Enantioresolution of 3-octanol, 6-methyl-5-hepten-2-ol (sulcatol), and 1-octen-3-ol was conducted using (S)-(+)-2-methoxy-2-(1-naphthyl)propionic acid (MαNP acid) and (S)-(+)-2-methoxy-2-(9-phenanthryl)propionic acid (M9PP acid). In each case, the diastereomeric esters obtained were readily separated by HPLC. The stereochemistry of the esters could be assigned from their respective 1H NMR analyses. Solvolyses of the esters gave enantiopure alcohols and acids. MαNP and M9PP acids displayed almost equivalent properties in 1H NMR anisotropy. The chiral resolving ability of M9PP acid was slightly superior to that of MαNP acid in HPLC.  相似文献

2-Methoxy-2-(9-phenanthryl)propionic acid was synthesized as a novel chiral resolving agent. The absolute configuration of (+)-2-methoxy-2-(9-phenanthryl)propionic acid was determined to be S by using X-ray structural analysis of the (1R,2S,5R)-menthyl ester. In the crystal, the methoxyl and carbonyl groups of the ester are in a syn-periplanar position. The syn-periplanar conformations of (1R,2S,5R)-menthyl esters were also observed by the NMR analyses in CDCl3. The utility of (S)-(+)-2-methoxy-2-(9-phenanthryl)propionic acid was exemplified by the resolution of (±)-3-octanol.  相似文献

An enantioresolution of 2-methoxy-2-(1-naphtyl)propionic acid (MαNP acid) using the diastereomeric salt with chiral (R)-phenylethylamine was achieved to give enantiopure (R)-MαNP acid in 29% yield with >99% ee based on rac-MαNP acid. X-ray crystallographic analysis of diastereomeric salt revealed that (R)-MαNP acid was tightly arranged by four independent hydrogen bonds and one CH–π interaction with (R)-phenylethylamine.  相似文献

3-Methoxy-4-aryl-furan-2,5-dicarboxylic acid (8) is selectively converted into its C-5 methylester (6) by treatment with methyl chloroformate followed by decarboxylation in one flask. Acylation of the resulting half ester with a 7-substituted indole was performed under mild conditions to afford 3-aryl-5-(1H-indole-3-carbonyl)-4-methoxy-2-furoic acid (11). The synthetic utility of the resulting furoic acids as a skeleton in the synthesis of potential insulin receptor activators is established.  相似文献

Reaction of the tert-butyl ester of 3-methoxy-2-pyrrole carboxylic acid with singlet oxygen yields a peroxidic intermediate which undergoes coupling with a range of nucleophiles to yield 5-substituted pyrroles. Efficient stereospecific synthesis of (S,S)-3-methoxy-4-methylaminopyrrolidine, an important intermediate for a novel quinolone antitumor agent AG-7352 is presented. Starting from either d- or l-tartaric acid, a stereospecific synthesis of the chiral pyrrolidine was achieved via two SN2 displacement reactions. From the results of this synthetic study, the absolute structure of AG-7352 was chemically determined.  相似文献

Mass spectra of previously unknown isomeric 1-isopropyl-3-methoxy-2-methylsulfanylpyrrole, 5-methoxy-2,2-dimethyl-6-methylsulfanyl-2,3-dihydropyridine, and 3-methoxy-7-methyl-2-methylsulfanyl-4,5-dihydro-3H-azepine were studied for the first time. Fragmentation of all heterocyclic compounds under electron impact begins with elimination of methyl radical, and the subsequent decomposition of the [M ? Me]+ ion (m/z 170) is specific for each isomers, which ensures their reliable identification in reaction mixtures. The corresponding linear precursors, methyl N-isopropyl-2-methoxybuta-2,3-dienimidothioate and 2-methoxy-N-(1-methylethylidene)-1-methylsulfanylbuta-1,3-dien-1-amine undergo isomerization and decomposition even under very mild temperature conditions, so that their mass spectra could not be recorded.  相似文献

2-Alkyl-7-methoxy-5-nitrobenzo[b]furan, 2-alkyl-9-methoxy-7-nitro-3-oxo-2,3-dihydro-5H-benzo[e][1,4]-dioxepin-5-yl acetate and 2-alkyl-5-hydroxy-9-methoxy-7-nitro-5H-benzo[e][1,4]-dioxepin-3-one were formed as a result of the cyclization of 2-(2-formyl-6-methoxy-4-nitrophenoxy)alkanoic acids under classical Perkin reaction conditions. The products were characterized by spectroscopic methods and the mechanism of the cyclization is discussed.  相似文献

Oxygenates are used in gasoline to increase the octane number and reduce carbon monoxide emission. 2-methoxy-2,4,4-trimethylpentane (TOME) is a tertiary ether which can potentially be used in addition with current oxygenates. This compound can be produced by etherification of diisobutylene with methanol. During the etherification, water is formed due to the dehydration of methanol. The appearance of water can cause (liquid + liquid) phase split in the production process. In this work, several physical properties of systems containing water, methanol and TOME are studied for the first time. The liquid density of 2-methoxy-2,4,4-trimethylpentane is presented from T = (298.15 to 408.16) K. Excess enthalpies are reported for the binary system of (methanol + 2-methoxy-2,4,4-trimethylpentane) at (T = 298.15 K). The (liquid + liquid) equilibrium (LLE) for (water + 2-methoxy-2,4,4-trimethylpentane) from T = (283.15 to 318.15) K is determined. The LLE is also reported for the ternary system of (water + methanol + 2-methoxy-2,4,4-trimethylpentane) at T = (283.15 and 298.15) K. The UNIQUAC parameters were regressed to model VLE, excess enthalpy and LLE for the binary and ternary data with one set of parameters.  相似文献

1-(4-Methoxy-3,5-dimethylphenyl)-2-methylpropan-1-ol reacted with nitriles [MeSCN, PhCN, MeCN, EtOC(O)CH2CN] in the presence of concentrated sulfuric acid to give both 1-R-3,3,7,9-tetramethyl-2-azaspiro[4,5]deca-6,9-dien- and -1,6,9-trien-8-ones and 1-R-7-methoxy-3,3,6,8-tetramethyl-3,4-dihydroisoquinolines. The reaction with 3,4-dimethoxyphenylacetonitrile afforded 10,11-dimethoxy-1,3,6,6-tetramethyl-1,5,6,12b-tetrahydrodibenzo[d,f]indole-2,8-dione. Three-component condensation of 2-methoxy-1,3-dimethylbenzene with isobutyraldehyde and nitriles led to the formation of spirocyclic systems and 3,4-dihydroisoquinoline derivatives in lower yield.  相似文献

A DMAP (4-dimethylaminopyridine) catalyzed cyclization of methyl 2-alkynylbenzimidates has been developed, which affords 3-methoxy-1-methyleneisoindoles with excellent Z-stereoselectivity under mild and transition-metal-free conditions. The (Z)-3-methoxy-1-methyleneisoindole products can be converted to corresponding 3-amino-1-methyleneisoindoles, 3-methoxy-isoindoles, 3-methyleneisoindolinones and isoindolinones with high efficiency.  相似文献

The enantioselective hydrogenation of (E)-2-(4-methoxy-3-(3-methoxypropoxy)-benzylidene)-3-methylbutanoic acid (1) to (R)-2-(4-methoxy-3-(3-methoxypropoxy)-benzyl)-3-methylbutanoic acid (2)—a key intermediate in the synthesis of the pharmacologically important renin inhibitor Aliskiren—is described. The stereochemistry of the catalytic transformation has been studied using a number of homogeneous chiral Rh(I) and Ru(II) complexes bearing ferrocene-based phosphine ligands. The highest enantioselectivity for the homogeneous hydrogenation of 1 (up to 95% ee) was achieved with a [Rh(NBD)2]BF4 pre-catalyst (substrate/catalyst ratio 100:1, 10 bar H2, 40 °C, in MeOH). To bring the enantioselectivity to perfection an effective method for the isolation of the enantiopure carboxylic acid is suggested likewise.  相似文献

A number of nitrogen heterocycles reacted with 4-methoxy-3-nitrobenzyl chloride in dimethyl-formamide in the presence of potassium carbonate to give the corresponding N-(4-methoxy-3-nitrobenzyl) derivatives. The reaction of 5-fluoro-1,3-bis(4-methoxy-3-nitrobenzyl)pyrimidine-2,4(1H,3H)-dione with aqueous methylamine afforded N,N′-bis(4-methylamino-3-nitrobenzyl)urea, whereas analogous reaction with 1-(4-methoxy-3-nitrobenzyl)-2-(methylsulfanyl)-1H-benzimidazole resulted in substitution of the methoxy group by methylamino.  相似文献

1,3-Dinitro-5-trichloromethylbenzene (2) was reacted with sodium methoxide in an attempt to prepare 4-methoxy-3,5-dinitrobenzaldehyde (7) via a reported tele nucleophilic aromatic substitution. The product from this reaction was methyl 3,5-dinitrobenzoate (5) and not the methoxy aldehyde as had been reported. Eicosapentaenoic acid is converted in good overall yield to an α,β-ethylenic epoxide derivative. The oxidative cleavage of the epoxide ring with periodic acid in ether proceeded in part with acid-catalyzed rearrangement of the vinyl epoxide moiety prior to cleavage. Among the products were 2E- and 2Z, 5Z, 8Z, 11Z-tetradecatetraenal, 4-hydroxy-2E,6Z,9Z,12Z-pentadecatetraenal and (all-Z)-2-methoxy-3,6,9,12-pentadecatetraenal.  相似文献

The X-ray diffraction study of the potassium salt of 2-methoxy-4-dimethylamino-6-dinitromethyl-1,3,5-triazine is carried out. The crystals are triclinic; C7H9N6O 5 ? ·2K+·2H2O; a = 7.645(7) Å, b = 8.230(7) Å, c = 12.435(9) Å; α = 99.99(8)°, β = 91.52(7)°, γ = 113.86(8)°; V = 701(1) Å3, ρcalc = 1.58 g/cm3, Z = 2, space group P-1. The compound crystallizes as a crystalline hydrate with two water molecules. Two planar fragments of dinitromethyl and 2-methoxy-4-dimethylamino-1,3,5-triazine construct the anion. Their geometrical parameters are explored. The coordination of potassium cations and numerous hydrogen bonds are found which result in the development of a complex 3-D framework.  相似文献
