Condensations of chiral diamines 11a-c with benzotriazole and formaldehyde gave benzotriazolyl intermediates 12a-c; similar condensations of alpha-amino-amides 10a-c with benzotriazole and paraformaldehyde gave 14a-c. Subsequent treatment of 12a-c and 14a-c with AlCl(3) led to enantiopure tricyclic 1,2,3,5,10,10a-hexahydroimidazo[1,5-b]isoquinolines 1a-c and 2,3,10,10a-tetrahydroimidazo[1,5-b]isoquinolin-1(5H)-ones 15a-c, respectively, via Lewis acid promoted iminium cation cyclizations.  相似文献

Metabolites of 1,5-Dihydroimidazo[2,1-b ]quinazolin-2(3H)-ones. Preparation and Reactions of Some 1,5-Dihydro-3-hydroxyimidazo[2,1-b]quinazolin-2(3H)-ones Hydroxylated 1,5-dihydroimidazo[2,1-b]quinazolin-2(3H)-ones 2–4 and 6 were isolated as metabolites of imidazoquinazolinones 1a and 1b , respectively. The synthesis of 1,5-dihydro-3-hydroxy-3-methylimidazo[2,1-b]quinazolin-2(3H)-ones 3 , 4 , and 6 , and the preparation of some derivatives thereof is described.  相似文献

Magnetic circular dichroism spectra of s-triazolo[ 1,5-b]isoquinolin-5(H)ones, 1,2-dihydro-6H-s-tetrazino[2,3-b] isoquinolin-6-ones and realted compounds were measured. The molecular symmetry of these compounds is so low that the MCD spectra contain only the Faraday parameter B. It appears that the magnitude and sign of the B term depends on the nature of the subslituents and of electron migration from endo- or exo-nitrogen to oxygen, respectively.  相似文献

发展了一种从3-(2-溴代苯甲酰基)-吲哚出发在无过渡金属参与下合成茚并吲哚酮类化合物的新方法。在甲苯/四氢呋喃(2∶1)混合溶剂中,3-(2-溴代苯甲酰基)-吲哚、正丁基锂和碘化锌原位生成的2-吲哚锌在碘化锂辅助下与芳基溴化物发生分子内加成-环化反应,合成了一系列取代的茚并-[1,2-b]吲哚-10(5H)-酮类化合物,且均获得了较好的收率。考察了溶剂、卤化锌、添加剂对产率的影响。  相似文献

The alkylation of benzimidazo[1,2-b]isoquinolin-11(5H)-one has been studied. This occurs at N(5) or C(6) depending on the type of alkylating agent and the reaction conditions. It was shown that C(6) alkylation is effected in reactions with reactive alkyl halides. A repeat alkylation occurs preferentially at the same position. Interaction with o-xylylidene dibromide leads to spiro[benzimidazo[1,2-b]-isoquinolin-6,2'-indan]-11-one and 1,6-dihydro-11H-6a,11b-diazobenzo[b]benzo[5,6]cyclohepta[1,2,3-l,m]fluoren-11-one, which are derivatives of new heterocyclic systems.  相似文献

The reaction of benzimidazo[1,2-b]isoquinolin-11(5H)-one with activated olefins has been studied. The derivatives of 3,10-dioxo-3H,10H-benzimidazo[1,2,3-ij]benzo[c][1,8]naphthyridine formed are the result of an initial Michael reaction at C(6) followed by intramolecular heterocyclization.  相似文献

A study has shown that the acylation of benzo[4,5]imidazol[1,2-b]isoquinolin-11(5H)-one occurs at N(5) or at C(6) depending on the nature of the acylating reagent and the reaction conditions. It was found that principally C-acylation takes place in the absence of base. The reaction with -halo-substituted carboxylic acid chlorides leads to the formation of C-acylated products which are converted to derivatives of the novel heterocyclic system 7H-2a,6b-diazabenzo[b]cyclopenta[l,m]fluorene-1,7(2H)-dione in the presence of base.  相似文献

A convenient and sustainable synthesis of pyrazolo[1,5-a]quinazolin-5(4H)-ones and [1,2,4]triazolo[1,5-a]quinazolin-5(4H)-one through copper-catalyzed cascade reactions of 2-bromobenzoates with 1H-pyrazol-5-amines or 1H-1,2,4-triazol-5-amine under ligand-free conditions in water is presented. It is notable that aqueous medium turned out to be crucial for the chemoselective formation of the title compounds. Compared with literature protocols, this new method showed advantages such as simple and sustainable procedure, commercially available starting materials, and convenient reuse of the reaction medium together with the copper catalyst.  相似文献

3-Aminocoumarin undergoes the Skraup reaction to give a new ring system, 5H[l]benzo-pyrano[3,4-b]pyridin-5-one (IVa). When 3-aminocoumarin was condensed with the ethoxy methylene derivatives of active methylene compounds, ethyl acetoacetate, and dimethyl acetylenedicarboxylate, the intermediates Vla-VIf were formed which on thermal cyclizations afforded other derivatives of 5H[l ]benzopyrano[3,4-b]pyridin-5-one (IVb-IVf). Three-component condensation of 3(5)-aminoazoles with benzaldehydes and dimedone in acetonitrile in the presence of montmorillonite KSF as heterogeneous catalyst regioselectively afforded 5,6,7,9-tetrahydroazolo[5,1-b]quinazolin-8(4H)-one derivatives in good yields. Oxidation of the condensation products with ceric ammonium nitrate in acetone gave the corresponding 6,7-dihydroazolo[5,1-b]quinazolin-8(5H)-ones.  相似文献

Tetraheterocyclic benzimidazo[1,2-a]quinazolin-4(1H)-one and tetrahydro-1,2,4-triazolo[5,1-b]quinazolin-8(4H)-one were synthesized in relatively high yields by the condensation reaction of an aldehyde and a cyclic beta-diketone with 2-aminobenzothiazole, 2-aminobenzimidazole or 3-amino-1,2,4-triazole without using any catalyst under solvent-free conditions.  相似文献

The synthetic routes of 6,7-dihydrobenzo[4,5]cyclohept[1,2-b]indol-12(5H)-one 5 from either 1-methyl or 1-sulfonylindole-2-carboxaldehyde 1 or ethyl 1,2-dimethylindole-3-carboxylate 6 are reported. 以2-溴甲基-6,7-亚甲二氧基-3-喹啉酸乙酯(1) 为原料,经三步合成了2-叔丁基-4-氟-苯并氧杂卓并[3,4-b]喹啉并[9,11]二氧戊环-14(6H)-酮(4).其中,6-(2-氟-4-叔丁基-苯氧甲基)-[1,3]二氧戊环并[4,5-g]喹啉-7-羧酸是经一锅法合成的.它的分子内闭环反应是在Eaton's reagent(P2O5-CH3SO3H)的作用下,在较温和的条件下进行的.所得新化合物3和4的结构经IR、1H NMR和元素分析得以证实.  相似文献

利用L-脯氨酸催化的5-氯水杨醛(1)与6-甲基-4-羟基吡喃酮(2)的缩合反应及硫酸铜催化下与1,3-二甲基-5-氨基吡唑(3)的串联反应,合成得到了10-氯-1,3-二甲基-5-(2-氧代丙基)色烯并[4,3-d]吡唑并[3,4-b]吡啶-6(3H)-酮(4)和10-氯-5-二乙氧基甲基-1,3-二甲基色烯并[4,3-d]吡唑并[3,4-b]吡啶-6(3H)-酮(5).化合物5的结构通过单晶X射线衍射法确定:晶体属于三斜晶系,空间群P-1;相对分子质量Mr=803.68;晶胞参数a=1.03160(10)nm,b=1.42900(13)nm,c=1.44268(15)nm;V=1.9448(3)nm~3;Z=2;晶胞密度Dc=1.372g/cm~3;吸收系数μ=0.228mm-1;单胞中电子的数目F(000)=840.晶体结构用直接法解出,经全矩阵最小二乘法对原子参数进行修正,最终的偏离因子为R=0.0681,w R=0.2051.在晶体结构中色烯环与吡啶环及吡唑环近似于共平面.  相似文献
