase I with IC(50) at 0.16 micromol ml(-1). 1, 2, and 4 showed no activity to KB cells.  相似文献

(w-6), an endophytic fungus associated with Acanthus ilicifolius. The complete (1)H and (13)C NMR assignments for these compounds were carried out using (1)H, (13)C, DEPT, COSY, HMQC, and HMBC NMR experiments. Three new xanthone derivatives, yicathin A (1), yicathin B (2), and yicathin C (3), and three known anthraquinone derivatives, alatinone (4), 1,5‐dihydroxy‐3‐methoxy‐7‐methylanthraquinone (5), and 5‐hydroxy‐1,3‐dimethoxy‐7‐methylanthraquinone (6), were isolated from the cultures of Aspergillus wentii pt‐1, an endophytic fungus isolated from the marine red alga Gymnogongrus flabelliformis. Their structures were unambiguously elucidated by NMR and mass spectroscopic methods as well as quantum chemical calculations. A new indole diterpene, named penicindopene A (1), together with seven known compounds (2??8), was isolated from the deep-sea fungus Penicillium sp. YPCMAC1. The structure of penicindopene A was elucidated by extensive spectroscopic analyses (1?D and 2?D NMR, and HRESIMS data), in addition to the ECD calculations for the assignments of its absolute configuration. Penicindopene A represented the first example of indole diterpenes possessing a 3-hydroxyl-2-indolone moiety, and it exhibited moderate cytotoxicities against A549 and HeLa cell lines with IC50 values of 15.2 and 20.5?µM, respectively.  相似文献

Two new xanthones, 8-(methoxycarbonyl)-1-hydroxy-9-oxo-9H-xanthene-3-carboxylic acid (1) and dimethyl 8-methoxy-9-oxo-9H-xanthene-1,6-dicarboxylate (2) and one known xanthone methyl 8-hydroxy-6-methyl-9-oxo-9H-xanthene-1-carboxylate (3) were isolated from the culture broth of the mangrove fungus Penicillium sp. (ZZF 32#) collected from the South China Sea. Their structures were established by comprehensive analysis of one-dimensional (1D) and two-dimensional (2D) NMR data. The structure of compound 3 was confirmed by X-ray crystallography, which led to the suggestion that janthinone (4) might have the same structure as 3. Compounds 1-3 were inactive against KB or KBv200 cells during cytotoxicity evaluations.  相似文献

Four new prenylindole derivatives, (R)‐6‐(2,3‐dihydroxy‐3‐methylbutyl)indole (1), (R)‐6‐(2,3‐dihydroxy‐3‐methylbutyl)indolin‐2‐one (2), and an unseparated mixture of (Z)‐6‐(4‐hydroxy‐3‐methylbut‐2‐en‐1‐yl)indolin‐2‐one (3a) and (E)‐6‐(4‐hydroxy‐3‐methylbut‐2‐en‐1‐yl)indolin‐2‐one (3b) with a ratio of 3 : 2, were isolated from the culture broth of a streptomycete isolated from Ailuropoda melanoleuca feces. Their structures were elucidated on the basis of 1D and 2D NMR spectroscopic techniques. The absolute configuration of 1 was determined by Mosher's method. The complete (1)H and (13)C NMR assignments are reported for the novel natural product Bacillosporin D together with the known compound Bacillosporin C. These compounds containing seven rings were isolated from the mangrove endophytic fungus SBE-14 from the South China Sea. 1D and 2D NMR experiments, including COSY, HMQC and HMBC were used to the determination of the structures and NMR assignments. It is proposed that 1 was biogenetically produced by transforming 2. Transforming a lactone to an anhydride is unusual in nature.  相似文献

Abstract

Two new compounds, (7R, 2E, 5E)-3,5,7-trimethyl-2,5-octadienedioic-8-methyl ester (1) and neovasipyridone G (3), together with a new natural product compound (7R,2E,5E)-3,5,7-trimethyl-2,5-octadienedioic acid (2), and six known compounds (4–9) were isolated from Penicillium sp. SYPF7381. Their structures were elucidated on the basis of extensive spectroscopic analysis, and the absolute configurations of compounds 1 and 2 were determined by optical rotation. The absolute configuration of compound 3 was determined by means of electronic circular dichroism (ECD) calculation. In addition, the in vitro anti-inflammatory activities of all compounds were assayed in RAW 264.7 cells by assessing LPS-induced NO production. Furthermore, the structure-antiinflammation activity relationships for these isolated compounds were summarized based on the experimental as well as the docking results.  相似文献
